전도성 액체기반 정상표면탄성파(CL-SSAW)를 이용한 백혈구로부터의 말라리아 기생충 분리 (Malaria Parasite Separation from White Blood Cells Using Conductive Liquid-Based Standing Surface Acoustic Wave (CL-SSAW))

  • An acoustofluidic device using conductive liquid-based electrodes was developed for malaria parasite separation from white blood cells. In this device, the electrode channels filled with a conductive liquid were used to generate standing surface acoustic waves (SSAWs) in a fluidic channel, which can overcome the limitation of conventional patterned metal electrodes. Separation performance of the device was evaluated using fluorescent polystyrene particles with two different sizes (2 and $10{\mu}m$ diameters), which were successfully separated. In addition, a mixture of malaria parasites and white blood cells were also efficiently separated with high purity of ~98% in the CL-SSAW device at the flow rate of $12{\mu}l/min$.

  • Mansam, the root of Codonopsis pilosula (Campanulaceae) has a action on blood metabolism. It has been known to possess anti leukemic effect by increasing of red blood cell, at same time, by decreasing white blood cell, with this connection, present study is aimed to clarify such potential anti leukenic effect by testing ethanol extract and chloroform extract against murine tumor model, lymphocytic leukemia P388. The data indicated that these extracts appeared to be inactive against this tumor line.

  • It has been observed that white loght can suppress both cell growth and protein secretion in Baker's yeast. This effect was explored in batch liquid fermentations. Possible applications of this phenomenon are (a) use as a tool for pre-concentrating excreted enzymes prior to subsequent purification and (b) an engineering variable for regulation yeast fermentations.

Aconitine 및 가열처리한 Aconitine이 Mouse의 면역반응에 미치는 영향 (Effect of Aconitine and Heated Aconitine on the Immune Response in Mice)

  • Dose-dependent, immune modulatory effects of aconitine and heated aconitine were studied in mice. Mice, administered aconitine and heated aconitine intraperitoneally every other day for 4 weeks, were sensitized and challenged with sheep red blood cells. Serum antibody titer, foot pad swelling and rosette forming cell number were measured to evaluate hurmoral and cell mediated immune responses. The results show that Humoral immune response was suppressed by aconitine 0.05 mg/kg and heated aconitine but increased by aconitine 0.10 mg/kg administration. Cell mediated immune response was suppressed in all groups. Especially heated aconitine administration significantly suppressed the cell mediated immune response. The number of peripheral circulating white blood cell was reduced by aconitine but was not affected by heated aconitine.

siMacro: A Fast and Easy Data Processing Tool for Cell-Based Genomewide siRNA Screens

  • Growing numbers of studies employ cell line-based systematic short interfering RNA (siRNA) screens to study gene functions and to identify drug targets. As multiple sources of variations that are unique to siRNA screens exist, there is a growing demand for a computational tool that generates normalized values and standardized scores. However, only a few tools have been available so far with limited usability. Here, we present siMacro, a fast and easy-to-use Microsoft Office Excel-based tool with a graphic user interface, designed to process single-condition or two-condition synthetic screen datasets. siMacro normalizes position and batch effects, censors outlier samples, and calculates Z-scores and robust Z-scores, with a spreadsheet output of >120,000 samples in under 1 minute.

Quercetin이 마우스의 면역반응에 미치는 영향 (Effects of Quercetin on the Immune Responses in Mice)

  • Effects of quercetin on the specific and non-specific immune responses were studied in vivo. Quercetin at a dose of 2.5, 5, 10, 20 and 40 mg/kg were orally administered to ICR male mice once daily for 28 consecutive days. Cyclophosphamide was injected intraperitoneally to ICR mice with a single dose of 5 mg/kg 2 days before secondary immunization. Mice were sensitized and challenged with sheep red blood cells (S-RBC). Immune responses were evaluated by humoral and cellular immune reponses and non-specific immune response. The results of this study were summarized as followings; 1. Quercetin significantly decreased the body weight, and introduced the atrophy of liver, spleen and thymus gland dose-dependently, but increased the numbers of white blood cell. 2. Querectin significantly depressed the hemagglutination titer, Arthus reaction and hemolytic plaque forming cell. 3. Quercetin significantly depressed the delayed type hypersensitivity and rosette forming cell. 4. Quercetin at a dose of 2.5, 5 and 40 mg/kg significantly depressed phagocytic activity. 5. Quercetin at a dose of 10 and 20 mg/kg significantly increased natural killer cell activity.

  • Dominant white와 Extended black 은 닭의 깃털 색깔에 관여하는 유전자위로서 각각 PMEL l7과 MC1R 유전자를 암호화하며 이들의 염기서열 다형은 닭의 깃털 색깔 양상과 매우 밀접하게 연관되어있다. 본 실험은 이러한 점에 착안하여 PMEL17과 MC1R 유전자의 염기서열에서 Barred Plymouth rock, Korean Ogol Chicken and White Leghorn의 품종 특이적인 염기서열 다형을 확인하였다. PMEL17 유전자에서 8개, MC1R 유전자에서 4개씩 모두 12개의 다형이 확인되었다. White Leghorn은 모든 염기서열 다형 위치에서 다른 두 종의 닭들과 다른 염기를 가지고 있었다. 그러나 Barred Plymouth Rock과 Korean Ogol Chicken은 모든 염기서열 다형 위치에서 같은 염기를 가지고 있었다.

