Bioequivalence Assessment of Acephyll® Capsule to Surfolase® Capsule (Acebrophylline HCl 100 mg) by Liquid Chromatography Tandem Mass Spectrometry

  • A sensitive and specific liquid chromatographic method coupled with tandem mass spectrometry (LC-MS/MS) was developed for the analysis of ambroxol (active moiety of acebrophylline). After acetonitrile precipitation of proteins from plasma samples, ambroxol and the domperidone (internal standard, IS) were eluted on a C18 column. The isocratic mobile phase was consisted of 10 mM ammonium acetate and methanol (10 : 90, v/v), with flow rate at 0.2 mL/min. A tandem mass spectrometer, as detector, was used for quantitative analysis in positive mode by a multiple reaction monitoring mode to monitor the m/z 379.2${\rightarrow}$264.0 and the m/z 426.2${\rightarrow}$175.1 transitions for ambroxol and the IS, respectively. Twenty four healthy Korean male subjects received two capsules (100 mg ${\times}$ 2) of either the test or the reference formulation of acebrophylline HCl in a 2 ${\times}$ 2 crossover study, this was followed by a 1week washout period between either formulation. $AUC_{0-t}$ (the area under the plasma concentration-time curve) was calculated by the linear trapezoidal rule. $C_{max}$ (maximum plasma drug concentration) and $T_{max}$ (time to reach $C_{max}$) were compiled from the plasma concentration-time data. The 90% confidence intervals for the log transformed data were acceptable range of log 0.8 to log 1.25 (e.g., log 0.8964 - log 0.9910 for $AUC_{0-t}$ log 0.8690 - log 1.0750 for $C_{max}$). The major parameters, $AUC_{0-t}$ and $C_{max}$ met the criteria of Korea Food and Drug Administration for bioequivalence indicating that Acephyll$^{(R)}$ capsule (test) is bioequivalent to Surfolase$^{(R)}$ capsule (reference).

A simple creep constitutive model for soft clays based on volumetric strain characteristics

  • The soft clays are widely distributed, and one of the prominent engineering problems is the creep behavior. In order to predict the creep deformation of soft clays in an easier and more acceptable way, a simple creep constitutive model has been proposed in this paper. Firstly, the triaxial creep test data indicated that, the strain-time (𝜀-t) curve showing in the 𝜀-lgt space can be divided into two lines with different slopes, and the time referring to the demarcation point is named as tEOP. Thereafter, the strain increments occurred after the time tEOP are totally assumed to be the creep components, and the elastic and plastic strains had occurred before tEOP. A hyperbolic equation expressing the relationship between creep volumetric strain, stress and time is proposed, with several triaxial creep test data of soft clays verifying the applicability. Additionally, the creep flow law is suggested to be similar with the plastic flow law of the modified Cam-Clay model, and the proposed volumetric strain equation is used to deduced the scaling factor for creep strains. Therefore, a creep constitutive model is thereby established, and verified by successfully predicting the creep principal strains of triaxial specimens.

  • In this study, to investigate the fatigue crack retardation behavior and the variability of retardation cycles, fatigue crack growth tests were conducted on 7075-T6 aluminum alloy under single tensile overload. A retardation coefficient, D was introduced to describe fatigue crack retardation behavior and a random variable, Z to describe the variability of fatigue crack growth. The retardation coefficient was separately formulated according to retardation behavior which is composed of delayed retardation part and retardation part. The random variable, Z was evaluated from experimental data which was obtained from fatigue crack growth tests under constant amplitude load. Using these variables, a probabilistic model was developed on the basis of the modified Forman's equation, and retardation behavior and cycles were predicted under certain overload condition. The predicted retardation curve well agrees with the trend of experimental crack retardation behavior. And this model well predicts the scatter of experimental retardation cycles.

Population Ecology of Blackthroat Seaperch Doederleinia berycoides in the Southern Seas of Korea (한국 남해 연안에 분포하는 눈볼대(Doederleinia berycoides)의 자원생태학적 특성치)

  • We investigated the population ecology of blackthroat seaperch, Doederleinia berycoides, from samples collected in the southern seas of Korea from January to December, 2006. Population ecological parameters included survival rate, the instantaneous coefficient of natural and fishing mortality, and age at first capture. The survival rate (S) of blackthroat seaperch was estimated as 0.4966 using the catch curve method. The instantaneous coefficient of total mortality (Z) was 0.8598/year. The instantaneous coefficient of natural mortality (M) was estimated as 0.4694/year. From the estimates of Z and M, the instantaneous coefficient of fishing mortality (F) was calculated as 0.3904/year. The age at first capture ($t_c$), based on the Pauly method, was 2.87 years.

  • The electrical characteristics of the soil where a ground electrode is buried vary with regions, seasons and environmental factors. Electrical discharge in the vicinity of the ground electrode will occur differently and significantly affect the performance of the grounding system. It is necessary to analyze discharge and ionization characteristics of soils when the grounding system is designed. The aims of this investigation are to understand correlation between the soil ionization and the transient ground impedance. This paper presents the experimental results on the soil ionization parameters and the transient ground resistance due to the soil ionization around a hemispherical ground electrode stressed by lightning impulse voltages.

  • A toroidal-field (TF) coil with a pure tension D-shape curve is designed for the confinement of high-temperature plasmas in the SNUT-79, which is a tokamak being built at Seoul National University. A toroidal assembly of 16 D-shape TF coils is designed to produce the magnetic field of up to 3T, of which ripples appear to be below 4% of the average toroidal field in the plasma region. Exact positions and currents in six equilibrium coils distributed symmetrically in the z=0 plane are found by the solution of a set of linear equations which is transformed from a Fredholm integral equation of the first kind. The decay indices resulted from equilibrium field indicate that the stability condition for vertical and horizontal displacements is satisfied.

