The Effect of Fermented Diet with Whole Crop Barley Silage on Fecal Shape and Odorous Compound Concentration from Feces in Pregnant Sows (청보리 사일리지 첨가 발효사료가 임신돈의 분 성상 및 악취물질 농도에 미치는 효과)

  • The study was conducted to investigate the effect of feeding fermented diet including whole crop barley silage on the odor reduction and microbial population change in feces, and the constipation prevention of pregnant sows. The concentration of phenol was not detected in tapioca, beet pulp, wheat bran and lupin seeds, while that of p-cresol was ranged between 9.62 and 52.11 mg/L showing that lupin was highest and tapioca was lowest. It was determined that tapioca and beet pulp were useful feed ingredients to reduce odor due to their lower contents of phenol and indole compounds. Ten pregnant sows were allocated to control group and fermented diet group in 5 sows in each group. They were fed 3.0 kg DM/d of diets for 28 days. Feces was examined and showed that the feces from the fermented diet group was observed with the higher moisture content and the lower hardness than that of the control diet group and the population of E. coli was decreased and the population of lactobacilus was higher than that of the control diet group. The concentrations of p-cresol and skatole were lower than the detection levels at 33% and 67% among the samples of feces of the control group and at 67% and 100% among the samples of the feces of fermented diet group respectively. Thus it is expected that the odor from the feces of pregnant sows fed the fermented diet could be reduced compared with that of control group. Therefore, it is suggested that feeding fermented whole barley diet to pregnant sows improve the function of intestine and reduce the rate of occurrences of constipation and odor levels.

Cryopreservation of in vitro Grown Shoot Tips of Korean Potato Varieties by Droplet-vitrification

  • Potatoes are the world's 4th major food crop after maize, rice, and wheat and also are a staple food for 1.3 billion people. Due to their wide adaptability to various environmental conditions, their yeild capacity, and high commercial value, potatoes have contributed to global food security. Many potato germplasms are commonly preserved as whole plants in fields or in storage to maintain their particular genetic combinations. However, field maintenance is expensive and has the risk of potential losses from diseases, pests, plant ageing and climate change. Over the past four decades, meaningful efforts have been made toward the safe long-term conservation of potatoes through cryopreservation methods such as droplet-vitrification. In this study, we tested 4 Korean potato varieties('Golden Egg', 'Golden Ball', 'Ja-Young' and 'Ha-Ryeong') with the modified potato droplet -vitrification protocol. Potato shoot tips are precultured in a sucrose-enriched medium(0.3 and 0.7M for 7 and 17hrs, respectively) and submitted to a loading step with C4 solution for osmoprotection. The treated explants were dehydrated with Plant Vitrification Solution(PVS)2 which is 80% A3 solution in ice for 30 minutes. Thawing and unloading steps were performed with 0.8M sucrose solution for 30 sec(40℃) followed by 30min(25℃, room temperature). In a potato post-culture medium(MS+0.1 mg·L-1 GA3+0.1 mg·L-1 kinetin), we obtained a survival rates of post-thawed explants ranging 16.1-82.2%. The results suggest that modified and optimized protocols are required dependinig on every cultivar, genetic and ecological types. To achieve higher survival and regeneration rates, each step within the cryoprocedure must be carefully optimized.

Meridic Diets for Rearing of Spodoptera frugiperda Larvae (열대거세미나방 유충 사육을 위한 반합성 인공사료)

  • Two meridic diets, N4 and N6, containing pinto bean, wheat germ, soybean, whole milk and sucrose as main nutrients were developed for rearing Spodoptera frugiperda (Noctuidae) larvae. Six larval instars were observed when neonate larvae were individually raised on these diets in small petri-dishes (ø 50 × 10 mm, 19.6 ㎤) at 25℃ and 15:9 h (light:dark) photoperiod. The average pupation rate of 97.8% on the N4 diet was significantly higher than the rate of 85.6% on N6 diet. The emergence rates were 92.0% on N4 diet and 93.5% on N6 diet, with a non-significant difference. The larval periods were 17.9 and 17.7 days for females, and 18.7 and 18.5 days for males, for N4 and N6 diets, respectively, with non-significant differences between diets and sexes. The pupal periods on both diets were identical (11.1 days for females and 12.8 days for males), despite differences between sexes. The pupal weights of both sexes on N4 diet were identical with a value of 257 mg, whereas those on N6 diet were 256 and 263 mg for females and males, respectively, with a non-significant difference. The longevity of female adults that emerged on N6 diet was 13.8 days and longer than 8.6 days on N4 diet. The pre-oviposition periods were 5.0 and 4.2 days on the N4 and N6 diets, respectively, with a non-significant difference, however, the oviposition period of 6.5 days on N6 diet was longer than the period of 3.9 days on N4 diet. The effective fecundity on N6 diet was 1,392 eggs (maximum 1,776) and was higher than that of 942 eggs (maximum 1,694) on N4 diet, with a non-significant difference. The egg hatching rates on N4 and N6 diets were 79.2 and 79.8%, and egg periods were 3.0 and 2.9 days, respectively, with non-significant differences.

