Effect of Ginseng on the Relaxation of the Penile Corpus Cavernosal Smooth Muscle in Rabbits (인삼이 토끼 음경해면체 평활근의 이완작용에 미치는 영향)

  • The effect of ginseng on the reactivity of penile corpus cavernosal smooth muscle strips of rabbits was investigated to support the clinical application of ginseng for the treatment of erectile dysfunction. New Zealand white rabbits were randomly divided into two groups, normal diet group (n=9) and ginseng saponin diet group (n=5). Then, each group was fed normal diet and Korean red ginseng saponin diet (50 mg/kg/day) for 10 weeks. We measured concentration dependent relaxation of corporal smooth muscle to acetylcholine (10-8 M to 10-4M) in organ chamber. The degree of relaxation was expressed as percentage of maximal relaxation obtained by papaverine (10-4M). Dose dependent relaxation of corpus cavernosal smooth muscle to acetylcholine, at the concentration of 10-8 M to 10-4M by half log increment, was 4.06$\pm$0.00, 4.30$\pm$1.30, 5.32$\pm$0.68, 11.64$\pm$1.74, 16.24$\pm$1.61, 23.33$\pm$ 2.29, 26.45$\pm$2.25, 30.43$\pm$2.40 and 33.41 $\pm$2.48 (%), respectively in normal diet group and 9. 83$\pm$4.15, 20.60$\pm$4.62, 24.1815.12, 35.75$\pm$5.71, 43.35$\pm$6.11, 51.30$\pm$6.22, 56.33$\pm$6.22, 54.30$\pm$4.17 and 51.98$\pm$3.92 (Vc), respectively in ginseng group. These data suggest that ginseng enhances ondothelium-dependent acetylcholine-induced relaxation of penile corpus cal.ernosal smooth muscle in rabbits.

  • The aim of this study was to investigate the expression of PPAR-α, -β/δ, -γ, PGC-1α, GLUT-4 and p-AMPK-α2 protein in the skeletal muscle of diabetic induced-rats by endurance training combined with rosiglitazone. The expression of PPAR-α, -β/δ, -γ, PGC-1α, GLUT-4 and p-AMPK-α2 protein in red and white gastrocnemius by western blotting. The body weight was higher in diabetic induced-rats compared to the normal rats and after the treatment of exercise combined with rosiglitazone was significantly reduced in the all group. The levels of blood glucose was higher in diabetic induced-rats compared to the normal rats and after the treatment of exercise combined with rosiglitazone was significantly reduced in the all group. The expression of PPAR-α, -γ, PGC-1α in skeletal muscle of diabetic induced-rats were increased all groups and increased significantly in the group with exercise combined with rosiglitazone. The expression of GLUT-4 and p-AMPK-α2 protein in the skeletal muscle of diabetic induced-rats were increased all groups and increased significantly in the group with exercise combined with rosiglitazoneI. These results suggest that exercise training and rosiglitazone may act as complementary therapies for the treatment of insulin rasistance.

Isolation, Expression Pattern, Polymorphism and Association Analysis of Porcine TIAF1 Gene

  • TIAF1 is a TGF-${\beta}$1-induced anti-apoptotic factor that plays a critical role in blocking TNF (tumor necrosis factor) cytotoxicity in mouse fibroblasts and participates in TGF-${\beta}$-mediated growth regulation. In this study, we obtained the full-length cDNA sequence of the porcine TIAF1 gene. Real-time PCR further revealed that the TIAF1 gene was expressed at the highest level in liver and kidney with prominent expressions detected in uterus, and lower levels detected in heart, spleen, lung, stomach, small intestine, skeletal muscle and fat of Large White pigs. Sequence analysis indicated that a 6 base-pair deletion mutation existed in the exon of the TIAF1 gene between Meishan and Large White pigs. This mutation induced deletion of Gln and Val amino acids. PCR-RFLP was used to detect the polymorphism in 394 pigs of a "Large White${\times}$Meishan" $F_{2}$ resource population and four purebred pig populations. The frequencies of the A allele (with a 6 bp deletion) were dominant in Chinese Meishan and Bamei pigs, and the frequencies of the B allele (no 6 bp deletion) were dominant in Large White and Landrace pigs. Association analyses revealed that the deletion mutation had highly significant associations (p<0.01) with meat marbling score of the thorax-waist longissimus dorsi (LD) muscle (MM1) and intramuscular fat percentage (IMF), and significant associations (p<0.05) with carcass length (CL). The results presented here supply evidence that the 6 bp deletion mutation in the TIAF1 gene affects porcine meat quality and provides useful information for further porcine breeding.

