본 연구에서는 알긴산올리고당의 일부 방사선 방어효과를 규명하기 위해 마우스에 3 Gy 선량의 방사선을 1회 전신 조사하여 방사선 유도 nitric oxide를 측정하였다. 측정결과 방사선조사 대조군에서 nitric oxide 생성이 하강된 반면 방사선조사 전 3일간 처치군 및 방사선조사 후 3일간 처치군에서 유의한 생성 증가를 보였고, 특히 방사선조사 전 3일간 처치군에서 가장 큰 차이를 보였다(P<0.001). 결론적으로, 항산화효과가 탁월한 알긴산올라고당은 nitric oxide의 생성 증가를 촉진시킴으로써 일부 방사선 방어작용을 수행한다는 사실을 규명하였고, 화학적 독성이 적은 자연산생물이 방사선방어제로 활용될 수 있음을 확인하였다.
1. Within experimental chromatin, the total protein: DNA ratio did not vary in the same organs of control and irradiated rats. However, the amount of RNA and total protein associated with the DNA varied considerably among the different types of chromatin. In particular, the content of chromatin was the control tissue. RNA and total protein ratio of chromatins from brtain, liver, testis and spleen declined with experimental I organs. 2. There was the same quantitative relationship between the amount of RNA and the amount of histone-protein associated with DNA in chromatin. 3. RNA: DNA ratio of chromatin showed 1.5-2 times increas in the irradiated organs except brain. However, RNA: DNA ratio was decreased in chromatin by irradiation. 4. Histone-protein:residual protein ratio was greatly varied among the organs. However, the effect was not found by irradiation. 5. Priming activity of chromatin showed a higher value in testis and the activity was greater in organs with higher metabolic activity: 6. Inhibition of Actinomycin D is observable in chromatin from testis, liver, spleen and brain declined without relationship between irradiated and non-irradiated conditions. Ammonium sulfate showed increased priming activity by the electrostatic dissociation of DNA and histone in chromatin on the stimulation depending on property of chromatins. 7. It is suggested that the results support a proposal that testis and spleen of highly sensitive to irradiation should an increase in the priming activity whereas brain and liver of lower sensitivity decreased in the activity.
In the event a population of animals is exposed to ionizing radiation, proper disposition of animals will minimize loss to the farmer and protect the public from unwholesome meat and dairy products. Clinical response is an important factor in considering salvage of animals for food following exposure to ionizing radiation whether the dose is lethal or not. It is the purpose of this report to present the discussions of blood picture and growth of y ung growing rabbits exposed to cobalt-60 gamma-irradiation. The experimental animals were Hyrnalayan rabbits of 65 days old. The body weight for all animals of the time of irradiation was approximately 450g. The 96 rabbits used in the experiment were allotted t groups of eight. Rabbits in one group served as controls and the others were exposed to single doses of 100, 200, 300, 400, 500, 600 and 700 Roentgens (R). The exposure does rate averaged 15 R per minute. Central blood was obtained by heart puncture. Erythrocyte and leukocyte enumerations and, determinations of hemoglobin in blood and hematocrit value were made in the usual manner. Erythrocyte counts, concentrations of hemoglobin in blood, and hematocrit readings dropped to the lowest value between 1 and 4 weeks postirradiation, returning towards the control values. But recovery was not complete at the time of the last determination on 6 weeks postirradiation. There was a progressive decrease with increasing radiation does in the above values. Mean corpuscular volume, mean corpuscular hemoglobin and mean corpuscular hemoglobin concentration were not affected by the irradiation. After irradiation of each does the numbers of total leukocyte and the absolute numbers of various types of white cell began to decrease rapidly, and reached a minimum after 2 weeks, returning towards normal levels. But recovery was not complete at the time of the last determination on 6 weeks postirradiation. The reductions in total leukocyte and differential counts as the radiation does increases were apparent. Relative neutrophilia was observed in each irradiated group because of more expense of lymphocytes after irradiation. Growth rate of the rabbits was not affected by 100 R of whloe-body irradiation, while rabbits irradiated with 200, 300, 400 and 500 R showed marked depression of growth rate. As the radiation does increases, a depression of growth rate was apparent. The rabbits exposed to 600 and 700 R ceased from growing and recovery was not recognized during the experimental period.
