SCANNING ELECTRON MICROSCOPIC STUDY OF JUNCTIONAL EPITHELIUM ON THE TOOTH SURFACES IN PERIODONTAL POCKETS (치주낭내 치아표면에 부착된 치은상피세포의 주사전자 현미경적 연구)

  • The purpose of this study was designed to evaluate the existence of the junctional epithelia on the tooth surface in periodontal pocket, and what were the morphologic differences between the junctional epithelia on the healthy and advanced periodontitis tooth. Fifteen premolar teeth from patients of Yon Sei University, Dental Infirmary were selected for this study. After extration, the teeth were prepared and examined in Scanning electro microscope. The results were as follows. 1. The junctional epithelia from healthy tooth surface were irreguraly round, oval, polygonal and slightly elongated while those from periodontal pocket were so elongated that difficult to distinguished the individual cell boundary. 2. There were a lot of round space so called 'HOLE or WHORL' which seemed tunnel in periodontal pocket with advanced periodontitis. 3. Microvilli were going to destructed and disappeared on surfaces of junctional epithelia in periodontal pocket with advanced periodontitis. 4. There were a lot of Filopodia on Junctional epithelia from healthy surfaces. %. Junctional epithelia from periodontal pocket with advanced periodontitis contained more inflammatory cells than healthy junctional epithelia did.

Verticillium Wilt of Potato Caused by Verticillium albo-atrum in Daegwallyong Area in Korea

  • Verticillium wilt was first observed in 2001 on potatoes (Solanum tuberosum) cv. Superior at Daegwallyong area, one of the major seed potato producing areas in Korea. The wilted potato plants showed typical symptoms including gradual yellowing and interveinal necrosis. There was discoloration in the vascular tissues of the infected stems which turned light brown. Fungal isolates from discolored vascular tissues were whitish to creamy with folding on potato dextrose agar medium, where they used to produce resting dark mycelia but no micro-sclerotia. Conidiophores were septate with side branches, swelled at the base, and arranged in a whorl. Conidia were 2.5-11.2$\times$2.0-4.5 $\mu\textrm{m}$ um in size and were borne in small clusters at the tips of phialides. Optimal temperature range for mycelial growth was $25-30^{\circ}C$. Based on these cultural and morphological characteristics, the fungus was identified as Verticillium albo-atrum Reink & Berth. Pathogenicity tests by root dipping method revealed that the fungus caused the same symptoms as observed in naturally infected potato plants. This is the first report of Verticillium wilt on potato caused by Verticillium albo-atrum in Korea.

Morphological Characteristics of Conidiogenesis in Cordyceps militaris

  • Conidial development of Cordyceps militaris was observed from germinating ascospores and vegetative hyphae through light and scanning electron microscopy (SEM). Ascospores were discharged from fresh specimens of C. militaris in sterile water as well as Sabouraud Dextrose agar plus Yeast Extract (SDAY) plates. We observed ascospore germination and conidial formation periodically. Under submerged condition in sterile water, most part-spores germinated unidirectionally and conidia were developed directly from the tips of germinating hyphae of part-spores within 36 h after ascospore discharge, showing microcyclic conidiation. First-formed conidia were cylindrical or clavate followed by globose and ellipsoidal ones. Germination of ascospores and conidial development were observed on SDAY agar by SEM. Slimy heads of conidia on variously arranged phialides, from solitary to whorl, developed 5 days after ascospore discharge. Besides, two distinct types of conidia, elongated pyriform or cylindrical and globose, were observed in the same slimy heads by SEM. Conidia were shown to be uninucleate with 4,6-diamidino-2-phenylindole staining. Conidiogenous cells were more slender than vegetative hyphae, having attenuated tips. Microcyclic conidiation, undifferentiated conidiogenous hyphae (phialides), polymorphic conidia and solitary, opposite to whorled type of phialidic arrangement are reported here as the characteristic features of asexual stage of C. militaris, which can be distinguished from other Cordyceps species.

Study of a Tobacco MADS-Box Gene Triggering Flower Formation

  • Recently, we have reported a rice MADS-box gene, OsMADS1, as a molecular factor triggering flower formation; this has been well studied in a heterologous system (Chung et al., 1994). In order to study whether the OsMADS1 homolog exists in other plant species, the OsMADS1 cDNA was used as a probe to screen a tobacco cDNA library, and a potential homolog, NtMADS3, was isolated. Sequence analysis revealed that the gene shares 56.1% identity in whole amino acids with OsMADS1. Like OsMADS1, the NtMADS3 gene starts to express at a very early stage of flower development, and the expression continues up to flower maturation. In the tobacco flower, the gene is expressed in whorl 2,3 and 4, corresponding to the petal, stamen, and carpel, respectively. Upon ectopic expression in the homologous system, NtMADS3 caused a trasition from inflorescence shoot meristem into floral meristem, reducing flowering time dramatically. These phenotypes strongly suggest the NtMADS3 gene is the OsMADS1 homolog of tobacco. Hybrids between the OsMADS1 and the NtMADS3 plants were also generated. The hybrids flowered even earlier than these two transgenic plants. The detailed studies are discussed here.

A Directional Feature Extraction Method of Each Region for the Classification of Fingerprint Images with Various Shapes (다양한 형태의 지문 이미지 분류를 위한 영역별 방향특징 추출 방법)

  • In this paper, we propose a new approach to extract directional features based on directional patterns of each region in fingerprint images. The proposed approach computes the center of gravity to extract features from fingerprint images of various shapes. According to it, we divide a fingerprint image into four regions and compute the directional values of each region. To extract directional features of each region from a fingerprint image, we spilt direction values of ridges in a region into 18 classes and compute frequency distribution of each region. Through the result of our experiment using FVC2002 DB database acquired by electronic devices, we show that directional features are effectively extracted from various fingerprint images of exceptional inputs which lost all or part of singularities. To verify the performance of the proposed approach, we explained the process to model Arch, Left, Right and Whorl class using the extracted directional features of four regions and analyzed the classification result.

