Ziziphus jujuba mill. Extract Promotes Myogenic Differentiation of C2C12 Skeletal Muscle Cells

  • Ziziphus jujuba Mill. (ZJM), a traditional folk medicine and functional food in South Korea and China, has been reported to having pharmacological activities against anti-cancer, anti-oxidative, and anti-obesity. However, the effect of ZJM related to myoblast differentiation has not been known. In this study, we investigated the effects and mechanism of ZJM on myogenic differentiation of C2C12 cells. ZJM promotes myogenic differentiation and elevates the formation of multinucleated myotube compared to the control group. ZJM significantly increased the mRNA and protein expression of MyHC1, myogenin and MyoD in dose- and time-dependent manner. Interestingly, ZJM significantly inhibited the mRNA and protein expression of protein degradation markers, atrogin-1 and MuRF-1, in dose- and time-dependent manner. Taken together, our data suggest that ZJM is a potential functional candidate for muscle growth and strength by promoting myogenic differentiation.

흑임자 첨가량을 달리한 구기자흑임자죽의 품질특성 (Quality Characteristics of Gugija.Heukimja Jook Containing Different Levels of Black Sesame Powder)

  • This study was to investigated optimum preparation conditions for Gugija Heukimja jook by response surface methodology (RSM). The important materials used for the Gugija Heukimja product were Lycium chinese, Seasamum indicum L., Ziziphus jujuba Mill. and rice. Moisture content, crude protein, crude fat, crude ash and crude fiber content were 15.62, 16.60, 13.62, 4.93 and 11.17% for Lycium chinese, 3.62, 20.94, 43.56, 5.67 and 10.72% for Seasamum indicum L., 24.12, 5.38, 0.73, 2.30 and 1.25% for Ziziphus jujuba Mill., and 12.36, 6.98, 0.46, 0.43 and 0.41% for rice, respectively. Optimal extraction conditions of brix, color, pH and total acidity in the materials for Gugija Heukimja jook were temperature $80^{\circ}C$, water volume 850 mL and extraction time of 63 min. The spreadability of jook with Lycium chinese and Ziziphus jujuba Mill. extracts was the highest when black sesame was 12 g pH was the lowest when black sesame was 9 g. PH showed a significant difference when the added black sesame ratio changed. The 'value L' of chromaticity was lower with an increase of the black sesame ratio, 'value a' was higher with an increase of the black sesame ratio, 'value b' was lower with an increase of the black sesame ratio. Brix was lower when black sesame in jook was added. Total acidity was highest (7.27%) when black sesame was 12 g. The degree of gelatinization tended to be lower with additional black sesame. Higher scores of sensory evaluation were found for the product containing 9 g black sesame with Lycii furctus and Ziziphus jujuba Mill. extracts ompared to the other samples with 3 and 6g. No significant difference in taste occurred for Gugija Heukimja jook prepared with over 9 g black sesame, while a great difference showed in color, pH and total acid. Therefore, the Gugija Heukimja jook prepared under the optimal conditions of 9 g black sesame with Lycium chinese and Ziziphus jujuba Mill. extracts exhibited higher qualities. Gugija Heukimja jook can use the easier for Yacksun jook. It is expected to become functional in the product's development.

대추 아임계수 추출물의 폴리페놀 함량 및 산화방지 평가 (Antioxidant activities of phenolic compounds from Ziziphus jujuba Mill extract using subcritical water)

  • 아임계수 추출 기술을 이용하여 대추 추출물을 얻고 총 페놀 함량, DPPH 라디칼 소거능과 ABTS 라디칼 소거능을 이용하여 추출조건에 따른 페놀 성분 함량 변화 및 산화 방지효과를 확인하였다. 아임계수 추출물은 총 페놀 함량, DPPH 라디칼 소거능, ABTS 라디칼 소거능의 결과 값이 모두 메탄올 및 에탄올을 이용하여 추출한 결과 보다 우수하게 나타났다. 아임계수 추출법은 메탄올 및 에탄올을 이용하여 추출하는 방법 보다 추출 시간도 20분 이내로 짧았고, 유기용매를 사용하지 않고 친환경적으로 추출할 수 있었다. 아임계수 추출물의 총 페놀 함량과 산화방지 효과의 결과는 높은 상관관계를 보였는데, 항산화 효과가 있는 페놀류가 많이 추출될수록 산화방지 효과도 우수하게 나타났다. 본 연구를 통해, 아임계수 추출 방법은 오직 물을 사용하여 유효성분을 추출하는 친환경적이고 안전한 신가공기술이며 잠재성과 우수성을 확인하였다. 아임계수 추출기술은 앞으로도 다양한 유효성분 추출 및 소재 가공으로 식품 산업에 응용하여 사용될 수 있을 것이다.

