Muscle Strength Activity of Herbal Medicine Extracts of Acanthopanax Sieboldianus and Allium Hookeri (오가피와 삼채 등 생약재 추출물의 근력 활성 효과)

  • We were selected oriental medicinal herbs known to be effective in strengthening muscles, improving sarcopenia, treating arthritis and osteoporosis. Amino acids have been reported to be associated with the above-mentioned effects. During the manufacturing process of water extracts of ginseng and samchae, the content of arginine, a basic amino acid, was significantly reduced. The decrease was 99.48mg/100ml for steamed red ginseng compared to 114.07mg/100ml for dried white ginseng, and 54.69mg/100ml for steamed samchae compared to 118.90mg/100ml for dried samchae. The extracts of the wild roots of Acanthus apias and the above-ground twigs of Acanthus apias showed significantly higher DPPH and ABTS radical scavenging activities. Meanwhile, DPPH activity was higher in steamed red ginseng and steamed samchae compared to dried white ginseng and dried samche, but ABTS activity increased to 66.88% for steamed red ginseng compared to 41.96% for dried white ginseng, but was 77.48% for dried samchae and 77.52% for steamed samchae.

Observations of a endometrial stromal sarcoma in the rat uterus wall (Rat의 자궁벽에 발생된 stromal sarcoma의 관찰)

  • The present observation was focussed mainly on the histopathological and immunohistochemical finding of the tumor developing from the outside of rat uterus wall. In gross finding, this tumor were a oval form mass and 0.75cm and 1cm in greatest demension. The sectioned surface showed uniform, solid, gray-white. In histopathological finding, the tumor showed the form of lymphoid nodule-like structure and was composed of the cells with karryorrhexis, karyolysis and necrosis in the central zone, mature cells in the paracentral zone, and concentric layers of compressed, flattened and atrophied smooth muscle fibers by the proliferating of the cells closely resembling endometrial stromal ells in the peripheral zone and a few smaller nodule-like structure in certain areas of the peripheral zone. In immunohistochemical observations, the proliferating cells presents only among the concentric layers of the smooth muscle fibers in the peripheral zone. Labeling index of proliferating cells in the peripheral zone was $4.34{\pm}1.28%$. These tumor cells did not metastasized in another orgns and labeling index was low, So it was concluded that this tumor was benign stomal sarcoma.

Synchronization of Synovial Chondromatosis and Mycobacterium intracellurae Infection in Olecranon Bursitis: A Case Report

  • A 73-year-old woman presented with a recurrent cystic mass around her left olecranon. She had a history of 8 steroid injections due to elbow pain beginning 3 years ago and twice had undergone aspiration of olecranon bursitis that developed two months prior to presentation. She had been taking medications for hypertension and diabetes with no pertinent past history. On magnetic resonance imaging (MRI), there were multiple nodules in the olecranon bursa, which were isointense to muscle on T1-weighted images and hyperintense to muscle on T2-weighted images. Our initial diagnosis was synovial chondromatosis. On bursoscopy, masses of gray-white colored nodules were observed in the bursa. Finally, synovial chondromatosis and non-tuberculous mycobacterial infection were concurrently diagnosed. In conclusion, uncalcified synovial chondromatosis and rice bodies can have similar visual and MRI characteristics; therefore, we suggest that clinicians should be aware of the possibility of other infections in cases of this type.

Differences in bedding material could alter the growth performance of White Pekin ducks raised for 42 days

