Dietary Supplementation of Fat Increased Milk Fat Percentage without Affecting Ruminal Characteristics in Holstein Cows in a Warm Tropical Environment

  • The purpose of this experiment was to investigate the effect of the diets supplemented with lard or prilled fat ($Carolac^{(R)}$) on lactation performance, plasma constituents and ruminal characteristics of Holstein cows under a warm climate. In trial 1, 18 Holstein cows, 14 primiparas at 43 DIM and 4 multiparas at 55 DIM, were randomly assigned into six $3{\times3}$ Latin squares, in which three dietary treatments were isoproteinous but varying in energy contents and three 21 d periods. The treatments were basal diet (Control), basal diet supplemented with 2.5% lard (LD), and basal diet supplemented with 2.5% commercial Prilled fat (PF). In trial 2, three rumen canulated pregnant nonlactating Holstein cows with 550 kg average body weight were allotted into a $3{\times}3$ Latin square design with diets same as in trial 1 were fed to the cows at the level of 1.5% body weight on dry matter (DM) basis. The results indicated that the DM intake did not differ among the treatments. Milk yield and 4% FCM yield were greater (p<0.05) in PF than in Control. LD and PF resulted in greater milk fat percentage. Protein, lactose and solid contents in milk were not different among the three dietary treatments. The concentration of nonesterified fatty acids (NEFA) in plasma was significantly greater in LD and PF than that in Control. However, the concentrations of triglycerides, urea nitrogen, and cholesterol in plasma were not significantly different among the three treatments. Although the ruminal molar percentage of isobutyrate in LD and PF was greater, no significant difference was observed in ruminal pH, NH3-N concentration and VFA production among the three treatments. Diet supplemented with fat can improve milk yield and milk fat percentage without resulting in disadvantages of ruminal characteristics in cows at early lactation and under warm climate.

  • In this study a sediment yield is compared by IUSG, IUSG with Kalman filter, tank model and tank model with Kalman filter separately. The IUSG is the distribution of sediment from an instantaneous burst of rainfall producing one unit of runoff. The IUSG, defined as a product of the sediment concentration distribution (SCD) and the instantaneous unit hydrograph (IUH), is known to depend on the characteristics of the effective rainfall. In the IUSG with Kalman filter, the state vector of the watershed sediment yield system is constituted by the IUSG. The initial values of the state vector are assumed as the average of the IUSG values and the initial sediment yield estimated from the average IUSG. A tank model consisting of three tanks was developed for prediction of sediment yield. The sediment yield of each tank was computed by multiplying the total sediment yield by the sediment yield coefficients; the yield was obtained by the product of the runoff of each tank and the sediment concentration in the tank. A tank model with Kalman filter is developed for prediction of sediment yield. The state vector of the system model represents the parameters of the tank model. The initial values of the state vector were estimated by trial and error.

Effect of Graded Levels of Green Grass Supplementation on Nutrient Digestibility, Rumen Fermentation and Microbial Nitrogen Production in Cattle Fed Rice Straw Alone

  • On an absolute straw diet, the effect of graded levels of green grass supplementation on intake, nutrient digestibility, rumen fermentation pattern and microbial N yield has been studied in cattle. Of the two trials conducted, 16 intact growing bulls of 304 kg weight and 32 months old, randomly allocated to four treatments in a completely randomized design in the 1st trial. While, in the, 2nd trial, four rumen cannulated local bulls of about 60 months old and 400 kg weight were used in a $4{\times}4$ Latin square design with four treatments in four periods. In both the trials, in addition to a mineral mixture, animals were supplemented with graded levels of naturally grown green grass of 0 kg (T1), 2 kg (T2), 4 kg (T3) or 6 kg (T4) to an ad libitum rice straw diet In the 1st trial, measurements were made on intake digestibility, growth rate, N balance and microbial N yield. While in the 2nd trial, in addition to the above parameters (except growth rate), rumen parameters were also studied. All levels of grass supplementation decreased the straw DM intake and increased the substitution rate. The rumen $NH_3-N$ concentration increased with the increase in grass level and ranges from 8-46 mg/l. The rumen pH and the rate and extent of DM degradability of straw were not affeceted by different rumen environments created by different levels of grass inclusion. At 48 h, straw DM degradability were 42, 44, 44 and 43% respectively for 0, 2, 4 and 6 kg grass supplementation daily. The whole gut digestibilities of DM, OM and ADF increased significantly (p < 0.05) only at 6 kg level daily. The microbial N yield was not affected by the levels of grass supplemented. The mean microbial N yield was 10 (SD 3.7) g/kg DOM apparently fermented in the rumen. The estimated minimum N loss and thus the maintenance requirement of tissue protein was 303 mg/kg $W^{0.75}/d$. All the animals lost live weight but 6 kg grass supplementation gave positive energy and N balances. Small amount of green grass supplementation is often recommended for optimization of rumen environment of a straw diet However, under the present experimental condition, no such beneficiary effect observed up to 6 kg (26% of DM intake) level of supplementation.

