Water Soluble Cyclosporine Monomethoxy Poly(ethyleneglycol) Conjugates as Potential Prod rugs

  • The highly water-soluble monomethoxypoly(ethyleneglycol) (mPEG) prod rugs of cyciosporin A(CsA) were synthesized. These prod rugs were prepared by initially preparing intermediate in the form of carbonate at the 3'-positions of CsA with chloromethyl chloroformate, in the pres-ence of a base to provide a 3'-carbonated CsA intermediate. Reaction of the CsA intermediate with mPEG derivative in the presence of a base provides the desired water-soluble prod rugs. As a model, we chose molecular weight 5 kDa mPEG in the reaction with CsA to give water soluble prodrugs. To prove that the prod rug is decomposed in the body to produce CsA, the enzymatic hydrolysis test was conducted using human liver homogenate at $37^{\circ}C$. The prodrug was decomposed in human liver homogenate to produce the active material, CsA, and the hydrolysis half-life ($t_{1/2}$) of the prodrug, KI-306 was 2.2 minutes at $37^{\circ}C$. However, a demon-stration of non-enzymatic conversion in pH 7.4 phosphate buffer was provided by the fact that the half-life ($t_{1/2}$) is 21 hours at 37$^{\circ}C$. The hydrolysis test in rat whole blood was also conducted. The hydrolysis was seen with half-life ($t_{1/2}$) of about 9.9, 65.0, 14.2, 3.4, 2.1 9.5, and 1.6 minutes for KI-306, 309, 312, 313, 315, 316, and 317, respectively. This is the ideal for CsA prodrug. The pharmacokinetic study of the prodrug, KI-306, in comparison to the commer-cial product (Sandimmune Neoral Solution) was also carried out after single oral dose. Each rat received 7 mg/kg of CsA equivalent dose. Especially, the prodrug KI-306 exhibits higher AUC and $C_{max}$ than the conventional Neoral. The AUC and $C_{max}$ were increased nearly 1.5 fold. The kinetic value was also seen with $T_{max}$ of about 1.43 and 2.44 hours for KI-306 and Neoral, respectively.

  • This paper proposes an attitude control system to keep the balance for a two-wheeled mobile manipulator which consists of a mobile platform and a three D.O.F. manipulator. In the conventional control scheme, complicated dynamics of the manipulator need to be derived for balancing control of a mobile manipulator. The method proposed in this paper, however, three links are considered as one body of mass and the dynamics are derived easily by using an inverted pendulum model. One of the best advantage of a sliding mode controller is low sensitivity to plant parameter variations and disturbances, which eliminates the necessity of exact modeling to control the system. Therefore the sliding mode control algorithm has been adopted in this research for the attitude control of mobile platform along the pitch axis. The center of gravity for the whole mobile manipulator is changing depending on the motion of the manipulator. And the orientation variation of center of gravity is used as reference input for the sliding mode controller of the pitch axis to maintain the center of gravity in the middle of robot to keep the balance for the robot. To confirm the performance of controller, MATLAB Simulink has been used and the resulting algorithms are applied to a real robot to demonstrate the superiority of the proposed attitude control.

Acute Radiation Syndrome in an Irradiated Minipig Model for Patients with Radiation Exposure

  • Background: Acute radiation syndrome (ARS) primarily refers to damage to the hematopoietic system, myeloid system, and gastrointestinal (GI) system caused by radiation exposure. Such damage progresses to become life-threatening. In particular, as the syndrome develops very rapidly-within several hours from radiation exposure-prompt and accurate diagnosis and treatment are needed, as is further research into appropriate diagnostic and treatment modalities. Materials and Methods: Minipigs, which display human-like properties, underwent whole-body irradiation at 2 or 4 Gy (doses causing hematopoietic ARS) or at higher doses of 7 or 12 Gy. Changes in the blood cells and clinical symptoms were analyzed and we performed a necropsy when the animals succumbed to ARS. Results and Discussion: The minipig irradiated with 2 Gy showed a decrease in white blood cells, including neutrophils, lymphocytes, and platelets in the early stages. However, the blood cell counts gradually increased and returned to normal values. The minipig irradiated with 4 Gy succumbed due to hematopoietic ARS. In contrast, the minipigs irradiated with 7 or 12 Gy exhibited clinical symptoms of combined GI damage and hematopoietic syndrome. Moreover, a characteristic pattern of platelet changes was observed in the 7 and 12 Gy irradiated minipigs. Conclusion: The changes in the platelet count caused by radiation exposure observed in minipigs, which are hematologically and pathohistologically similar to humans, suggest that they can be used as a novel diagnostic criterion.