ISSR 표지를 이용한 국내 재배 대추나무의 유전특성 분석 (Analysis of Genetic Characteristic of Jujube (Ziziphus jujuba Mill.) Cultivated in Korea Revealed by ISSR Markers)

  • 대추나무는 갈매나무과에 속하며 과실에 높은 영양가를 지니고 있어 전통 한의학에서 널리 사용되고 있는 경제적으로 중요한 종이다. 국내에서 재배되고 있는 대추나무들의 품종실태를 파악함으로서 우량개체 선발과 육종계획 수립에 유용한 유전정보를 얻고자 보은, 경산 등 대추 주산지 6곳에서 수집된 대추나무 270개체를 대상으로 ISSR 표지 분석을 수행하였다. 그 결과 유전적 다양성을 보여주는 S.I.값은 0.107, 유전적 유사도는 0.935로 높게 나타나 연구에 사용된 대추나무들이 특정 품종 또는 개체에 편중되어 있었다. 또한 270개체 중 67%인 180개체가 '복조'와 동일한 유전자형을 나타내는 것으로 관찰되었다. 본 연구를 통해 국내 재배 대추나무 개체들의 유전적 다양성이 매우 낮은 것으로 파악되었다. 이에 외부 교란에 취약할 가능성이 큰 것으로 예상되며, 시장에 판매되고 있는 대부분의 대추가 단위결실을 통해 생산되는 것으로 판단된다.

  • Many plant species are used in China in tradtional medicine for the prevention and treatment of cancer. This paper presents some of these species with defals on other pharmacologic evaluation and its tradional Chinese meditional uses. The known bioaktivities and some chemical constituents of each of the species given. Information on bioactivities of each species resulting from tests on experimental animals are given. Many of the known chemical constituents of each species are given. Various species of plants elective for various types. 배, 맥아, 대추, 익모초, 황금 및 이들 혼합물의 생리활성 (Physiological activities of Pyrus pyrifolia Nakai, malt, Ziziphus jujuba Mill., Leonurus japonicus Houtt., Scutellaria baicalensis, and their mixtures)

  • 본 연구는 염증을 개선하고 소화에 도움을 주는 최적 혼합물 개발을 위한 소재 발굴을 위해 11종류 식물 추출물을 이용하여 항염증, 트립신 활성 및 항산화 효과를 알아보았다. LPS로 염증이 유도된 RAW 264.7 대식세포에서 대추(ZJ), 익모초(LJ), 황금(SB), 도라지 그리고 참취 추출물은 과분비 되는 NO와 TNF-α 함량을 감소시켜 뛰어난 항염증 효과가 있었다. 11개의 시료 중 맥아(MA), 배(PP), 무 그리고 도라지 추출물은 트립신 활성이 높았다. DPPH 라디칼 소거작용으로 항산화 효과를 알아본 결과 SB, PP, ZJ 그리고 MA 추출물이 높은 항산화 효과가 있었다. 따라서 염증을 개선하면서 소화에 도움을 주는 최적 혼합물 개발을 위해 배, 맥아, 대추, 익모초 그리고 황금을 선택하였다. LPS로 자극한 RAW 264.7 대식세포에서 PP, MA, ZJ, LJ 및 SB(1:1:2:1:2, w/w)을 함유하는 혼합물의 추출물은 각각의 PP, MA, ZJ, LJ 및 SB의 추출물보다 NO 생산을 현저하게 억제하였다. 혼합물 추출물의 DPPH 라디칼 소거작용은 PP, MA, ZJ 및 LJ의 각각의 추출물보다 현저하게 높았다.

