Changes of Enzyme Activities and Inorganic Nutrient Contents Associated with Flower Development in Tulip (Tulipa gesneriana) (튤립(Tulipa gesneriana) 꽃의 발달단계에 따른 효소 활성 및 미량요소 함량의 변화)

  • This study was carried out to investigate the changes of enzymes and micro inorganic nutrients that is associated with flower senescence during flower development in tulip cultivars, ‘Apeldoorn’ and ‘Golden Apeldoorn’. Ribonuclease, peroxidase and protease activities were gradually increased from the stage of early flowering to later Polyphenol oxidase showed the highest activity at stage 5, which the flower was in full bloom indicating that it acts at an initial stage of flower senescence. The protease activity was different in the petal extracts during flower development between the cultivars ‘Apeldoorn’ (red petal) and ‘Golden Apeldoorn’ (yellow petal). This result suggested that protease might relate to pigment biosynthesis in petal of tulip. In contrast to the decrease of inorganic nutrients K, Mn, Zn and P contents during floral development, Ca, Mg and Fe showed the gradual increasement that is similar with ribonuclease, peroxidase and protease. It suggests that they have some interaction during flower senescence.

  • The Nectar Ritual Painting(Jeung7551) in National Museum of Korea is estimated to have been produced in the 16th century. Ezimagodo who is the head priest of Ruganji in Kyoto, Japan donated this painting in 2010. Overall, damages were serious, such as missing, staining, folding, insects of the face and mounting silk, therefore, it need to conservation treatment. The treatment of conservation in painting was removing surface stains and repairing missing area. It was mounted by following the original form. The analysis result of textile in mounting and support fiber used silk and hemp in microscope. Pigments in paint were divided into six color types. This painting was used that vermilion(cinnabar) and red lead for red color, litharge(massicot) and gold for yellow color, lead white for white color, malachite for green color, azurite for blue color, and carbon black(chinese ink) for black color.

Interpretation of Coloring Materials Recorded in Ceremonial Writing of the Hanging Painting of Chiljangsa Temple (Five Buddhas) (칠장사 오불회 괘불탱 화기에 기록된 채색 재료의 해석)

  • This study aimed to interpret the coloring materials recorded in the ceremonial writing with the scientific investigation results of the Hanging Painting of the Chiljangsa Temple (Five Buddhas). The results confirm that the Jinboon, Joohong, Hwangdan, Hwanggeum and Seokjahwang are clearly connected to lead white, cinnabar, minium, gold, and orpiment, respectively. Danghayeop and samrok are related to Copper trihydroxychloride, while Daecheong and Joongcheong are azurite, and they seems to be classified by the particle size. Yeonji and Chunghwa are organic dyes in red and blue, respectively, with blue confirming the existence of the side, but Yeonji differs from the names of the ancient texts and Chinese characters; it is unclear whether it is a commonly used Yeonji because of differences in the names of the ancient texts. The presence of Sootohwang has not been confirmed in the gwaebultaeng, but it can be extracted from the soil as a yellow-colored material but the possibility of Deunghwang cannot be ruled out.

A Scientific Analysis of Ancient Pigments on Wall Paintings at Yeongsanjeon in Tongdo Temple Using a Field-XRF (휴대용X선형광분석기를 이용한 통도사 영산전 벽화 안료의 과학적 성분분석)

  • In ancient period, a variety of inorganic or organic pigments had been used as colorants in various kinds of religious and secular paintings such as tomb paintings and wall and scroll paintings in buddhist temples, and danchung(cosmic patterns) for the surface of wooden buildings. This study discusses the results obtained from an analysis of the pigments on the wall paintings of Yeongsanjeon(Hall of Vulture Peak) in Tongdo temple by a qualitative analysis using a field-XRF. The results can be briefly summarized as follows. Firstly, assuming from the major components examined from F-XRF analysis, raw materials of pigment of each color are: red to be Cinnabar(HgS) or Hematite($Fe_2O_3$); white to be White Lead[$2PbCO_3{\cdot}Pb(OH)_2$] in most cases and Calcite($CaCO_3$) or Chalk($CaCO_3$), Kaolin($Al2O_3{\cdot}SiO_2{\cdot}4H_2O$) in some cases; yellow to be Yellow Ocher[$FeO(OH){\cdot}nH_2O$]; black to be carbon(C); green on the painted surface to be Celadonite[$K(Mg,Fe^{2+})(Fe^{3+},Al)(Si_4O_{10})(OH)_2$] in most cases; dark green on the halo of figures to be Malachite[$CuCO_3{\cdot}Cu(OH)_2$], Copper Green[$2CuO{\cdot}CO_2{\cdot}H_2O$] or Atacamite[$Cu_2Cl(OH)_3$]. Secondly, incarnadine and pink were made by mixing with more than two pigments such as red and white for making various tone of colors. The qualitative analysis of pigments on the wall paintings of Yeongsanjeon, in conclusion, displays that the all pigments for ancient periods are inorganis pigments. However, it has the limitation to identify a definite kinds of mineral for each pigment because it was not possible to collect samples from cultural heritage for conducting a crystalline analysis of XRD.