Effect of Dietary Energy Level on Nutrient Utilization, Insulin-like Growth Factor-I and Insulin-like Growth Factor Binding Protein-3 in Plasma, Liver and Longissimus dorsi Muscle in Growing-finishing Pigs Using Soybean Oil as an Energy Source

  • Two experiments were carried out to study the effects of dietary energy level on nutrient digestion, nitrogen (N) utilization, growth performance, insulin-like growth factor-I (IGF-I), and insulin-like growth factor binding protein-3 (IGFBP-3) in plasma, liver and longissimus dorsi muscle in growing-finishing pigs. In experiment 1 (Exp 1), 15 castrated male pigs (Duroc${\times}$Landrace${\times}$Large White) (Body weight, BW, 55.6${\pm}$1.8 kg) were divided into three groups and fed rations containing 13.33, 14.87 and 17.35 MJ digestible energy (DE)/kg as treatments I, II and III, respectively, using soybean oil as an energy source. The experiment lasted 8 days and faecal and urinary samples were collected during the last 3 days. The results showed that the digestibility of dry matter (DM), energy and N was increased from treatments I to III (p<0.01). N-retention and N-retention rate were not influenced by dietary DE level (p>0.05). In experiment 2 (Exp 2), 36 female pigs (Duroc${\times}$Landrace${\times}$Large White) (BW 41.5${\pm}$3.8 kg) were divided into three groups. The pigs were fed with the same three rations used in Exp 1 for 60 days. At the end of Exp 2, eight pigs were selected from each group for blood sampling and 4 pigs for slaughter trial. The results indicated that average daily feed intake (ADFI) and N-intake were significantly decreased (p<0.01), and DE intake (p<0.01) and average daily gain (ADG) (p<0.05) were increased. IGF-I and IGFBP-3 in plasma were increased (p<0.05). No significant differences in IGF-I and IGFBP-3 in liver and longissimus dorsi muscle were found between different treatments. It was concluded that higher dietary DE level improved nutrient digestibility, ADG and feed/gain ratio when soybean oil was used as an energy source in the ration of growing-finishing pigs. No significant differences were found in Nretention and IGF-I and IGFBP-3 in liver and longissimus dorsi muscle between different treatments.

Changes in Contents of Some Taste Compounds of Fish Meat by Heating Conditions (어육의 가열조건에 따른 몇가지 정미성분 함량의 변화)

  • The changes of nucleotide and their related compounds, TMAO, TMA and total creatinine content of mackerel, pacific saury, yellow croaker and brown sole were investigated on heating conditions. The results were as follows. The content of inosine was the highest in all fish commonly, and white muscle fish showed higher than that of red muscle fish. The content of hypoxanthine showed increase by heat treatment but other compounds showed decrease. The contents of TMAO showed 31.1, 26.2, 49.4 and 58.5 mg% in each sample of raw materials, and brown sole showed the highest content than other fishes and TMAO contents showed decrease by heat treatment. The contents of TMA showed 3.7, 5.8, 22.9 and 16.9 mg% in each sample, and pacific saury showed the highest content than other fishes but TMA contents showed increase by heat treatment. The contents of total creatinine showed 341.2, 469.8, 52.3 and 87.6 mg% in each sample, and red muscle fish showed higher than that of white muscle fish and the highest content was shown in yellow croaker. All fish species showed decrease in contents of flavoring by heat treatment.

