Effect of Mixture of Gastrodiae rhizoma on Blood Amelioration in High Cholesterol-diet Rats (고콜레스테롤 식이 랫드에서 천마혼합액의 혈액개선 효과)

  • This study was carried out to investigate the effect of mixture of Gastrodiae rhizoma (GM) on blood amelioration in high cholesterol-diet rats. Sprague-Dawley male rats were randomly assigned to one normal diet and three high cholesterol-diet groups which contained 1 % (w/w) cholesterol diet. The groups of high cholesterol-diet were classified to control (high cholesterol-diet only), GM-1 (high cholesterol-diet and GM) and ST-1 (high cholesterol-diet and Statin drug). The body and organs weight were not significantly changed among the tested groups. Contents of serum total cholesterol and LDL-cholesterol were significantly increased in cholesterol-diet groups compared with normal diet group but significantly decreased in the group of GM-1. Morphology of red blood cell in GM-1 group was similar to normal diet group but the control group had many crystals of cholesterol. Hepatic xanthine oxidase activity in the rats of high cholesterol-diet was decreased up to the levels of normal diet group according to oral administration of GM. The results of the present study demonstrate that the orally injection of GM can ameliorate the status of total cholesterol and LDL-cholesterol, and repress xanthine oxidase in liver in high cholesterol-diet rats. These finding suggest that GM is expected to be an effective tea for the blood amelioration in high cholesterol-diet rats.

Antioxidative Effect of Suaeda japonica Ethanol Extract and Solvent Partitioned Fractions (칠면초 추출물의 항산화 효과)

  • We studied the antioxidant activity of Suaedajaponica, found in Korean mud flats. A 70% ethanol extraction yielded 9.74%. The 70% ethanol extract was further fractionated with hexane, chloroform, ethyl acetate, butanol, and water. The water fraction resulted in the highest extraction yields, but the ethyl acetate fraction resulted in the highest total phenol content and also total flavonoid content. The antioxidative activity of the fractions was evaluated using several methods and compared to commercial antioxidants. The scavenging activities of the hydroxyl radical, hydrogen radical, hydrogen peroxide, and xanthine oxidase of the ethyl acetate fraction were equivalent to that of BHA. The scavenging activities of the hydroxyl radical, hydrogen peroxide, and xanthine oxidase of the ethanol extracts were approximately 80~90% of that of BHA.

Effects of Cordyceps Sinensis on Antioxidation in the Livers of Hydrocortisone Acetate-Treated Rats (동충하초(冬蟲夏草)가 hydrocortisone으로 유발시킨 양허(陽虛) 동물모형(動物模型)에서 항산화(抗酸化) 작용(作用)에 미치는 영향(影響))

  • Objectives : Cordyceps Sinensis (CS) was tested for the effects of antioxidant enzymes and lipid peroxidation in the liver. Methods : We measured the changes in body weight, enzyme activity, lipid peroxide and the death rate in the hydrocortisone acetate-treated rats. Results : In vitro, CS didn't effect levels of lipid peroxide. the activities, and the ratio of type conversion of xanthine oxidase. In the hydrocortisone acetate-treated rats, lipid peroxide, the activities, the ratio of type conversion of xanthine oxidase, and the death rate all increased. But, glutathione peroxidase and superoxide dismutase decreased. In vitro, after CS was administered to hydrocortisone acetatetreated rats, the levels of lipid peroxide in the liver, and the death rate decreased. However, the activities, and the ratio of type conversion of xanthine oxidase decreased. The body weight, glutathione peroxidase, and superoxide dismutase in+creased. The effects of Sinensis Cordyceps Broth did better than the effects of Sinensis Cordyceps Mycelia. Conclusions : These results suggest that CS decrease the activities of free radical generating enzymes which form lipid peroxide and increase the activities of oxygen free radical scavenging enzymes.

