Effects of Single Total Body Irradiation (TBI) on the Peripheral Blood of Piglets (돼지에서 단일 전신 방사선 조사 용량에 따른 혈액변화 관찰)

  • We examined total body irradiation (TBI)-induced effects by complete blood count (CBC) and fluorescence-activated cell sorter analysis (FACS) in the piglet following radiation irritation. A CBC included red blood cell count, white blood cell count, and platelet cell count. Four piglets were examined in this study and each piglet was divided by irradiation dose, two piglets with 4 Gy, two with 6 Gy, one with 8 Gy. All piglets showed leukopenia, thrombocytopenia after irradiation. In 6 and 8 Gy group, three piglets showed severe hemostatic disorder and gastrointestinal disorder suchas diarrhea and anorexia, and they died between 10 and 15 days after radiation irritation. In 4 Gy, two piglets showed no clinical sign after radiation injury, but persistent leukopenia was shown in blood examination. We suggest that a single TBI dose less than 6 Gy is adequate for conditioning piglet for bone marrow transplantation.

Siamese Crocodile White Blood Cell Extract Inhibits Cell Proliferation and Promotes Autophagy in Multiple Cancer Cell Lines

  • Cancer represents one of the most significant threats to human health on a global scale. Hence, the development of effective cancer prevention strategies, as well as the discovery of novel therapeutic agents against cancer, is urgently required. In light of this challenge, this research aimed to evaluate the effects of several potent bioactive peptides and proteins contained in crocodile white blood cell extract (cWBC) against LU-1, LNCaP, PC-3, MCF-7, and CaCo-2 cancer cell lines. The results demonstrate that 25, 50, 100, and $200{\mu}g/ml$ cWBC exhibits a strong cytotoxic effect against all investigated cell lines ($IC_{50}$ $70.34-101.0{\mu}g/ml$), while showing no signs of cytotoxicity towards noncancerous Vero and HaCaT cells. Specifically, cWBC treatment caused a significant reduction in the cancerous cells' colony forming ability. A remarkable suppression of cancerous cell migration was observed after treatment with cWBC, indicating potent antimetastatic properties. The mechanism involved in the cancer cell cytotoxicity of cWBC may be related to apoptosis induction, as evidenced by typical apoptotic morphology features. Moreover, certain cWBC concentrations induced significant overproduction of ROS and significantly inhibited the $S-G_2/M$ transition in the cancer cell. The molecular mechanisms of cWBC in apoptosis induction were to decrease Bcl-2 and XIAP expression levels and increase the expression levels of caspase-3, caspase-8, and p53. These led to a decrease in the expression level of the cell cycle-associated gene cyclin-B1 and the arrest of cell population growth. Consequently, these findings demonstrate the prospect of the use of cWBC for cancer therapy.

Effects of Egg White Consumption on Allergy, Immune Modulation, and Blood Cholesterol Levels in BALB/c Mice

  • We previously demonstrated that water-soluble egg yolk extract is not related to elevation of serum immunoglobulin E, which can initiate allergic reactions; however, it increases the level of high density lipoprotein (HDL)-cholesterol and the activity of B lymphocytes. In this study, egg white (EW) was fed to BALB/c mice to determine its influence on growth efficiency, immune modulation, and changes in serum lipid levels. A total of 50 five-wk-old BALB/c male mice were divided into 5 groups, 4 of which were fed 0, 10, 50, or 100 mg/d EW for 4 wk. Mice with an uptake of 10, 50 and 100 mg/d EW showed no significant changes in daily weight gain, feed efficiency rate, or populations of white blood cells. However, the activities of both B and T lymphocytes were significantly increased in all three EW groups at the final week of treatment. Interestingly, serum levels immunoglobulin E were not altered by EW consumption, but the IgG level was significantly increased in the 100 mg/d EW group. Serum lipid profile analyses showed no significant changes in total cholesterol, HDL, low density lipoprotein, or triglyceride levels by EW consumption. Taken together, these data demonstrate that consumption of EW promotes immune cell activities and the upregulation of serum IgG levels. However, we found no changes in serum lipid profiles and IgE levels. Therefore, our study suggests that consumption of EW might not be related to the risk of food allergy, but could be an excellent candidate for the maintenance of physiological homeostasis.

