Effect of Active Ceramic Resources in Cutaneous Wound Healing and Expression of its Related Cytokines

  • Cutaneous wound healing is a highly orchestrated process comprising a number of well-defined temporally overlapping phase. These include the migration of inflammatory cells to the wound site, and re-establishment of an epithelial barrier (re-epithelization), and the deposition of matrix by fibroblast [1]. We attempt to find out that active ceramic resources has effects on cutaneous wound healing. And furthermore, we investigate its related mechanism and expression of cytokines of cutaneous wound healing which is accelerated by active ceramic resources. (omitted)

  • Purpose: Many clinical trials have shown the effectiveness of platelet releasate on chronic wounds. However, a large volume of blood must be aspirated from a patient and a platelet separator is required. Here, we hypothesized that platelet concentrate obtained from a blood bank (PCBB) would be also effective at stimulating wound healing. The purpose of this study was to investigate the effectiveness of PCBB on accelerating healing of diabetic wounds in vivo. Methods: Round wounds of 5 mm diameter were made at four sites(two wounds on the left and two on the right side) on the backs of nine diabetic mice. Three hundred million platelets suspended in 0.05 ml fibrinogen were dispersed on each wound on left sides. Same amount of fibrinogen without platelets was dispersed on right side control wounds. Thereafter, 0.05 ml thrombin was applied to the each wound. Ten days after wound treatment, healed wounds were excised and the extent of wound healing in each group was compared. Results: Quantitative histologic analysis of epithelial gap distances revealed that PCBB treatment had greatly accelerated wound healing. Mean epithelial gap distances for PCBB treated and control wounds were $2.5{\times}0.6mm\;and\;3.6{\times}0.5mm$, respectively(p<0.05). Conclusion: Our results suggest that PCBB has potential to accelerate the healing of diabetic wounds.

혈소판 농축재제를 이용한 창상치유의 촉진 (Acceleration of Wound Healing Using Adipose-derived Stem Cell Therapy with Platelet Concentrates: Plateletrich Plasma (PRP) vs. Platelet-rich Fibrin (PRF))

  • Purpose: Although platelet-rich plasma (PRP) potentiate the wound healing activity of adipose-derived stem cells (ADSCs), its effect cannot be sustained for a prolonged period of time due to short duration of action. This led us to design and produce platelet-rich fibrin (PRF), in an effort to develop a tool which lasts longer, and apply it on wound healing. Methods: Two symmetrical skin defects were made on the back of seven nude mice. ADSCs were applied to each wound, combined with either PRP or PRF. The wound area was measured over 14 days. By day 16, the wound was harvested and histologic analysis was performed including counting of the blood vessel. Results: The healing rate was more accelerated in PRP group in the first 5 days (p<0.05). However, PRF group surpassed PRP group after 6 days (p<0.05). The average number of blood vessels observed in the PRF group was $6.53{\pm}0.51$, compared with $5.68{\pm}0.71$ for the PRP group. Conclusion: PRF exerts a slow yet pervasive influence over the two-week course of the wound healing process. Thus, PRF is probably more beneficial for promoting the activity of ADSCs for a sustained period of time.

The effect of Korean Red Ginseng on full-thickness skin wound healing in rats

  • Background: Panax ginseng is regarded as one of the best compounds for promoting health, and it has been used traditionally as a medicinal herb. Recently, Korean Red Ginseng (RG) has been shown to protect skin from aging and wrinkling; it can also relieve atopic dermatitis and allergy symptoms. This study aimed to evaluate RG's effects on the regeneration of the full-thickness skin wounds in rat. Methods: Full-thickness skin wounds were generated in rats, and then RG was administered either orally or topically. The wound-healing effects of RG were investigated by assessing wound size, mRNA expression patterns of genes related to wound healing, histological staining, and measurements of lipid, moisture, and elasticity in skin tissues. Results: The wound size was smaller, and tissue regeneration rate was faster in the RG-treated group than that in the control group on days 15 and 20 after initiating treatment. On postoperative day 20, skin lipid and moisture content had increased significantly in the RG-treated group. Significant increases in the gene expression levels of transforming growth $factor-{\beta}1$ and vascular endothelial growth factor were found in the RG group during the early stages of wound healing. Matrix metalloproteinase-1 and matrix metalloproteinase-9 showed significant increases in gene expression levels on day 20. Conclusion: The results suggested that RG may promote healing of full-thickness skin wounds in rats. They also provided basic insights into the effects of RG on skin regeneration, supporting its use as a dressing material for wound treatment and its development as a functional food.