  • Recently, there have been a lot of researches on the whole face replacement system, but it is not easy to obtain stable results due to various attitudes, angles and facial diversity. To produce a natural synthesis result when replacing the face shown in the video image, technologies such as face area detection, feature extraction, face alignment, face area segmentation, 3D attitude adjustment and facial transposition should all operate at a precise level. And each technology must be able to be interdependently combined. The results of our analysis show that the difficulty of implementing the technology and contribution to the system in facial replacement technology has increased in facial feature point extraction and facial alignment technology. On the other hand, the difficulty of the facial transposition technique and the three-dimensional posture adjustment technique were low, but showed the need for development. In this paper, we propose four facial replacement models such as 2-D Faceswap, OpenPose, Deekfake, and Cycle GAN, which are suitable for the Cox platform. These models have the following features; i.e. these models include a suitable model for front face pose image conversion, face pose image with active body movement, and face movement with right and left side by 15 degrees, Generative Adversarial Network.

  • Animal models have been used to elucidate the pathophysiology of varying diseases and to provide insight into potential targets for therapeutic intervention. Although alternatives to animal testing have been proposed to help overcome potential drawbacks related to animal experiments and avoid ethical issues, their use remains vital for the testing of new drug candidates and to identify the most effective strategies for therapeutic intervention. Particularly, the study of metabolic diseases requires the use of animal models to monitor whole-body physiology. In line with this, the National Institute of Food and Drug Safety Evaluation (NIFDS) in Korea has established their own animal strains to help evaluate both efficacy and safety during new drug development. The objective of this study was to characterize the response of C57BL/6NKorl mice from the NIFDS compared with that of other mice originating from the USA and Japan in a chemical-induced diabetic condition. Multiple low-dose treatments with streptozotocin were used to generate a type-1 diabetic animal model which is closely linked to the known clinical pathology of this disease. There were no significantly different responses observed between the varying streptozotocin-induced type-1 diabetic models tested in this study. When comparing control and diabetic mice, increases in liver weight and disturbances in serum amino acids levels of diabetic mice were most remarkable. Although the relationship between type-1 diabetes and BCAA has not been elucidated in this study, the results, which reveal a characteristic increase in diabetic mice of all origins are considered worthy of further study.

Brown preadipocyte transplantation locally ameliorates obesity

  • Background Brown adipose tissue (BAT) is a potential target for anti-obesity treatments. Previous studies have shown that BAT activation causes an acute metabolic boost and reduces adiposity. Furthermore, BAT and BAT-derived cell transplantation reportedly help treat obesity by regulating glucose and fatty acid metabolism. However, since BAT transplantation leads to whole-body weight loss, we speculated that earlier approaches cause a generalized and unnecessary fat tissue loss, including in breast and hip tissues. Methods We transplanted white adipose tissue-derived or BAT-derived preadipocytes prepared from C57BL/6 mice into one side of the inguinal fat pads of an obese mouse model (db/db mice) to examine whether it would cause fat loss at the peri-transplant site (n=5 each). The same volume of phosphate-buffered saline was injected as a control on the other side. Six weeks after transplantation, the inguinal fat pad was excised and weighed. We also measured the concentrations of glucose, triglycerides, fatty acids, and total cholesterol in the peripheral blood. Results BAT-derived preadipocytes showed abundant mitochondria and high levels of mitochondrial membrane uncoupling protein 1 expression, both in vivo and in vitro, with a remarkable reduction in weight of the inguinal fat pad after transplantation (0.17±0.12 g, P=0.043). Only free fatty acid levels tended to decrease in the BAT-transplanted group, but the difference was not significant (P=0.11). Conclusions Our results suggest that brown adipocytes drive fat degradation around the transplantation site. Thus, local transplantation of BAT-derived preadipocytes may be useful for treating obesity, as well as in cosmetic treatments.

ALARA 개념(槪念)에 의한 기체상방사성물질(氣體狀放射性物質)의 환경방출한도(環境放出限度) 설정(設定) (Establishment of Release Limits for Airborne Effluent into the Environment Based on ALARA Concept)

