A Study on the Long-Term Effects of Dietary Protein Level on Ca and Skeletal metabolism in Ovariectomized Rats (장기간의 고.저단백식이섭취가 난소절제쥐의 Ca 및 골격대사에 미치는 영향)

  • This study was performed to investigate the effect of dietary protein level on the metabolic changes of Ca and skeletons in postmenopausal women, using ovariecotomized rats as an animal model. The female rats of 200∼250g were fed either 8%(L) or 50%(H) casein diet for 15 weeks(1st experiment). At 15th week, the rats of each diet group were undergone ovariectomy or sham-operation and they were continued to feed the same experimantal diet for 9 more months(2nd experiment). Ca metabolism, kidney function and bone composition were determined at the end of 1st experiment, 3rd and 9th month of 2nd experiment. After 1st experiment, high protein group showed higher urinary Ca and protein excretion, however, there was no difference in GFR and urinary hydroxyproline excretion. The weights, ash and Ca content of femur, scapular and vertebra tended to be higher in high protein groups which tells that high protein promoted skeletal growth. In 2nd experiment, high protein group showed higher urinary Ca and protein excretion and lower Ca absorption and balance. GFR was not affected by dietary protein and ovariectomy but increased with time, as well as kidney weight which shows the continuous development of kidney at this age of 15 month in rats. There were no difference in urinary hydroxyproline, serum ALP, and PTH among experimental groups. The weights of femur, scapular, 4th vertebra increased with time, showing the skeleton continues to grow at this age in rats. However, Ca contents, Ca/wt, Ca/ash were decreased with time and tended to be lower in high protein group especially in femur. In conclusion, prolonged feeding of high protein diet deteriorated Ca metabolism and induced bone loss as time after menopause is extended.

Effect of Dietary Protein and Fat Levels on the Utilization of Nitrogen and Energy and Body Composition in Rats (식이중 단백질과 지방수준이 흰쥐의 성장, 질소와 에너지 이용 및 체조성에 미치는 영향)

  • In order to investigate the effect of dietary protein and fat levels on the growth and the utilization of nitrogen and energy and body composition in rats, Sprague\ulcornerDawley 48 male rats of 8 weeks old weighing approxijIlately 215-220g were subjected to feeding trials for 8 weeks and then subsequently to metabolic trials for 2 weeks. Four dietary protein levels (4, 8, 16, 32%) and each protein level contained two fat levels(3.9, 11.7%=1O,3J% of 3600kcal ME/kg) by addition of an appropriate amount of carbo\ulcornerhydrate and the following results were obtained. The body weight gain and food efficiency ratio of the rats to which a diet of 16% protein and 3.9% fat was fed were significantly higher than in either case of 8% pro\ulcornertein diet or of 32% protein diet. The digestibility of protein in the experimental diets was 73.3 -93.4%. The digesti\ulcornerbility of energy ( energy absorption) in the experimental diets was 83.2 -91.5%. The utilization of protein and the metabolic energy efficiency in the experimental diets was highest at the diet of 8% protein and 3.9% fat. The analysis of the body composition after feeding trials for 8 weeks has shown that the content of body water and protein were not affected by protein and fat levels in diet. The content of body fat in the rats to which 3.9% fat diet was fed was high\ulcornerer than that in those to which 11.7% fat diet was fed. From the above experimental results it may be suggested that the best formula of diet for the 8 weeks old rats may be composed of the 8% protein and 3.9% fat.

The Effect of Dietary Protein Source and Sulfur Amino acid Content on bone Metabolism in Growing Rats (식이 단백질의 종류와 함황아미노산 함량이 성장기 쥐의 골밀도에 미치는 영향)