Study on the hemodynamics in the cows diseased with mastitis (유방염에 이환된 젖소에 있어서 혈액 동태학에 관한 연구)

  • Hemodynamic values were assessed in the cows diseased with mastitis. Hemodynamic testes were performed for white blood cell(WBC), red blood cell(RBC), packed cell volume(PCV), hemoglobin concentration, monocyte, eosinophil, neutrophil, lymphocyte, prothrombin time, activated partial thromboplastin time, thrombin time, fibrinogen, platelet concentrations, antithrombin-III and plasminogen activities. Significant alterations were observed in the mean values of most analytes. The numbers of monocytes, eosinophil, and neutrophil, and prothrombin time were increased while the number of lymphocyte, activated partial thromboplastin time, thrombin time, fibrinogen concentration, plasminogen activity and platelet concentration were decreased. The number of RBC, PCV, hemoglobin and antithrombin-III activity were unchanged compared with normal mean values. These data indicated that activation of hemodynamic mechanisms was initiated either directly by the endotoxin-releasing or indirectly by the inflammatory mediators released by response to etiologic agents. We suspected that the changes of hemodynamic values in the cows diseased with mastitis were very similar to those of experimental endotoxin-induced mastitis.

Effect of Cyclophosphamide Administration after Stimulation With Phytohemagglutinin on Immune Response in Mice (Phytohemagglutinin 자극후(刺戟後) Cyclophosphamide 투여(投與)가 마우스의 면역반응(免疫反應)에 미치는 영향(影響))

  • This study was undertaken to elucidate the mechanism of the cyclophosphamide(CY)-induced potentiation of cell-mediated immune response by observing the effect of the phytohemaggllutinin(PHA) treatment before the CY administration into mice. Cy administration reduced the circulating white blood cells especially lymphocyte. PHA pretreatment before CY administration enhanced the depressing effect of CY administration on white blood cells. CY administration suppressed both the antibody formation to sheep red blood cells(SRBC) and rosette formation on the spleen cells with SRBC severely. On the other hand, CY administration potentiated the delayed-type hypersensitivity(DTH) strongly. Injection of PHA into mice slightly inhibited both the antibody formation and the DTH. PHA pretreatment before CY administration into mice suppressed not only humoral immune response but also cell-mediated immune response and the degrees of suppression were most remarkable when the PHA pretreatment was performed 5 days before CY administration. This depression of DTH caused by PHA pretreatment before CY administration may be the result that PHA stimulation make the helper cell sensitive to CY. The potentiation of cell-mediated immune response by CT may be due to the destruction of CY-sensitive suppressor T cell.

  • 백혈구는 형태상으로 임파구, 중성구, 반핵구 등 7~8 종의 정상적인 백혈구 종류가 있으며 비정상적인 백혈구는 변형으로 인하여 수 십 가지가 되어 분류시 많은 어려움이 있다. 백혈구는 질환에 대한 많은 정보를 가지고 있어 질병 유무 및 상태 판단에 절대적으로 필요한 검사로서 현재는 전문가에 의해 백혈구 크기, 색상, 내부 핵유무, 핵의 모양 및 boundary 모양 등을 개인적 판단 기준으로 검사하고 있어 많은 어려움이 있다. 이에 질환별 백혈구 형태 분류 알고리즘과 이에 따른 백혈구 영상 정보 확보 및 검색 시스템을 설계 구현하였다.

  • Objective: A case report on the Leukopenia Patients Using Ortho-Cellular Nutrition Therapy (OCNT) Methods: A Korean female in her 50s suffering from the rare side effect of leukopenia after taking athlete's foot medicine (Terbinafine HCl) Results: The results revealed that the white blood cell count, which had dropped before the OCNT, had improved to a normal level. Conclusion: The application of the OCNT can help change health-related figures in patients with relevant problems.

Influences of Plant Extract Supplementation on Performance and Blood Characteristics in Weaned Pigs

  • One hundred and twenty crossbred pigs [(Duroc$\times$Yorkshire)$\times$Landrace] were used to determine the effects of plant extract (PE) supplementation on performance and blood characteristics in weaned pigs fed a corn-dried whey-SBM based diet. Treatments were 1) NC (antibiotic free basal diet), 2) PC (NC diet+100 ppm apramycin and 100 ppm oxytetracycline), 3) PE 0.1 (NC diet+0.1% plant extract), 4) PE 0.2 (NC diet+0.2% plant extract) and 5) PE+AB (PC diet+0.1% plant extract). Through the entire experimental period, ADG of pigs fed PC (300 vs. 281 g/d), PE 0.2 (310 vs. 281 g/d) and PE+AB (306 vs. 281 g/d) diets was higher than that of pigs fed NC diet (p<0.05). However, no differences were found among the treatments for ADFI and gain/feed. At day 2 after the onset of the experiment, fecal consistency score of pigs fed PC, PE 0.1, PE 0.2 and PE+AB diets was lower than that of pigs fed NC diet. There were no significant differences in red blood cell, white blood cell, lymphocytes, neutrophils and monocytes concentrations of blood among the treatments. In conclusion, PE can be used to replace antibiotics in diets for weaned pigs without negative affects on performance. Optimal PE levels seemed to be 0.2% and the results obtained point out to a synergic effect of the combination of PE and antibiotic on performance in weaned pigs.