  • The effect of different commercially available bedding materials on the growth performance and carcass characteristics of ducks for 42 days was investigated. 336 one-day-old White-Pekin ducklings (60.48 ± 0.16 g) were randomly allocated into 24-floor pens with one of the three beddings namely i) coco peat, ii) rice husks, or iii) sawdust. 14 ducklings per pen and 8 replicate pens per bedding material were used. Birds were fed a starter diet from days 1-21 and a grower diet from days 22-42. Weekly growth performance evaluation was conducted for the average body weight, weight gains, daily feed intake, and feed conversion efficiency. One bird per pen was sacrificed on day 42 for the evaluation of carcass characteristics including the carcass, breast, and leg muscle percentages. Breast and leg muscle samples were then collected and analyzed for their proximate and pH values. Higher body weights (p < 0.05) were noticed with rice husks on day 42 only. Improved daily gains (p < 0.05) were also noticed for birds raised with rice husks over the entire period (days 1-42). Concerning feed intake, higher values (p < 0.05) were similarly noted with rice husks for the grower phase (days 22-42), and the entire experimental period (days 1-42). Marginally improved feed intake values were also noted with the use of rice husks as the bedding materials on day 42 (p = 0.092). Improved feed efficiency (p < 0.05) was noticed with rice husks on day 35, the grower period, and the entire 42-day period. However, no significant differences were noticed for most of the carcass characteristics that were evaluated. Nevertheless, higher (p < 0.05) pH values for the breast muscle were noticed with the use of coco peat and sawdust as the bedding. Conclusively, the bedding type could have a significant impact on the growth performance of ducks without adverse effects on carcass characteristics. The use of rice husks as bedding might be advantageous and is therefore recommended.

Correlation between Heart-type Fatty Acid-binding Protein Gene Polymorphism and mRNA Expression with Intramuscular Fat in Baicheng-oil Chicken

  • This study aims to determine the polymorphism and mRNA expression pattern of the heart-type fatty acid-binding protein (H-FABP) gene and their association with intramuscular fat (IMF) content in the breast and leg muscles of Baicheng oil chicken (BOC). A total of 720 chickens, including 240 black Baicheng oil chicken (BBOC), 240 silky Baicheng oil chicken (SBOC), and 240 white Baicheng oil chicken (WBOC) were raised. Three genotypes of H-FABP gene second extron following AA, AB, and BB were detected by polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) strategy. The G939A site created AA genotype and G956A site created BB genotype. The content of IMF in AA genotype in breast muscle of BBOC was significantly higher than that of AB (p = 0.0176) and the genotype in leg muscle of WBOC was significantly higher than that of AB (p = 0.0145). The G939A site could be taken as genetic marker for higher IMF content selecting for breast muscle of BBOC and leg muscle of WBOC. The relative mRNA expression of H-FABP was measured by real-time PCR at 30, 60, 90, and 120 d. The IMF content significantly increased with age in both muscles. The mRNA expression level of H-FABP significantly decreased with age in both muscles of the three types of chickens. Moreover, a significant negative correlation between H-FABP abundance and IMF content in the leg muscles of WBOC (p = 0.035) was observed. The mRNA expression of H-FABP negatively correlated with the IMF content in both breast and leg muscles of BOC sat slaughter time.

"Dietary supplementation of L-tryptophan" increases muscle development, adipose tissue catabolism and fatty acid transportation in the muscles of Hanwoo steers

  • This study investigated the effects of dietary rumen-protected L-tryptophan (TRP) supplementation (43.4 mg of L-tryptophan kg-1 body weigt [BW]) for 65 days in Hanwoo steers on muscle development related to gene expressions and adipose tissue catabolism and fatty acid transportation in longissimus dorsi muscles. Eight Hanwoo steers (initial BW = 424.6 kg [SD 42.3]; 477 days old [SD 4.8]) were randomly allocated to two groups (n = 4) of control and treatment and were supplied with total mixed ration (TMR). The treatment group was fed with 15 g of rumen-protected TRP (0.1% of TMR as-fed basis equal to 43.4 mg of TRP kg-1 BW) once a day at 0800 h as top-dressed to TMR. Blood samples were collected 3 times, at 0, 5, and 10 weeks of the experiment, for assessment of hematological and biochemical parameters. For gene study, the longissimus dorsi muscle samples (12 to 13 ribs, approximately 2 g) were collected from each individual by biopsy at end of the study (10 weeks). Growth performance parameters including final BW, average daily gain, and gain to feed ratio, were not different (p > 0.05) between the two groups. Hematological parameters including granulocyte, lymphocyte, monocyte, platelet, red blood cell, hematocrit, and white blood cell showed no difference (p > 0.05) between the two groups except for hemoglobin (p = 0.025), which was higher in the treatment than in the control group. Serum biochemical parameters including total protein, albumin, globulin, blood urea nitrogen, creatinine phosphokinase, glucose, nonesterified fatty acids, and triglyceride also showed no differences between the two groups (p > 0.05). Gene expression related to muscle development (Myogenic factor 6 [MYF6], myogenine [MyoG]), adipose tissue catabolism (lipoprotein lipase [LPL]), and fatty acid transformation indicator (fatty acid binding protein 4 [FABP4]) were increased in the treatment group compared to the control group (p < 0.05). Collectively, supplementation of TRP (65 days in this study) promotes muscle development and increases the ability of the animals to catabolize and transport fat in muscles due to an increase in expressions of MYF6, MyoG, FABP4, and LPL gene.

