Studies on the supercritical fluid extraction of taxol from yew tree (초임계 유체를 이용한 주목 수피로부터 taxol의 추출에 관한 연구)

  • Studies were carried out to examine some factors affecting the supercritical carbon dioxide extraction of taxol from the bark of Taxus cuspidata using a continuous packed bed extractor. The factors investigated in this study were pressure, temperature, volume of carbon dioxide, and co-solvent. It was found out that the amount of taxol extracted was not significantly affected by the operating pressure in the absence of a co-solvent although it increased by about 20% at 5500 psig. With $24\ell$ of carbon dioxide the saturated amount of taxol was extracted at 318K and 5500psig. Methanol was found to be the most effective co-solvent in terms of amount of taxol extracted among six different co-solvents used. When methanol was used as a co-solvent the effect of operating pressure became significant; approximately 50% increase in the amount of taxol extracted was observed at 3000 psig as compared to at 2500 or 3500psig. The optimum methanol concentration in supercritical fluid was 13% (w/w)at 308K, 3000psig.

Taxol Produced from Endophytic Fungi Induces Apoptosis in Human Breast, Cervical and Ovarian Cancer Cells

  • Currently, taxol is mainly extracted from the bark of yews; however, this method can not meet its increasing demand on the market because yews grow very slowly and are a rare and endangered species belonging to first-level conservation plants. Recently, increasing efforts have been made to develop alternative means of taxol production; microbe fermentation would be a very promising method to increase the production scale of taxol. To determine the activities of the taxol extracted from endophytic fungus N. sylviforme HDFS4-26 in inhibiting the growth and causing the apoptosis of cancer cells, on comparison with the taxol extracted from the bark of yew, we used cellular morphology, cell counting kit (CCK-8) assay, staining (HO33258/PI and Giemsa), DNA agarose gel electrophoresis and flow cytometry (FCM) analyses to determine the apoptosis status of breast cancer MCF-7 cells, cervical cancer HeLa cells and ovarian cancer HO8910 cells. Our results showed that the fungal taxol inhibited the growth of MCF-7, HeLa and HO8910 cells in a dose-and time-dependent manner. IC50 values of fungal taxol for HeLa, MCF-7 and HO8910 cells were $0.1-1.0{\mu}g/ml$, $0.001-0.01{\mu}g/ml$ and $0.01-0.1{\mu}g/ml$, respectively. The fungal taxol induced these tumor cells to undergo apoptosis with typical apoptotic characteristics, including morphological changes for chromatin condensation, chromatin crescent formation, nucleus fragmentation, apoptotic body formation and G2/M cell cycle arrest. The fungal taxol at the $0.01-1.0{\mu}g/ml$ had significant effects of inducing apoptosis between 24-48 h, which was the same as that of taxol extracted from yews. This study offers important information and a new resource for the production of an important anticancer drug by endofungus fermentation.

Determinants of Choice of Surgery in Asian Patients with Early Breast Cancer in A Middle Income Country

  • Background: Breast-conserving surgery (BCS) plus radiotherapy is equivalent to modified radical mastectomy (MRM) in terms of outcome. However there is wide variation in mastectomy rates dependent both on tumour and patient characteristics. Objective: This study aimed to assess the determinants of surgery choice in Asian patients with early breast cancer in a middle-income country. Materials and Methods: 184 patients with early breast cancer treated between Jan 2008 and Dec 2010 were recruited to complete a questionnaire. Chi-square test was used to analyze the association between surgery choice and demographic and tumour factors, surgeon recommendation, family member and partner opinions, fear of recurrence, avoidance of second surgery, fear of disfigurement, interference with sex life, fear of radiation and loss of femininity. Results: 85 (46%) had BCS while 99 (54%) had mastectomy. Age >60, Chinese ethnicity, lower education level, and larger tumour size were significantly associated with mastectomy. Surgeon recommendation was important in surgery choice. Although both groups did not place much importance on interference with sex life, 14.1% of the BCS group felt it was very important compared to 5.1% in the mastectomy group and this was statistically significant. There was no statistical difference between the two groups in terms of the other factors. When analyzed by ethnicity, significantly more Malay and Indian women considered partner and family member opinions very important and were more concerned about loss of femininity compared to Chinese women. There were no statistical differences between the three ethnic groups in terms of the other factors. Conclusions: When counseling on surgical options, the surgeon has to take into account the ethnicity, social background and education level, age and reliance on partner and family members. Decision-making is usually a collective effort rather than just between the patient and surgeon, and involving the whole family into the process early is important.

