Gene Expression study of human chromosomal aneuploid

  • Chromosomal copy number changes (aneuploidies) are common in human populations. The extra chromosome can affect gene expression by whole-genome level. By gene expression microarray analysis, we want to find aberrant gene expression due to aneuploidies in Klinefelter (+X) and Down syndrome (+21). We have analyzed the inactivation status of X-linked genes in Klinefelter Syndrome (KS) by using X-linked cDNA microarray and cSNP analysis. We analyzed the expression of 190 X-linked genes by cDNA microarray from the lymphocytes of five KS patients and five females (XX) with normal males (XY) controls. cDNA microarray experiments and cSNP analysis showed the differentially expressed genes were similar between KS and XX cases. To analyze the differential gene expressions in Down Syndrome (DS), Amniotic Fluid (AF)cells were collected from 12 pregnancies at $16{\sim}18$ weeks of gestation in DS (n=6) and normal (n=6) subjects. We also analysis AF cells for a DNA microarray system and compared the chip data with two dimensional protein gel analysis of amniotic fluid. Our data may provide the basis for a more systematic identification of biological markers of fetal DS, thus leading to an improved understanding of pathogenesis for fetal DS.

X-linked Gene Expression Profiles by RNAi-Mediated BRCA1 Knockdown in MCF7 Cells

  • Germ-line mutations of the BRCA1 gene confer an increased risk for breast and ovarian cancers. BRCA1 in female cells is directly related with the maintenance of the inactive X chromosome (Xi). The effect by the loss of the BRCA1 function on the X chromosome gene expression remains unclear in cancer cells. We attempted to investigate the expression pattern of the X-linked genes by performing BRCA1 knockdown via RNA interference in the MCF7 breast cancer cell line. The transcriptional and translational levels of BRCA1 were decreased over 95% in the MCF7 cells after BRCA1 knockdown. The expression patterns of one hundred ninety X-linked genes were profiled by the X chromosome-specific cDNA arrays. A total of seven percent of the X-linked genes (14/190) were aberrantly expressed by over 2-fold in the MCF7-BRCA1 knockdown cells, which contained two up-regulated genes (2/190, 1 %) and 12 down-regulated genes (12/190, 6.3%). It is interesting that 72% of the aberrantly expressed X-linked genes were located on the Xq (10/14,) region. Our data suggests that BRCA1 may not be important to maintain X chromosome inactivation in cancer because the BRCA1 knockdown did increase the expression of the only one percent of X-linked genes in the human breast cancer cells.

한국산 감자바이러스병에 관하여 (Studies on potato virus disease in Korea)

  • The potato viruses, as possible potato virus X(PVX), potato virus Y(PVY), potato virus S(PVS) are isolated from potato tuber, which collected from eleven areas (Table 1) in Korea. These viruses are isolated by single lesion isolation, Aphid transmission and inoculating methods through the many species of the different plants. The PVX is identified by host range, symptoms, physical properties, serological reaction and electron micrography. The other two viruses are identified by the first two methods mentioned above. The results of the above experiments are as follows. The total value of these viruses infection is 81%. The value of PVX infection is higher than the other two viruses. The properties of PVX are marked as local lesions on Comphrena globosa. The dilution end point is $10^{-6}$, the thermal inactivation point is $70^{\circ}C$ and the size of virus particles is around 13 x 60 $m\mu$.

  • 담배거세미나방 핵다각체병바이러스에 섭식촉진물질, 다각체 침전방지제 및 전착물질과 자외선 차단제를 첨가하여 만든 3종의 바이러스 살충제를 담배거세미나방 유충에 먹여 제제별로 살충 효과를 비교하였다. 바이러스 살충제에 설탕농도 1-5% 첨가로 섭식촉진이 증대되어 살충효과가 높았으며, 바이러스의 불활화 차단효과도 있었다. 살충제 제형에 따른 바이러스 침전방지는 polyvinylalcohoI 0.5%, 식물체 전착성은 Triton X -100 0.1 %에서 효과가 좋았다. 자외선차단제의 양은 white carbon은 100 L 당 800 g, molasses는 30%첨가에서 바이러스의 불활화가 감소되었으며 white carbon-molasses 혼합제는 white carbon 500 g, molasses 10%의 비율에서는 살충효과가 약간 감소하였으나 세 제제 모두 살포 3일후 95%이상의 살충율을 보였다. White carbon 체제는 molasses 제제보다 다각체 포매가 뚜렷하여 잔류효과가 증대되었으나 유충의 섭식은 molasses제제에서 촉진되었다.

