Effects of Enzyme Treatments on Yield and Flavor Compounds of Garlic Extracts (효소처리에 의한 마늘 착즙액의 수율 및 향미성분변화)

  • In this study, attempts were made to develop a garlic juice extraction method that would result in minimum changes in quality. Protopectinase and a mutienzyme containing cellulase, pectinesterase, ${\beta}-glucanase$, etc. were applied to garlic residue after first extraction, and the yields of garlic juice and the flavor component changes of the juices were investigated. Enzyme concentrations of 0.04, 0.08, and 0.12% which were based on pulp weight before extraction were added and allowed to hydrolyze for 30, 60, 90 and 120 min,. respectively. Increase in the garlic juice yield was observed according to the amount of enzyme added and the reaction time until reaching a maximum point. When 0.12% protopectinase was applied to the garlic residue for 90 min, the yield increased by 13.8%. Under the same conditions, when multienzyme was applied to the garlic residue, the yield increased by 14.5%, which was considered the maximum. The flavor compounds decreased when compared with the total GC peak area of garlic juice prepared without enzymes(control). The volatile flavor compounds in garlic juice prepared with 0.12% protopectinase for 60 min decreased by 6%. The free sugars profile of the garlic juice prepared with 0.12% protopectinase for 60 min was similar to that of the control. The type of enzyme used did not affect the free amino acid profile of the garlic juice. These results indicate that the optimum conditions for extraction of garlic juice are hydrolyzing the residue with 0.12% protopectinase for 60 min, extracting garlic juice from the hydrolyzed reside, and then combining the extracted juice with the first extraction.

Discrimination study between carcass yield and meat quality by gender in Korean native cattle (Hanwoo)

  • Objective: The aim of this study was to identify a distribution pattern of meat quality grade (MQG) as a function of carcass yield index (CYI) and the gender of Hanwoo (bull, cow, and steer) to determine the optimum point between both yield and quality. We also attempted to identify how pre- and post-deboning variables affect the gender-specific beef quality of Hanwoo. Methods: A total of 31 deboning variables, consisting of 7 pre-deboning and 24 post-deboning variables from bulls (n = 139), cows (n = 69), and steers (n = 153), were obtained from the National Institute of Animal Science (NIAS) in South Korea. The database was reconstructed to be suitable for a statistical significance test between the CYI and the MQG as well as classification of meat quality. Discriminant function analysis was used for classifying MQG using the deboning parameters of Hanwoo by gender. Results: The means of CYI according to 1+, 1, 2, and 3 of MQG were 68.64±2.02, 68.85±1.94, 68.62±5.88, and 70.99±3.32, respectively. High carcass yield correlated with low-quality grade, while high-quality meat most frequently was obtained from steers. The classification ability of pre-deboning parameters was higher than that of post-deboning parameters. Moisture and the shear force were the common significant parameters in all discriminant functions having a classification accuracy of 80.6%, 71%, and 56.9% for the bull, cow, and steer, respectively. Conclusion: This study provides basic information for predicting the meat quality by gender using pre-deboning variables consistent with the actual grading index.

Effects of Water Stress on Growth and Yield of Paeonia lactiflora Pallas (수분스트레스가 작약의 생육과 수량에 미치는 영향)

  • The study was carried out to find effects on growth and yield by drought in Paeonia lactiflora Pallas. The water stress treatment was imposed artificially on April 1 to May 30(top part growth high stage), June 1 to July 30(root growth high stage), August 1 to September 29(late growth stage) and control(below 50kPa) in rain shelter. In drought periods of April 1 to May 30, stem length and number of stem were 67.5cm and 10.4 ea/plant. It showed poor growth compared with control, and root length and diameter also decreased to 27.2cm and 24.5mm, respectively. In root yield, drought treatment of June 1 to July 30 showed lower root yield as $1,809kg\;10a^{-1}$, $1,902kg\;10a^{-1}$ for drought period of August 1 to September 29, compared with $2,039kg\;10a^{-1}$ of control, resulting in 11% and 7% reduction, respectively. Optimum irrigation times (50kPa) was 17 days after continuos rain-off in Spring(April 1 to May 30), 9 days in summer(June 1 to July 30). It could be estimated that effective irrigation point was at least 13 days in after August(Aug. 1 to Sep. 29).

