The Carcinogenic Liver Fluke Opisthorchis viverrini is a Reservoir for Species of Helicobacter

  • There has been a strong, positive correlation between opisthorchiasis-associated cholangiocarcinoma and infection with Helicobacter. Here a rodent model of human infection with Opisthorchis viverrini was utilized to further investigate relationships of apparent co-infections with O. viverrini and H. pylori. A total of 150 hamsters were assigned to five groups: i) Control hamsters not infected with O. viverrini; ii) O. viverrini-infected hamsters; iii) non-O. viverrini infected hamsters treated with antibiotics (ABx); iv) O. viverrini-infected hamsters treated with ABx; and v) O. viverrini-infected hamsters treated both with ABx and praziquantel (PZQ). Stomach, gallbladder, liver, colonic tissue, colorectal feces and O. viverrini worms were collected and the presence of species of Helicobacter determined by PCR-based approaches. In addition, O. viverrini worms were cultured in vitro with and without ABx for four weeks, after which the presence of Helicobacter spp. was determined. In situ localization of H. pylori and Helicobacter-like species was performed using a combination of histochemistry and immunohistochemistry. The prevalence of H. pylori infection in O. viverrini-infected hamsters was significantly higher than that of O. viverrini-uninfected hamsters ($p{\leq}0.001$). Interestingly, O. viverrini-infected hamsters treated with ABx and PZQ (to remove the flukes) had a significantly lower frequency of H. pylori than either O. viverr-iniinfected hamsters treated only with ABx or O. viverrini-infected hamsters, respectively ($p{\leq}0.001$). Quantitative RT-PCR strongly confirmed the correlation between intensity H. pylori infection and the presence of liver fluke infection. In vitro, H. pylori could be detected in the O. viverrini worms cultured with ABx over four weeks. In situ localization revealed H. pylori and other Helicobacter-like bacteria in worm gut. The findings indicate that the liver fluke O. viverrini in the biliary tree of the hamsters harbors H. pylori and Helicobacter-like bacteria. Accordingly, the association between O. viverrini and H. pylori may be an obligatory mutualism.

  • Morphological characters of section Acutae of the genus Carex (Cyperaceae) were reexamined. The epidermal patterns of perigynium, achene and leaf were investigated using by a scanning electron microscope (SEM) and a light microscope (LM). Morphological characters such as length and width of stem. leaf, bract, spike, scale, perigynium and achene, and shape of cross-sectioned stem, spike, scale, apex of scale, perigynium, beak of perigynium and achene, and epidermal pattern of perigynium, achene and leaf (shape of fundamental epidermal cell and cell wall, type of silica body, shape of beak epidermal cell and cell wall in perigynium, subsidiary cell shape. size and frequency of stomatal complex of leaf were useful for the identification of the observed 11 taxa. C. cinerascens, C. heterolepis and C. forficula have been confused due to similar morphological characters. C. cinerascens, C. heterolepis and C. forficula were distinct. however with respect to from width of leaf, shpae of scale, apex of scale, perigynium, beak of perigynium, and epidermal patterns of achene and leaf.

Effects of Feeding Earth Worm Meal on the Performance of Laying Hens and Fatty Acids composition in Egg Yolk (지렁이 분말의 급여가 산란계의 생산성 및 난황의 지방산 조성에 미치는 영향)

  • A study was conducted to investigate the effect of supplementing earthworm meal (EWM) on the performance of laying hens and fatty acid composition in egg yolks. A total of 360 laying hens at 55 weeks of age were fed the experimental diets containing 0.0% (Control), 0.1% and 0.2% of EWM for 5 weeks. Eggs were collected and weighted every day and egg production and feed conversion were recorded every weeks during the experimental period. However fatty acid composition of egg yolk were measured at last week of experimental period. Amount of feed intake tended to increase by supplemental EMW, but feed conversion ratio of birds fed EWM was not different among three groups. Average egg production seemed to increase and significantly improved (P<0.05) when fed a 0.1% EWM and 0.2% EWM, respectively. Average egg weight was prone to decrease when fed a 0.1% EWM compared to that fed a 0% (control) or 0.2% EWM. Average daily egg mass tended to improve by the addition of EWM. It was more increased in 0.2% EWM treatment than 0.1% EWM. The ratio of egg yolk n-6/ n-3 fatty acids contents was 5:1 fed a 0.1% and 0.2% EWM. But these ratio was 10:1 in control group. It is concluded that 0.2% earthworm meal supplementation in the 55 weeks old laying hens diet, improves the laying performance and ratio of egg yolk n-6/ n-3 fatty acids contents (P<0.05).

