Generally, carrier chickens contaminate the processing plant equipment to such an extent that negative chickens procell afterwards result in contaminated. meat. This study was performed to investigate the prevalence of Complliobacter jejuni in two chicken procelling plants. Altogether two hundred samples were collected from cloaca, carcasses, chilling water, and evis-cerationknives at different processing stages during the period of June to September 1987. The isolated organisms were tested for distribution of biotype, serotype. The results obtained were summarized as follows: 1. C. jejuni was isolated from 41(34.2%) of 120 chicken feces, 9(45.0%) of 20 carC888eS before chilling, 11(55.0%) of 20 carcasses after chilling, 12(60.0%) of 20 eviscerationlmives. The evilceration knives and chilling water were considered as major means of croll contamination. 2. In biotyping 82 isolates of C. jejuni, 64(78.1%) were cl888ified as biotype I, and 18(21.9%) belonged to biotype II. 3. In serotyping 82 isolates of C. jejuni, 64(78.1%) were identified as serotype LIO 37, and 18(21.9%) were untypable.
Effects of filtration and centrifugation on the oxidative stabilities of sesame oils during storage at $70^{\circ}C$ were studied by combination of determining peroxide values and conjugated dienoic acid values of oils and measuring the hexanal formation using headspace gas chromatography. Crude sesame oil from roasted seeds contained more free fatty acids, conjugated dienes, and metals (Fe, Cu, Mg and Zn); on the other hand, higher contents of moisture and ${\gamma}-tocopherol$ were found in the filtered or centrifuged oil. Only filtered oil contained more peroxides than the crude oil in spite of the color advantage of the highest L and b values among three oils. All the oils showed the tendency of increasing in total color difference during storage at $70^{\circ}C$, fatty acid compositions were relatively constant except for decreasing tendency of linoleic acid in filtered oil. No significant difference at 1% in the oxidative stabilities was observed between centrifuged oil and crude oil with higher susceptibility to the oxidation in the filtered oil. Centrifuged sesame oil was the best in the aspect of both oxidative stability and the oil color.
The effects of temperature and flow rate of eluent on the separation of adjacent lighter lanthanide pairs (La : Ce, Ce : Pr, Pr: Nd) have been studied with displacement chromatography. Two serial columns packed with Amberlite 120 cation exchange resin are used for loading and separation. The retaining ion is $H^+$ ion and the eluent is 0.012M and 0.015M of EDTA solution. The columns and the eluent are maintained at the temperature of 90$^{\circ}C$ and pressurized to reduce vaporizing in the ion-exchange resin column. The eluated solution is analyzed directly with ICP-AES. The separation factors of the lanthanide pairs, La: Ce, Ce :Pr, and Pr: Nd, are 4.6, 2.8, and 1.9, respectively and are higher than that from theoretical calculation at 25$^{\circ}C$. When the flow rate is reduced from 2.5 ml/min to 1.5 ml/min, the HETP is reduced from 1.60 cm to 0.88 cm. The separation efficency can be improved at lower flow rate of eluent and higher operating temperature. The recoveries of pure lanthanides than 99.9% are 49∼77% from this separation.
A bacterial strain capable of hydrolyzing xylan and locust bean gum (LBG) was isolated from farm soil by enrichment culture using mixture of palm kernel meal (PKM) and wheat bran as carbon source. Nucleotide sequence of 16S rDNA amplified from the isolate YB-1107 showed high similarity with those of genus Cellulosimicrobium strains. Xylanase productivity was increased when the Cellulosimicrobium sp. YB-1107 was grown in the presence of wheat bran or oat spelt xylan, while mannanase productivity was increased drastically when grown in the presence of PKM or LBG. Particularly, maximum mannanase and xylanase activities were obtained in the culture filtrate of media containing 0.7% PKM or 1% wheat bran, respectively. Both enzyme activities were produced at stationary growth phase. Mannanase from the culture filtrate showed the highest activity at $55^{\circ}C$ and pH 6.5. Xylanase activity was optimal at $65^{\circ}C$ and pH 5.5. The predominant products resulting from the mannanase or xylanase hydrolysis were oligosaccharides for LBG or xylan, respectively. In addition, the enzymes could hydrolyze wheat bran and rice bran into oligosaccharides.
The measurement of trace amounts of uranium(VI) and thorium(VI) in the solutions containing high concentration of dissolved salts was carried out. The procedure using reversed phase liquid chromatography with trace enrichment techniques has been developed to cope with the high salt content of the samples. 2 ml of sample were passed through a small C_{18}$ reversed phase enrichment column with ${\alpha}$-HIBA eluent (0.11 M, pH 5.5) where the uranium and thorium were separated from other constituents and concentrated. The uranium and thorium were then backflushed from the column onto a deactivated C_{18}$ reversed phase analytical column where furthe separation was achieved with a mixed eluent (pH3.0, 0.17M ${\alpha}$-HIBA/0.0038 M 1-pentanesulfonate). The separated species were determined spectrophotometrically by postcolumn reaction with Arsenazo III, the chromogenic reagent. Detection limits were found within 1 ppb range for both species.
