대하새우로부터 분리한 WSBV의 게놈서열 분석 (Partial genomic sequence of baulovirus associated with white spot syndrome (WSBV) isolated from penaeid shrimp P. chinensis)

  • 새우의 갑각에 흰점을 유발하는 특징을 가진 WSBV는 Baculovirus의 일종으로 여러 종류의 새우에 높은 치사율을 보이는 병원체로서 새우양식에 막대한 피해를 주고 있다. 본 연구에서는 국내에서 양식중인 대하에 질병을 유발한 WSBV의 특성을 알아내고자 치사한 새우로부터 바이러스의 게놈을 클로닝하여 재조합클론(E3)을 분자생물학적으로 분석하였다. E3의 염기서열을 분석한 결과, 이 클론은 AcNPV를 포함한 지금까지 알려진 어떠한 바이러스와도 뚜렷한 상동성(60%)을 보이지 않아 WSBV가 기존의 바이러스와 구분되는 새로운 바이러스임을 알 수 있다. E3의 염기서열에 기초하여 한쌍의 PCR 프라이머를 작성하였다. 병든 새우로부터 분리한 DNA를 30회 증폭한 결과, 예상크기의 산물을 얻을 수 있어 이 방법은 바이러스의 감염여부를 알아낼 수 있는 진단법으로도 활용가능하다.

White Sport Syndrome Virus Disease of Shirmp and Diagnostic Methods

  • Since 1993, the White Spot Syndrome Virus (WSSV) disease occurred in China among cultured shrimps resulting in mass mortality. Epizootiological surveys undertaken during the outbreak period of 1993-1994 indicated that all stages of Penaeus chinensis, P. japonicus and P. monodon were infected. Consequent to the transport of contaminated shrimp seedlings and seawater, the disease spread all over the farms of China. The disease was more rapidly transmitted at temperatures above $25^{\circ}C$. Challenge experiments showed the causative agent was highly virulent. White spots appeared on the carapace of both span-taneous and experimentally infected shrimps. Moribund shrimps contained turbid hemolymph, hypertrophied Iymphoid organ and a necrotic mid-gut gland. Electron microscopy showed the presence of viral particles in the gills, stomach, lymphoid organ, and epidermal tissue of the infected shrimp. The visions were slightly ovoid with an envelope and averaged 350 $\times$ 150 nm; nucleocapsids measured 375 $\times$ 157 nm. With discontinuous sucrose gradient of 35, 50 and 60% (w/v), the virus was separated from hemolymph of the infected shrimp. The estimated molecular weight of genomic DNA was 237 Kb with EcoR I, 247 Kb with Hind III and 241kb with Pst I. A total of 9 hybridoma colones secreting monoclonal antibodies (MAbs) were produced from mouse myeloma and spleen cells immunized with WSSV. The immunofluorescence assay of gill tissue showed that the MAbs reacted with diseased but not with healthy shrimp. The MAbs belonged to IgGl, IgG2b subclass and IgM class, all with kappa light Immune-electron-microscopy with colloidal gold marker showed the presence of 5 MAbs epitopes on the envelope and one on the capsid of the virus. Baculoviral mid-gut gland necrosis showed the specificity of the MAbs produced. For diagnosis 5 different methods were selected. Using Kimura primers for PCR, or MAbs for immunoblot, ELISA or FAT method, in situ hybridization was carried out to show the gene. All these methods detected WSSV in the organ samples of the diseased shrimp but not in healthy one.

Detection of infectious hypodermal and hematopoietic necrosis virus and white spot syndrome virus in whiteleg shrimp (Penaeus vannamei) imported from Vietnam to South Korea

  • In this study, whiteleg shrimp (Penaeus vannamei) imported from Vietnam were collected from South Korean markets, and examined for 2 viruses: infectious hypodermal and hematopoietic necrosis virus (IHHNV, recently classified as decapod penstyldensovirus-1), and white spot syndrome virus (WSSV). Among 58 samples, we detected IHHNV in 23 samples and WSSV in 2 samples, using polymerase chain reaction and sequencing analyses. This is the first report of IHHNV and WSSV detection in imported shrimp, suggesting that greater awareness and stricter quarantine policies regarding viruses infecting shrimp imported to South Korea are required.

  • Because of the great concern over the possibility of contamination from the rod-shaped nuclear virus (PRDV) from Japan and white spot virus (WSSV) from Taiwan, most eggs used in Korean shrimp farms are currently obtained from local broodstock. In addition, the screening of imported broodstock for any viral presence at the National Fisheries Research and Development Institute is also mandatory. Nonetheless, massive mortality from white spot syndrome continues in Korea. In the present study, we present an improved PCR method to use tissue-extracted DNA instead of viral DNA extracted from a purified virus based on a sucrose density gradient, and produced results within 8 h. In 1998, this modified PCR method was able to detect that diseased Penaeus japonicus were infected within 8 h. In 1998, this modified PCR method was able to detect that diseased Penaeus japonicus were infected only with PRDV, while Fenneropenaeus chinensis were infected with both PRDV and WSSV. In 1999, PRDV and WSSV were detected in F. chinensis with signs of infection, but not with WSSV alone.

  • White spot syndrome virus (WSSV) is a prevalent and virulent pathogen affecting cultured whiteleg shrimp (Litopenaeus vannamei) in Korea. In this study, seven monoclonal antibodies (mAbs) (10A12, 16C3, 17G4, 21G5, 22C4, 23B6 and 24G6) were produced by using purified WSSV. The reactivity of these mAbs was analysed by Western blot (WB), indirect immunofluorescence (IIF), and lateral flow immunochromatographic assay (LFIA). WB analysis demonstrated that three mAbs (17G4, 22C4, and 23B6) reacted specifically to VP28 with an approximate molecular weight of 24 kDa, mAb 16C3 reacted with approximately 17 kDa. IIF analysis demonstrated specific fluorescence signals on gill tissues of WSSV-infected shrimp, with five mAbs (10A12, 16C3, 22C4, 23B6, and 24G6), pleopods from WSSV-infected shrimp were used for LFIA, where, two mAbs (21G5 and 22C4) exhibited positive reaction. In conclusion, it can be inferred that the mAbs usage and specificity depends on the nature of assay used for diagnosis.