흑 마늘 첨가에 따른 마늘 젖산 발효물의 물성 및 기능성평가 (Effects of Black Garlic on the Rheological and Functional Properties of Garlic Fermented by Leuconostoc mesenteroides)

  • 흰 마늘/흑 마늘 첨가에 따른 김치 젖산균을 이용한 발효물을 제조하여 품질 및 기능성 평가를 수행하였다. 고형분 함량은 17% 흰 마늘 습식분쇄물과 22.63% 흑 마늘 습식분쇄물이다. 마늘 혼합물에 Leuconostoc mesenteroides 균주를 이용하여 24시간 동안 젖산 발효시킨 발효물의 생균수와 점도는 흑 마늘의 첨가농도가 증가함에 따라 감소하였으며, 흑 마늘을 10% 수준으로 첨가한 경우에 생균수는 $1.6\times10^{11}$ cfu/mL, 점도는 9.31 $Pa{\cdot}s^n$를 나타내었다. 마늘 발효물의 물과 70% EtOH 추출물의 polyphenol 함량은 각각 6.29 mg/mL, 5.99 mg/mL이며, DPPH 소거 활성은 $IC_{50}$ 값이 1.03 mg/mL, 1.07 mg/mL, ACE 저해 활성의 $IC_{50}$값은 68.54 mg/mL과 72.78 mg/mL이다. ACE 저해활성은 흑 마늘이 많이 첨가될수록 높게 나타났으며 그중에서 70% EtOH 추출보다 물 추출에서 높은 활성을 나타내는 것으로 나타났다. 마늘 발효물의 sucrose 전환율은 흑 마늘 첨가 농도가 낮을수록 급격하게 증가하면서 흑 마늘 20%, 10%, 0% 첨가에서 각각 20%, 65%, 85%를 보였다. 흑 마늘 10% 함유 발효물의 저장 중 점도는 완만하게 감소하였으며, 생균수는 냉장저장 1주 후 $1.6\times10^{11}$ cfu/mL에서 $6.5\times10^{10}$ cfu/mL로 감소하였으며, 냉동저장이 냉장저장보다 높은 생균수를 나타내었다.

Siamese Crocodile White Blood Cell Extract Inhibits Cell Proliferation and Promotes Autophagy in Multiple Cancer Cell Lines

  • Cancer represents one of the most significant threats to human health on a global scale. Hence, the development of effective cancer prevention strategies, as well as the discovery of novel therapeutic agents against cancer, is urgently required. In light of this challenge, this research aimed to evaluate the effects of several potent bioactive peptides and proteins contained in crocodile white blood cell extract (cWBC) against LU-1, LNCaP, PC-3, MCF-7, and CaCo-2 cancer cell lines. The results demonstrate that 25, 50, 100, and $200{\mu}g/ml$ cWBC exhibits a strong cytotoxic effect against all investigated cell lines ($IC_{50}$ $70.34-101.0{\mu}g/ml$), while showing no signs of cytotoxicity towards noncancerous Vero and HaCaT cells. Specifically, cWBC treatment caused a significant reduction in the cancerous cells' colony forming ability. A remarkable suppression of cancerous cell migration was observed after treatment with cWBC, indicating potent antimetastatic properties. The mechanism involved in the cancer cell cytotoxicity of cWBC may be related to apoptosis induction, as evidenced by typical apoptotic morphology features. Moreover, certain cWBC concentrations induced significant overproduction of ROS and significantly inhibited the $S-G_2/M$ transition in the cancer cell. The molecular mechanisms of cWBC in apoptosis induction were to decrease Bcl-2 and XIAP expression levels and increase the expression levels of caspase-3, caspase-8, and p53. These led to a decrease in the expression level of the cell cycle-associated gene cyclin-B1 and the arrest of cell population growth. Consequently, these findings demonstrate the prospect of the use of cWBC for cancer therapy.

Effects of Media on the Callogenensis and Cell Mass Production in Cell Cultures of Panax vietnamensis

  • This study was initiated to investigate the impacts of media types and other components on the callogenensis and cell mass production of Panax vietnamensis in the first step of the cell biomass procedure. Four media were checked: Murashige-Skoog (MS), White, Gamborg and Nitch-AII. All the four media were shown potential media for Panax vietnamensis callogenensis and cell mass production, in which the MS medium showed the best results: the successful callogenensis ratio and cell mass formation were 30% and 62,93 ${\pm}$ 3,63 mg (DW) respectively, the Nitch medium showed the lowest results: the successful callogenensis ratio and cell mass formation were 15% and 27,10 ${\pm}$ 2,24 mg (DW) respectively. The results showed that the MS medium is the most suitable medium for Panax vietnamensis callogenensis and cell mass production.