  • This study was performed to evaluate the effect of dietary protein source and sulfur amino acid content on bone metabolism in ra. Thirty male rats (body weight 145$\pm$2g) were divided into three groups. The rats in the first group were fed on casein 20% diet as animal protein source and those in the second group were fed on soy 20% diet as plant protein source. Sulfur amino acid ratio of these group was 1.07:1. The rats in the third group were fed on soy 20% diet and the sulfur amino acid were supplemented with the amount contained as much in the soy 20% diet. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks, The total body, spine, femur bone mineral density and bone mineral content were measured using Dual Energy X-ray Absorptiometry Calcium, phosphate, pyridinoline, creatinine in urine and calcium, phosphate, alkaline phosphatase, osteocalcin in serum were measured. During the experimental period, plant protein (soy protein) group had a lower urinary Ca excretion, urine pyridinoline & crosslinks value and had a higher Ca efficiency in total bone and femur bone mineral density than animal protein (casein) group. There were no significant differences in serum calcium, phosphate, alkaline phosphatase and osteocalcin among the three groups of the rats. The findings from this study demonstrated that plant protein (soy protein) is beneficial of bone mineral density because it had a higher Ca efficiency in total bone and femur bone mineral density than animal protein (casein). However, the supplementation of sulfur amino acid on soy results were consistent with prior studies that dietary sulfur amino acid load had a negative effect on calcium balance. The rats fed sulfur amino acid supplementation diet increased urinary calcium excretion and decreased calcium efficiency for total and femur mineral density.   • The feasibility of foam separation to remove protein produced from fish culture water was investigated, By assuming foam separation column as a single well-mixed pool, a simplified model for foam separator conditions was alse studied under the batch operation. The model indicated that the protein removal could be described as a first-order reaction whose rate increases with both superficial air velocity and protein concentration in the bulk solution. from ,the results of an experimental study on the effects of superficial air velocity, the protein concentration, temperature, and pH on protein removal, the superficial air velocity and initial protein concentration in bulk solution were found to be important operational factors. Other experimental results important that foam separator operated under batch conditions would be considered as a completely mixed reactor. The protein removal rate by foam separation process was increased proportionally with the superficial air velocity, and the adsorptive removal rate of protein was found to obey Langmuir adsorption formula. The preposed simplified model was verified with the experimental data obtained from this study. Under the experimental range used, both temperature and pH did not affect the protein removal.

  • Defatted sesame flour is the by-products obtained after oil extracting process. Although this flour has high quality and quantity of protein its use is limited only for animal feed and fertilization. Sesame seeds contain antinutrients such as oxalate, phytate and phenol compounds and these compounds lower their nutritive value. recently, ultrafiltration(UF) has been used to concentrate protein from various food sources. This study was carried out to examine the effects of UF with different membrane pore size on the components of sesame protein concentrates including antinutrients and to compare with that of conventional acid-precipitated sesame protein isolate. The protein contents of sesame protein concentrates prepared by JF using 10K, 30K, 100K were 84.2%, 82.7%, 76.4% and the protein yields were 36.44%, 34.69, 31.43% and the protein contents was 88.7% Alkali extraction process at pH 9.0 followed by UF technique reduced oxalate and phytate content. There were 85% and 94% reduction of oxalate and phytate content by UF with membrane pore size of 100K daltons, respectively. However, the content of total phenol compounds was not reduced by this method. About 99% of calcium and 50% of zinc were removed by UF with membrane of 100K daltons. total essential amino acid contents of sesame protein concentrates prepared by UF were decreased slightly when compared with acid-precipitated sesame protein concentrate.

  • A human gene has been reported that may encode the enzyme acetohydroxyacid synthase. Previously this enzyme was thought to be absent from animals although it is present in plants and many microorganisms. In plants, this enzyme is the target of a number of commercial herbicides and the use of these compounds may need to be reassessed if the human enzyme exists and proves to be susceptible to inhibition. Here we report the construction of several plasmid vectors containing the cDNA sequence for this protein, and their expression in Escherichia coli. High levels of expression were observed, but most of the protein proved to be insoluble. The small amounts of soluble protein contained little or no acetohydroxyacid synthase activity. Attempts to refold the insoluble protein were successful insofar as the protein became soluble. However, the refolded protein did not gain any acetohydroxyacid synthase activity. In vivo complementation tests of an E. coli mutant produced no evidence that the protein is active. Incorrect folding, or the lack of another subunit, may explain the data but we favor the interpretation that this gene does not encode an acetohydroxyacid synthase.

  • The effects of the protein fraction of Panax ginseng on primary cultured chicken embryonic brain cells and DRG cultured with a deficient medium were studied. The protein fraction was further fractionated into four groups according to the molecular weight; larger than 10,000 dalton(fraction A), between 5,000 and 10,000 daltons(fraction B), between 1,000 and 5,000 daltons(fraction C), between 500 and 1,000 daltons(fraction D). All four protein fractions at the concentration of $100\;{\mu}g/ml$ significantly increased the number of the brain cells which promoted the neurite outgrowth. The activity of PDHC in the brain cells was elevated significantly by the protein fraction B at the concentration of $100\;{\mu}g/ml$. It was noted that $100\;{\mu}g/ml$ protein fraction C and D significantly enhanced the synthesis of protein in the brain cells. At the concentration of $100\;{\mu}g/ml$, the protein fraction B enhanced RNA synthesis and the protein fraction A significantly enhanced DNA synthesis in the brain cells. The protein fractions B, C, and D significantly promoted the neurite outgrowth of DRG at the concentration of $100\;{\mu}g/ml$.

