Evolutionary Optimization of Pulp Digester Process Using D-optimal DOE and RSM

  • Optimization of existing processes becomes more important than the past as environmental problems and concerns about energy savings stand out. When we can model a process mathematically, we can easily optimize it by using the model as constraints. However, modeling is very difficult for most chemical processes as they include numerous units together with their correlation and we can hardly obtain parameters. Therefore, optimization that is based on the process models is, in turn, hard to perform. Especially, f3r unknown processes, such as bioprocess or microelectronics materials process, optimization using mathematical model (first principle model) is nearly impossible, as we cannot understand the inside mechanism. Consequently, we propose a few optimization method using empirical model evolutionarily instead of mathematical model. In this method, firstly, designing experiments is executed fur removing unecessary experiments. D-optimal DOE is the most developed one among DOEs. It calculates design points so as to minimize the parameters variances of empirical model. Experiments must be performed in order to see the causation between input variables and output variables as only correlation structure can be detected in historical data. And then, using data generated by experiments, empirical model, i.e. response surface is built by PLS or MLR. Now, as process model is constructed, it is used as objective function for optimization. As the optimum point is a local one. above procedures are repeated while moving to a new experiment region fur finding the global optimum point. As a result of application to the pulp digester benchmark model, kappa number that is an indication fur impurity contents decreased to very low value, 3.0394 from 29.7091. From the result, we can see that the proposed methodology has sufficient good performance fur optimization, and is also applicable to real processes.

An Immunoserological Study of Vaccine Against Haemorrhagic Fever with Renal Syndrome (신증후출혈열 백신의 면역혈청학적 연구)

  • Since $Hantavax^{TM}$, formalin inactivated Hantaan virus vaccine (10,240 ELISA units/ml), has been developed in 1990 to prevent against haemorrhagic fever with renal syndrome (HFRS) caused by Hantaan or Seoul virus, it has been commercially available in Korea. Twenty-one healthy people were booster shot once and twice after primary basic vaccination with $Hantavax^{TM}$. Seroconversion rates were measured by immunofluorescent antibody technique (IFAT), enzyme-linked immunosorbent assay (ELISA), high density composite particle agglutination (HDPA), and plaque reduction neutralization test (PRNT). Seroconversion rates of 21 vaccinees at one year after primary basic vaccination were 52.3%, 95.2%, 0.0%, 47.6%, and 28.6%, and 13 vaccinees at one month after 1st booster vaccination were 100%, 100%, 30.7%, 100% and 100% by IFAT, ELISA (IgG, IgM), HDPA and PRNT, respectively. Seroconversion rates declined slightly by twenty months, and they were 84.6%, 92.3%, 0.0%, 84.6% and 69.2% by IFAT, ELISA (IgG, IgM), HDPA and PRNT, respectively. Seroconversion rates of 9 vaccinees at three months after 2nd booster vaccination were 100%, 100%, 0.0%, 100%, and 88.9%, and 16 vaccinees at one year after the 2nd booster vaccination were 87.5%, 93.8%, 0.0%, 87.5% and 81.3% by IFAT, ELISA (IgG, IgM), HDPA and PRNT, respectively. Based on the above result $Hantavax^{TM}$ has proved a vigorous anamnestic response after the 1st and the 2nd booster vaccination and has persisted higher fluorescence, agglutination and neutralizing antibody titers in vaccinees.

Detection and Characterization of Enteroviral RNA in Paraffin-embedded Heart Tissues from Patients with Dilated Cardiomyopathy

  • The aim of this study was to investigate viral etiology in dilated cardiomyopathy (DCM) by polymerase chain reaction (PCR) or nested reverse transcription PCR (RT-PCR), and characterize the enteroviral RNA presented in the clinical specimens. Twenty-eight paraffin-embedded heart tissue samples were assayed to detect cytomegalovirus, herpes simplex virus type 1, type 2, parvovirus, adenovirus, and enterovirus (EV) with each specific primer. Of these 28 patients (mean age: 27, M: 24, F: 4), 26 were histologically diagnosed as DCM and 2 as myocardial infarction (MI). Nested RT-PCR detected enteroviral RNA in 7 (26.9%) of 26 patients with DCM, and none of patients with MI. And none of DNA viruses tested were detected from the samples. Amplified products were also genotyped by single-strand conformation polymorphism (SSCP). Three subtypes can be differentiated from 7 clinical specimens. Furthermore, direct sequence analysis was performed to determine whether genetic variation of EV is present in the explanted heart tissues from patients with DCM. Although most of the sequences among the wild isolates have the greatest similarity to those of coxsackievirus B3, there are specific regions of variable sequences (no 490 - no 510). The data suggest that enterovirus may be a major viral pathogen for the DCM in Korea and nucleotide sequence data indicate that coxsackievirus B3 may be a leading etiologic agent of DCM.