Effect of Antibiotic Fermentation Residues on Rice and Tomato Growth (항생물질 발효 부산물이 수도 및 토마토 생육에 미치는 영향)

  • In order to evaluate the applicability of two kinds of antibiotic fermentation residues on rice and tomato growth, yield, yield components, and some indicators for plant growing status were checked including analysis of physico-chemical properties of these two antibiotic fermentation residues. The results obtained are as follows: 1) These two antibiotic fermentation residues contain high organic matter ($21.6{\sim}24.2%$), phosphorus ($2900{\sim}4600 ppm$) and exchangeable cations ($55.4{\sim}138.3 meq/100 g$,), showing their pH values of $7.0{\sim}8.0$ range. 2) Both have developed net positive charge rather high and stiffly that exhibits high negative ion adsorption capacities, accordingly showing higher zero point of charges($pH 7.0{\sim}8.0$) than those of common soils. 3) The effect of the two kinds of antibiotic fermentation residues on rice growth was more or less the same comparable to the effect of the other fertilizers applied, showing the maximum yield at the application rate of 40 ㎏/10a. 4) The effect of these antibiotic fermentation residues on tomato growth was also similar to effects on rice plant showing the yield increment upon fertilizer application including two antibiotic fermentation residues but no significant differences among fertilizers. 5) According to the plant growing status, plant height, dry matter, number of effective tillers and grain number per panicle of rice and plant height and fresh weight of plant of tomato showed similar trend with yield of both plants.

Extraction Yield and Quality Attributes of Agar from Domestic Seaweeds According to Various Pretreatments (국산원조(國産原藻)의 전처리(前處理)과정에 따른 한천(寒天)의 수율(收率) 및 품질특성(品質特性))

  • Domestic seaweeds Gracilaria verrucosa collected from coastal areas of Namhae, Wando and Yeosoo, Korea were subjected to the examination of yield and quality of agar prepared according to various pretreatment conditions. In alkali treatment at high temperature, higher alkali concentration for one-hour period at $90^{\circ}C$ gave rise to higher yield of agar. In acid treatment, higher yield was obtained by higher acid concentration and longer treating period. Alkali treatment at room temperature gave rise to a slightly decreased yield at higher alkali concentration and longer treating time. Total nitrogen and crude ash of agar samples were greatly decreased by pretreatments. Jelly strength, gelation ability, gelation point and viscosity of agar samples tended to increase as the alkali-treating condition of seaweed became stronger. It was shown that sulfur content of agar had a high negative correlation with jelly strength of its gel. Various alkali treatments of seaweed at room temperature showed no marked difference in agar quality and did not exhibit any good effect comparable to alkali treatment at high temperature.

Study on Evaluation of Plastic Deformation Zone at Crack Tip for the Multi-Passed Weld Region of the Pressure Vessel Steel Using Nondestructive Method (비파괴법에 의한 압력용기 강 다층용접부의 균열선단에서 소성변형 역성장거동 평가에 관한 연구)

  • The purpose of this study is to evaluate the behaviour of the plastic deformed zone at crack tip on the standard Charpy specimens which were taken from the multi-passed weld block of the pressure vessel steel. Notch was machined on the standard Charpy test specimens and pre-crack which was located around the fusion line was made under the repeat load. Four point bend and acoustic emission tests were carried out simultaneously. The size of plastic region at crack tip was calculated using stress intensity factor. Relationships between characteristics of acoustic emission and plastic zone size at crack tip were discussed through the cumulative AE energy. Regardless of the specimens, AE signals were absent within the elastic region almost and most of AE signals were produced at the plastic deformation region from yield point to the mid-point between yield and maximum load. More AE signals for the weldment were produced compared with the base-metal and PWHT specimen. Relations between plastic deformed zones at crack tip and cumulative AE energy for the weldment and PWHT specimen were different quietly from the base-metal. Besides, number of AE counts for the weldment was the larger than those of the base-metal and PWHT specimen.