  • 농축인자법(濃縮因子法)(Concentration Factor Method)을 이용(利用)하여 결정피폭경로(決定被曝經路)를 분석(分析)한 후(後) 표준원전(標準原電)의 기체상방사성대기방출물(氣體狀放射性大氣放出物)에 대한 유도방출한도(誘導放出限度)(Derived Release Limits, DRL's)를 계산(計算)하였다. 이 방출한도(放出限度)는 핵시설(核施設) 주변(周邊)의 결정군구성개인(決定群構成個人)에 대(對)한 방사선(放射線) 피폭(被曝)을 관련피폭한도이하(關聯被曝限度以下)로 유지(維持)시키는 양(量)이다 본(本) 연구(硏究)에서는 1985년(年) 초(初) 미국(美國)의 환경보호청(環境保護廳)(EPA)에서 새로 권고(勸告)한 피폭한도(被曝限度)를 채택(採擇)하여 계산(計算)을 수행(遂行)하였다. 유도방출한도(誘導放出限度)(DRL)의 계산(計算)은 미국(美國)의 원자력(原子力) 규제위원회(規制委員會)(USNRC)가 규제지침(規制指針)(Reg. Guide) 1.109에서 제시(提示)하고 있는 선양평가(線量評價)모델을 표준(標準)모델로 사용(使用)하여 수행(遂行)하였으나, 동(同) 모델의 피폭경로분석(被曝經路分析)에서 우유(牛乳) 및 육류(肉類)의 섭취경로(攝取經路)는 국내(國內)의 현실상(現實狀) 무시가능(無視可能)한 것으로 고려(考慮)하여 본(本) 연구(硏究)에서 제외(除外)시켰다. 계산(計算)에서 고려(考慮)한 방출선원항(放出線源項)은 희유기체(稀有氣體), 요오드, 입자상원소(粒子狀元素) 및 삼중수소기체(三重水素氣體)였으며, 방출원(放出源)에서 북(北)쪽으로 1.3 km 거리에 위치(位置)하고 있는 주민영구거주지역(住民永久居住地域)을 대상(對象)으로 계산(計算)을 수행(遂行)하였다. 본(本) 연구(硏究)에서는 표준원전(標準原電)의 대상(對象)으로 고리원전(古里原電) 1호기(號機)를 선정(選定)하여 동원전(同原電)의 연간방출(年間放出)에 대(對)한 유도한도(誘導限度)를 계산(計算)하였으며, 1982년도(年度)의 실방출률(實放出率)과 비교(比較) 검토(檢討)하였다. 검토결과(檢討結果), 고리원전(古里原電) 1호기(號機)의 1982년도(年度)의 실방출률(實放出率)은 본(本) 연구(硏究)에서 구(求)해진 유도방출한도(誘導放出限度)보다 낮았으며, 방출물(放出物)에 의한 시설주변(施設周邊) 개인(個人)의 년간피폭선량(年間被曝線量)은 EPA에서 권고(勸告)하는 피폭한도이하(被曝限度以下)로 유지(維持)되었음을 알 수 있었다. 또한 본(本) 연구(硏究)에서 가장 결정적(決定的)인 피폭경로(被曝經路)는Co-60와 Cs-137과 같은 입자상침적방사핵종(粒子狀沈積放射核種)으로 오염(汚染)된 토양(土壤)에 의한 전신외부(全身外部) 피폭경로(被曝經路)였다.

$[^{51}Cr]Cr(III)$-EDTA 착물 합성 및 $[^{51}Cr]Cr(III)$-EDTA 주사후 두경부 방사능 계측에 의한 사구체 여과율 측정 (Synthesis of $[^{51}Cr]Cr(III)$-EDTA Complex and Measurement of Glomerular Filtration Rate br Radioactivity Counting of Head and Neck Region)

  • 원자력병원에 설치된 MC-50 싸이클로트론에서 생산된 무담체 $^{51}Cr$-EDTA의 합성법 및 정상 성인 남자자원자를 대상으로 무담체 $^{51}Cr$-EDTA 주사후 사구체여과율을 측정하고, 이 검사 중 피검자가 받는 방사능 흡수선량을 MIRD system의 계산법으로 계산하여 다음과 같은 결과를 얻었다. 1) $^{51}Cr$-EDTA는 bicarbonate 촉매하에 합성하였으며 이 방법은 기존의 방법보다 짧은 시간내에, 상온에서 반응이 완결되는 장점이 있으며 이 연구를 통하여 국내에서도 비방사능이 월등히 높은 무담체 $^{51}Cr$-EDTA의 사용이 가능하게 되었다. 2) GFR 측정시 $^{51}Cr$-EDTA 주사후 두경부 방사능 계측으로 혈액채취를 대신 할 수 있고, 경험적 공식이 아닌 2-compartment model에 의한 계산법을 이용할 수 있어 간편하게 임상이용이 가능하며 한국인의 체형에 맞는 새로운 공식의 유도가 가능할 것으로 기대된다. 3) $50{\mu}Ci$ $^{51}Cr$-EDTA의 전신방사능 흡수선량은 무시할 수 있는 정도임을 알 수 있었으며, MIRD system은 아주 적은 방사능 오염의 피폭 선량측정에 유용함을 알 수 있었다.

