  • The purpose of this research was to investigate the effect of Eunkyo-San(EKS) on the immune system. Administration of EKS(500 mg/kg) enhanced viability of splenocytes and thymocytes in BALB/c mice, and also EKS increased of splenic B, T lymphocytes and thymic T lymphocytes, significantly increased CD4 positive TH cells. EKS markedly enhanced the production of -interferon in mice serum. EKS accelerated the production of nitric oxide in peritoneal macrophages. These results suggest that EKS have an immune-enhancing activity.

  • Hepatitis E virus (HEV) infections cause epidemic or sporadic acute hepatitis, which are mostly self-limiting. However, viral infection in immunocompromised patients and pregnant women may result in serious consequences, such as chronic hepatitis and liver damage, mortality of the latter of which reaches up to 20-30%. Type I interferon (IFN)-induced antiviral immunity is known to be the first-line defense against virus infection. Upon HEV infection in the cell, the virus genome is recognized by pathogen recognition receptors, leading to rapid activation of intracellular signaling cascades. Expression of type I IFN triggers induction of a barrage of IFN-stimulated genes, helping the cells cope with viral infection. Interestingly, some of the HEV-encoded genes seem to be involved in disrupting signaling cascades for antiviral immune responses, and thus crippling cytokine/chemokine production. Antagonistic mechanisms of type I IFN responses by HEV have only recently begun to emerge, and in this review, we summarize known HEV evasion strategies and compare them with those of other hepatitis viruses.

  • Kamiinsamyangyoung-tang(KIT) has been widely used to treat amenorrhea and bradymenorrea caused from vital energy and blood deficit. KIT was composed of Insamyangyoung-tang, Cervi Cornu Pantotrichum and Hominis Placenta. The aim of this study is to investigate effect of KIT on Immune response and Blood formation. We investigated thymocytes, splenocytes population, ${\gamma}$-interferon, IL-2, IL-4, NO and phagocytic activity. We found that KIT had no effect on the Th and Tc cell population of thymocytes, Th cell population of splenocytes and ${\gamma}$-interferon quantity was decreased. KIT decreased the formation of Nitric Oxide from abdominal macrophage, on the other hand, it had no influence on the quantity of IL-2, IL-4.

  • A teratogenic study was carried out on New zealand White rabbits in order to examine the teratogenic potentiality of the recombinant human interferon-${\alpha}$A(rHuIFN-${\alpha}A$), an available therapeutic agent. The rHuIFN-${\alpha}A$ was intravenously administered at dose sevels of $1{\times}10^5$, $4{\times}10^5$ and $1.2{\times}10^6$ I.U/kg/day for a period of13 days from day 6 to day 18 of gestation. Two-thirds of the pregnant females in each group were sacrificed on day 29 of gestation and their fetuses were examined. The remaining dams were allowed to litter naturally, and the postnatal develpment of the offsprings was observed. The administration of rHuIFN-${\alpha}A$ during a period of organogenesis produced no embryotoxic and teratogenic effects.

  • The purpose of this research was to investigate the effects of Insamyangyoung-tang water extract (IYT) on the specific immune response in BALB/c mice. When IYT (500mg/kg) was administerd p.o. once a day for 7 days to BALB/c mice, the cell viability of thymocytes and splenocytes was increased. Also, IYT increased the population of CD4/sup +/ cells in thymocytes and the population of Thy1/sup +/ cells and CD4/sup +/ cells in splenocytes. In addition, IYT increased the production of γ -interferon and interleukin-2 from thymocytes and the production of γ -interferon from splenocytes. These results suggest that IYT enhances the specific immune response via activation of Th1 cells in thymocytes and splenocytes.

  • Interferon (IFN) alpha has been the first line therapy of chronic hepatitis B in children, but HBeAg seroconversion occurred in 26% of treated children compared to 11% of controls in multinational randomized controlled study. Recently, lamivudine was shown to be a potent inhibitor of Hepatitis B virus (HBV) reproduction both in HBeAg positive and in HBeAg negative (the pre-core mutant form) chronic hepatitis in randomized studies worldwide. Lamivudine therapy led to considerable improvement in the seroconversion rate of HBeAg in children with chronic hepatitis B, though long-term therapy resulted in the expansion of lamivudine-resistant mutant viruses. Combination therapy with lamivudine plus alpha-IFN does not seem to improve HBe Ag seroconversion. Above all, the most effective way to prevent hepatitis B is universal HBV vaccination.

  • Analysis of interferon alpha (IFN) modified with high molecular weight branched PEG was performed by capillary electrophoresis (CE) and MALDI-TOF mass spectrometry (MALDI-TOF MS). IFN was modified by the reaction of amine residues with an active ester of monomethoxy polyethylene glycol at various molar ratios. As a CE method. capilary electrophoresis sodium dodecyl sulfate nongel sieving (CE-SDS NGS) was performed using an uncoated capilary filled with a hydrophilic replaceable polymer network matrix. • The purpose of this study was to investigate the effects of Kami boatang(KBT) on the immune cells in Balb/c mice. KBT (500mg/kg) was administerd p.o. once a day for 7 days. KBT enhanced the proliferation of thymocytes and splenocytes. KBT enhanced the subpopulation of helper T(Th) cells, but did not affect the subpopulation of Thyl/B220 cells and Th/Tc cells in splenocytes. KBT enhanced the production of ${\gamma}$-interferon and interleukin-2 in thymocytes, but decreased the production of interleukin-4. KBT enhanced the production of ${\gamma}$-interferon, interleukin-2 and interleukin-4 in splenocytes and serum. KBT suppressed the production of nitric oxide, and enhanced the phagocytic activity in peritoneal macrophages. These results suggest that KBT has a potent activity on the immune response via the increase of the production of cytokines and phagocytic activity in vivo.