세포 이동능력 분석을 위한 96-Well Plate 전용 Lab-Made Wound Maker (A Lab-Made Wound Maker for Analysis of Cell Migration in a 96-Well Plate)

  • Cell migration은 embryogenesis 혹은 cancer metastasis 이외에, 물리적 손상에 의한 상처의 수복을 위해서 손상된 부위로 세포가 이동하는 매우 흔하게 관찰되는 현상 중 하나 이다. Wound healing assay는 in vitro의 이차원 평면상에서 세포의 이동을 관찰할 수 있는 기본적인 연구 기법이다. In vitro상에서 물리적 손상을 재현하는 가장 손쉬운 접근법으로서, 세포의 confluent monolayer 표면에 날카로운 도구를 이용하여 기계적인 스크레치를 내는 방법이 사용되고 있다. 완충 스프링이 탑재된 금속 핀을 96-well plate를 기반으로 하는 wound maker에 장착하여 multi-well plate 바닥 표면의 고르지 못한 굴곡과 스크레칭 팁 사이에 직각을 이루는 접촉면에서의 미세한 조절이 가능하도록 하였다. 실험용 팁으로 confluent monolayer위에 스크래치를 내었을 때에는 다양한 지그재그 패턴이 그려진 반면에, 직접 제작한 wound maker에서는 동일한 형태의 선형 wounds가 fibroblast가 seeding된 96-well plate의 각 well의 중심부에 그려짐을 확인하였다. 상용화 되어있는 몇몇 multi-well plate가 본 실험에서 제작된 wound maker와 호환되는 것을 고려하여 보았을 때에, 실시간 wound healing을 관찰하는 high content screening (HCS)실험에 있어서의 활용적인 측면에서 기존의 전형적인 polypropylene 파이펫 팁을 이용한 스크래칭 방법보다 더욱 용이한 방법임을 알 수 있다.

New techniques for wound management: A systematic review of their role in the management of chronic wounds

  • Debridement is a crucial component of wound management. Recent technologies such as hydrosurgery (Versajet), ultrasound therapy (the MIST therapy device), or plasma-mediated bipolar radio-frequency ablation therapy (Coblation) seem to represent interesting alternatives for wound debridement. The purpose of this systematic review was to describe, evaluate, and compare these three recently developed methods for the management of chronic wounds. In January 2016, an electronic database search was conducted of MEDLINE, PubMed Central, and Embase for articles concerning these three innovative methods for the management of chronic wounds. A total of 389 references were identified by our search strategy, and 15 articles were included. We extracted data regarding the number and age of patients, indications, operating time, number of procedures, costs, wound healing time, decrease in exudation, perioperative blood loss, bacterial load, and the occurrence of complications. The 15 articles included studies that involved 563 patients who underwent hydrosurgery (7 studies), ultrasound therapy (6 studies), or Coblation (2 studies). Six randomized controlled trials were included that compared the use of a scalpel or curette to hydrosurgery (2 studies) or ultrasound therapy (6 studies). Hydrosurgery, in addition to being a very precise and selective tool, allows significantly faster debridement. Ultrasound therapy provides a significant reduction of exudation, and improves the wound healing time. No comparative study dedicated to Coblation was identified. Despite the obvious clinical interest of the topic, our review of the current literature revealed a lack of prospective randomized studies comparing these devices with each other or with standard techniques, particularly for Coblation and hydrosurgery.

