White Matter Damage and Hippocampal Neurodegeneration Induced by Permanent Bilateral Occlusion of Common Carotid Artery in the Rat: Comparison between Wistar and Sprague-Dawley Strain

  • In order to reproduce chronic cerebral hypoperfusion as it occurs in human aging and Alzheimer's disease, we introduced permanent, bilateral occlusion of the common carotid arteries (BCCAO) in rats (Farkas et al, 2007). Here, we induced BCCAO in two different rat strains in order to determine whether there was a strain difference in the pathogenic response to BCCAO. Male Wistar and Sprague-Dawley (SD) rats (250-270 g) were subjected to BCCAO for three weeks. Kluver-Barrera and cresyl violet staining were used to evaluate white matter and gray matter damage, respectively. Wistar rats had a considerably higher mortality rate (four of 14 rats) as compared to SD rats (one of 15 rats) following BCCAO. Complete loss of pupillary light reflex occurred in all Wistar rats that survived, but loss of pupillary light reflex did not occur at all in SD rats. Moreover, BCCAO induced marked vacuolation in the optic tract of Wistar rats as compared to SD rats. In contrast, SD rats showed fewer CA1 hippocampal neurons than Wistar rats following BCCAO. These results suggest that the neuropathological process induced by BCCAO takes place in a region-specific pattern that varies according to the strain of rat involved.

Chronic cerebral hypoperfusion and plasticity of the posterior cerebral artery following permanent bilateral common carotid artery occlusion

  • Vascular dementia (VaD) is a group of heterogeneous diseases with the common feature of cerebral hypoperfusion. To identify key factors contributing to VaD pathophysiology, we performed a detailed comparison of Wistar and Sprague-Dawley (SD) rats subjected to permanent bilateral common carotid artery occlusion (BCCAo). Eight-week old male Wistar and SD rats underwent BCCAo, followed by a reference memory test using a five-radial arm maze with tactile cues. Continuous monitoring of cerebral blood flow (CBF) was performed with a laser Doppler perfusion imaging (LDPI) system. A separate cohort of animals was sacrificed for evaluation of the brain vasculature and white matter damage after BCCAo. We found reference memory impairment in Wistar rats, but not in SD rats. Moreover, our LDPI system revealed that Wistar rats had significant hypoperfusion in the brain region supplied by the posterior cerebral artery (PCA). Furthermore, Wistar rats showed more profound CBF reduction in the forebrain region than did SD rats. Post-mortem analysis of brain vasculature demonstrated greater PCA plasticity at all time points after BCCAo in Wistar rats. Finally, we confirmed white matter rarefaction that was only observed in Wistar rats. Our studies show a comprehensive and dynamic CBF status after BCCAo in Wistar rats in addition to severe PCA dolichoectasia, which correlated well with white matter lesion and memory decline.

Polyacetylene 화합물들의 투여량에 따른 독성 (The Toxicological Parameter Assessment in Experimental Animals for Various Dosages of Polyacetylene Compounds)

  • 정상 wistar rat strain의 체중 및 장기무게 변화 그리고 사료, 물 섭취량의 변화 등을 조사함으로써 polyacetylene 화합물들의 투여량에 따른 독성변수(toxicological parameter)의 범위를 추정하고자 하였다. 또한, ICR mouse strain의 Sarcoma-180으로 유도되는 고형암의 억제와 관련해서 polyacetylene 화합물들의 투여량에 따른 독성변수(toxicological parameter)와의 관계를 관찰하고자 하였다. Panaxydol 40$\mumoles/kg b.w.을 3일간 연속 복강 투여했을때 wistar rat의 체중증가율은 대조군보다 약 17% 감소하였으나, polyacetylene 의 투여를 중지하면 약 4일 경과후부터 체중증가율이 정상으로 회복되었다. 이 투여량을 20$\mumoles/kg b.w.로 줄이면 대조군과의 체중증가율의 차이는 약 10% 정도로 완화되며 그 체중감소율은 panaxydol, panaxynol, panaxytriol의 순으로 증가하였다. 이때 장기무게의 관찰 결과는 대조군과 polyacetylene 화합물들 투여군 사이에 의미있는 차이가 없는 것으로 나타났다. 또한 정상 mice의 간세포에 대한 electron micrograph 관찰결과, panaxydol 40$\mumoles/kg b.w./day 투여시에도 대조군에 비하여 별다른 미세조직의 변화가 관찰되지 않았다.