Anti-oxidative, Nitric Oxide Inhibitory Activities and Irritation Test of the Fermented Opuntia humifusa Cladodes

  • Opuntia humifusa is a member of the Cactaceae family. In the present study, the antioxidant, nitric oxide(NO) inhibitory activities and potential irritation response of the fermented Opuntia humifusa cladodes(FOH) were investigated for cosmetic use. Antioxidant activities were tested using 1,1-diphenyl-2-picrylhydrazyl(DPPH) and xanthine oxidase assay, we found that FOH could scavenge DPPH free radicals and inhibit xanthine oxidase activity in a dose dependent manner, with $IC_{50}$ of 2599.46${\mu}g/ml$ and 721.38${\mu}g/ml$, respectively. To investigate the possible anti-inflammatory effects of FOH, RAW 264.7 macrophages were pretreated with FOH($0{\sim}400{\mu}g/ml$) for 30 min and then treated with LPS for 24 h. We found that cell number did not vary significantly with the treatment of FOH, and FOH did not show any inhibitory effects on LPS-induced NO production. After application of FOH to rabbits for skin and eye irritation test, the experimental sites did not show any response compared to the control. FOH were considered to be a non-irritant to the skin and eye. Based on the above information, we suggest that FOH can be considered to be a non-irritant base cosmetic material for safely use.

Effect of Testosterone on Free Radical Generating Enzyme and Lipid Peroxidation (지질과산화 반응과 Free Radical 생성계 효소활성에 미치는 Testosterone의 영향)

  • Sex hormones not only regulate external sexual characteristics but several internal biochemical processes. It is well accepted that life-span of female is longer than that of male. Life-span is closely related with aging process in which free radicals are known to be involved. We investigated the effect of testosterone on free radical generating systems and lipid peroxidation based on the sexual difference. Lipid peroxide levels of male and female mouse were increased proportionately with age, especially in male mouse. Increase in enzyme activity of aldehyde oxidase with age was observed in male mouse, while no siginificant change in enzyme activity was found in female mouse. Enzyme activity of xanthine oxidase also showed similar results. It, however, was not significant statistically. Lipid peroxide level and xanthine oxidase type conversion ratio of male and female mouse liver homogenate incubated at $37^{\circ}C$, increased remarkably in proportion to incubation time, especially in male mouse. Lipid peroxide level and aldehyde oxidase activity were measured in normal male mouse, castrated mouse and testosterone treated-castrated mouse. Castrated mouse group showed decrease in lipid peroxide level and aldehyde oxidase activity compared with normal group. Treatment of castrated mouse with testosterone, however turned the level of lipid peroxide and aldehyde oxidase activity back to normal. From the above results, it might be concluded that testosteron could increase the activities of free radical generating enzymes which would result in the formation of lipid peroxide, consequently leading to aging.

Antioxidant Activities and Phenolic Compounds Composition of Extracts from Mulberry (Morus alba L.) Fruit

  • The objective of this research was to evaluate the ability of water and ethanol extracts from mulberry fruit (Morus alba L.) to influence the inhibitory activity of angiotensin converting enzyme (ACE) and xanthine oxidase(XOase). The total phenol contents and sixteen phenolic compounds were investigated in water and ethanol extracts. In order to understand the factors responsible for the potent antioxidant and antihypertensive ability of mulberry, it has been evaluated for anti-oxidative activity using Fenton's reagent/ethyl linoleate system and for free radical scavenging activity using the 1,1-diphenyl-2-picryl hydrazyl free radical generating system. The total phenol contents and total of phenolic compounds in ethanol extract showed higher levels than water extract in mulberry fruit six phenolic compounds (chlorogenic acid, narigin, syringic acid, quercetin, naringenin, kampferol) has a higher individual phenolic compound content in the 60% ethanol extraction than 80% ethanol extract. The inhibitory activity on angiotensin converting enzyme (ACE) were highest in 80% ethanol extract (9.0%). Also, activity of xanthine oxidase(XOase) inhibition appeared highest in 80% ethanol extracts and correlated well with the total phenolic content, which was modulated by the concentration of individual phenolic compounds. This result revealed, that strong biological activity was caused by specific phenol compound contents. Utilization of water and ethanol extracts from mulberry fruit are expected to be good candidate for development into source of free radical scavengers and anti-hypertentive activity