Effects of Ionizing Radiation on Postharvest Fungal Pathogens

  • Postharvest diseases cause losses in a wide variety of crops around the world. Irradiation, a useful nonchemical approach, has been used as an alternative treatment for fungicide to control plant fungal pathogens. For a preliminary study, ionizing radiations (gamma, X-ray, or e-beam irradiation) were evaluated for their antifungal activity against Botrytis cinerea, Penicillium expansum, and Rhizopus stolonifer through mycelial growth, spore germination, and morphological analysis under various conditions. Different fungi exhibited different radiosensitivity. The inhibition of fungal growth showed in a dose-dependent manner. Three fungal pathogens have greater sensitivity to the e-beam treatment compared to gamma or X-ray irradiations. The inactivation of individual fungal-viability to different irradiations can be considered between 3-4 kGy for B. cinerea and 1-2 kGy for P. expansum and R. stolonifer based on the radiosensitive and radio-resistant species, respectively. These preliminary data will provide critical information to control postharvest diseases through radiation.

무병종서 생산을 위한 감자X바이러스 및 엽권바이러스에 관한 연구 (Studies on the Potato Virus X and Potato Leaf Roll Virus for Disease-free Seed Potato Production)

  • A series of experiment was carried out to study on the production of disease-free seed potatoes at the Alpine Experiment Station from 1960 to 1968, which initiated a study of comparison on degeneration of plain warm region and high altitude products and the effect of latent potato virus X (PVX) and potato leaf roll virus(PLRV) on degeneration. Particular observations were made on some aspect of the nature of potato virus disease and its control such as concentrations of PVX, range of host plants, physical properties such as concentrations of PYX, range of host plants, physical properties and carrying effect of insects, by investigating 9 different areas of the main potato producing regions (Kimhae, Taegu, Choongju, Taejoen, Suwon, Kwangju, Chonju, Cheju and Chinju). Highly purified anti-serum was separated and tested for control of the virus disease and also various method of prevention and control of PLRV were observed, using cultivation of sprouted seed tubers, early harvesting method, and systemic chemicals. The results obtained are summarized as follows; 1. Potato yield in the plain region decreased by 32.8~66.3% in the first year cultivation of seed potatoes from colder region, and the rate of virus infection was 92.9 to 95.4%. 2. Plants of three families including, 20 species were susceptible to the PVX, and among the plants Salvia officinalis of a habits only was the carrier while the symptom of Digitalis purpurea of Screphulariaceae was masked. Necrosis and ring spot was occurred in most pJants of the Solanaceae and ring spot symptom also was observed in Nicotiana tabacum L. var. White Burley and in N. glutinosa. 3. The 8$C_2$ strain of virus had the following physical properties; thermal inactivation point, 68-$72^{\circ}C$ : dilution inactivation point, above 1, 000, 000 dilution: ageing in vitro, 240-360 days: and ageing in dry plant tissue, 30 days. 4. Myzus persicae and Oxya spp. did not transmit the 8$C_2$ strain of potato virus. 5. Virus was purified through the ammonium sulphate isolating method, and higher titer value, 1/2048 was obtained through anti-serum test. 6. Inhibition Chenopodiacae on the virus infection of potato was remarkable, and inhibition of local lesion host also was observed. 7, By earlier planting of sprouted seed tubers, growth period could be prolonged by 10 to 12 days. 8. Earlier harvest decreased much the rate of virus infection of seed potatoes. 9. According to the results of aphid control trial using systemic soil insecticides at Kangnung and Taekwanlyung, PSP 204, Disyston and Thimet was effective to aphid control. In particular, control effect of twice treatments of PSP 204 was great. 10. Treatmental effect of those chemicals lasted about 60-70 days. However, single foliar application of emulsified chemicals was not effective to potato virus control. 11. The effect of PSP 204, Disyston, and Thimet on the control of potato leaf roll virus was great, particularly in the case of two treatments of PSP 204, at Kangnung as well as at Taekwanlyung. Higher negative correlationship between the control effect of potato leaf roll virus and potato yield was observed showing the value r=-0.85 at Kangnung, and r=-0.87 at Taekwanlyung. 12. Differences in the control effects among PSP 204, Disyston, and Thimet was not noticed.