The present study intends to characterize the DNA damage-inducible responses in Caenorhabditis elegans. To study UV-inducible responses in C. elegans, two UV-inducible cDNA clones were isolated from C. elegans by using subtration hybridization method. To investigate the expression of isolated genes, UV100 and UV150, the cellular levels of the transcript were determined by Northern blot analysis after UV-irradiation. The transcripts of isolated gene increased rapidly and reached maximum accumulation after UV-irradiation. Compared to the message levels of control, the levels of maximal increase were approximately 2 folds to UV-irradiation. These results implied that the effects of damaging agents are complex and different regulatory pathways exist for the induction of these genes. To study the function of UV100 and UV150 gene in response to UV irradiation, we carried out a RNAi experiment and investigated the UV sensivity. This result indicated that UV100 gene involved in stage-specific repair pathway or regulated by development.
A new hydrogen sulfide-oxidation bacterium, Thiobacillus sp. was isolated from waste coal mine water around Hawsun in Chunnam province. The isolate was motile gram-negative rod shape, formed spore and grew up to be aerobically facultative chemolithotroph by using energy released from the oxidation of reduced inorganic sulfur compounds. It could assimilate various kinds of organic compounds and grew well upon thiosulfate-supplemented basal medium. To the lelvel of 32 mM in thiosulfate concentration, thiosulfate in itself was utilized as energy source for growth. However, from those of the higher concentration than 32 mM, thiosulfate functioned specifically as the substrate inhibitor rather than as the energy source. It was found that the optimum thiosulfate concentration for growth was 32 mM. The G+C content of the DNA was 65.0 mol%. The isolate had 16 : 1 + 17$_{cyc}$, 16 : 0 as their major non-hydroxylated cellular fatty acids, 3-OH 12 : 0 as a hydroxylated fatty acid and also contained unidentified $C_{18}$ branched fatty acid. The ubiquinone system in the respiratory chain was Q-9. Based on the physiological and biochemical characteristics, the isolate was assigned to a novel species of the genus Thiobacillus sp. iw.
Present paper reports a new method to separate the electrolyte membranes and carbon paper without using ultrasonic waves and stirring. In this method, these were separated from fuel cell membrane-electrode assembly(MEA) using the distilled water, butanol and surfactant by dipping method without the dispersion of catalyst particles. Separated carbon paper catalysts and fuel cell Pt/C catalysts were heated in aqua regia at $80{\sim}85^{\circ}C$ and added to precipitant. After calcination, Pt metal was recovered which might be used in fabricating new fuel cells.
The effects of CRP purified from human ascites fluid on phagocytic activity of the human macrophage were investigated. CRP was purified using affinity chromatography including absorption on p-diazonium phosphocholine or C-polysaccharide coupled sepharose 4B and gel filtration on hydroxylapatite column chromatography. Macrophage was separated ficoll hypaque gradient density and absorption method, and then was confirmed phagocytic uptake test using latex method. CRP was able either to inhibit or to enhance phagocytic activity of human macrophage against bacteria in vitro. The effects of CRP on phagocytic activity of human macrophage were in time and dose-dependent manners. The additional sequence of reaction mixture against bacteria in vitro shows a threshold stimulus on the activation of phagocytic response upon the CRP.

말징버섯 Calvatia craniformis의 culture broth 40 liter를 여과하여 얻은 균사채를 열수 추출하여 진한 갈색 건조분말(Fr. A) 13.1g을 분리하였다. Fr. A에 대하여 DEAE-cellulose ion chromatography를 시행하여 중성분획인 흰색 분말 (Fr. B) 2.50g을, 산성분획인 갈색 분말 (Fr. C) 3.50g을 각각 분리하였다. Fr. B 500mg에 대하여 Sepharose CL-4B gel filtration chromatography를 시행하여 흰색분말 Fr. D(Calvatan) 350mg을 분리하였다. Calvatan 350mg에 대하여 Con A-Sepharose 4B affinity chromatography를 적용하여 미흡착 분획인 Fr. F($\alpha$-form)와 흡착 분획인 Fr. E ($\beta$-form)로 정제하였다. 항암작용의 기전을 구명하고자 마우스에 대하여 면역학적 실험을 시행하였다. macrophage의 활성에 대한 영향을 조사하였던 바, 투여군의 활성화된 macrophage에 의해 유리되는 superoxide anion의 양은 대조군에 비해 1.4배 증가하였고 Calvatan 투여군의 용혈반 형성세포(PFC)는 대조군에 비해 3.1배 증가하였다. 화학 분석에 의해, Calvatan은 다당체 87.2%, 단백질 1.8% 및 hexosamine 1.3%로 구성되어 있었다. 따라서 항암성 분획들은 protein-bound polysaccharide임을 알 수 있었다. 또한 각 분획의 다당체를 구성하고 있는 단당류는 glucose, galactose, mannose 및 xylose 였으며 단백질 부분은 16종의 아미노산으로 구성되어 있었다.