A STUDY ON TEMPOROMANDIBULAR JOINT DYSFUNCTION USING MAGNETIC RESONANCE IMAGING (자기 공명 영상을 이용한 악관절 기능 장애에 관한 연구)

  • The temporomandibular joint was evaluated using magnetic resonance imaging using a urface coil in 11 patients having reciprocal clicking or locking and compared with the normal joint in five subjects. Serial multisection 3㎜-thick parasagittal, paracoronal, and axial image on both closing and opening mouth were obtained with a 1.5 Tesla MR system and surface coil using CSMEMP, GRASS, MPGR, powerful extensions of fast imaging that is currently under clinical evaluation. MR images obtained were analized correlating with the theory of internal derangement. The obtained results were as follows: 1. The serial findings of structures in joint were determined on the serially sectioned images of joint with reciprocal clicking or locking by CSMEMP and MPGR on closing mouth. 2. The delta shaped white images of synovial fluid in the glenoid fossa and on the posterior surface of condyle were revealed on the parasagittal images by MPGR on opening mouth as in the normal joints. 3. The white image of joint fluid surrounding meniscus was recognized on the paracoronal image by GRASS on opening mouth as in the normal joints. 4. In joints having temporomandibular dysfunction the smooth image of displaced meniscus was recognized, but otherwise in the normal joints the image of muscle was noted on the paracoronal image sectioned at the anterior portion of condyle by GRASS. 5. The more thickened fascial plane between superior and inferior belly of lateral pterygoid muscle was not recognizable in joints having temporomandibular dysfunction than in the normal joints.

Differential Effects of Cod Proteins and Tuna Proteins on Serum and Liver Lipid Profiles in Rats Fed Non-Cholesterol- and Cholesterol-Containing Diets

  • Fish muscles are classified into white and red muscles, and the chemical composition of the two fish muscles have many differences. Few reports have assessed the health-promoting functions of white fish muscle proteins (WFP) and red fish muscle proteins (RFP). We therefore evaluated the mechanisms underlying the alteration of lipid profiles and cholesterol metabolism following the intake of WFP prepared from cod and RFP prepared from light muscles of tuna. Male Wistar rats were divided into six dietary groups: casein (23%), WFP (23%), and RFP (23%), with or without 0.5% cholesterol and 0.1% sodium cholate. Compared to the WFP-containing diet, the RFP-containing diet supplemented with cholesterol and sodium cholate significantly increased serum and liver cholesterol contents. However, in the RFP groups, an alteration in cholesterol metabolism including an increased tendency to excrete fecal sterols and hepatic cholesterol $7{\alpha}$-hydroxylase was related to the reduction of hepatic cholesterol contents. This phenomenon might be related to the tendency of an increased food intake in RFP-containing diets. These results highlight the differential effects of WFP and RFP on serum and liver lipid profiles of Wistar rats fed non-cholesterol- or cholesterol-containing diets under no fasting condition.

Sequence Characterization, Expression Profile, Chromosomal Localization and Polymorphism of the Porcine SMPX Gene

  • The full-length cDNA of the porcine SMPX gene was obtained by the rapid amplification of cDNA ends (RACE). The nucleotide sequences and the predicted protein sequences share high sequence identity with both human and mouse. The promoter of SMPX was sequenced and then analyzed to find the promoter binding sites. The reverse transcriptase-polymerase chain reaction (RT-PCR) revealed that SMPX has a high level of expression in heart and skeletal muscle, a very low expression in lung and spleen and no expression in liver, kidney, fat and brain. Moreover, SMPX has a differential expression level in skeletal muscle, the expression in 65-day embryos being higher than other stages. The porcine SMPX was mapped to SSCXp24 by using a somatic cell hybrid panel (SCHP) and was found closely linked to SW1903 using the radiation hybrid panel IMpRH. An A/G single nucleotide polymorphism (PCR-RFLP) in the 3'-untranslated region (3'-UTR) was detected in eight breeds. The analysis of allele frequency distribution showed that introduced pig breeds (Duroc and Large White) have a higher frequency of allele A while in the Chinese indigenous pig breeds (Qingping pig, Lantang pig, YushanBlack pig, Large Black-White pig, Small Meishan) have a higher frequencies of allele G. The association analysis using an experimental population (188 pigs), which included two cross-bred groups and three pure-blood groups, suggested that the SNP genotype was associated with intramuscular fat content.