Development of the stable liquid formulation of Burkholderia cepacia YC5025, a biocontrol agent for cucumber anthracnose

  • A new and effective formulation using antagonistic bacteria, Burkholderia cepacia YC5025 in vegetable oil was developed for the biocontrol of anthracnose. The bacterial population in the formulation was maintained to 5x10/sup7/ cfu/ml upto 60 days at room temperature. Control efficacy of the formulation for anthracnose was over 80% by spraying of diluted suspension(x1,000) in growth chamber tests. On the contrary, the bacterial suspension in distilled water or bacterial culture broth containing same number of spores as the formulation had low control efficacy around 40% even 2-weeks storage after preparation. The shelf-life of the formulation was longer than that of bacterial preparation using clay minerals such as talc or bentonite. The mechanisms of newly developed bacterial formulation are possibly the formation of water film on the surface of cucumber leaves and inactivation of the bacteria in the vegetable oils during storage. Further field tests and improvements with new liquid bacteiral formulation need to be done for practical application.

전기자극 하에서 알콜 산화효소의 안정화연구; 당, 히드로젤, 계면활성제 효과 (Stabilization of Alcohol Oxidase under Electrostimulation; Sugars, Hydrogels and Surfactants Effect)

  • 효소는 환경과 조건에 따라 상이한 활성도를 나타내며, 전기 자극에 의하여 비활성화된 알콜 산화효소는 당, 계면활성제, 히드로젤 들에 의하여 안정화되었다. 이들 효소는 trehalose, TritonX-100, Brij에서 보다 안정하였으며, 이들 효소의 안정도는 Hansenula sp., P. pastoris, C. boidinii에서 분리된 효소가 순서적으로 보다 안정하였다. 전기자극하에서 효소의 안정화에 대한 이 연구는 생물공학, 단백질공학, 의료공학 분야에 널리 이용될 수 있으리라 사료된다.

Female Carriers of Duchenne Muscular Dystrophy

  • Dystrophinopathy, caused by mutations in the DMD gene, presents with variable clinical phenotypes ranging from the severe Duchenne muscular dystrophy (DMD) to the milder Becker muscular dystrophy(BMD) forms. DMD is a recessive X-linked form of muscular dystrophy. Two-thirds of mothers of affected males are thought to be DMD carriers. Approximately 2.5-7.8% of female DMD carriers have muscle weakness and are categorized as manifesting DMD carriers. The symptoms of female carriers of DMD range from mild muscle weakness to severe gait problems. The most commonly presented symptom is mild proximal muscle weakness, which is often asymmetric and progressive, but shows variable clinical spectrum with BMD of more severe DMD-like phenotype. Atypical presentations in manifesting carriers are myalgia or cramps without limb weakness, isolated cardiomyopathy and camptocormia. Multiplex PCR and MLPA analysis are common techniques to identify mutations in the DMD gene. Relationship between X-chromosome inactivation and clinical severity is not clear. Female carriers of DMD are not less common, and they have an important role of birth of a male DMD.

Inactivation of the Wall-Associated De-N-acetylase (PgdA) of Listeria monocytogenes Results in Greater Susceptibility of the Cells to Induced Autolysis

  • Several species of Gram-positive bacteria have cell wall peptidoglycan (syn. murein) in which not all of the sugar moieties are N-acetylated. This has recently been shown to be a secondary effect, caused by the action of a peptidoglycan N-acetylglucosamine deacetylase. We have found that the opportunistic pathogen Listeria monocytogenes is unusual in having three enzymes with such activity, two of which remain in the cytoplasm. Here, we examine the enzyme (PgdA) that crosses the cytoplasmic membrane and is localized in the cell wall. We purified a hexa-His-tagged form of PgdA to study its activity and constructed a mutant devoid of functional Lmo0415 (PgdA) protein. L. monocytogenes PgdA protein exhibited peptidoglycan N-acetylglucosamine deacetylase activity with natural substrates (peptidoglycan) from both L. monocytogenes and Escherichia coli as well as the peptidoglycan sugar chain component N-acetylglucosamine, but not with N-acetylmuramic acid. As was reported recently [6], inactivation of the structural gene was not lethal for L. monocytogenes nor did it affect growth rate or morphology of the cells. However, the pgdA mutant was more prone to autolysis induced by such agents as Triton X-100 and EDTA, and is more susceptible to the cationic antimicrobial peptides (CAMP) lysozyme and mutanolysin, using either peptidoglycan muramidases or autolysis-inducing agents. The pgdA mutant was also slightly more susceptible than the wild-type strain to the action of certain beta-lactam antibiotics. Our results indicate that protein PgdA plays a protective physiological role for listerial cells.