Alterations of Antioxidant Status and Mitochondrial Succinate Dehydrogenase Activity in the Liver of Wistar Strain Albino Rats Treated with by Ethanol Extracts of Annona senegalensis Pers (Annonaceae) Stem Bark

  • Numerous ethnomedicinal uses have been attributed to different parts of Annona senegalensis (ASE), including its uses as food and food additives. The present study investigated toxicological and antioxidant effects of 28 days administration of ethanol extracts of ASE stem bark to Wistar strain albino rats. Acute toxicity test was done to determine lethal dose in Wistar rats while sub-acute toxicity test was conducted on rats divided into four groups (A - control, B - 50 mg/kg, C - 100 mg/kg, D - 150 mg/kg, respectively and treated for 28 days. Oxidative stress markers in liver and kidney as well as hepatic succinate dehydrogenase activity in the mitochondrial and post mitochondrial fractions (PMF) were evaluated. The $LD_{50}$ value of ASE was > 2,000 mg/kg. White blood cell counts gradually increased, but red blood cell counts and haematocrits level decreased significantly (p < 0.05) by about 50%. Liver enzymes in the serum and mitochondrial succinate dehydrogenase activity increased significantly (p < 0.05). Superoxide dismutase and catalase activities also increased in liver mitochondria and PMF while malondialdehyde (MDA) and reduced glutathione levels increased only in the PMF. Furthermore, only MDA levels increased significantly in the kidney after 28 days extract administration. Histopathological examination showed hepatic necrosis and no obvious signs of nephrotoxicity. Anona senegalensis is relatively safe, but prolonged ingestion could induce oxidative stress and impair ATP synthesis through the modulation of the activity of mitochondrial succinate dehydrogenase.

Digital Autoradiographic System을 이용한 선천성고혈압에서의 Muscarinic Cholinergic Receptor 분포 및 특성 (Distribution of the Muscarinic Cholinergic Receptors and Characterization in the Brain of Wistar Rats and Spontaneously Hypertensive Rats (SHR Strain) by Digital Autoradiography)

  • Using in vitro autoradiography with a digital autoradiography system and radioreceptor assay, the distribution and the binding characteristics of the muscarinic cholinergic receptors (mAChR) were studied in regions of rat brain. Radioreceptor assay revealed that mAChR could be measured with saturation binding assay in the brain and heart homogenates: No difference in Kd or Bmax of the brain or heart was found between the normal Wistar rats and SHR rats. Specific binding of $^3H$ quinuclidinyl benzilate (QNB) increased and saturation was reached by 2 hours after incubation with slide-mounted brain tissue. The distribution of mAChR was heterogeneous along the fields of brain. Affinity (Kd) of mAChR was not different significantly among cortex, hippocampus and caudate-putamen. No difference was found between normal rats and SHR strain. More receptors (Bmax) were found in the cortex and hippocampus than in the caudate-putamen in normal rats. More receptors were found in the cortex and caudate-putamen in SHR rats than in normal rats. Radioreceptor assay and digital autoradiographic analysis of affinity and number of mAChR gave the same results. With the above findings, we concluded that we could use digital autoradiographic system with $^3H$-QNB in the characterization of mAChR of rats and that the cortex and caudate-putamen of SHR strain rats have more receptors than those of normal rats.

