오미자(Schizandra chinensis) 열매 추출물의 Lactobacillus plantarum 젖산발효를 통한 고농도 GABA 함유 발효음료 제조 (Production of fermented Omija (Schizandra chinensis) beverage fortified with high content of gamma-amino butyric acid using Lactobacillus plantarum)

  • 오미자 열매를 이용하여 기능성물질인 GABA를 강화시킨 오미자 젖산발효음료를 개발하기 위해서 L. plantarum EJ2014에 의한 젖산발효 최적화를 수행하였다. 오미자 열매 추출액을 젖산 발효시킨 경우 발효 2일 동안 생균수 $4.0{\times}10^7CFU/mL$수준을 유지하면서 젖산균 증식이 미비하였지만 영양성분 YE와 MSG를 함께 첨가한 경우 50배 정도 증가된 $2.2{\times}10^9CFU/mL$생균수를 나타내었다. 특히 젖산발효 3일 동안에 대조군, MSG, YE를 각각 첨가군에서 초기 pH 3-4 수준에서 유지되었다. 반면에 YE와 MSG를 함께 첨가한 발효물은 pH가 증가되면서 pH 5.0 이상을 나타내었으며, 초기 산도 1.74%에서 발효 3일에 0.56% 수준으로 감소하였다. 오미자 열매 추출물인 대조군과 2% MSG 또는 0.5% YE만을 첨가한 조건에서는 GABA로 소량 전환되는 것으로 나타났다. 반면에 MSG와 YE를 함께 첨가한 경우 젖산발효 1일째부터 GABA가 생성되면서 3일째에는 MSG 2%를 거의 소진하는 것으로 나타났으며, HPLC 정량분석으로 0.92% GABA 생성이 확인되었다. MSG와 YE함께 첨가한 발효조건에서는 젖산발효 1일째부터 발효물은 붉은 색이 퇴색되면서 황색으로 변화하였으며, 발효 초기에 적색을 나타내는 a값이 15.74에서 발효 후에 5.39로 크게 감소하는 것으로 나타났다. 색, 맛, 향 등을 포함한 전반적인 기호도를 나타낸 관능평가에서 오미자 착즙 원액과 발효물 100%인 실험군에서 점수가 각각 4.50, 4.33을 기록하여 상대적으로 낮은 기호도를 나타낸 반면에 발효물 50% 첨가한 실험군이 6.33으로 가장 높은 기호도 점수를 나타내었다. 오미자 열매 추출액에 오미자 발효물을 50% 첨가함으로서 기호성이 우수하며 고농도 GABA와 probiotic을 함유한 오미자 발효음료의 제조가 가능하였다.

  • ${\ulcorner}Volume\;1{\lrcorner}$ of ${\ulcorner}$Medical Records as a Guide to Clinical Works${\lrcorner}$, written by Ye Tian Shi, showed some clinical cases of dizziness. In this study, his diagnosis and treatment was studied with 16 clinical cases of dizziness with pathogenic factor, pathogenesis and symptoms of dizziness. Ye Tian Shi thought that phlegm, fire, wind and insufficiency were the causes of dizziness and phlegm-fire, phlegm-fire-wind, wind-phlegm and insufficiencyfire-wind were the causes of dizziness, clinically. Dizziness is caused when the body is in condition of excess in the upper and deciency in the lower. The acompanying clinical symptoms of dizziness are endogenous wind, fire of deficiency type, phlegm wind and phelegm fire. For the treatment of dizziness, Ye Tian Shi used the combination of medicines with some modifications by the cases for phlegm, fire, wind and insufficiency. He also encouraged the mental therapy for the treatment of dizziness. He emphasized the early treatment of dizziness to prevent hemiplegia after apoplexy. It can be postulated from Volume 1 of ${\ulcorner}$Medical Records as a Guide to Clinical Works${\lrcorner}$, diagnosis and treatment of symptoms and illness of Ye Tian Shi was strictly based on actual clinical cases.

비스듬히 입사하는 음장에 대한 유한 길이의 탄성 원통 쉘의 음향 산란 (Acoustic scattering of an obliquely incident acoustic field by a finite elastic cylindrical shell)

  • 본 연구에서는 무한 유체에 놓여있는 유한 길이의 탄성 원통 쉘에 외부에서 비스듬히 평면파가 입사할 때 발생하는 음향 산란 현상을 이론적으로 연구했다. 유한 길이의 원통 쉘에서는 해석적인 산란 해가 존재하지 않기 때문에, Kirchhoff 가정을 적용한 Ye의 산란 기법[Z. Ye, J. Acoust. Soc. Am. 102, 877-884 (1997)]을 사용했다. 탄성 원통 쉘의 특성은 3차원 탄성파 이론을 적용하여 구현했으며 원통 쉘 내부의 유체를 고려했다. 유도된 해석 해를 이용하여 내부 유체가 산란 음장에 미치는 효과, Rayleigh 변수에 대한 산란 음장, 탄성 재질의 변화에 따른 먼 거리 산란 함수를 살펴보았다.