Growth and development of Metorchis orientalis in chicks and its adult morphology (Metorchis orientalis의 병아리내 성장발육 및 성충의 형태)

  • In order to observe the infectivity, growth and development and adult morphology of Metorchis orientalis, a total of 40 chicks were experimentally infected with 100 metacercariae respectively, collected from Pseuderasbora larva. The worms of various developmental stages were recovered from chicks at 1.5, 3, 5, 7, 9, 11, 1:k and 21 days after infection, and they were prepared for morphological observations and measurements. All of the worms were found in the gallbladders of chicks, and their recovery rate was 32% in average. The growth of the body was rapid from 9 to 11 days after infection. The genital primordia appeared in 1.5 and 3-day old worms, and ovary and testes were first observed in 5-day old worms. Thereafter, genital organs gradually matured and completed up to 11 days after infection. The adult worm was leaf-like, and possessed a con- voluted tubular seminal vesicle, an ovoid ovary, a sac-like seminal receptacle, 2 lobed-testes and follicular vitellaria. Eggs were $31.9{\times}15.3{\;}\mu\textrm{m}$ in average size, ellipsoid to elliptical in shape and possessed abopercular thickenings. From the above results, it is concluded that M. orientalis grows in sigmoid pattern in chicks, and their genital organs fully matul.e between days 9 and 11. It is also confirmed that a chick is a new definitive host of M. orientalis.

Epidemiological Studies on Ascaris lumbricoides Reinfection in Rural Communities in Korea II. Age-specific Reinfection Rates and Familial Aggregation of the Reinfected Cases (한국 농촌지역의 회충재감염에 대한 역학적 조사연구 ll. 연령별 재감염률 및 재감염의 가족집적성)

  • Epidemiological studies on the reinfetion pattern of Ascaris lumbricoides were undertaken by means of blanket mass chemotherapy and worm collection in a rural village in Korea, during 1977~1980. The study objectives were to determine the age (sex)-specific reinfection rate during 2, 4, 6 and 12 months through repeated mass chemotherapy with pyrantel pamoate, and to observe the familial aggregation tendency of the reinfected cases. The results obtained are as follows: 1. The age (sex)-reinfection curve revealed that the reinfection rate is much higher in younger individuals than in olders in all of 4 kinds of interval chemotherapy groups. The highest reinfection rate and the highest burden of reinfected worms were observed in preschool children, followed by primary school students. Such fluctuation in the age-specific reinfection rates was more pronounced in males than in females. 2. There was noted a significant tendency of familial aggregation among the reinfected cases. It is suggested that reinfection occurs never randomly but preferably to the members of certain household families. From these reinfection analyses, it is inferred that the principal mode of A. lumbricoides transmiSSIOn in the surveyed rural area is likely to be of 'dooryard type', in which case children and certain family members are more preferably reinfected. It is also suggested that the preschool children should be included in the primary targets of mass control programme.

On the Applicability of Partially Purified Antigenic Preparations of Paragonimus westermani (부분정제 폐흡충 항원의 유용성 검토)

