Anitoxidative components of pueraria root (칡뿌리의 항산화 성분)

  • As a search for natural antioxidants, antioxdative fractions in Pueraria roots were extracted, identified using column chromatography, thin layer chromatography or high performance liquid chromatography. The components which have most effective antioxidative activities were further identified by IR and GC-MS. Separated antioxidative components were then added to four different oils to examine their antioxidative activities. Yield of extract obtained from pueraria root powder by solvent extraction using four step solvent systems was 2.54%. Antioxidative activity of the extracts was as effective as that of 100 ppm ${\delta}$-tocopherol addition, when 0.1% of the extracts was added to linoleic acid. The strongest antioxidative component of methanol extract of pueraria root was identified as puerarin. Aunioxidative activity of puerarin on lard was more effective than ${\alpha}$-tocopherol, but less effective than ${\delta}$-tocopherol. When the puerarin was added to edible oil and heat treated at $145^{\circ}C$, the acid value was lowest in lard and was highest in soybean oil. Antioxidative activity in terms of carbonyl value, thiobarbituric acid value and anisidine value was most high in palm oil and least in soybean and cottonseed oil.

Change Vector Analysis : Change detection of flood area using LANDSAT TM Data (LANDSAT TM을 이용한 홍수지역의 변화탐지 : Change Vector Analysis 방법을 중심으로)

  • Change detection and analysis is a powerful application of remote sensing, in that the spectral resolution of multi-band sensors can be used to advantage in monitoring both significant and subtle land cover changes over time. In this study, the LANDSAT TM data was used to detect the change areas affected by flood from a heavy rainfall. The study area is the Nakdong River located in the Korea peninsular. Among the several change detection techniques, change vector analysis(CVA), principle component analysis(PCA) and image difference approach are utilized in this paper. CVA uses any number of spectral bands from multi-date satellite data to produce change image that yield information of the magnitude and direction of differences pixel values. And accuracy assessment was carried out with a change image produced from three techniques. In result, CVA was found to be the most accurate for detecting areas affected by flood. CVA with the overall accuracy and Kappa coefficient of 97.27 percent and 94.45 percent, respectively.

  • The presenilin 1 (PS1) or PS2 is an essential component of the ${\gamma}$-secretase complex, which mediates the intramembrane proteolysis of selected type-I membrane, including the ${\beta}$-amyloid precursor protein (APP) to yield A${\beta}$. Familial Alzheimer's disease (FAD)-associated mutations in presenilins give rise to an increased production of a highly amyloidogenic A${\beta}$42. In addition to their well-documented proteolytic function, the presenilins play a role in calcium signaling. We have previously reported that presenilin FAD mutations cause highly consistent alterations in intracellular calcium signaling pathways, which include deficits in capacitative calcium entry (CCE), the refilling mechanism for depleted internal calcium stores. However, molecular basis for the presenilin-mediated modulation of CCE remains to be elucidated. In the present study, whole-cell patch clamp method was used to identify a specific calcium-permeable ion channel current(s) that is responsible for the CCE deficits associated with FAD-linked PS1 mutants. Unexpectedly, both voltage-activated and conventional store depletion-activated calcium currents I(CRAC), were absent in HEK293 cells, which were stably transfected either with wild-type or FAD mutant (L286V, M146L, and delta E9) forms of PS1. Recently, magnesium-nucleotide-regulated metal cation current, or I(MagNum), has been described and appears to share many common properties with I(CRAC) including calcium permeability and inhibitor sensitivity (e.g. 2-APB). We have detected I(MagNum) in all 293 cells tested. Interestingly, FAD mutant 293 cells developed only about half of currents compared to PS1 wild type cells.