Effect of the Toluene Administration on the Liver Damage in Rats Fed Low or Standard Protein Diet (저 및 표준단백식이로 성장시킨 흰쥐에 Toluene 투여가 간 손상에 미치는 영향)

  • To study the effect of toluene administration on the liver damage, rats were previously fed a low (casein 7%, LP) or standard(casein 20%, SP) protein diet and for four days toluene(50% in olive oil) was given at 0.2ml/100g body weight/day to the male rats, and then the degree of liver damage in toluenetreated animals fed LP were compared with those fed SP. The increasing rate of liver weight/body weight and the serum levels of xanthine oxidase to the control group were higher in rats fed SP than those fed LP. The decreasing rate of protein contents in cytosol, mitochondria and glycogen, glutathione contents of liver to the control group were higher in rats fed SP than those fed LP. In histopathological findings, the swelling of hepatic cell around the central vein was demonstrated in all the two groups toluene-treated rats. But the degree of swelling severity in hepatocytes was somewhat higher in rats fed SP than those fed LP. Therefore it is assumed that the degree of liver damage severity in toluenetreated animals was higher in rats fed SP than those fed LP.

  • Objectives : This study was investigated to analyze the anti-oxidant activity of water and ethanol extracts from the Magnoliae Flos. Methods : The effects on electron donating ability (EDA), SOD-like activity and xanthine oxidase inhibition have been investigated the anti-oxidant activity for utilization as a natural material of cosmetics. Results : This study was investigated to analyze the anti-oxidant activity of water and ethanol extracts from the Magnoliae Flos. In the electron donating ability test, water and ethanol extract from Magnoliae Flos showed an effect of 75.6% and 83.5% at 1,000 ppm concentration, respectively. In the superoxide dismutase (SOD)-like activity, 1,000 ppm of water extract showed an effect of 45.2% and ethanol extract showed an effect of 64.2%. Xanthine oxidase showed a meaningful inhibitory effect of 68.8% in water extract and 23.2% at 1,000 ppm in ethanol extract. Conclusion : From these results, Magnoliae Flos showed the development possibility as a natural material of anti-oxidant cosmetics.

  • To evaluate an effect of liver damage on the dermal oxygen free radical metabolizing enzyme activities, the $CCl_4$ (0.1 ml/ 100 g body wt., 50% $CCl_4$ in olive oil) was intraperitoneally given to the rats every other day for 2 weeks. Based on the histopathological findings, liver weight (%), serum alanine aminotransferase, xanthine oxidase activities and hepatic lipid peroxide contents, the animals were induced severe liver damage. In the present liver damaged animal, all the activities of dermal scavenging enzymes such as superoxide dismutase, catalase and glutathione peroxidase were significantly decreased compared with central. And under the cytochemical electron microscopy the more coarse granules of cerrous perhydroxide were found compared with the control. In conclusion, the $CCl_4$-induced liver damage may influence upon the activities of some dermal oxygen free radical scavenging enzymes.

Evaluation of physiological functionalities of Codonopsis lanceolata root extracts on the storage temperatures and durations

  • The effects of bioactivity, binding polyphenolic contents, DPPH radical scavenging activity, ABTS radical scavenging activity, xanthine oxidase and immune activity on the storage temperature and durations of Codonopsis lanceolata root were evaluated in vitro. The contents of total polyphenol and flavonoid content by storage temperature and storage period was no significant difference according to the storage period, but it was found that the content was higher at lower storage temperature. The DPPH free radical scavenging activity at six different concentrations, 500, 1000, 2500, 5000, 10000 and 20000 mg/L are measured, the scavenging activity according to different storage temperature and storage period showed relatively the higher the activity in the shorter storage period or the lower storage temperature. The ABTS radical scavenging activity did not show a significant difference under various storage temperature and storage period conditions. The xanthine oxidase inhibitory activity of C. lanceolata extract tended to decrease more, depending on the longer storage period or the higher storage temperature. In different storage period and storage temperature conditions, the immune cell growth of C. lanceolata extract promoted a concentration-dependent manner in both human T cell and B cell, and did not show a significant difference. These results of this study suggested that the root of C. lanceolata may assist in the potential biological activities, and can be used as a source of human health products.