  • In order to obtain more specific antigenic preparation for the diagnosis of human paragonlmiasis, crude saline extract of whole worm (=PwWWE), secretory.excretory components (PwSEC) and secretion-excretion-free somatic extract (PwSM) of 12 week-old ParagoninBus westermani were filtrated through Sephadex G-200 gel column. The adult Paragonimus worms were obtained from expevimentally infected doge. A total of 11 antigenic solutions was Iyophilised or diluted to adjust protein content of 1mg/ml. To evaluate the antigenicity of crude antigens and fractions, micro-ELISA was done with the sera from P westermani in(ected cases, C. sinensis infected cases and non-infected control cases to detect Paragonimus specific IgG antibody. The results were as follows: 1. When the PwWWE was filtrated through Sephadex G-200 gel, it was separated into three fractions; PwWWE Fr. 1, PwWWE Fr. 2 and PwWWE Fr. 3. The percentage of protein content was 28.0%, 21.6% and 50.4% respectively. The PwSM was also. separated into three fractions; PwSM Fr. 1, PwSM Fr. 2, PwSM Fr. 3. and their percentage of protein content was 41.3%, 38.6% and 20.1%. However, the PwSEC showed different fractionation pattern; i.e. fraction 1 (=PwSEC Fr.1) and 3 (PwSEC Fr. 3) without fraction 2. The percentage of protein content was 14.0% in PwSEC Fr. 1 and 86.0% in PwSEC Fr. 3. 2. When the antigenicity of each Paragonimus crude antigen and fractionated antigen was evaluated for specific IgG aritibody by micro-ELISA in 10 human paragonimiasis sera, PwSEC Fr, 1 was the most potent antigen showing the mean absorbance 1.98. The PwWWE Fr. 1, PwSEC, PwWWE were next to that: their mean absorbance were 1.72, 1.38 and 0.83 respectively. The antigenicity of fractions 2 and 3 was much weaker in binding specific IgG antibody. 3. When the antigens were reacted in micro-ELISA with 10 human clonorchiasis sera, most antigens showed weak reactivity. Each fraction 1 of crude antigens reacted higher than other fractions or crude antigens; the mean absorbance was 0.17 in fraction 1, but in others the absorbances were about 0.06. 4. With non-infected control sera, the result of micro-RLISA revealed almost same pattern with those of the clonorchiasis sera. From the above results, it became apparent that PwWWE Fr. 1, especially PwSEC Fr. 1 was the most potent antigen reacted with Paragonisfaus specific IgG antibody.

Studies on Intestinal Trematodes in Korea IX. Recovery Rate and development of Firbricola seoulensis in Experimental Animals (한국의 장흡충에 관한 연구 IX. 실험동물내에서의 Fibricola Seoulens양 충체위수률 및 발육)

  • An experimental study was carried out to observe the susceptibility of several kinds of laboratory animals to Fibricola seoulensis infection, a diplostomatid fluke of mammals. The metacercariae were obtained from the viscera of the snakes, Natrix tigrina lateralis and 50~2,000 in number each was artificially fed to a total of 127 animals; albino rats, mice, dogs, cats, rabbits and chickens. After 3 days to 8 weeks the animals were sacrificed and the recovery rate of worms as well as their maturity was observed. The results are as follows: 1. The overall worm recovery rates throughout the experimental period was highest in albino rats (40.0%) followed by mice (33.9%), cats (20.9%), dogs (11.4%), rabbits (0.05%) and chickens (0%). However, the recovery rates in the same host decreased as infection progressed longer and variable by the amount of metacercariae given. 2. From albino rats and mice, the highest recovery rates were obtained in 1,000 and 200-metacercariae infection groups respectively, and it is considered that such amount should be the optimum dose for experimental infection of these animals. 3. The main location of F. seoulensis in experimental animals was small intestine especially the duodenum. 4. The maturity index (No. mature worms/ No. examined) was 100% in albino rats and mice, while only 22.7% or 0% in dogs or cats respectively. From the results, it is concluded that albino rats and mice are the most susceptible hosts for F. seoulensis infection among six kinds of laboratory animals examined.