  • Gorwth factors such as TGF-beta, PDGF and FGF are thought to play important roles in wound healing. However, thier biological activity and signal transduction during wound repair remain poorly understood. Growth factors are often ligands for receptor tyrosine kinase and receptor serine/threonine kinases. With recent advances in signal transduction by receptor kinases, we are beginning to understand the underlying mechanism of how growth factors may regulate cutaneous wound repair. In this paper, we will describe the pharmacological effects of growth factors on wound healing, and dscuss the potential underlying signaing mechanisms. thus, we hope to provide the basis for designing more specific therapeutics for wound healing in the near future.

In Vitro and in Vivo Wound Healing Properties of Plasma and Serum from Crocodylus siamensis Blood

  • The plasma and serum of Crocodylus siamensis have previously been reported to exhibit potent antimicrobial, antioxidant, and anti-inflammatory activities. During wound healing, these biological properties play a crucial role for supporting the formation of new tissue around the injured skin in the recovery process. Thus, this study aimed to evaluate the wound healing properties of C. siamensis plasma and serum. The collected data demonstrate that crocodile plasma and serum were able to activate in vitro proliferation and migration of HaCaT, a human keratinocyte cell line, which represents an essential phase in the wound healing process. With respect to investigating cell migration, a scratch wound experiment was performed which revealed the ability of plasma and serum to decrease the gap of wounds in a dose-dependent manner. Consistent with the in vitro results, remarkably enhanced wound repair was also observed in a mouse excisional skin wound model after treatment with plasma or serum. The effects of C. siamensis plasma and serum on wound healing were further elucidated by treating wound infections by Staphylococcus aureus ATCC 25923 on mice skin coupled with a histological method. The results indicate that crocodile plasma and serum promote the prevention of wound infection and boost the re-epithelialization necessary for the formation of new skin. Therefore, this work represents the first study to demonstrate the efficiency of C. siamensis plasma and serum with respect to their wound healing properties and strongly supports the utilization of C. siamensis plasma and serum as therapeutic products for injured skin treatment.

저에너지 레이저 조사가 창상치유에 미치는 영향 (The effect of low energy laser irradiation on wound healing)

  • This study was performed to determine the optimal dose of laser energy for wound healing. The skin wound with 8mm diameter was induced over the lumbar vertebrae of the rats, and wound squares, scab hardness, hematologic findings and histopathlogic findings according to irradiation of laser energy were studied. 1. Wound square was significantly reduced at Day 1 (p<0.01), 2 (p<0.01), 3 (p<0.05), 7 (p<0.01) and 8 (p<0.05), respectively in experimental groups, especially group II, compared with control group. 2. Scab hardness was significantly increased at Day 1 (p<0.01), 2 (p<0.01), 3 (p<0.05), 5 (p<0.01) and 7 (p<0.01), respectively in experimental groups, especially group II, compared with control group. 3. In hematological findings, red blood cells and white blood cells in experimental group were increased according to the lapse of days, but they were not significant. 4. In histopathologic findings, experimental groups, especially group II, revealed early scab formation, early appearance of phagocytes and fibroblast, rapid growth of granulation tissue and collagen, and promotion of wound healing in the result.

개에서 Allium cepa L.의 창상치유효과 (Wound Healing Effects of Allium Cepa L. in Dogs)

  • This study was performed to investigate the wound healing effects of Allium cepa L. on Staphylococcus intermedius contaminated wound. 4 full-thickness skin wounds ($2\;cm{\times}2\;cm$) were made in the back and Staphylococcus intermedius was inoculated in seven dogs. In each dog, the wounds were treated with onion extract (experimental group) and normal saline (control group), respectively. The wound contraction rate of the experimental group was higher than that of the control group during the experimental period. The occupation rate showed that the wounds of the experimental groups contracted more vigorously than that of the control group in the early period of wound healing. The mean value of tensile strength on the 22 day after wounding was $4,741{\pm}848\;g/cm^2$ in the experimental group and $2,400{\pm}397\;g/cm^2$ in the control group. The number of S. intermedius in the experimental groups was decreased more rapidly than that of the control group. These results suggest that application of activated onion extract in like full-thickness skin wounds promote wound healing.