Development of environmentally friendly inorganic fluorescent pigments, A3V5O14 (A = K and Rb) and Cs2V4O11: Crystal structure, optical and color properties (친환경 무기 형광 안료 A3V5O14 (A = K and Rb) and Cs2V4O11 개발: 결정구조, 광학적 특성 및 착색 특성)

  • To develop the bright-vivid red- and yellow-inorganic fluorescent pigments with high luminescence properties, A3V5O14 (A = K and Rb) and Cs2V4O11 inorganic pigments were synthesized by a water assisted solid state reaction (WASSR) method and a conventional solid state reaction method. Although impurity peaks corresponding to the AVO3 and AV3O8 (A = K, Rb, and Cs) were observed in all samples prepared, the trigonal structure A3V5O14 (A = K and Rb) and orthorhombic structure Cs2V4O11 were successfully obtained as a main phase. These inorganic pigments showed the broad absorption band (under 550 nm) originated from CT transitions of VO4 polyhedron, and the strong broad red- and green-emission bands due to 3T21A1 and 3T11A1 transitions of the [VO4]3- group. The A3V5O14 (A = K and Rb) and Cs2V4O11 pigments showed a bright-vivid red- and yellow-body color, where the a* values of the A3V5O14 (A = K and Rb) were +35.5 and +45.9, respectively, and b* value of Cs2V4O11 pigments was +50.3. The L* values of the A3V5O14 (A = K and Rb) and Cs2V4O11 inorganic pigments were over +45. These results indicate that the A3V5O14 (A = K and Rb) and Cs2V4O11 inorganic pigments could be an attractive candidate as a bright-vivid red- and yellow inorganic pigments.

The Stability Appraisement on Cultural Property Material with the Replacing Fumigation Gas of Methyl Bromide (Methyl Bromide를 대체하는 훈증 가스의 문화재 재질 안정성 평가)

  • Methyl Bromide that was used as fumigation gas turned out to be the substance of destroying the ozone layer. For that reason, at the Montreal Protocol in 1987 the use of methyl bromide was forbidden starting 2005 in the advanced country. Also according to the 2007 Bali Protocolly methyl bromide is expected to be forbidden and therefore the purpose of this study is to find out the effects of substitution fumigation gas (Ethylene Oxide+HFC 134a, Methyl Iodide, Cyanogen and Argon) on the metal(silver, copper and iron), wood(oregon pine), pigment(yellow, red, blue, white and black), textile(hemp, ramie, jute, silk1 and silk2 / indigo, safflower and cork) and paper. After the fumigation test, ethylene oxide+HFC 134a did not have changes in the weight and color of the material itself before and after the experiment. On exterior alteration, color change occurred partly on paper and metal. Also, in most materials color change extent was 0.5 to 1.5 on the average and showed scanty difference. The materials after the fumigation test with methyl iodide did not show weight changes after the test. However, color changes more than 1.0 was shown in most of the materials especially in dyed textile material. In blue pigment, the discoloration on the surface could be seen by naked eyes. Fumigation test with cyanogen gas did not show weight changes and discoloration is more than 1.5 before and after the test. The weight changes of test materials with the argon gas was decreased about 3 to 6%. It can be observed that discoloration on paper was generated. Color changes can be seen on jute dyed with safflower and cork for two weeks with argon gas and the extent was 6.3 and 6.0.

Scientific Study on Materials and Painting Techniques of Portrait of Sim Huisu (심희수 초상의 재료와 제작기법에 대한 과학적 조사)

  • Portrait of Sim Huisu is a seventeenth-century Joseon portrait of a meritorious vassal. The National Museum of Korea currently owns two portraits of Sim Huisu, which are the eldest son's family and by the eldest grandson of the family's second eldest son. Both were donated in 1980. Portraits were still in its original mounting, but the supporting silk had been damaged and stained in a flood. Conservation treatment was undertaken to restore the original style, and scientific analysis, such as, X-ray, XRD, XRF and Graff "C" stain, was conducted to study the materials and painting techniques. The support silk was found to be refined fibroin and a plain weave consisting of two weft threads and one warp thread. The lining papers were found to be bamboo fiber paper of first layer in China and Korean traditional mulberry paper in second. Various pigments were identified in the painting, including white lead, cinnabar, atacamite, ink stick, azurite, silver, and gold. The study also confirmed the use of the back painting, with colors such as white White Lead, green Atacamite, orange Minium, black Ink Stick, and yellow Dye. Also, it was found that stick ink or dye was used with white lead.