A New Yellow Waxy Corn Hybrid with High Yield "Daehakchal Gold 1" for Edible

  • A new yellow waxy corn hybrid "Daehakchal Gold 1" was developed from single cross between Yeongdeok Jaera and Okchen Jaera at Chungnam National University in 2007. Inbred CNU57 derived from Yeongdeok Jaera was used as the seed parent of Daehakchal Gold 1, and inbred CNU 27 derived from Okchen Jaera as the pollen parent. Tasseling date of this hybrid was seven day later than that of check hybrid, Chalok 1. Daehakchal Gold 1 was 19.7cm in ear length and 4.5cm in ear diameter. The yield of Daehakchal Gold 1 and check hybrid in dry matter were 146.5g and 113.9g per plant, respectively. On yield trial, which were increased 29.7% compared with a check hybrid, Chalok 1. Especially, Daehakchal Gold 1 had yellow kernels and good eating quality. The ratio of kernel set length/ear length was similar to Chalok 1. It is moderately resistant to Bioporalis maydis and corn borer. The yields of Daehakchal Gold 1 in fresh ear weight and in number of fresh ear were 16% and 8%, respectively, higher than those of a check hybrid in regional yield trials for three years. Seed production of this hybrid has gone well due to good match during crossing between seed and pollen parents.

  • To select the highest yield soiling type forage, the yield performance of selected sorghum-sudangrass hybrids imported in 1992 have been evaluated at Chonnam region. The soiling type hybrids used in this experiment were TE-haygrazer, FP-5 Sudan, Sweet sioux V, and X -78050. Three different cuttings were done on July 6, August 6 and September 16 in 1992. This trial was arranged as a randomized complete block design with three replications under taken over a period 5 months from May to September in 1992. The results of this experiment were summarized as follows: The hybrids tested were not significantly different by plant height, leaf blade length, leaf blade width and stem diameter. In the first cutting, dry matter yield of FP-5 Sudan was the highest. In the second and third cutting, dry matter yield of Sweet sioux V was the highest among hybrids. The contents of crude protein, hemicellulose, relative feeding value(RFV), and amino acid were the highest by the first cutting, but NDF and ADF were shown the lowest.

Yield Response of Bushbean (Phaseolus vulgaris L.) to Doses of Fertilizers and Sowing Times in Bangladesh

  • The present study was initiated to explore the yield potential of bushbean by exploiting the interaction of variety, sowing time, and fertilizers containing N, P, and K Two varieties namely BARI Bushbean 1 and BARI Bushbean 2, three levels of fertilizer viz. control ($N_{0}P_{0}K_{0}$), medium ($N_{20}P_{30}K_{30}$), high ($N_{40}P_{60}K_{60}$) and three sowing times (November 15, December 1 and December 15) were considered as treatment variables. Among the varieties, BARI Bushbean 2 always showed better performance for most of the yield and yield attributes duly attributed by the application of fertilizers ${\circleda}$ N: P: K = 40: 60: 60 respectively. The highest seed yield (1375.17 kg/ha) was recorded when the crop was sown on the $15^{th}$ November with the supplemented soil nutrition status as above. The influence of sowing times indicate that there is a gradual trend of decreasing in seed yield and other associated parameters after first sowing ($15^{th}\;November$). The results of this study suggest that the multi-locational trial under varied sowing times with soil nutrition might potentially increase the long-term adaptation of bushbean in Bangladesh.

Effect on Milking Performance of Vitamin-Trace Element Supplements to Early Lactation Italian Brown Cows Grazing Ryegrass (Lolium multiflorum) Pasture

  • The objective of this study was to examine the effects of concentrates containing different levels of a vitamin-trace elements premix on milk yield and composition of dairy cows. The trial, which lasted 14 weeks, was conducted from January to March and used 45 multiparous Brown cows in the early phase of lactation. Cows (n = 15 per treatment) were randomly allocated to three dietary treatments: the first group (control, C-0) was fed pelleted concentrate containing background vitamins and trace elements that supplied 1.0 times cows' daily requirements; the second group were fed the same concentrate, but containing 2.5 g/kg of vitamin and trace mineral premix per kg of concentrate (C-2.5); the third group were fed the same concentrate, but containing 5 g/kg of vitamin and trace mineral premix per kg of concentrate (C-5). The daily ration included ad libitum chopped oat hay, and the cows also had 8 h/d grazing on a ryegrass (Lolium multiflorum) pasture. During the performance trial, cow milk yield was daily recorded and individual milk samples were analysed for milk composition and to determine milk renneting properties. Cows fed the intermediate premix level (C-2.5) in diet showed the highest fat-corrected milk production (p<0.05) compared to other groups. None of the milk quality parameters studied were influenced by dietary treatment, except for milk rheological parameters (rennet clotting time and curd firmness) that were positively improved in cows fed the C-2.5 diet (p<0.05). The findings from this study show that intermediate level of vitamin-trace elements premix in concentrate can be advantageously used in grazing dairy cows without negative effects on yield and quality of milk produced.