  • Ceramic femoral heads are now widely used in Total Hip Replacement (THR). Due to their high biocompatibility and low ductility, ceramic femoral heads are considered to be suitable for young and active patients. However, as in testing the mechanical stability of the femoral head, the conventional proof test (standard ISO 7206-10) has its limit of showing axisymmetric stress distribution on the contact surface, while non-uniformed stress distribution is expected after THR. Since non-uniformed stress distribution can result in the increased probability of ceramic femoral head fracture, it is considerable to evaluate the stress distribution in vivo-like conditions. Therefore, this study simulated the ceramic femoral heads under in vivo-like conditions using finite element method. The maximum stress decreased when increasing the size of the femoral head and stress distribution was concentrated on superior contact surface of the taper region.

Effects of Dietary Bamboo Charcoal on the Carcass Characteristics and Meat Quality of Fattening Pigs

  • The purpose of this study was to investage the effects of dietary bamboo charcoal on the carcass characteristics and meat quality of fattening pigs. Fifty four crossed pigs of $61.0{\pm}1.0$ kg body weight (BW) were grouped and housed in 6 animals (3 barrows and 3 gilts) per pen and 3 replications per treatment. The basal diet (C) was supplied with 0.3% bamboo charcoal as treatment 1 (T1) and 0.6 % as treatment 2 (T2). The pigs were fed that experimental diet for 42 days, thereafter 10 longissimus dorsi (LD) per treatment were randomly collected at the time the pigs reached an average weight of $110.0{\pm}5.0$ kg. The carcass weight, backfat thickness, and the carcass grade were better (p<0.05) in the pigs fed bamboo charcoal than in C. The crude fat concentration of LD was higher (p<0.05) in T1 than in C. While the composition of stearic acid and arachidonic acid was lower (p<0.05) in treatments than in C, the composition of oleic acid and linoleic acid of treatments was higher (p<0.05) than C. The physico-chemical characteristics, such as meat color and amino acid composition of LD were not affected (p>0.05) by the supplemented bamboo charcoal. In conclusion, dietary supplementation with bamboo charcoal improved the carcass grade and fatty acids composition of pork meat from fattening pigs, where the composition of unsaturated fatty acids was increased, but that of saturated fatty acids was decreased.

Effect of the Pattern Making Method on the Silhouette of the Flared Skirt (패턴 제작 방법이 플레어스커트의 실루엣에 미치는 영향)

  • The purpose of this study is to provide the basic knowledge for the pattern development of a flared skirt. Two flat pattern (FP) methods, the concentric circle method and the corrected concentric circle method, and a draping were used for pattern making. Using these patterns, skirts were made with lengthwise grain in the centerline. The influence of pattern making method on the silhouette of the flared skirt was evaluated by the hemlines formed at the skirt worn in a dress form. Moreover, the combined influence of pattern making method and cutting direction on the silhouette of the flared skirt was examined with the draping pattern (DP) skirt and a FP skirt with $45^{\circ}$ bias grain in the centerline. The DP skirt had more uniform nodes and formed a stable wave form than the FP skirts throughout the whole hemlines. The number of nodes was reduced with lining in the two FP skirts, whereas the number of nodes was not changed in the DP skirt. The unit wave form of all the skirts by three patterns showed long loop form, and the slope angles of the unit wave form of the two FP skirts were higher than that of the DP skirt. The silhouette of the flared skirt was highly influenced by the shape of the waist circumference line in patterns. The DP skirt with lengthwise grain in the centerline showed good silhouette with uniform nodes and high marking efficiency, compared to the FP skirt with $45^{\circ}$ bias grain in the centerline.