Study on Oil Production from Pyrolysis of Mixed Plastic Waste Using Multidimensional Chromatography (Multidimensional Chromatography/Mass Spectrometry를 이용한 혼합 폐플라스틱의 열분해 오일 특성 평가에 관한 연구)

  • The total hydrocarbon distribution of oil products obtained from the pyrolysis of four kinds of mixtures of polyethylene-polystyrene waste has been studied by multidimensional chromatography(high performance liquid chromatography followed by capillary gas chromatography)/mass spectrometry. Saturated, unsaturated and aromatic hydrocarbons in oil products were selectively pre-separated according to structural groups by HPLC and the weight fraction of each group was estimated by analysis of each component using GC-FID response factors. The hydrocarbon distribution of aliphatic fraction consists of $C_{5}$ to $C_{25}$ saturated and unsaturated hydrocarbons. And that of aromatics fraction consists of benzene, toluene, xylene, styrene, propenyl benzene, naphthalene, and some of derivatives. Pyrolysis temperature did not affect the ratio of total weight fraction of aliphatic over aromatic hydrocarbon distribution in case of PS only and PE-PS mixtures (1:1 and 1:4 wt. ratio) as a feed while affected the ratio of total wt. fraction in case of PE only. The optimal temperature for the maximum oil production was $600^{\circ}C$ for pyrolysis of PS and 1:1 and 1:4 mixtures of PE and PS. The optimal condition for aromatic recovery was $600^{\circ}C$ with 1:1 mixture of PE and PS. In this condition, aromatic was produced up to 90% of total oil product. The maximum yield of toluene, xylene, styrene, and propenyl benzene were 8.6, 8.9, 51.0 and 7.4% of feed for pyrolysis PS at $700^{\circ}C$, respectively. However, only 1.3% naphthalene was recovered at $700^{\circ}C$ with 1:1 PE:PS(by wt.).

Extraction, Component Analysis and Antibacterail Activity of Panax ginseng Absolute Essential Oil (인삼 앱솔루트 정유의 추출, 성분 분석 및 항균 활성)

  • Interests in natural products have been gradually increasing as well-being era has arrived. Panax ginseng was chosen since it represents Korean traditional medicine as its effect on many age-related symptoms. Panax ginseng absolute essential oil was prepared and analyzed for its main components. Ginseng absolute oil was extracted from Panax ginseng using organic solvent of hexane and ethanol with the yield of 0.146%. The oil was analyzed by GC-MS and main components are sesquiterpenes such as neoclovene, panasinsene, and calarene and more than 110 components were identified from it. The oil was tested for antibacterial activity against general bacteria(E. Coli, S. aureus and P. aeruginosa), yeast, acne germ(Propionibacterium acne) and dandruff germ(Pityrosporum ovale). Panox ginseng absolute essential oil showed the prominent antibacterial activity against Propionibacterium acnes ACCC 6919.

Effects of Sweet Bee Venom on the Central Nervous System in Rats -using the Functional Observational Battery- (Sweet BV 시술이 Rat의 중추신경계에 미치는 영향 - 기능관찰 종합평가를 이용하여-)

  • Objectives: This study was performed to analyse the effects of Sweet Bee Venom(Sweet BV-pure melittin, the major component of honey bee venom) on the central nervous system in rats. Methods: All experiments were conducted at Biotoxtech Company, a non-clinical studies authorized institution, under the regulations of Good Laboratory Practice (GLP). Male rats of 5 weeks old were chosen for this study and after confirming condition of rats was stable, Sweet BV was administered in thigh muscle of rats. And checked the effects of Sweet BV on the central nervous system using the functional observational battery (FOB), which is a neuro-toxicity screening assay composed of 30 descriptive, scalar, binary, and continuous endpoints. And home cage observations, home cage removal and handling, open field activity, sensorimotor reflex test/physiological measurements were conducted. Results: 1. In the home cage observation, there was not observed any abnormal signs in rats. 2. In the observation of open field activity, the reduction of number of unit areas crossed and rearing count was observed caused by Sweet BV treatment. 3. In the observation of handling reactivity, there was not observed any abnormal signs in rats. 4. In the observation of sensorimotor reflex tests/physiological measurements, there was not observed any neurotoxic signs in rats. 5. In the measurement of rectal temperature, treatment of Sweet BV did not showed great influences in the body temperature of rats. Conclusions: Above findings suggest that Sweet BV is relatively safe treatment in the central nervous system. But in the using of over dose, Sweet BV may the cause of local pain and disturbance of movement. Further studies on the subject should be conducted to yield more concrete evidences.