소금 섭취량을 달리한 정상 및 고혈압쥐에서 Atrial Natriuretic Peptide가 혈압, Renin-Aldosterone 및 신배설에 미치는 영향 (Effect of Sodium Intake on Responses of Blood Pressure, Renin-Aldosterone and Renal Excretions to Atrial Natriuretic Peptide in Spontaneously Hypertensive Rats)

  • 장기적으로 소금 섭취량을 달리 한 정상 및 고혈압쥐에서 atrial natriurectic peptide (ANP) 가 혈압, renin-aldosterone 및 신배설에 미치는 효과를 비교하였다. 생후 6주된 spontaneously hyper-tensive rate (SHR)와 Wistar 숫쥐를 각각 저염, 정상염 및 고염 (2, 10, 25 mmol NaCl/100g diet) 군으로 나누어 실험 식이를 6주동안 먹었다. 실험 당일 아침에 쥐를 ether로 마취시킨 상태에서 대퇴 동맥과 정맥 및 방광에 catheter를 삽입한 다음에 쥐를 restraining cage에 넣어 고정시켰다. 마취가 깨고 혈압이 안정된 후에 정맥관으로 0.9% saline을 0.02ml/min의 속도로 80분간 주입하고 안정시 뇨와 혈액을 채취하였다. 그후 ANP를 380 ng/kg/min의 속도로 20분간 주입하였으며 그동안 10분 간격으로 요를 채취한 다음 채혈하였다. 혈장 aldosterone 농도와 renin 활성도 (PRA)를 방사면역법으로 측정하였다. ANP를 주입하기 전에 SHR 고염군의 평균동맥압은 정사염이나 저염군보다 유의하게 높았으나, Wistar의 혈압은 소금군 사이에서 차이가 없었다. ANP 주입전의 혈장 aldosterone 농도는 소금 섭취량이 많을수록 유의하게 낮았으며, Wistal 군이 SHR 보다 높은 값을 보였다. 혈장 renin 활성도는 소금군 간에 차이가 없었으며, Wistar와 SHR 간에도 차이가 없었다. 요량이나 Na, K 배설률은 소금군 간에 유의한 차이가 없었으나, SHR이 Wistar 보다 높은 경향을 보였다. Hematocrit 값도 소금군 간에 차이가 없었으나, SHR의 값이 Wistar 보다 유의하게 높았다. ANP를 주입하는 동안 혈압이 점진적으로 감소하여 20분후에는 $20{\sim}30\;mmHg$ 정도 감소하였으나, 각 소금군사이에 차이가 없었다. 그러나, ANP에 의한 혈압 강하 정도는 SHR이 Wistar 보다 유의하게 높았다. ANP 주입 후에는 모든 군에서 aldosterone과 혈장 renin 활성도가 유의하게 감소하였는데, aldosterone의 감소 정도는 저염군에서 가장 크게 나타났다. ANP의 이뇨 및 Na 배설 항진효과는 Wistar에서는 소금군 간에 차이가 없었으나, SHR 에서는 고염군의 Na 배설률이 저염군보다 유의하게 높았다. ANP의 혈압강하 효과나 Na 배설 항진효과가 SHR에서 Wistar 보다 유의하게 높게 나타났으나, 이뇨반응, renin 및 hematocrit의 변화에는 차이가 없었다. 이상의 결과에서, 장기적으로 소금 섭취량이 다름에 따라 ANP의 효과 중에서 특히 aldosterone 분비나 SHR의 Na 배설에 차이가 나타났는데 그 작용 기전은 확실치 않다.

  • There studies were conducted using inbred ICR mice to examine the sex of preimplantation mouse embryo. The morphological normality of mice embryos treated with the culture medium containing rat H-Y antiserum(10%, v/v) plus complement(20%,v/v) was observed and also the sexing of embryos was investigated by chromosomal analysis. The results obtained were summarized as follows: 1. The viability of preimplantation mouse embryos, which were incubated in vitro with different media condition, was scored 68.9-85.5% in control group. However, 151 embryos normally developed up to blastocyst and 160 embryos were retarded growth or destroyed out of total 311 embryos treated in the medium containing H-Y antiserum(10%, v/v) plus complement(20%,v/v). 2. H-Y antiserum was prepared from inb red rats (Wistar and Donryu strain) with different immunization times (4, 5 and 6th) to examine the specific titer of embryos by the number of immunization. Precentage of normally developed embryos incubated either in the medium containing the antiserum of Wistar plus complement or Donryu plus complement was revealed 50.9, 47.4 and 50.0% (4, 5 and 6th immunization and 47.8, 41.2 and 48.7%, respectively. 3. Twenty two females and five males were identified out of fourty-eight normally developed embryos incubated in the medium containing H-Y antiserum plus complement by chromosomal analysis.