Impact of Expanded Small Alkyl-Binding Pocket by Triple Point Mutations on Substrate Specificity of Thermoanaerobacter ethanolicus Secondary Alcohol Dehydrogenase

  • Site-directed mutagenesis was employed to generate five different triple point mutations in the double mutant (C295A/I86A) of Thermoanaerobacter ethanolicus alcohol dehydrogenase (TeSADH) by computer-aided modeling with the aim of widening the small alkyl-binding pocket. TeSADH engineering enables the enzyme to accept sterically hindered substrates that could not be accepted by the wild-type enzyme. The underline in the mutations highlights the additional point mutation on the double mutant TeSADH introduced in this work. The catalytic efficiency ($k_{cat}/K_M$) of the ${\underline{M151A}}$/C295A/I86A triple TeSADH mutant for acetophenone increased about 4.8-fold higher than that of the double mutant. A 2.4-fold increase in conversion of 3'-methylacetophenone to (R)-1-(3-methylphenyl)-ethanol with a yield of 87% was obtained by using ${\underline{V115A}}$/C295A/I86A mutant in asymmetric reduction. The ${\underline{A85G}}$/C295A/I86A mutant also produced (R)-1-(3-methylphenyl)-ethanol (1.7-fold) from 3'-methylacetophenone and (R)-1-(3-methoxyphenyl)-ethanol (1.2-fold) from 3'-methoxyacetophenone, with improved yield. In terms of thermal stability, the ${\underline{M151A}}$/C295A/I86A and ${\underline{V115A}}$/C295A/I86A mutants significantly increased ${\Delta}T_{1/2}$ by $+6.8^{\circ}C$ and $+2.4^{\circ}C$, respectively, with thermal deactivation constant ($k_d$) close to the wild-type enzyme. The ${\underline{M151A}}$/C295A/I86A mutant reacts optimally at $70^{\circ}C$ with almost 4 times more residual activity than the wild type. Considering broad substrate tolerance and thermal stability together, it would be promising to produce (R)-1-(3-methylphenyl)-ethanol from 3'-methylacetophenone by ${\underline{V115A}}$/C295A/I86A, and (R)-1-phenylethanol from acetophenone by ${\underline{M151A}}$/C295A/I86A mutant, in large-scale bioreduction processes.

  • Green Marketing, a response to the serious environment problems of the 21st century, is an important point for trends in both art and fashion. It is a strategy for reminding consumers of the threatening environment problems and to recognize that everyone must take responsibility for these problems. Green marketing is a concept that is the point at issue and social reform movement. The purpose of this study was to find examples of Green Marketing in fashion and classify them into four groups. The results of this study were as follows. First, the terms related to green marketing as related to fashion can be categorized into four areas: 1. Eco-friendly product marketing: product merchandising that usually uses organic materials and green manufacturing processes. 2. Green-communication marketing: PR strategies that use green slogans through deep impression advertising or special promotional events relating reducing, reusing and recycling, etc., with the brand's name. 3. Green environmental-marketing: marketing strategies that is an enlightenment campaign for environmental preservation and sustainable fashion design such as reusing, recycling, and reducing. 4. Cause-related marketing: marketing plans that return profits to society and that take an active part in the causes of the community. It was expected that these types of green marketing strategies would yield positive effects in enhancing brand value and consumer confidence as well as increasing profits.

  • Most engineering optimization are based on numerical linear and nonlinear programming methods that require substantial gradient information and usually seek to improve the solution in the neighborhood of a starting point. These algorithm, however, reveal a limited approach to complicated real-world optimization problems. If there is more than one local optimum in the problem, the result may depend on the selection of an initial point, and the obtained optimal solution may not necessarily be the global optimum. This paper describes a new harmony search(HS) meta-heuristic algorithm-based approach for structural size optimization problems with continuous design variables. This recently developed HS algorithm is conceptualized using the musical process of searching for a perfect state of harmony. It uses a stochastic random search instead of a gradient search so that derivative information is unnecessary. Two classical space truss optimization problems are presented to demonstrate the effectiveness and robustness of the HS algorithm. The results indicate that the proposed approach is a powerful search and optimization technique that may yield better solutions to structural engineering problems than those obtained using current algorithms.

  • The radiation-induced graft polymerization of methacrlic acid and methyl methacrylate onto a polyester fabric was investigated with ${\gamma}$-ray as the radiation source, and the rate of grafting was examined. When acrylic acid, methacrylic acid, and methyl methacrylate were grafted onto a polyester fabric, grafting efficiency was depened upon the dielectric constant of the solvent in the monomer mixture. The yield of the graft polymerization was related to the total dose, the concentration of the monomer, and the concentration of the swelling agent. The melting point and the glass transition temperature of MA and MMA grafted copolymers were analysed by means of DTA. Physical properties, such as the moisture regain, the antistatic property, and the wicking time were measured.