  • With the advent of hemodialysis, the success of renal transplants in the 1960s and the wide use of continuous ambulatory peritoneal dialysis at the end of the 1970s, children with renal failure now enjoy an extended life span. As a result, several children experience renal osteodystrophy and growth retardation. Renal osteodystrophy is induced by phosphorus retention, hypocalcemia, low vitamin D levels and hyperparathyroidism. The pharmacologic interventions are used to prevent bone deformities and to normalize growth velocity. But surgical intervention is required sometimes whorl osteodystrophy is severe and poorly controlled. We report an eight-year-old boy with ctironic renal failure who developed severe bone deformities and needed osteotomy.

Morphological analysis of Tripylina stramenti(Nematoda: Enoplida: Trischistomatidae) firstly recorded in Korea

  • The genus Tripylina Brzeski, 1963 is a group of terrestrial nematodes that inhabit soil and semi-wet biomes, most of which have been reported from Europe, Asia, America, Africa, and New Zealand. Tripylina stramenti (Yeates, 1971) Tsalolikhin, 1983, belonging to the family Trischistomatidae Andrassy, 2007, is newly discovered from Korea. The specimens were collected from the edge of the valley of Ulleung-gun, Gyeongsangbuk-do, South Korea. Tripylina stramenti described herein shows typical morphological characters of the genus Tripylina, including six and four setae in a single whorl, present dorsal tooth and subventral teeth, prodelphic reproductive system of female, absence post-uterine sac, narrow sickle-shaped spicule of male and generally S-shaped tail. The specimens differ in their body width (from the New Zealand populations), outer labial setae length, cuticles thickness(from the West African populations), and nerve ring position (from Indian populations). This species is distinguished from other Tripylina species by its relatively large body, two anterior and subventral teeth, and single cervical seta. In this study, morphological characteristics and morphometric information of the Korean T. stramenti isolate are described, illustrated, and compared with the same species from other geographic origins.

Morphological Studies on the Epithelium of the Midgut of the Nymph and the adult in the Grasshopper, Oxya sinuosa (우리벼메뚜기 유충과 성충의 중장 점막 상피에 관한 형태학적 연구)

  • In order to know the morphological characteristics and changes according to the developmental stage, the comparative observations have been studied by light and electron microscope on the midgut(gastric caecum, stomach) of the grasshopper(Oxya sinuosa Mistshenko). The results obtained are summarized as follow : In light microscopic level, the cuboidal shaped cells of the gastric caecum in the 5th instar nymph are differentiated the columnar epithelium in the adult. A number of pigment granules are appeared in the gastric caecum of the 5th instar nymph, however the pigment granules were absent in the adult. Indistinct or undifferentiated folds of the epithelial layer were appeared in the 5th instar nymph, whereas the well-developed folds were appeared the gastric caecum in the adult. The well-developed muscular layers are seen in the 5th instar numph, however in the adult the muscular layers are appeared thin or a few layers. In electron microscopic level, in the midgut epithelium, a number of well-developed rER, a few lipid droplets, multi vesicular bodies, small vesicles and glycogen granules were found. Two types of the pigment granules in the gastric caecum of the 5th instar nymph were appeared, whereas the whorl-membrane, a few secretory granules and one type of the gastro-entero-endocrine cell were found in the adult midgut. The light and dark cells could be distinguished in the stomach epithelim of the 5th instar nymph.

Occurrence of Bluish Green Mold of Pleurotus eryngii by Penicillium corylophilum (Penicillium corylophilum에 의한 큰느타리버섯(Pleurotus eryngii) 푸른곰팡이병의 발생)

  • Bluish green mold of Pleurotus eryngii caused by Penicillium was observed in Seungju on Feburary 4, 1999. The causal fungus was identified as Penicillium corylophilum based on its cultural and morphplogical characteristics. Conidiophores one-stage branched, terminating in a whorl of $2{\sim}4$ metulae. Metulae mostly $10{\sim}18{\times}2.5{\sim}3.0\;{\mu}m$. Phialides flask-shaped, $9{\sim}13{\times}2.2{\sim}2.8\;{\mu}m$. Conidia subglobose, $2.5{\sim}3.2{\times}2.5{\sim}3.0\;{\mu}m$. Colonies on Czapek agar mostly attaining a diameter of 21 mm within 7 days at $25^{\circ}C$. Colour blue-green soon becoming grey-green. Pathogenicity test by wound inoculation revealed that Penicillium corylophilum caused the same symptoms as observed in the field at $6{\sim}7$ days after inoculation.

A Study on the Measurement of New Concept for the Contact Force between Rail and Wheel (신개념의 레일.차륜간 접촉력 측정에 관한 연구)

  • The derailment is defined as phenomena in which the wheels run off the rail due to inordinate lateral force generated when wheel flange contacts with the rail. Derailment coefficient is typical standard assessing running safety and derailment. The traditional method measuring by strain gage adhered to wheels is very complicated and easy to fail. It also requires too much cost and higher measurement technique. Therefore it can hardly ensure safety because we can't confirm at which time we need to identify safety. In this paper, we principally researched the method measuring easily wheel load generated by contacts between wheel flange and the rail, and lateral force. Correlation of vibration and displacement which was related physical amounts of wheel load and lateral force, was investigated and analyzed through analysis, experiment and measurement. And it is presents new measurement method of derailment coefficient which can estimate derailment possibility only by movement of vibration and displacement, by which we understand the rate for acceleration and displacement to contribute wheel load and lateral force and compare actual data of wheel load and lateral force measured from wheel.