Construction of a Protein-Protein Interaction Network for Chronic Myelocytic Leukemia and Pathway Prediction of Molecular Complexes

  • Background: Chronic myelocytic leukemia is a disease that threatens both adults and children. Great progress has been achieved in treatment but protein-protein interaction networks underlining chronic myelocytic leukemia are less known. Objective: To develop a protein-protein interaction network for chronic myelocytic leukemia based on gene expression and to predict biological pathways underlying molecular complexes in the network. Materials and Methods: Genes involved in chronic myelocytic leukemia were selected from OMIM database. Literature mining was performed by Agilent Literature Search plugin and a protein-protein interaction network of chronic myelocytic leukemia was established by Cytoscape. The molecular complexes in the network were detected by Clusterviz plugin and pathway enrichment of molecular complexes were performed by DAVID online. Results and Discussion: There are seventy-nine chronic myelocytic leukemia genes in the Mendelian Inheritance In Man Database. The protein-protein interaction network of chronic myelocytic leukemia contained 638 nodes, 1830 edges and perhaps 5 molecular complexes. Among them, complex 1 is involved in pathways that are related to cytokine secretion, cytokine-receptor binding, cytokine receptor signaling, while complex 3 is related to biological behavior of tumors which can provide the bioinformatic foundation for further understanding the mechanisms of chronic myelocytic leukemia.

Effect of Succinylation on Functional Properties of Aspergillus fumiagtus Cell Protein (숙시닐화가 Aspergillus fumiagtus 균체단백질의 기능적 특성에 미치는 영향)

  • The influence of succinylation on several functional properties of fungal protein (Aspergillus fumigatus) was investigated. Fungal protein was succinylated to 20.7 and 85.3% by addition of 2.5 and 10% succinic anhydride, respectively. Succinylated fungal protein decreased the absorbance at 260nm, nucleic acid and carbohydrate, but increased the proteinous nitrogen and protein extraction in fungal protein. Succinylation had an enhancing effect on the functional properties as much as the degree of it was increased. Oil retention of succinylated fungal protein was higher about from two to five times than those of milk casein. Nitrogen solubility of succinylated fungal protein was increased to 32 and 51% than that of milk casein and soy flour. Emulsifying activity and stability were increased in proportion to the succinylated degree of fungal protein. As the result of succinylation increase more than 80%, emulsifying activity increased about 8.4 times. In conclusion, succinylated fungal protein improved functional properties, compared with nonsuccinylated fungal protein, milk casein and soy flour.

UTILIZATION OF LUPIN (Lupinus angustifolius) SEEDS SUPPLEMENTED WITH AMINO ACIDS BY CHICKS

  • The utilization of feeding white sweet lupin (Lupinus angustifolius cv. Uniwhite) seeds supplemented with the limiting amino acids were investigated in day-old single comb White Leghorn male chicks. These were fed a commercial chick mash for the first 10 days and on a semi-synthetic protein-free diet for the next 6 days. For the subsequent 6 days of experimental feeding period, the birds were fed on the protein-free diet, basal diet containing 9.31% of lupin seed meal (LSM) protein, diets supplemented with methionine, methionine + tryptophan or methionine + tryptophan + lysine in the basal diet, and diet containing 9.84% of soybean meal (SBM) protein. When the LSM protein was supplemented with methionine, protein intake, body weight gain, protein efficiency ratio (PER) and net protein ratio (NPR) were increased (p<0.05). The birds excreted lower urinary nitrogen and fecal nitrogen per protein comsumption, had improved apparent (AD) and true (TD) digestibility but did not alter biological value (BV) of the protein. Metabolizability (MEn/GE) and heat production (HP) per MEn intake (HP/MEn) was lowered while energy retention (ER) was highered (p<0.05) compared with those of the basal diet. Also the body weight gain, PER, NPR and ER was increased but the BV and HP/MEn was lowered compared with those of the SBM protein. The results indicated that lupin seed supplemented with methionine increase body weight gain and energy rentention but did not alter biological value compared with those of lupin seed and soybean meal.