  • To determine the optimum harvesting date with good grain yield and quality, six different harvesting dates were tested from 25 to 50 days heading with five days intervals, using two varieties of Baekdong, naked barley and Olmil, wheat. The dates when grain yield, 1,000 grain weight and grain/whole spike weight ratio were reached to the highest were 35 and 40 days after heading for Baekdong and Olmil, respectively. Various grain qualities such as unhulled grain, plumpness, protein content and whiteness of grain were also very good at that times. One day earlier harvesting before the optimum dates resulted in yield decreases by 3.9% for Baekdong and 4.4% for Olmil. And it might be delayed for each 5 days with Combine harvesting.

Protein Fractionation of Whole Crop Silages, and Effect of Borate-phosphate Buffer Extraction on In vitro Fermentation Characteristics, Gas Production and Degradation (사료작물 사일리지의 단백질 분획 및 Borate-phosphate Buffer 추출이 In vitro 발효성상, Gas 발생 그리고 분해율에 미치는 효과)

  • Protein fractionation was evaluated from whole crop silages of rye (RS), wheat (WS), triticale (TS), oat (OS), barley (BS), and rice straw silage (RSS), and in vitro trial was carried out to examine the effect of silage and extraction of soluble protein on fermentation characteristics, total gas production and degradation. Soluble protein of silages was extracted with borate-phosphate buffer, and fermentation characteristics, gas production and degradation of silages were estimated by incubating anaerobically the mixed solution of strained rumen fluid and artificial saliva (1:1, v/v) containing dried and ground silages placed in nylon bag at $39^{\circ}C$ up to 48h. Soluble protein (SP) content was lowest for RSS as 2.11% in total CP compared to those for other silages. Highest A fraction (NPN) was observed from RS (74.33% of total CP) while those from TS and RSS were relatively low (48%). B2 fraction was relatively higher for RS, RSS and WS than for TS and BS. $B_3$ fraction was lowest in WS among silages. C fraction (27.07) in RSS was higher than in other silages (1.40~9.93%). pH in incubation solution was increased (P<0.01~P<0.001) for extracted silages up to 12h but decreased (P<0.01) at 48h for non-extracted ones. Contents of ammonia-N (P<0.001) and total VFA (P<0.01~P<0.001) were higher for non-extracted silages than for extracted ones. Acetate proportion was increased (P<0.001) in buffer extracted silages while those of propionate and butyrate were decreased (P<0.001) up to 24h incubation. Increased (P<0.001) total gas production was obtained from non-extracted silages up to 12h while gas production was increased (P<0.01) in extracted ones thereafter. In vitro degradation of dry matter and CP was increased (P<0.001) in non-extracted silages but that of neutral detergent fiber was increased (P<0.001) in extracted ones without difference among silages. Difference in mean values of degradability for each silage prior to- and post extraction with borate buffer, however, was not found among silages. It may be concluded that high NPN content of silages may reduce the protein availability in silages and borate buffer soluble components in silages can stimulate the early stage of fermentation.

Global Warming on Double Cropping in North Korea (지구온난화에 따른 북한의 두벌농사 특성 평가)

  • In order to evaluate the benefits of global warming on the double cropping with staple crops in North-Korea, four aspects such as the increasing rate of air temperature, the wintering temperatures for winter crops, the causing temperature of cool injury to rice and the securing of accumulated temperature for the double cropping in the different agricultural climate zones were analyzed by comparing the differences between the past 22 years from 1973 to 1994 and the recent 5 years from 2002 to 2006. The warming rate in recent daily mean air temperature of $8.96^{\circ}C$ in North Korea was higher by $0.64^{\circ}C$ than that in the past with large regional variations ranging from $1.06^{\circ}C$ in Samjiyeon of northern inland semi-alpine zone to $12.26^{\circ}C$ in Jangjeon of east central coastal zone. With the accumulated temperatures of more than $3,150^{\circ}C$ and $2,650^{\circ}C$, it was possible to apply the double cropping patterns with winter wheat and for cropping patterns with spring potato, respectively, to the whole region except for the northern inland semi-alpine zone. However, the wintering temperature higher than $-15^{\circ}C$ of average daily minimum air temperature of January, cropping patterns were impossible to northern inland semi-alpine zone and most regions of the northern mountainous zone. The days passed by below $17^{\circ}C$ in daily mean air temperature, causing the spikelet sterility at meiotic stage of rice in July, were a lot recorded from 21 to 29 days in northern inland semi-alpine zone and from 2 to 10 days in east-northern coastal zone, respectively. Therefore, a reasonable utilization of heat / temperature resources would relieve the limiting factors in double cropping for stable production of staple crops in North-Korea.