Epidemiological studies on bovine tuberculosis in mass outbreak region (우결핵 집단 발생직역에 대한 역학적 고찰)

  • The epidemiological survey of mass outbreak region of bovine tuberculosis from January of 2007 through May in 2009. The results were enumerated as follows. The results of tuberculin skin test are: 7 (0.4%) out of 1,697 in 2007, 61 (2.8%) out of 2,163 in 2008, 80 (4.9%) out of 1,639 in 2009. The sex and age distribution among the incidence of positive: 135 (91.8%) out of 147 in female, 12 (8.2%) in male. Among female, age 1: 6.1%, age 2: 30.6%, 3: 38.8%, 4: 14.2%, 5: 0.7% and 6: 1.4%. Among male, age 1: 4.1%, 2: 1.4%, 3: 2.7% and more frequent occurrence in age 3, 38.8% in female and 2.7% in male. The rate of recurrence by farms: recurrence 1: 6 (35.3%), 2: 9 (52.9%), 3: 1 (5.9%), 6: 1 (5.9%), The recurrence rate of 2 or more was 64.7%. The ELISA test result among 114 heads over 14 farms: 75 (65.8%) showed positive and 39 (34.2%) negative. Geographical distribution of recurrence is characterized as concentrated along the major traffic and stream crossing the village, and spread from the high elevation to downward area.

Prevalence of antibodies against bovine viral infectious diseases in farmed deer and wild water deer in Jeonbuk province (사육사슴 및 야생고라니의 소 바이러스성 전염병에 대한 혈청학적 연구)

  • Farmed deer could be susceptible carrier to bovine viral infectious disease. But unfortunately, there has not been an overall study over this subject in Korea so far. Therefore, a study was conducted to see serum antibodies to bovine leukosis, food and mouth disease, bovine viral diarrhea and infectious bovine rhinotracheitis in deer using the sera of farmed deer. As a result, two deer in a farms showed positive in bovine leukosis antibodies, using ELISA. For wild water deer, no antibodies were found for those diseases. As a result, it can be assumed that deer were relatively low rate of exposure to highly contagious disease such as viral bovine infectious disease in Korea. As this study was conducted over limited in number of subject and regions, continued study should be carried out in order to prevent and control the interspecies transmission in the future.

Development of a Finite Element Program for Determining Mat Pressure in the Canning Process for a Catalytic Converter (촉매변환기를 캐닝할 때 발생하는 매트의 압력분포 유한요소해석 프로그램의 개발)

  • The catalytic converter in the front part of an automobile's exhaust system converts toxic exhaust gas into nontoxic gas. The substrate in the central part of the converter has a circular or oval-shaped cross section and fine lattice-shaped walls. In the canning process, the substrate is wrapped in mats and inserted into a can. During this process, mat pressure is induced, which may cause brittle fracturing in the substrate. In this paper, a finite element program for determining the mat pressure distribution was developed to avoid these fractures. The program was created in Microsoft EXCEL, so the input and output procedures are relatively simple. It was assumed that the substrate is rigid, the mat is material nonlinear, and the can is linear elastic. The can is modeled as a beam element to resist both bending and uniform tension/compression. The number of elements is fixed to 35, and the number of iterations, to 20. The solutions are compared to ABAQUS solutions and found to be in good agreement.

Effects of a Dietary Fermented Mushroom (Flammulina velutipes) By-Product Diet on Pork Meat Quality in Growing-Fattening Berkshire Pigs

  • This study was carried out to investigate the effects of fermented mushroom (Flammulina velutipes) by-products on meat quality characteristics in fattening Berkshire pigs. The fermented diet mainly contained 40.0% mushroom by-products, 26.0% rice bran, and 20.0% formula feed and was fermented for 5 d. The basal diet for the control (C) was substituted with 10% (T1), 30% (T2), 50% (T3), and 70% (T4) fermented diet. Warner-Bratzler shear forces (WBSF) were significantly lower (P < 0.05) in treatments than that in C. The meat color (lightness, redness, and yellowness) was significantly lower (P < 0.05) in treatments than that in C, whereas fat color (redness and yellowness) was significantly higher in treatments than that in C (P < 0.05). The compositions of palmitoleic acid and arachidonic acid were significantly higher (P < 0.05) in T4 than that in C. The amino acid composition of longissimus dorsi (LD) and the sensory evaluation of cooked meat were not affected by diet type. In conclusion, a diet of fermented mushroom by-products increased pH and backfat color, but decreased cooking loss, WBSF, and meat color of LD in growing-fattening Berkshire pigs.