국내 여성용 인대 사용 실태 및 만족도에 관한 연구 (A Study on the Actual Conditions of and Satisfaction with the Existed Female Dress Forms Usage)

  • 콘크리트포장에 초기균열을 일으키는 중요한 인자 중 하나는 콘크리트 내부의 초기온도이다. 따라서 콘크리트포장의 초기균열 발생원인을 연구하기 위해서는 초기온도를 계측하여 분석하는 일이 필요하다. 본 논문에서는 초기균열이 발생하는 슬래브 장소와 초기균열의 발생시간이 초기온도패턴에 어떤 영향을 받는지를 검증하였고 더불어, 줄눈부에서 발생하는 균열의 발생시점과 시공시간과의 관계도 알아보았다. 본 논문을 위해서 "중부내륙고속도로 여주-충주간 제 1공구 시험도로 건설공사구간 STATION 1+400$\sim$1+700" 지점에서 시험시공이 이루어졌으며, 시공 후 72시간 동안 i-Button(온도계측센서)을 이용하여 온도계측을 시행하였으며, 초기균열의 거동은 Demec gauge를 사용하였으며, 초기균열 및 줄눈부 균열은 육안으로 확인하였다. 초기온도패턴과 초기균열의 분석 결과, 콘크리트의 초기온도패턴은 슬래브에 초기균열이 발생하는 위치와 시각에 영향을 주는 것으로 나타났다 초기균열균열은 온도낙차폭이 가장 큰 슬래브에서 발생하였으며, 그 시각은 슬래브의 온도가 급강하하는 새벽이었다. 또한, 콘크리트 슬래브의 거동이 인근 줄눈부에 발생한 초기균열에 따라 영향을 받으며. 줄눈부에 발생한 균열의 발생시기가 서로 다를 경우에 균열의 거동이 달라질 수 있다는 가능성이 제시되었다. 그 외에도, 오전에 시공한 슬래브에서의 균열 발생률이 오후에 시공한것보다 더 큰 것으로 나타났으며, 균열의 발생 간격이 큰 균열이 그렇지 않은 균열보다 더 큰 균열틈을 보였다.

Three Dimensional Measurement of Ideal Trajectory of Pedicle Screws of Subaxial Cervical Spine Using the Algorithm Could Be Applied for Robotic Screw Insertion

  • Objective : To define optimal method that calculate the safe direction of cervical pedicle screw placement using computed tomography (CT) image based three dimensional (3D) cortical shell model of human cervical spine. Methods : Cortical shell model of cervical spine from C3 to C6 was made after segmentation of in vivo CT image data of 44 volunteers. Three dimensional Cartesian coordinate of all points constituting surface of whole vertebra, bilateral pedicle and posterior wall were acquired. The ideal trajectory of pedicle screw insertion was defined as viewing direction at which the inner area of pedicle become largest when we see through the biconcave tubular pedicle. The ideal trajectory of 352 pedicles (eight pedicles for each of 44 subjects) were calculated using custom made program and were changed from global coordinate to local coordinate according to the three dimensional position of posterior wall of each vertebral body. The transverse and sagittal angle of trajectory were defined as the angle between ideal trajectory line and perpendicular line of posterior wall in the horizontal and sagittal plane. The averages and standard deviations of all measurements were calculated. Results : The average transverse angles were $50.60^{\circ}{\pm}6.22^{\circ}$ at C3, $51.42^{\circ}{\pm}7.44^{\circ}$ at C4, $47.79^{\circ}{\pm}7.61^{\circ}$ at C5, and $41.24^{\circ}{\pm}7.76^{\circ}$ at C6. The transverse angle becomes more steep from C3 to C6. The mean sagittal angles were $9.72^{\circ}{\pm}6.73^{\circ}$ downward at C3, $5.09^{\circ}{\pm}6.39^{\circ}$ downward at C4, $0.08^{\circ}{\pm}6.06^{\circ}$ downward at C5, and $1.67^{\circ}{\pm}6.06^{\circ}$ upward at C6. The sagittal angle changes from caudad to cephalad from C3 to C6. Conclusion : The absolute values of transverse and sagittal angle in our study were not same but the trend of changes were similar to previous studies. Because we know 3D address of all points constituting cortical shell of cervical vertebrae. we can easily reconstruct 3D model and manage it freely using computer program. More creative measurement of morphological characteristics could be carried out than direct inspection of raw bone. Furthermore this concept of measurement could be used for the computing program of automated robotic screw insertion.