Interspecies Comparison of the Oral Absorption of Itraconazole in Laboratory Animals

  • The oral absorption and disposition of itraconazole were studied in rats, rabbits and dogs. Serum levels of itraconazole and its active metabolite, hydroxyitraconazole, were determined by a validated HPLC method. The absorption of itraconazole was relatively rapid in rats and dogs but was slower in rabbits. The terminal elimination half-life ($T_{1/2,{\lambda}z}$), time to the peak concentration ($T_{max}$), dose and weight normalized area under the curve (AUC) and the peak concentration ($C_{max}$) of itraconazole found in the dog were comparable to those reported in humans. As in humans, the metabolite to parent drug AUC ratios in rats and dogs were greater than unity but was less in rabbits. The dog appears to be an appropriate animal model while the rat, not the rabbit, may be used as an alternative animal model in predicting the oral absorption of itraconazole in humans.

  • In order to furnish the fundamental data for the domestic production of magnetic compass and the prescription of standardization about it in Korea, authors made the helmholtz coil and investigated the characteristics of them. Subsequently, the damping curves of T190 and T165 compasses in the helmholtz coil were measured and analyzed the performance of those compass. The results obtained are as follows; 1. The relation between electric current (I sub I) to flow in the helmholtz coils, that the thickness of coil is 1mm, diameter 1m, winding number 117, and intensity of magnetic field is presented as follows. Vertical magnetic force: Z(Gauss)=0.34+1.506 I sub(i) Horizontal magnetic force: H(Gauss)=0.183+1.506 I sub(i) 2. Period of T190 compass is longer than T165 compass in all horizontal magnetic force. In the amplitude, the former is larger than the latter above 0.08 Gauss, but this phenomenon is opposed to that below 0.08 Gauss. 3. As the porizontal magnetic force is intensive, period of magnetic compass is short, amplitude is large, and damping degree and damping factor are small. The time elapsed to the principal points of damping curve is proportional to the -0.65 power of the horizontal magnetic force.

Bioequivalence Study of Toriem® Tablet to Motilium-M® Tablet (Domperidone Maleate 12.72 mg) Evaluated by Liquid Chromatography/Tandem Mass Spectrometry

  • The aim of the present study was to evaluate the bioequivalence of two domperidone maleate tablets, Motilium-$M^{(R)}$ Tablet (Janssen Korea Ltd., reference product) and $Toriem^{(R)}$ Tablet (Daewon Pharm. Co., Ltd., test product). Domperidone was extracted by liquid-liquid extraction using tert-butyl methyl ether and separated in less than 3 min on $C_{18}$ reverse-phase column using an isocratic elution. A tandem mass spectrometer, as detector, was used for quantitative analysis in positive mode by a multiple reaction monitoring mode to monitor the m/z $426.1{\rightarrow}119.1$ and the m/z $837.4{\rightarrow}158.2$ transitions for domperidone and the internal standard (roxithromycin), respectively. Calibration curves, from $0.05{\sim}50$ ng/mL of domperidone, showed correlation coefficients (r) higher than 0.9941. Intra day and inter day precision (C.V. %) for quality control were ranged from 10.04 to 16.09% and from 10.87 to 18.69%, respectively. The lower limit of quantification (LLOQ) of domperidone was 0.05 ng/mL. The method described is precise and sensitive and has been successfully applied to the study of bioequivalence of domperidone in 24 healthy Korean volunteers. Twenty-four healthy male Korean volunteers received a single dose of each medicine ($2{\times}12.72\;mg$ domperidone maleate) in a $2{\times}2$ crossover study. There was a one-week washout period between the doses. Plasma concentrations of domperidone were monitored for over a period of 24 hr after the administration. $AUC_{0-t}$ (the area under the plasma concentration-time curve) was calculated by the linear trapezoidal rule. $C_{max}$ (maximum plasma drug concentration) and $T_{max}$ (time to reach $C_{max}$) were compiled from the plasma concentration-time data. The 90% confidence intervals for the log transformed data were within acceptable range of log 0.8 to log 1.25 (e.g., $log\;0.92{\sim}log\;1.05$ for $AUC_{0-t}$, $log\;0.81{\sim}log\;1.05$ for $C_{max}$). The major parameters, $AUC_{0-t}$ and $C_{max}$ met the criteria of KFDA for bioequivalence indicating that $Toriem^{(R)}$ tablet is bioequivalent to Motilium-$M^{(R)}$ tablet.

Development of the Electronic compass for Automatic Correction do Deviation (自動自差修正이 가능한 電子컴퍼스의 개발에 관한 연구)

  • The Electronic compass made as a pilot model in this research is comprised of a three axis magnetic sensor, an accustar clinometer, and a fiber optic gyro sensor. The results confirming the output character, performance, and the accuracy of the deviation corrects of each sensor are as follows: 1) As for the output character of the three axis magnetic sensor, the magnetic field showed a cosine curve on the X axis, a - sine curve on the Y axis, and constant figures on the Z sensor. The horizontal component H and the vertical component V of the terrestrial magnetism calculated from the output voltage were 33.2${\mu}$T and 23.95${\mu}$T respectively. 2) When the fiber optic gyro sensor is fixed on the electromotive rotation transformation and has made a clockwise rotation with the speed of 10/sec, 20/sec, and 30/sec, the relationship between the output and the rotation angle of the fiber optic gyro sensor showed proportionally constant values. 3) When the magnetic field was induced with a magnet, the deviation before the correction was significant at a high of 25. However, the deviation after the correction using Poisson correction was in the 2 range, significantly lower than before the correction. It was confirmed that automatic deviation corrects are possible with the electronic compass made as a pilot model in this research.