거대핵세포(megakaryocyte)에서 유래된 혈소판은 백혈구와 더불어 방사선에 민감하여 가장 먼저 감소하므로 방사선장해의 지표로 활용된다. 따라서 혈소판이 부족하게 되면 점상출혈(dot hemorrhage)이 계속되고 혈구 수의 감소와 면역기능 저하로 이어진다. 특히, 전신조사(whole body irradiation)로 4~6 Gy를 받을 경우, 3주 후 혈소판이 최하로 감소되어 출혈과 빈혈에 의한 조혈기계 장해의 사망원인이 된다. 따라서 본 연구는 혈소판 장해의 메카니즘을 밝히고 방어효과를 알아보고자 하였다. 실험에 사용한 방어물질은 마늘 성분의 알리인(Alliin)을 사용하였으며, rat의 꼬리 정맥에 투여한 후 전자현미경(TEM)으로 관찰하였다. 연구 결과, 5 Gy 방사선조사(Irradiation) 후 10일 경과된 혈소판 조직에서는 세포막이 심하게 손상되어 풍선모양으로 부풀어 올랐으며, 당질층(glycocalyx)이 비후되었다. 조사 후 20일 경과된 혈소판에서는 소기관들의 형태를 알아볼 수 없을 정도로 손상되었으며, 세포질의 내용물이 간질부위로 유출되어 손상이 매우 심하였다. 또한 과립들의 형태도 관찰되지 않았다. 30일 경과된 혈소판에서는 가운데 부분에서 공포(vacuole)가 생기고 조직의 손상이 관찰되었다. 하지만 Alliin을 정맥 투여한 실험군에서는 과립(granules)들의 형태가 대조군과 같이 일정하게 유지되고 있었으며, 세포막의 손상도 확인되지 않았다. 따라서 Alliin의 혈관투여시 혈소판의 방사선방어 효과를 확인할 수 있었다.
본 연구는 감마선 조사 후 원시반포의 퇴화과정에서 나타나는 형태학적인 변화를 알아보기 위하여 시행되었다. ICR 계통의 생후 3 주된 미성숙 생쥐에 $LD_{80(30)}$의 선량인 8.3 Gy를 2 시간 동안 전신조사하였다. 방사선 조사 후 0 시간, 3 시간, 6 시간, 그리고 12 시간에 난소를 적출하여 조직절편을 제작한 후 가장 큰 절편을 관찰하였다. 정상 대 퇴화 난포의 비율은 조사 후 6 시간에 유의하게 감소하였다. 6 시간에 퇴화된 원시난포의 수가 증가하였으며, germinal vesicle이 관찰되지 않거나 지질방울이 증가하였으며, 난막이 사라진 것을 알 수 있었다. 과립세포의 모양은 둥글게 변하였으며, 세포자연사하는 세포가 관찰되었다. 대조군의 경우 정상 대 퇴화 원시난포의 비율이 62.50% 이었으나, 이 비율은 0 시간, 3 시간, 6 시간, 12 시간에 각각 51.61 %, 48.97 %, 11.11 %, 7.14 %로 감소하였다. 결론적으로 방사선에 의해서 윈시난포에 퇴화가 유발되는데 그 양상은 세 가지 유형을 나타내었다. 과립세포의 모양은 정상적이나 난자의 세포자연사, 난자의 형태는 정상적이나 한 개 혹은 그 이상의 과립세포가 세포자연사하는 경우, 혹은 난자와 과립세포 모두가 세포자연사 과정을 겪는 경우를 확인할 수 있었다. 본 연구를 통해 원시난포 퇴화를 형태학적으로 규명할 수 있는 단서가 제시되었다.
The efficacy of the radioprotective effect of Callophyllis japonica ethyl acetate (CJEA) extract was studied by comparing it to that of amifostine, a well-known radioprotective agent, and by evaluating the dose reduction factor, an indicator of radioprotective efficacy. Pretreatment with CJEA extract (100 mg/kg body weight) prior to receiving 12 Gy irradiation significantly improved the survival of jejunal crypts at 3.5 day post-irradiation, but attenuated the level of malondialdehyde compared to vehicle alone (P < 0.01). A similar gastroprotective effect was also obtained in the amifostine-treated irradiated group (P < 0.01). The efficacy of the radioprotective effect was further confirmed by the dose reduction factor, 1.41. Collectively, these results suggest that CJEA extract is a useful radioprotectant whose efficacy is similar to that of amifostine and whose radioprotective mechanism is in part the reduction of lipid peroxidation caused by gamma irradiation.