Supplemental effects of biofloc powder on growth performance, innate immunity, and disease resistance of Pacific white shrimp Litopenaeus vannamei

  • An 8-week feeding trial was conducted to study the effect of dietary supplementation of a biofloc powder on growth performance and non-specific immune response of Litopenaeus vannamei. Seven experimental diets were prepared with supplementation of graded levels of dried biofloc powder by 0, 0.5, 1.0, 2.0, 4.0, 6.0, and 8.0% (designated as Con, BF0.5, BF1, BF2, BF4, BF6, and BF8, respectively). Triplicate groups of shrimp ($1.01{\pm}0.01g$) were hand-fed with one of the diets four times a day. At the end of the feeding trial, significantly (P<0.05) higher growth performance and feed utilization were obtained in BF4 groups compared to those fed the Con diet. The innate immunity of shrimp was improved by the dietary supplementation of biofloc. Dietary inclusion of biofloc at the level of 4.0% significantly increased disease resistance of shrimp against Vibrio harveyi. The results indicate that biofloc might be used as a dietary supplement for growth performance, innate immunity and disease resistance of Pacific white shrimp.

Effect of nanobubbles (oxygen, ozone) on the Pacific white shrimp (Penaeus vannamei), Vibrio parahaemolyticus and water quality under lab conditions

  • This study assessed the effects of oxygen and ozone nanobubbles on gill morphology, weight gain, and mortality of Pacific white shrimp (Penaeus vannamei), as well as the level of Vibrio parahaemolyticus and water quality of shrimp culture tanks under lab conditions. Two experiments were carried out with oxygen macrobubble, ozone macrobubble, oxygen nanobubble, ozone nanobubble, and control treatments (air-stone macrobubble). Experiments were done in triplicate in 100 L tanks with 15‰ saline water, and 20 shrimp per tank. Tanks in Experiment 1 were not inoculated with bacteria; tanks in Experiment 2 were inoculated with V. parahaemolyticus at a concentration of 106 CFU/mL. The results revealed that short treatments with ozone nanobubbles had minimal impact on shrimp gills, mortality, and growth rates, reduced V. parahaemolyticus concentration in water compared to the other groups, and improve water quality. These laboratory results indicate that ozone nanobubble treatment may be useful for controlling V. parahaemolyticus. More work is needed to find the best protocol to apply the technology on a commercial scale.

Substitution of Fish Meal by Mealworm (Tenebrio molitor) in Practical Diets for Pacific White Shrimp, Litopenaeus vannamei

  • Mealworm (Tenebrio molitor) was evaluated to investigate the effect of partial or total replacement of fish meal in diets for white shrimp, Litopenaeus vannamei. Experimental groups of shrimp with average initial body weight (2.43 ± 0.54 g) were fed each with 4 isonitrogeneous (38% crude protein) diets formulated to include 0, 25, 50 and 100% (diets 1 to 4, respectively) of fish meal substituted with mealworm. After eight weeks of feeding trials, shrimp fed with diet 3 and 4 revealed the highest values for live weight gain(8.01 ± 2.51 and 7.93 ± 1.12), specific growth rates (2.70 ± 1.12 and 2.59 ± 0.51) as well as better feed conversion ratio (2.69 ± 0.09 and 2.72 ± 0.19) compared to the control group with statistically significant manner (p<0.05). Survival range was 98% in all the treatments. An increase in weight gain and other growth associated parameters was observed with higher replacement. These results clearly indicate that 50% and 100% of fish meal protein in shrimp diet can be replaced by mealworm not only without any adverse effect but also the effect of promoting growth performance.

The epizootic started in 1992, and spread through east and south east Asia and into other shrrimp growing countries of the region. Determination of optimal dietary valine concentrations for improved growth performance and innate immunity of juvenile Pacific white shrimp Penaeus vannamei

  • A study was conducted to evaluate dietary valine (Val) requirement for Pacific white shrimp (Penaeus vannamei). Five isonitrogenous (353 g/kg) and isocaloric (4.08 kcal/g) semi-purified diets containing graded levels of Val (2.7, 5.1, 8.7, 12.1 or 16.0 g/kg) were formulated. Quadruplicate groups of 12 shrimp (average body weight: 0.46 ± 0.00 g) were fed one of the experimental diets (2%-5% of total body weight) for 8 weeks. Maximum weight gain was observed in 8.7 g/kg Val group. However, the growth performance was reduced when Val concentration in diets were higher than 12.1 g/kg. Feed conversion ratio was significantly increased with 2.7 and 16.0 g/kg Val inclusion. Shrimp fed the diets containing 2.7 g/kg Val showed significantly lower protein efficiency ratio, whole-body crude protein and Val concentrations. Dietary inclusion of Val significantly improved the relative expression of insulin-like growth factor binding protein and immune-related genes (prophenoloxidase, lysozyme and crustin) in the hepatopancreas and 8.7 g/kg Val group showed highest expression among all the groups. The dietary requirement of Val for maximum growth of juvenile P. vannamei, estimated using polynomial regression analysis on growth, was 9.54 g/kg of Val (27.2 g/kg based on protein level) and maximum growth occurred at 9.27 g/kg of Val (26.2 g/kg based on protein level) based on broken-line regression analysis.