농업형질 및 생리활성 평가를 통한 황기 단간 계통 선발 (Selection of Short Stem Astragalus membranaceus Lines by Assessing Agronomic Characteristics and Biological Activity)

  • Background: Astragalus membranaceus belonging to the Leguminosae family is often utilized as a traditional medicine. The aim of this study was to elucidate the basic breeding information required to develop short stem A. membranaceus cultivars. Methods and Results: Roots of A. membranaceus advanced yield trial (AYT) lines were harvested in late October 2017. Root yield of six AYT lines were increased in a range of 8.9 - 74.8% compared with 'Aseong' as control (check variety). The height of seven AYT lines were shorter than that of 'Aseong'. In addition, stem diameter of nine AYT lines was thicker than that of 'Aseong'. Consequently, 1502-56, 1503-90, and 1510-80 were selected as elite lines for the development of short stem cultivars. HPLC analysis was perfromed to identify lines with high level active components such as calycosin-7-O-${\beta}$-D-glucoside and calycosin. The levels of both active components were higher in 1502-56, and 1503-90, but not in 1510-80 compared to 'Aseong'. In addition, 2,2-Diphenyl-1-picryl hydrazyl (DPPH) radical scavenging activity was higher in the 1502-56 compared to 'Aseong'. Considering these results, two AYT lines, 1502-56 and 1503-90 were selected as short stem lines with high calycosin-7-O-${\beta}$-D-glucoside and calycosin content. Conclusions: Taken together, Two short stem lines were identified in this study. In our future study, regional yield trial (RYT) will be conducted using these selected lines to develop new cultivars.

A Wheat Variety, "Hwanggeumal" with Good Bread Quality, Red Grain, Partial Waxy, Tolerance to PHS

  • A new winter wheat(Triticum aestivum L.) cultivar "Hwanggeumal" was developed by the NICS(National Institute of Crop Science), RDA(Rular Dvelopment Administraion) in 2019. It was derived from a cross of the "Jokyoung//Kauz/Rayon" and "Jopoom" in 2008. It had advanced generation through bulk and pedigree method for seven years and designated line name "Jeonju398" after AYT(Advance Yield Trial) test for two years. And "Hwangeumal" was designated variety name after RYT(Regional Yield Trial) test in eight locations around Korea for two years from 2018 to 2019. Its heading date was April 19 and maturity date was May 31, which were similar to Jokyoung. "Hwanggeumal" had shorter plant height(75 cm) and spike length(7.1 cm), spikes per m2(699) and lower 1,000 grain weight(44.2 g) than "Jokyoung"(78 cm, 8.2 cm, 776, 46.6 g, respectively). "Hwanggeumal" was showed weak to winter hardiness and susceptible to powdery mildew but tolerance to PHS(Pre-harvest sprouting). The average grain yield in the AYT was 6.2 ton/ha, which were 10% more than "Jokyoung" And in the RYT was 5.1 ton/ha in upland and 4.4 ton/ha in paddy field, which were lower than "Jokyoung", respectively. "Hwanggeumal"s flour yield (71.4%) and flour lightness (91.82) showed similar to "Jokyung" and higher protein content (14.0%) and gluten content (10.3%) and SDS-sedimentation volume (60.3ml). These result showed that the "Hwanggeumal" dough strength of flour is strong than "Jokyung". "Hwanggeumal"s HMW-GS(High molecular weight gluten subunits) composition are Glu-D1 (5+10), Granule-bound starch synthase(GBSS) composition are Wx-A1 (a), Wx-B1 (b), Wx-D1 (a) and composition of Puroindolines are Pina-D1(a), Pinb-D1(b).

Reliable preparation of [11C]GR205171, a selective NK1 radioligands for noninvasive imaging

  • [11C]GR205171, a Neurokinin 1 (NK1) radioligand, has been known as such a promising PET probe for quantitation of NK1 receptors in the brain by positron emission tomography (PET) imaging. First trial to synthesis of [11C]GR205171 was to use methylene chloride and tetrabutylammonium hydroxide for preactivation of precursor, but the result was not successful in radiochemical yield (0~25%) and unreliable. 7 years later, inorganic base (Cs2CO3) was tried to achieve higher radiochemical yield, and they showed higher yield (~53%). We have tried to repeat the same synthesis method, but it did not work properly, because there were the lack of the detail procedure and still reproducibility in radiochemical yield. Here we report the improved synthesis protocol to produce [11C]GR205171 in high yield via commercial automated synthesizer. The sonicator which combines water heating bath was used to activate desmethyl-GR205171, and this method showed high efficiency and reasonable yields (4.7 ± 0.6%, non-decay corrected from molecular sieve trap) with >95% radiochemical purity.