  • Damping is a property for materials and systems to dissipate energy during periodic deformations. Generally, damping causes stiff decrease in amplitudes and shifts in phases. Also, even at resonance, amplitudes are substantially attenuated. This phenomenon of damping helps in reducing stresses developed during vibrations and consequently improves fatigue lives of materials. In this work internal damping and complex moduli are experimentally determined. An impulse technique is utilized in experiments and cantilever beams are selected as test subjects for the measurements of flextural vibrations since the beams lend themselves easily as simplistic ideal models. A resonance method is employed to determine resonance frequencies which are utilized to compute storage moduli. Also, loss moduli are evaluated from damping capacities and storage moduli. The storage and loss moduli combined yield complex moduli. Finally internal damping is evaluated from bandwidth technique, the real component of the transfer function.

Recovery of Lactic Acid from Fermentation Broth Using Precipitation and Reactive Distillation (발효액으로부터 침전과 반응증류를 이용한 젖산의 회수)

  • Precipitation and reactive distillation were employed to recover lactic acid from fermentation broth. Lime was initially added to fermentation broth in order to convert soluble lactic acid to an insoluble calcium lactate form. Drowning-out crystallization was used to decrease the solubility of calcium lactate by adding ethanol as a co-precipitant. In the ideal solution of organic acids as well as fermentation broth, precipitation experiments were performed with varying amounts of ethanol. Precipitation process was followed by reactive distillation. Carboxylate salts formed in the previous precipitation process were mixed with carbon dioxide and triethylamine to precipitate as calcium carbonate. The remaining liquid was distilled for 1 hr at different temperatures. Triethylamine and water were recovered from the top of the distiller, while organic acids, inducing lactic acid as a main component remained in feeding bottle. The yield of recovered lactic acid was 67.5% with the purity of 99.7%.

A Study on Development of High Functional Materials Producing Technique using By-products from Skate Processing (1) - Development of Chondroitin Sulfate Materials using Skate Cartilages (저이용 홍어 가공 부산물을 이용한 고기능성 물질의 생산기술 개발에 관한 연구(1) - 홍어연골을 이용한 콘드로이틴 황산 소재 개발)

  • For the purpose of reuse the wasted by-products from the skate process to the health functional food or medicinal material, chondroitin sulfate was extracted from the skate cartilage with the method of hydrolysis with protease enzyme, and the contents of chondroitin sulfate and hydrolyzed protein were measured qualitatively and quantitatively. The effects of chondroitin sulfate on body weight or liver weight changes, hepatotoxicity elimination and anti-inflammatory actions were measured from in vivo test with feed-treated mice. From the hydrolytic extraction of skate cartilage with the mixture of 1% alcalase and 1% protease for 4 hours, the extraction yield of chondroitin sulfate was about 32.55%. The content and molecular weight of chondroitin sulfate was 26.63% and $2.85{\times}10^5Da$., respectively and the content ratio of chondroitin sulfate to protein was measured to 1 to 2.76 with gel permeation chromatography. For the odor component, trimethylamine decreased about 30% but almost not ammonia from chondroitin sulfate with the treatment of activated carbon. From the feeding chondroitin sulfate to mice, the control effect of body and liver weights decrease was measured, anti-inflammatory action and hepatotoxicity elimination action were also measured. From these results, process operation conditions for manufacturing of chondroitin sulfate were suggested.

Input LC Fiter Design of Diode Rectifiers Considering Filter VA Rating Reduction (필터소자의 용량 저감을 고려한 다이오드 정류기의 입력LC필터 설계)

  • In this paper, input LC filter design of diode rectifiers considering filter V A rating reduction has been propoesd. It consisted of an input LC parallel resonent tank whose inductor and capacitor values are se$.$ lected so that the input filter presents an infinite impedance to harmonic input ac current component. The operation of proposed input filter has been analyzed in detail under steady state conditions. Performance evaluation and related design data have been provided on Per Unit basis for the proper implementation of diode rectification system. Finally, Detailed input and output current analysis has shown that the proposed input filter yield high quality input ac current waveforms, in particular, high input power factor values and more reliabilty which reducing the V A rating of passive components as compared to the standard type LC filter.filter.