Evaluation of accumulated particulate matter on roadside tree leaves and its metal content (가로수 수종별 잎의 미세먼지 축적량 및 금속 원소 함량 평가)

  • It is known that different plant species have ability to deposit different amounts of particulate matter (PM) on their leaves and plants can absorb heavy metals in PM through their leaves. Heavy metals in PM can have toxic effect on human body and plants. Therefore, PM on different roadside trees at Chungbuk national University including box tree (Buxus koreana), yew (Taxus cuspidate), royal azalea (Rhododendron yedoense), and retusa fringetree (Chionanthus retusa) was quantified based on particle size (PM>10 and PM2.5-10). The metal concentration in PM accumulated on leaves was analyzed using inductively coupled plasma-mass spectroscopy. In this study, the mass of PM>10 deposited on the surface of the tree leaves ranged from 6.11 to 32.7 ㎍/㎠, while the mass of PM2.5-10 ranged from 0 to 14.8 ㎍/㎠. The royal azaleas with grooves and hair on the leaf surface retained PM particles for longer time, while the yews and box trees with wax on leaf surfaces accumulated more PM. The PM contained elements in crustal material such as Al, Ca, Mg, and Fe and heavy metals including Cu, Pb and Zn. The concentration of elements in crustal material was higher in the coarser size, while heavy metal concentration was relatively higher in the finer size fraction. The Mn, Cd, Cu, Ni, Pb, and Zn concentrations of leaves and PM2.5-10 were significantly correlated indicating that PM was taken up through tree leaves.

  • Reefs are the marine structure that can give resting, inhabiting, feeding and nursing spaces for a variety of fishes. Usually artificial reefs are made of cement and steels respectively in Japan as well as in Korea. However since resources deficiency has been getting serious, other materials are taken into consideration for the basic bodies of artificial reefs. About 300 thousand tons of waste agricultural plastic films are generated every yew in Korea, but no effective recycling techniques have been developed. In this sense, artificial reefs made of waste agricultural plastic films are the most representative symbol of the recycled products in the Resource Recycling Era. In particular, since these reefs could be made of the semi-cleaned waste agricultural plastic films that still contain high portion of soil, it is very environmentally friendly not only in manufacturing process but also in using under water. Furthermore they have some evident advantages as follows; 1) high fish swarming effect 2) good initial attachment of the marine growths 3) extremely low corrosion to brine 4) easy adjustment of the gravity 5) economical manufacture, transportation and jettison 6) excellent safety to ecosystem caused by lower elution of toxic substances 7) good recyclable property after application and so on.

Nationwide Incidence Estimation of Uterine Cervix Cancer among Korean Women (한국 여성에서의 자궁경부암 발생률)

  • To estimate the incidence of uterine cervix cancer among Korean women, we have conducted a study using the claim data on the beneficiaries of Korea Medical Insurance Corporation (KMIC). All medical records of the potential cases with diagnosis of ICD-9 180, 181, 182, 199, 219, 233 in the claims sent by medical care institutions in the whole country to the KMIC from January 1988 to December 1989, were abstracted and Gynecology specialist reviewed the records to identify the new cases of uterine cervix cancer among the potential cases during the corresponding period. Using these data, the incidence of uterine cervix cancer among Korean women was estimated as of July 1, 1988 to June 30, 1989. The crude rate was estimated to be 17.34(95% CI: $16.76\sim17.92$) per 100,000 and the cumulative rates for the ages $0\sim64\;and\;0\sim74$ were 1.7% and 2.2%, respectively. The age-adjusted rate for the world population was 19.93 per 100,000 which was higher than those of other Asian countries including China and Japan in $1983\sim1987$. The truncated rate for ages $35\sim64$ was 52.05 per 100,000 which was one of the highest in the world. With increasing age, the incidence rate increased to 78.11 per 100,000 in women aged $55\sim59$ years, then it decreased in the older groups. This finding suggests that detecting rate of uterine cervix cancer may decrease in women aged 60 years or older due to inadequate medical care seeking behavior. In the geographical area, the SIR of Jeju province was significantly low but it might be due to statistical unstability by small case numbers.