The Karyotype of Fischoedeyius cobboldi (Poirier, 1883) from Korean Cattle (한국산 코볼드쌍구흡충의 핵형 분석)

  • As a series of systematic classification of paramphistomes, the worms in the rumen and reticulum of 310 Korean cattle slaughtered at Chonju abattoir were collected from February 1986 to June 1987 and were classified by morphology of the worms. Afterwards, the karyotype of Fischoederius cobboldi (Poirier, 1883), which is a very rare species in Korean cattle, was studied with germ cells of the worm by means of modified air-drying method. The chromosome numbers in the haploid and diploid cells of 315 F. cobboldi were n=9 and 2n=18, respectively. The meiotic divisions were observed frequently; 1,904 haploid and 49 diploid cells were recognized. Nine pairs of mitotic chromosomes were homologous in the metaphase stage and the chromosomes were composed of seven medium-sized metacentrics (m) or submetacentrics (sm) and two small-sized submetacentrics (sm). While, meiotic metaphases were composed of seven medium and two small·sized chromosomes. The 3rd, 4th, 2nd and 5th pairs of chromosomes was metacentric having centromere indices of 40.4%, 40.0%, 39.7% and 38.9%, respectively, and the remaining ones were submetacentric with centromere indices from 32,4% to 36.2%. As a series of C-banding method, C-band was shown in centromeric region from all of the haploid germ cells, except chromosome No. 1 which included heterochromatin at the tip region. Chromosomes No, 4, 6 and 9 showed remarkable C-band distinguished from others.

C3H/He Mice as an Incompatible Cholangiocarcinoma Model by Clonorchis sinensis, Dicyclanil and N-Nitrosodimethylamine

  • Clonorchis sinensis is a Group-I bio-carcinogen, associated with cholangiocarcinoma (CCA). The hamster is the only experimental model of C. sinensis-mediated CCA, but we oblige another animal model. The present study intended to develop a C. sinensis (Cs) mediated CCA model using C3H/He mice, co-stimulated with N-nitrosodimethylamine (NDMA) and dicyclanil (DC). The mice were divided into 8 groups with different combinations of Cs, NDMA, and DC. Six months later the mice were sacrificed and subjected to gross and histopathological examination. The body weights were significantly reduced among the groups treated with 2 or more agents (eg. Cs+NDMA, Cs+DC, NDMA+DC, and Cs+NDMA+DC). In contrast, liver weight percentages to body weight were increased in above groups by 4.1% to 4.7%. A Change of the spleen weight was observed only in Cs+NDMA group. Though C. sinensis infection is evident from hyperplastic changes, only 1 worm was recovered. Two mice, 1 from Cs and the other from Cs+DC group, showed mass forming lesions; 1 ($281.2mm^3$) from the Cs group was a hepatocellular adenoma and the other ($280.6mm^3$) from the Cs+DC group was a cystic mass (peliosis). Higher prevalence of gray-white nodules was observed in Cs group (42.9%) followed by Cs+NDMA+DC group (21.4%). The mice of the Cs+NDMA+DC group showed hyper-proliferation of the bile duct with fibrotic changes. No characteristic change for CCA was recognized in any of the groups. In conclusion, C3H/He mice produce no CCA but extensive fibrosis when they are challenged by Cs, NDMA, and DC together.

Nasitrema attenuata (Digenia: Nasitrematidae) Infection of Long-beaked Common Dolphin (Delphius capensis) in the East Sea, Korea

  • The trematodes of Genus Nasitrema are the most common infection in the heads of small odontocetes. Five species of Nasitrema have been described in Asia. But those parasites has been not surveyed in the East Sea of Korean peninsula. There are no evidence of pathology associated with Nasitrema attenuate. To determine the infection ratio of N. attenuata in the dolphin, it was surveyed 9 long-beaked common dolphin, Delphinus capensis, caught incidentally from Samcheok to Pohang, East Sea, Korea. Five males and four females, estimated to be between 1 to 10 years old, were examined. The quantity of N. attenuata harvested by aspiration from pterygoid sinuses ranged from 10 to 100 with a 100% (9/9) infection with older dolphins exhibiting higher parasite counts. The body of trematoda was elongated with tegumental patches resembling the leaves of lilies and oral sucker located ventrally at the anterior terminal. Esophagus was absent and intestinal bifurcation arose immediately from the posterior of pharynx. The undulated intestinal caeca transversed the entire body and terminated at the posterior extremity of the worm. The genital pore opened anterior to the ventral sucker. This report confirms the prevalence and morphology of N. attenuata in the dolphins from the East Sea, Korea.