Cellulomonas sp. YE-5가 생산하는 Cellulase의 정제 (Purification of Cellulase Produced from Cellulomonas sp. YE-5)

  • 토양과 퇴비 등에서 섬유소 자화력이 있는 세균을 분리하고 이 균주들 중 cellulase 생산능이 가장 우수한 균주를 선별하여 Cellulomonas sp.로 동정하였다. Cellulomonas sp. YE-5의 최적 배양조건은 pH6.5 Solka floc 0.8(w/v), urea 0.06(w/v), K2HPO4 0.1(w/v), bacto peptone 0.2(w/v), yeast extract 0.2(w/v) 그리고 MgSO4.7H2O 0.0(w/v) 이었으며, 이러한 배지를 사용하여 $30^{\circ}C$ 에서 48시간 배양하였을 때 avicelase, CMCase 그리고 Beta-glucosidase는 각각 0.350, 3.180, 0.882units/ml의 활성을 나타내었다.

"고려사" 예지(禮志)의 복식사적 검토 (Historiographical Study of Costume Depicted in the "Ye-Ji(禮志)" Section of the Book "Goryosa(高麗史)")

  • This study is about the history of costumes depicted on the book Goryosa(高麗史) Ye-Ji(禮志). Researches about the costumes of Goryo dynasty are very rare and are mostly based on Goryodogyung(高麗圖經) and Goryosa Yebok-Ji(與服志). However, records about suitable costumes for a special scene of a rite can also be found in Ye-Ji besides Yebok-Ji. Therefore, this study analyzes Ye-Ji and categorizes the results into character and situation. Characters that can be found are king, officials, crown prince, aristocrat, commoner and envoy. Situations that can be found, according to the original text, are Gilrye(吉禮), Hyoongrye(凶禮), Goonrye(軍禮) and Garye(嘉禮). The results of the study are as follows; $\blacktriangleright$ The king had to change clothes in order to suit the situation according to the precise sequence of rituals. Therefore the king had to wear different clothes in the same rite, optionally, depending on the situation. This also applies to the case of officials. $\blacktriangleright$ The crown prince and aristocrats generally wore the same clothes as officials. $\blacktriangleright$ In the important ritual of Garye(嘉禮), many cases can be found where officials wore Jobok(朝服) and hands-on worker wore Gongbok(公服). $\blacktriangleright$ It is remarkable that on the New Year's day, the winter solstice and Sungsoojul(聖壽節; the emperor's birthday) the envoy of Ming Sangbok(常服); whereas the king and the officials of Goryo wore the Myunbok(冕服) or Jobok(朝服).

Effect of salicylic acid and yeast extract on curcuminoids biosynthesis gene expression and curcumin accumulation in cells of Curcuma zedoaria

  • The aim of this study is to evaluate the effect of yeast extract (YE) and salicylic acid (SA) on the expression of curcuminoid-biosynthesis genes (CzDCS and CURS1-3), and accumulation of curcumin in Curcuma zedoaria cell cultures. The results showed that, in cells treated with YE or SA, the expression levels of curcuminoid genes were 1.14- to 3.64-fold higher than the control (untreated cells), in which the YE exhibited a stronger effect in comparison with SA. Curcumin accumulation also tended to be similar to gene expression, curcumin contents in YE- or SA-treated cells were 1.61- to 2.53-fold higher than the control. The SA treatment at the fifth day of culture stimulated the curcumin accumulation and expression in all four genes compared to that at the beginning. While the YE treatments gave different results, the CzCURS1 and CzCURS3 genes were expressed strongly in cells that were treated at the beginning. However, the CzDCS and CzCURS2 genes showed the opposite expression pattern, they were activated strongly in the treatments at day five of the culture. However, the content of curcumin reached its maximum value on the fifth day of culture in all investigations.

Cloning and DNA Sequence of Carboxymethylcellulase (CMCase) Gene from Cellulomonas sp. YE-5

  • CMCase positive clones were screened from Cellulomonas sp. YE-5 and named pCE1, pCE2 and pCE3. Among the positive clones pCE1 was used for this study, because it has the smallest insert and the highest CMCase activity among the 3 clones, and its nucleotide sequence was determined. The CMCase gene in pCE1 was composed of 1071 bp of nucleotides coding 357 amino acids. Computer analysis showed that the pCE1 has 65% sequence homology with the endoglucanase from Cellulomonas fimi.