  • Bone homeostasis is maintained by balance between bone resorbing-osteoclasts and bone forming-osteoblasts. Excessive osteoclastic bone resorption plays a critical role in bone destruction in pathological bone diseases such as osteoporosis, rheumatoid arthritis, and periodontal disease. Many compounds derived from natural products have pharmacological applications and have therapeutic value for treating or preventing several bone diseases characterized by excessive bone resorption. To discover new compounds that can act as anti-resorptive agents, we screened for natural compounds that regulate osteclast differentiation, and found that water extract of Ziziphus Jujuba Mill (WEZJ) has inhibitory effects on osteoclast differentiation. In this study, WEZJ clearly inhibits the osteoclast differentiation in the presence of receptor activator of nuclear factor kB (RANKL), macrophage colony-stimulating factor (M-CSF) without cytoxicity by blocking activation of nuclear factor of activated T cells (NFAT)c1, and c-Fos. In signaling pathway, the phosphorylation of Akt, p38, c-Jun N-terminal kinases (JNK), extracellular signal-regulated kinases (ERK) and the expression of osteoclast-associated receptor (OSCAR), tartrate-resistant acid phosphates (TRAP), Integrin av, Integrin b3, Cathepsin K are suppressed, too. These result suggest that WEZJ may have therapeutic value for treating or preventing several bone diseases characterized by excessive bone destruction.

Chemical Composition of Some Tropical Foliage Species and Their Intake and Digestibility by Goats

  • The chemical composition and water extractable dry matter (DM) of foliages from Erythrina (Erythrina variegata), Fig (Ficus racemosa), Jackfruit (Artocarpus heterophyllus), Jujube (Ziziphus jujuba Mill), Kapok (Ceiba pentandra) and Mango (Mangifera indica) and the feed intake, digestibility and N retention when feeding these foliages were studied in two experiments. In Experiment 1, 12 male goats, 3.5 months old and weighing 14.6 kg, were randomly allocated to a diet consisting of one of the foliages in a change-over design with three periods. The foliages were offered ad libitum at the level of 130% of the average daily feed intake. The Erythrina foliage had a low content of DM and condensed tannins (CT) and a high concentration of crude protein (CP) in leaves plus petioles (193 g/kg DM) and stem, while the Mango foliage had a low CP (69 g/kg DM) and high DM content. The other foliages were intermediate. High content of CT was found in the leaves plus petioles of Jackfruit foliage and in the stem of Fig and Mango foliage. There was a difference in feed intake, nutrient intake, apparent digestibility and N retention between the foliages, with Erythrina, Jackfruit and Kapok foliage being significantly higher in these parameters than Fig, Jujube and Mango foliage. The water extractable DM could be used to estimate N retention, but not DM digestibility in this study. In Experiment 2, 4 male goats weighing 13.4 kg and 6 months old were allocated to a 4${\times}$4 Latin square design. The treatments were: water spinach ad libitum and Fig, Jujube or Mango foliage ad libitum +0.5% of BW as water spinach DM. Feed intake, apparent digestibility and N retention were not significantly different among the foliage diets, but higher than for water spinach alone (p<0.05). Supplementation with water spinach to a diet consisting of low quality foliages such as Fig, Jujube and Mango, increased DM and CP intake, apparent digestibility and N retention, compared to feeding these foliages as sole feeds.

대추 식초 음료 생산을 위한 대추 추출액 발효 조건 최적화 (Optimization of Acetic Acid Fermentation for Producing Vinegar from Extract of Jujube (Zizyphus jujuba Mill.) Fruits)

  • 대추 추출물로부터 식초를 생산하기 위한 발효공정의 최적 조건을 연구하였다. 아세트산 세균을 이용한 발효 공정의 조절 변수로 초기 세균수, 에탄올의 농도, 대추 추출물의 농도, 온도, pH를 고려하여 반응 표면 분석법을 통해 분석하였다. FCD 기반의 통계 분석을 통해, 영향이 낮은 것으로 나타난 초기 세균수와 대추 추출물의 농도는 고려요소에서 제거한 다음, 아세트산 생성과 최대 산도를 얻을 수 있는 발효 공정 최적 조건을 탐색하였다. 최대 산도와 최대 아세트산 생성 조건에 대해 각각 $R^2$값이 0.71, 0.78로 나타난 반응 표면 회기식 분석을 통해 7.69% 에탄올을 함유한 25%의 대추 추출물로부터 $21.75^{\circ}C$에서의 아세트산 발효를 통해 52.76 mg/mL의 아세트산을 생산하는 것이 최대 효율 공정으로 예측되었으며 이를 실험을 통해 검증하였다.