Six week-old ICR mice were divided into four groups including NC, RC, RR and RS, then they were injected with sun ginseng (RS), red ginseng (RR) and saline (RC) intraperitoneally as an amount of 60mg/kg body weight at 1 hour, 12 hours and 36 hours before the whole body irradiation with 6.5Gy of X-ray. And the mice were sacrificed at nine days after the irradiation. The RS group were significant increase in the weight of spleen (p<0.05) and the numbers of endogenous spleen colony (p<0.001) and WBC (p<0.05) especially neutrophils, lymphocytes and monocytes in comparison with the RC group. The RR group were significant increase in the numbers of endogenous spleen colony (p<0.01) and WBC (p<0.05) especially neutrophils, lymphocytes and eosinophils in comparison with the RC group. The RS group exhibited a more increase in the weight of spleen and the number of endogenous spleen colony than the RR group. The values of ALT (alanine transaminase) and AST (aspartate transaminase) were significantly elevated (p<0.05) by radiation and they were significantly decreased (p<0.05) in the RS group to the values of the NC group. The value of TBARS(thiobarbituric acid reactive substance) was significantly elevated (p<0.01) by radiation and it was significantly decreased (p<0.05) in the RS group. And the values of SOD (superoxide dismutase) and CAT (catalase) were significantly increased (p<0.05) in the RS group in comparison with the RC group. Taken together the above results, sun ginseng demonstrated the protective effects on hepatocytes and immune and hematopoietic cells in mice exposed to medium dose of irradiation, and those radioprotective effects were a little higher or similar to the red ginseng.

Mercury, one of the most diffused and hazardous organ-specific environmental contaminants, exists in a wide variety of physical and chemical states. Although the reports indicate that the mercury induces a deleterious damage, little has been known from the investigations of its effects in living organisms. The purpose of this study is to evaluate the effects of mercury chloride and ionizing radiation. Prepubertal male F344 rats were administered mercury chloride in drinking water throughout the experimental period. Two weeks after whole body irradiation, organs were collected to analyze the induced injury. Serum levels of GOT, GPT, ALP, and LDH were checked in the experimental groups and the hematological analysis was accomplished in plasma. In conclusion, the target organ of mercury chloride seems to be urinary organs and the pattern of damage induced by mercury differs from that by irradiation.

This study deals with the radiation protection effect of the pretreatment of $\beta$-carotene and combination with selenium on the DNA damage in mice after whole body ${\gamma}$-irradiation. This was obtained the radioprotective effect by evaluation of DNA damage levels in mice spleen and blood after irradiation. Six-week-old ICR male mice were administrated with $\beta$-carotene and combination with selenium orally once a day for 5 days and then irradiated with 8.0 Gy of $\gamma$-ray at a dose rate of 1.0 Gy/min. A 59-year-old male patient had 5-month history of gait disturbance and memory impairment. His initial brain computed tomography scan showed $3.5{\times}2.8cm$ sized mass with high density in the pineal region. The tumor was hypointense on T2 weighted magnetic resonance images and hyperintense on T1 weighted magnetic resonance images with heterogenous enhancement of central portion. The tumor was totally removed via the occipital transtentorial approach. Black mass was observed in the operation field, and after surgery, histopathological examination confirmed the diagnosis of malignant melanoma. Whole spine magnetic resonance images and whole body 18-fluoro-deoxyglucose positron emission tomography could not demonstrate the primary site of this melanoma. Scrupulous physical examination of his skin and mucosa was done and dark pigmented lesion on his left leg was found, but additional studies including magnetic resonance images and skin biopsy showed negative finding. As a result, final diagnosis of primary pineal malignant melanoma was made. He underwent treatment with the whole brain radiotherapy and extended local boost irradiation without chemotherapy. His preoperative symptoms were disappeared, and no other specific neurological deficits were founded. His follow-up image studies showed no recurrence or distant metastasis until 26 weeks after surgery. Primary pineal malignant melanomas are extremely rare intracranial tumors, and only 17 cases have been reported since 1899. The most recent case report showed favorable outcome by subtotal tumor resection followed by whole brain and extended local irradiation without chemotherapy. Our case is another result to prove that total tumor resection with radiotherapy can be the current optimal treatment for primary malignant melanoma in the pineal region.
