Objective: Trimethylation of histone 3 (H3) at 4th lysine N-termini (H3K4me3) in gene promoter region was the universal marker of active genes specific to cell lineage. On the contrary, coexistence of trimethylation at 27th lysine (H3K27me3) in the same loci-the bivalent H3K4m3/H3K27me3 was known to suspend the gene transcription in germ cells, and could also be inherited to the developed stem cell. In galline species, throughout example of H3K4m3 and H3K27me3 ChIP-seq analysis was still not provided. We therefore designed and demonstrated such procedures using ChIP-seq and mRNA-seq data of chicken follicular mesenchymal cells and male germ cells. Methods: Analytical workflow was designed and provided in this study. ChIP-seq and RNA-seq datasets of follicular mesenchymal cells and male germ cells were acquired and properly preprocessed. Peak calling by Model-based analysis of ChIP-seq 2 was performed to identify H3K4m3 or H3K27me3 enriched regions ($Fold-change{\geq}2$, $FDR{\leq}0.01$) in gene promoter regions. Integrative genomics viewer was utilized for cellular retinoic acid binding protein 1 (CRABP1), growth differentiation factor 10 (GDF10), and gremlin 1 (GREM1) gene explorations. Results: The acquired results indicated that follicular mesenchymal cells and germ cells shared several unique gene promoter regions enriched with H3K4me3 (5,704 peaks) and also unique regions of bivalent H3K4m3/H3K27me3 shared between all cell types and germ cells (1,909 peaks). Subsequent observation of follicular mesenchyme-specific genes-CRABP1, GDF10, and GREM1 correctly revealed vigorous transcriptions of these genes in follicular mesenchymal cells. As expected, bivalent H3K4m3/H3K27me3 pattern was manifested in gene promoter regions of germ cells, and thus suspended their transcriptions. Conclusion: According the results, an example of chicken H3K4m3/H3K27me3 ChIP-seq data analysis was successfully demonstrated in this study. Hopefully, the provided methodology should hereby be useful for galline ChIP-seq data analysis in the future.
As the rapid extension of the game and film industry, studies on developing natural movements on a 3D characters are increasing. Rigging a character with joints is essential to create realistic movements on a 3D character. The rapid development of the CG industry, rigging technologies and workflow is becoming more sophisticated. Despite the progress and the growth of rigging operations, has shown the limitations of such repetitive tasks. For this study, analyzes the issues and inefficiency of the old method of skin weights and propose a better approach. First, need to understand the general process of an animation pipeline and learn the technology term of skin weights. Second, comparing the traditional ways of skinning a character and applying other deformers to work properly. Third, testing out new ways of weighting a character by applying deformers such as lattice and finally converting lattice weights back to skin weights. Forth, verifying effectiveness of the new method of skin weights by comparing with the traditional skin weighting process. The study shows that the new method of skin weights, reduced working hours and a better final weighting outcome. Expecting this study to enhance the method of skin weights and able to utilize this new skinning technology.
It has become possible for small scale laboratories to interpret large scale genomic DNA, thanks to the reduction of the sequencing cost by the development of next generation sequencing (NGS). De novo assembly is a method which creates a putative original sequence by reconstructing reads without using a reference sequence. There have been various study results on de novo assembly, however, it is still difficult to get the desired results even by using the same assembly procedures and the analysis tools which were suggested in the studies reported. This is mainly because there are no specific guidelines for the assembly procedures or know-hows for the use of such analysis tools. In this study, to resolve these problems, we introduce steps to finding whole genome of an unknown DNA via NGS technology and de novo assembly, while providing the pros and cons of the various analysis tools used in each step. We used 350Mbp of Toxocara canis DNA as an application case for the detailed explanations of each stated step. We also extend our works for prediction of protein-coding genes and their functions from the draft genome sequence by comparing its homology with reference sequences of other nematodes.

Quality management work at construction sites demands substantial time and effort, involving the preparation of documents in Excel, approval processes, and the execution of quality tests. These Excel-based tasks include printing quality test reports, performing quality tests, generating and approving test reports, and preparing management ledgers. This division of processes extends the duration of work and diminishes efficiency. Accordingly, a cloud-based construction site quality management system was developed to enhance the productivity of quality management work. The purpose of this study is to analyze the productivity of the construction site quality management system, which is in the early stages of implementation at construction sites. This study implemented the construction site quality management system at a road construction site and scrutinized subsequent alterations in the quality management workflow before and after the implementation. Additionally, a survey was conducted among quality control engineers to collect data on work time both before and after the introduction of the construction site quality management system. Based on the collected data, the Monte-Carlo simulation method was applied to analyze the productivity improvement effect of the construction site quality management system, and the results were presented. The results of this study can serve as foundational data for future research endeavors focused on the automation of quality management works.