  • After ablactation, wistar strain white male rats, weighing 270g and 340g, were fed with a diet of CLEA for three months. The whole daily excretion of each feces and urine were collected, and extracted with water($80^{\circ}C$ hot water). The combined extraction were filtered and the $B_2$ was determined with the parts of the filterates by the lumiflavin fluorometric method, and the FMN, FAD and FR with the rest of the filterates by paper chromatography. The following results were obtained; 1. $B_2$ contents in the feces were $27.52{\gamma}$ per 100 grams per body weight, and $83.93{\gamma}$ per each rat per day. 2. $B_2$ contents in the urine were $18.47{\gamma}$ per 100 grams per body weight, and $56.33{\gamma}$ per each rat per day. The total daily excretion of $B_2$ contents in the feces were 1. 5 times as much as in the urine. 3. Among the total daily $B_2$ excretion of one white wistar strain rat in the feces were the following ; FAD, 81.0% ; FMN, 14.9% ; FR, 3.3%. Therefore the order of the contents were FAD>FMN>FR.

Lactobacillus casei strain C1 attenuates vascular changes in spontaneously hypertensive rats

  • Hypertension can be caused by various factors while the predominant causes include increase in body fluid volume and resistance in the circulatory system that elevate the blood pressure. Consumption of probiotics has been proven to attenuate hypertension; however, the effect is much strain-dependent. In this study, a newly isolated Lactobacillus casei (Lb. casei ) strain C1 was investigated for its antihypertensive properties in spontaneously hypertensive rats (SHR). Lactic acid bacteria (LAB) suspension of 11 log colony-forming unit (CFU) was given to SHR (SHR+LAB, n=8), and phosphate buffer saline (PBS) was given as a control in SHR (SHR, n=8) and in Wistar rats as sham (WIS, n=8). The treatment was given via oral gavage for 8 weeks. The results showed that the weekly systolic blood pressure (SBP), mean arterial pressure (MAP), diastolic blood pressure (DBP) and aortic reactivity function were remarkably improved after 8 weeks of bacterial administration in SHR+LAB. These effects were mostly attributed by restoration of wall tension and tensile stress following the bacterial treatment. Although not statistically significant, the level of malondialdehye (MDA) in SHR+LAB serum was found declining. Increased levels of glutathione (GSH) and nitric oxide (NO) in SHR+LAB serum suggested that the bacterium exerted vascular protection through antioxidative functions and relatively high NO level that induced vasodilation. Collectively, Lb. casei strain C1 is a promising alternative for hypertension improvement.

랫트의 스트레인과 성 간의 간질에 대한 감수성의 차이 (Differences in susceptibility to infection with Fasciola hepatica between strains and sexes of the rat)

  • 랫트는 간질에 높은 감수성을 나타내기 때문에 간질의 연구에 실험실 모델로 많이 이용된다. 그러나 간질의 피낭유충을 랫트에 감염시킨 다음 랫트의 분변에서 충란을 검출해보면 충란의 검출률은 100%에 훨씬 못미쳐서 실험의 설계에 어려움을 겪는 경우가 흔하다. 이 연구에서는 우리나라에서 많이 사육되고 있는 4가지 스트레인(Sprague-Dawley, Fisher 344, the spontaneously hypertensive rats 그리고 Wistar-Kyoto)의 암 수 랫트에 간질의 피낭유충을 경구적으로 접종하고 분변검사와 부검을 실시하여 감염여부를 확인하였다. 충체회수율은 랫트의 성과 스트레인 간에 유의성 있는 차이를 보였는데 Sprague-Dawley계의 랫트가 가장 높은 충체회수율을 보였으며 평균 충체회수율은 암컷보다 수컷에서 높았다. 이 연구의 결과는 간질에 관한 연구 특히 간질구충제의 효능을 검정하는 시험에 유용한 지침이 될 것으로 사료된다.