Acute Oral Toxicity Study of Ethanol Extract of Curcuma longa L. in Mice (마우스에서 울금 에탄올 추출물의 단회 경구투여 독성에 관한 연구)

  • A yellow-colored pigment is found in turmeric, or Curcuma longa L. (Zingiberaceae), a perennial herb distributed mainly throughout tropical and subtropical regions. C. longa has potent antiviral, antimutagenic, anti-inflammatory, anticancer, and antioxidant properties. However, pharmacological mechanisms of ethanol extract derived from C. longa remain poorly understood. The aim of this study was to investigate the potential acute toxicity of C. longa (Curcuma longa L.) extract in BALB/c mice administered a single oral dose of 0, 20, 200, and 2,000 mg/kg by gavage. After the administration of the agent, signs of toxicity were observed every hour for the first 6 hr and every day for 14 days. No mortality, abnormal clinical signs, or pathological changes were observed compared to a control group, and there were no differences in the body weights of the control and treatment groups. Biological serum activities were not significantly changed in the treatment group compared to the control group. These results indicate that a single oral administration of C. longa extract does not exert any toxic effects at a dose of 2,000 mg/kg body weight and that the $LD_{50}$ of C. longa extract is greater than 2,000 mg/kg body weight. Accordingly, C. longa appears to have potential in various functional agents or foods, without toxicity.

Dermatophytosis of the Four-toed Hedgehog Caused by Trichophyton erinacei

  • Trichophyton erinacei is a dermatophyte pathogen that infects both humans and hedgehogs. A two-month old female four-toed hedgehog presented to the Chonbuk Animal Medical Center with pruritus, excoriation and crust on her face for ten days. The owner of the hedgehog also exhibited the clinical signs of scaly erythema with fine vesicles on her neck. A presumptive diagnosis of dermatophytosis was made based on the results of an acetate tape preparation in which hyphae and chains of arthroconidia were observed. The crusts from the lesions were then cultured on Sabouraud Dextrose Agar for identification. After 10 days of incubation, downy colored colonies that had a central umbo with a white granular surface and a yellow pigment ring in the reverse were observed. Microscopic analysis revealed the presence of numerous teardrop shaped microconidia singly attached to the sides of the hyphae. In addition, 2-6 roomed macroconidia that were somewhat irregular in shape and size were present, and abundant intermediate sized spores were observed between the micro and macro conidia. To confirm that the culture was T. erinacei, the internal transcribed spacer region of the 5.8S phase of the ribosomal RNA gene (ITS1-5.8S-ITS2 rDNA) was amplified by PCR and then sequenced. A 679-base pair fragment of DNA was then compared with sequences in GenBank and found to be 99% homologous with sequences of T. erinacei (Z97997 and Z97996. The clinical signs were resolved after four weeks of treatment with oral and topical ketoconazole and chlorhexidine. To the best of our knowledge, this represents the first case of T. erinacei isolated from a four-toed hedgehog in Korea.

A Study of Weather Resistance on Dancheong Ground Treatment of Tranditional Wooden Building in Korea (한국 목조건축물 단청 바탕처리에 대한 내후성 연구)

  • In this study, the method for processing glue and glue-alum sizing from the dancheong ground treatment was evaluated with respect to weather resistance, and its effect on the conservation of dancheong was analyzed. Viscosity and pH of the glue and glue-alum specimen were measured and classified into three categories(none layer, glue layer, and glue-alum layer), which were further classified according to low concentration(four times for 2%) and high concentration(once for 10%). The base layer formation was subsequently classified into three categories based on pigment adoption, namely, Noerok(celadonite), Seokganju(terra rossa), and Jangdan(red lead). The completed specimen was subjected to a changing-environment experiment for evaluating weather resistance and observing the surface. Color variations were analyzed before and after the experiment. The results indicate that glue-alum sizing comprising 5% alum or 7% alum has strong acidity that can affect the life of dancheong, and the high level of 7% alum makes it difficult to create a solid coating layer. After ultraviolet irradiation, the specimen with 7% alum changes its color to yellow. Furthermore, after moisture absorption and drying, cracks can be observed on the entire specimen surface that corroborate the physical change. Additionally, gas-based corrosion causes marginal surface changes. Hence, the formation of a stable coating layer can be achieved by incorporating a low concentration glue solution that is almost neutral, and the application of glue-alum sizing having 2% concentration can aid in the conservation of dancheong.