  • In order to investigate the effect of dietary fatty acids on lipids metabolism in adipose pads and superficial pectoral muscle of chicks, the present, experiment was carried out using signle comb White Leghorn male chicks fed a fat-free diet and diets containing margaric, stearic or linoleic acids. Total lipids of tissues were extracted with a chlorofrom-methanol solution. The lipid components were fractionated by thin layer chromatography and the fatty acid distribution of lipid fractions was determined by gas liquid chromatography. The neck adipose pads contained 34-62% total lipids, in which triglycerides, were dominant, being 97-98%. Margaric, stearic and linoleic acids were distributed at a relatively high rate in the adipose tissue when the corresponding acids were fed, and margaric acid feeding lowered palmitoleic acid distribution and linoleic acid feeding elevated stearic acid distribution. The wet superficial pectoral muscle contained 1.3-1.7% total lipids, of which 77-79% was phospholipids and 11-13% was free cholesterol. When margaric acid was fed, margaric and heptadecenoic acids appeared in the muscle lipids. When linoleic acid was fed, this acid was significantly highly distributed in every muscle lipid fractions, whereas, when stearic acid fed no elevation of stearic acid distribution was observed. In the muscle phospholipids, oleic acid was significantly highly distributed in the stearic acid fed chicks, and the linoleic acid feeding signigicantly lowered the distribution value of palmitic and oleic acids, but elevated the distribution value of stearic acid. And the linoleic acid feeding lowered the distribution value of eicosatrienoic acid and elevated the distribution value of arachidonic acid.

Study on Some Qualitative Features of Meat from Young Goat of Bulgarian Breeds and Crossbreeds of Goats Slaughtered at Various Ages

  • A comparative study on the quality of meat from three breeds of goats reared in Bulgaria: Local Aboriginal (LA); Bulgarian White Dairy (BWD) and crossbreeds of local aboriginal with Saanen goats (LA ${\times}$ S). Eight intact young male goats from each breed have been included in the experiment. The animals from the three breeds have been reared under similar conditions. The animals have been slaughtered at the age of 2 months (at weaning) and at the age of 6 months (upon reaching sexual maturity). The following tests have been performed on samples of m. longissimus dorsi between $5^{th}$ and $6^{th}$ ribs: pH 45 min post mortum, colorimetrically color of meat, water holding capacity and fatty acid composition of the fat. The thickness of muscle fibre and the proportion of muscle, connective and adipose tissue have been microscopically determined at m. longissimus dorsi, m. semitendinosus and m. iliopsoas. After deboning and separating the bigger tendons, the chemical composition of the meat from the left half of the carcass has been determined. The results reveal lack of statistically significant differences among the three breeds concerning pH, the water holding capacity, color of meat, thickness of the muscle fibre and fatty acid composition of the fat. The carcass meat of crossbreeds of LA ${\times}$ S contains significantly more fat than the other two breeds. This influences the proportion of muscle, connective and adipose tissue in m. longissimus dorsi, m. semitendinosus and m. iliopsoas, resulting in more adipose tissue in the LA ${\times}$ S compared to young goats of the LA or the BWD. There are differences in slaughtering 2-month-old goats (at weaning) and 6-month-old ones (upon reaching sexual maturity). At a greater age the content of fat in the carcass increases, as well as the quantity of the adipose tissue, the intensity of the colour of the meat and there is a tendency towards thickening of the muscle fibre and increasing the water holding capacity of meat. The moisture content in meat decreases due to the increase of fat.