Update on Phosphorylation-Mediated Brassinosteroid Signaling Pathways (단백질 인산화에 의해 매개되는 브라시노스테로이드 신호전달 연구의 최근 상황)

  • Protein phosphorylation is a universal mechanism that regulates cellular activities. The brassinosteroid (BR) signal transduction pathway is a relay of phosphorylation and dephosphorylation cascades. It starts with the BR-induced activation of the membrane receptor kinase brassinosteroid insensitive 1 (BRI1), resulting in the dephosphorylation of transcription factors such as BZR1/BES2 and BZR2/BES1 followed by BR-induced gene expression. Brassinosteroid signal transduction research has progressed rapidly by identifying the phosphorylation/dephosphorylation site(s) of the BR-regulated kinase and phosphatase substrates with a simultaneous pursuit of mutant phenotypes. Autophosphorylation, transphosphorylation, and serine/threonine and tyrosine phosphorylation of the receptor protein kinases BRI1 and BRI1-associated kinase (BAK1) have increased the understanding of the regulatory role of those kinases during physiological and developmental processes in plants. The phosphorylation event initiated by BR is also found in the regulation of receptor-mediated endocytosis and the subsequent degradation of the receptor. However, the basic molecular links of the BR signal transduction pathway are not well understood regarding this phosphorylation/dephosphorylation event. This review summarizes the current state of BR signal transduction research to uncover the phosphorylation/dephosphorylation networks and suggests directions for future research on steroid signal transduction to gain a more comprehensive understanding of the process.

  • For Singapore, 2017 was an uneasy year. The presidential election was fraught with controversy since the revised Presidential Election Act allowed only one candidate to be eligible for the election. The bitter feud between Prime Minister Lee Hsien Loong and his siblings shocked many Singaporeans. Succession planning for the next top leadership is still veiled in obscurity. The anti-globalization trend and the increasing pressure to raise the tax have become major challenges for Singapore's economy to overcome. China's continuous diplomatic pressure has called into the question Singapore's pragmatic foreign policy. Although its relations with China were back to normal, Singapore, the ASEAN chair in 2018, is still facing intractable problems in safeguarding ASEAN centrality in the growing US-China rivalry. In the meantime, Singapore has pursued its diversity and equality, heading toward a more matured multi-racial and multi-cultural society in 2017. The first female president, Halimah Yacob, served as a symbolic epitome of Singapore's emphasis on diversity and harmony among different ethnic groups and minorities. This great milestone, however, has largely been questioned by Singaporeans, as it seemed to be a political gesture that only utilized Halimah's double minority in the level of ideologies. The election of the Malay president has led Singaporeans to think about the real equity and equality among minorities, while strongly motivated to move toward a more inclusive society. In 2018, Singaporean leaders will try to resolve many challenging problems by reaffirming leadership succession planning, which is expected to lead Singapore to pursue a more integrated society.

A Study on the Characteristics of Chwibyeong in 『Jungli-Euiguae』 written Hangeul (『뎡니의궤(整理儀軌)』에 나타난 취병의 특성 연구)