A cold storage is a warehouse of a insulated building with cooling installations. It has many different types of cold rooms with temperatures below 0 degrees Celsius, and the sequential workflow such as receiving, picking and packing runs in that rooms. Recently, the cold storages have adopted RFID technology, and consequently, warehouse product management in them are becoming intelligent and network. However, information inconsistency in warehouses caused by physical and logical errors reduces reliability in the RFID cold storage management system and worsens their work efficiency. Therefore, it is necessary to develop an early detection system to identify errors. In this paper, we suggest a supervisory system detecting logical errors on business processes of the RFID cold storage. It is composed of a master supervisor and mobile supervisor. In the master supervisor, the manager can set the constraints conditions and get alerts, and in the mobile supervisor, the workers confirm and deal with these faults directly. The supervisory system improve reliability of the RFID cold storage management system by recognizing a failure to identify physically and logically using these constraint conditions. This paper shows that the supervisory system can reduce the average recovery time to improve reliability by decreasing the time for detecting and analyzing errors in the RFID cold storage management system.

3D printing is a process of producing 3d object from a digital file in STL format by joining, bonding, sintering or polymerizing small volume elements by layer. The various type of 3d printing is classified according to the additive manufacturing strategies. Among the types of 3D printer, SLA(StereoLithography Apparatus) and DLP(Digital Light Processing) 3D printer which use polymerization by light source are widely used in dental office. In the previous study, a full-arch scale 3d printed model is less precise than a conventional stone model. However, in scale of quadrant arch, a 3d printed model is significantly precise than a five-axis milled model. Using $3^{rd}$ Party dental CAD program, full denture, provisional crowns and diagnostic wax-up model are fabricated by 3d printer in dental office. In Orthodontics, based on virtual setup model, indirect bracket bonding tray can be generated by 3d printer. And thermoforming clear aligner can be fabricated on the 3d printed model. 3D printed individual drilling guide enable the clinician to place the dental implant on the proper position. The development of layer additive technology enhance the quality of 3d printing object and shorten the operating time of 3D printing. In the near future, traditional dental laboratory process such as casting, denture curing will be replaced by digital 3D printing.
Purpose. The purpose of this study was to evaluate the currently published literatures investigating the accuracy of computer-aided design and computer-aided manufacturing removable partial denture (CAD-CAM RPD) framework with different manufacturing techniques and methods. Materials and methods. A comprehensive search for literatures was conducted in PubMed database using specific keywords with the patient, intervention, comparison, and outcome (PICO) question, "Is there a difference in accuracy of RPD frameworks manufactured using digital workflow according to the manufacturing process and methods?" Results. A total of 7 articles were selected. Two studies compared intraoral scanning and laboratory scanning for RPD frameworks and had heterogenous results. In the studies using different manufacturing process, RPD frameworks had clinically acceptable accuracy in both subtractive and additive manufacturing. Polyetheretherketone (PEEK)-milled RPD frameworks showed higher fit accuracy than traditionally casted or 3D printed RPDs. Direct milling method showed a higher accuracy than indirect milling method. However, in rapid prototyping, indirect method showed higher accuracy than direct method. Conclusion. The RPD frameworks fabricated using CAD-CAM technology showed a clinically acceptable level of accuracy regardless of manufacturing process or techniques. Consistent results have not been reported regarding the digital impression methods, which were intra oral scanning or laboratory scanning, and further studies are needed.
A workflow is presented to estimate the size of a representative elementary volume and 3-D hydraulic conductivity tensor based on fluid flow analysis for a discrete fracture network (DFN). A case study is considered for a Cretaceous granitic rock mass at Gijang in Busan, Korea. The intensity and size of joints were calibrated using the first invariant of the fracture tensor for the 2-D DFN of the study area. Effective hydraulic apertures were obtained by analyzing the results of field packer tests. The representative elementary volume of the 2-D DFN was determined to be 20 m square by investigating the variations in the directional hydraulic conductivity for blocks of different sizes. The directional hydraulic conductivities calculated from the 2-D DFN exhibited strong anisotropy related to the hydraulic behavior of the study area. The 3-D hydraulic conductivity tensor for the fractured rock mass of the study area was estimated from the directional block conductivities of the 2-D DFN blocks generated for various directions in 3-D. The orientations of the principal components of the 3-D hydraulic conductivity tensor were found to be identical to those of delineated joint sets in the study area.

Even though MR can reveal excellent soft-tissue contrast and functional information, CT is also required for electron density information for accurate dose calculation in Radiotherapy. For the fusion of MRI and CT images in RT treatment planning workflow, patients are normally scanned on both MRI and CT imaging modalities. Recently deep-learning-based generations of CT images from MR images became possible owing to machine learning technology. This eliminated CT scanning work. This study implemented a CycleGan deep-learning-based CT image generation from MR images. Three CT generators whose learning is based on T1- , T2- , or T1-&T2-weighted MR images were created, respectively. We found that the T1-weighted MR image-based generator can generate better than other CT generators when T1-weighted MR images are input. In contrast, a T2-weighted MR image-based generator can generate better than other CT generators do when T2-weighted MR images are input. The results say that the CT generator from MR images is just outside the practical clinics and the specific weight MR image-based machine-learning generator can generate better CT images than other sequence MR image-based generators do.