Effects of protein content and the inclusion of protein sources with different amino acid release dynamics on the nitrogen utilization of weaned piglets

  • Objective: We aimed to investigate the effect of the differing amino acid (AA) release dynamics of two protein sources on the growth performance, nitrogen deposition, plasma biochemical parameters, and muscle synthesis and degradation of piglets when included in their diets at normal and low concentrations. Methods: Forty-eight piglets (Duroc×Landrace×Large White) with initial body weight of 7.45±0.58 kg were assigned to six groups and fed one of 6 diets. The 6 dietary treatments were arranged by 3×2 factorial with 3 protein sources and 2 dietary protein levels. They are NCAS (a normal protein content with casein), NBlend (a normal protein content with blend of casein and corn gluten meal), NCGM (a normal protein content with corn gluten meal), LCAS (a low protein content with casein), LBlend (a low protein content with blend of casein and corn gluten meal), LCGM (a low protein content with corn gluten meal). The release dynamics of AA in these diets were determined by in vitro digestion. The digestibility, utilization and biological value of nitrogen in piglets were determined by micro Kjeldahl method. Plasma insulin was measured by enzyme-linked immunosorbent assay kits. The protein expression of mediators of muscle synthesis and degradation was determined by western blotting. Results: Although the consumption of a low-protein diet supplemented with crystalline AA was associated with greater nitrogen digestion and utilization (p<0.05), the final body weight, growth performance, nitrogen deposition, and phosphorylation of ribosomal protein S6 kinase 1 and eIF4E binding protein 1 in the muscle of pigs in the low-protein diet-fed groups were lower than those of the normal-protein diet-fed groups (p<0.05) because of the absence of non-essential AA. Because of the more balanced release of AA, the casein (CAS) and Blend-fed groups showed superior growth performance, final body weight and nitrogen deposition, and lower expression of muscle ring finger 1 and muscle atrophy F-box than the CGM-fed groups (p<0.05). Conclusion: We conclude that the balanced release of AA from CAS containing diets and mixed diets could reduce muscle degradation, favor nitrogen retention, % intake and improve growth performance in pigs consuming either a normal- or low-protein diet.

cDNA Cloning, Tissue Expression and Association of Porcine Pleiomorphic Adenoma Gene-like 1 (PLAGL1) Gene with Carcass Traits

  • Pleiomorphic adenoma gene-like1 (PLAGL1) encodes a zinc-finger (ZF) protein with seven ZFs of the C2H2-type which is a regulator of apoptosis and cell cycle arrest, and also regulates the secretion of insulin. In both human and mouse, PLAGL1 is a candidate gene for tumor suppressor and transient neonatal diabetes mellitus (TNDM). In this study, a 2,238 bp fragment covering the complete coding region was obtained and deposited to GenBank (accession number: DQ288899). The reverse transcriptase-polymerase chain reaction (RT-PCR) indicated that PLAGL1 was expressed almost equally in heart, liver, spleen, lung, kidney, stomach, small intestine, skeletal muscle, fat, uterus and ovary. Comparing the sequences of Large White and Meishan pigs, a C-T transition in exon 6 was found. The polymorphism could be detected by TaqI and was genotyped in five purebreds (Large White, Landrace, Meishan, Tongcheng and Bamei). Association analysis was performed between the polymorphism and carcass traits in 276 pigs of a "Large White${\times}$Meishan" F2 resource population. As a consequence, significant associations of the genotypes with shoulder backfat thickness (SFT) and internal fat rate (IFR) were observed. Pigs with TT genotype had low SFT and high IFR compared with TC or CC genotypes.