  • Chwibyeong is a hedge made into a folding screen shape by setting up supports and placing plants on it. Traces of chwibyeong can be found in various paintings, such as the 『Donggwoldo painting of Changdeokgung Palace』, 『Gyujanggak painting』, 『Munhuimyo painting』, and 『Hwaseong Temporary Palace painting depicting Suwon Hwaseong Temporary Palace』. However, since the Chwibyeong has not survived to this day, the whereabouts of the Chwibyeong were estimated through ancient documents, old paintings, and photographs. In the case of a manned Chwibyeong using a support, a picture related to the composition of the Chwibyeong using a base can be Winnow willow. Methods for creating intoxicants using coniferous trees such as Chinese juniper and Rigid-branch yew have been studied. In existing old paintings, only the supports, parts of leaves, and branches were depicted, so the location of the Chwibyeong could not be confirmed through the drawings. However, in the 『Jungli-Euiguae』, written around 1797, a manned type of Chwibyeong using a support was depicted as the material of the Chwibyeong. By being able to confirm the type of tree used, it was possible to confirm the method of creating a Chwibyeong using Winnow willow introduced in the 『LimwonGyeongjeji』.   • Cleft lip and/or palate is one of the most common congenital craniofacial anomalies. According to previous epidemiologic studies, incidence of cleft lip and/or palate has been increasing nowadays. However, there is no report about epidemiologic study of cleft lip and/or palate patients who visited dept. of orthodontics in Korea. So the purpose of this study was to provide the epidemiological characteristics and important basic clinical data for the diagnosis and the treatment of the cleft lip and/or palate patients. With the orthodontic and cleft charts, diagnostic models and X-ray films from 250 patients with cleft lip and/or palate who visited Dept. of Orthodontics, Seoul National University Dental Hospital during the last 11 years, the authors investigated patient's visiting yew, types of cleft, patient's gender, and Angle's classification of malocclusion, and surgery timing. The results were as follows ; 1. The number of cleft patients who visited Dept. of Orthodontics, SNUDH increased during 1988-1990 and then it declined until 1992. From 1993 to 1996, it showed a stationary trend. After 1997 it showed an overwhelmingly increasing trend. 2. In the cleft type, the ratio of cleft lip cleft lip and alveolus cleft palate : cleft lip and palate was 7.6:19.2:9.6:63.6. In cleft position, unilateral clefts were more than bilateral ones (cleft lip 79:21, cleft lip and alveolus 77:23, cleft lip and palate 75.5:24.5). In cleft side, left clefts were mote than right clefts (cleft lip 53.3:46.7 cleft lip and alveolus 59.5:40.5, cleft lip and palate 59.2:40.8). 3. In gender ratio, males were more than females in cleft lip (57.9:42.1), cleft lip and alveolus (68.8:31.2) and cleft lip and palate (76.1:23.9). But in cleft Palate females were more than males as 41.7: 58.3. 4. In the age groups, 7-12 year group was the most abundant as $52\%$, and then 0-6 year group ($20.4\%$), 13-18 year group ($17.2\%$), more than 18 yew group ($10.4\%$) were followed as descending order. 5. Most of the cleft lip repair surgeries were operated in 0-3 month ($60.3\%$) and 4-6 month ($17.9\%$). 6. The cleft palate repair surgeries were done in 1-2 year ($31.7\%$), 0-1 year ($25.6\%$), 2-3 year ($12.1\%$), more than 5 year ($11.6\%$) as descending order. 7. The lip scar revision surgeries were done before admission at elementary school in $60\%$. (4-6 you ($27.5\%$), 6-8 year ($19.6\%$), more than 10 year ($19.6\%$), 2-4 year ($13.7\%$) as descending order) 8. The rhinoplasties were done before admission at elementary school in $51.7\%$. (0-2 year ($7.1\%$), 2-4 year ($14.3\%$), 4-6 year ($21.4\%$), 6-8 year ($14.3\%$)). 9. The pharyngeal flap were done at 6 Y (72.5 months) after birth on average and there was even distribution of surgery timing. 10. In relationship between Angle's classification of malocclusion and cleft types, Class I was most abundant and Class III, Class II were followed as descending order in cleft lip group. But Class III was most abundant and Class I, Class II were followed as descending order in cleft lip and alveolus group, cleft palate group, and cleft lip and Palate group. The percentage of frequency in Class III malocclusion was overwhelmingly higher in cleft lip and palate group than any other groups. 11. Because the frequency of class III malocclusion was most prevalent in all age groups, anterior crossbite was the most common chief complaint of cleft patients.
