  • Microfluidics and BioMEMStechnology has increasingly been used as a tool for studying small volumes oftissue and even individual cells. One of the most important benefits ofmicrofluidic technology is the potential to build devices that analyze and sortmammalian cells. The "sorting problem" typically requires that a fewcells be selected and isolated from a larger population of hundreds, thousandsor even millions of other cells. For example, cancer tumor cells may resideamong a large population of healthy cells, but it would be of great interest toidentify, isolate and study only the cancer cells. In another application, onemay want to determine the number of white blood cells within a sample of blood.We have developed microfluidic devices that enable researchers to select cellsfrom a population by a variety of methods, including antibody staining,dielectrophoretic selection, and physical size selection. These devices haveapplications in cancer research where cancer cells must be identified fromnormal tissue, but where only small samples of tissue are available. In thistalk, we will present some of our microfluidic cell sorting devices, discusstheir physical principles, and their use in biological applications.

과량의 부추 생즙 투여가 소형견의 혈액상에 미치는 영향 (Hematological Effect of Administration Excessive Amount of the Korean Native Chinese Chive Extracts in Small Dogs)

  • This study was conducted to observe the hematological effect of the small dogs when administrated excessive amount of the Korean native chinese chive(Allium tuberosum Rottler) extracts for a short period. In this experiment, clinically healthy dogs(n = 10, average weight = 4.17 kg) were used. They were inserted stomach tube and administrated the extracts(5ml/kg) for 7 days. The followings are the result of this experiment. The red blood cell counts, hemoglobin concentration and mean corpuscular hemoglobin concentration were significantly decreased. (P<0.05) The packed cell volume was slightly decreased. The mean corpuscular volume and methemoglobin concentration were significantly increased.(P<0.05) The white blood cell counts and reduced glutathione were slightly increased. Consequently, We determined that administrated excessive amount of the Korean native chinese chive extracts induced hemolytic anemia.

양파로부터 추출한 천연염료의 피부자극성 시험 (Skin Irritation of Natural Dyes Extracted from Onion (Allium cepa))

  • This study was conducted to investigate the skin irritation by transdermal administration of the three dyes. These dyes were originated from onion by using extraction method. By the order of extraction from onion, A-dye was obtained from onion by using water at 90-100$\circ$C. B-dye was extracted from A-dye with ethylacetate. After ethylacetate extraction from A-dye, the lower layer named as C-dye. Twenty-four New Zealand white rabbits were divided into three groups. The each groups was consisted of two subgroups according to high dose (extracted dyes) and low dose (the 100-fold dilutions of A-, Band C-dye). In primary skin irritation test of male New Zealand White rabbits, body temperature and weights were not significantly changed and blood cells were positioned in normal blood cell ranges of health rabbits. Primary irritation index was "0" in the test and control sites of all animals used in this study. By the results obtained in the present test, all dyes were evaluated as a non-irritant on the basis of the criteria of Draize.of Draize.

Vitamin A가 마우스의 면역반응에 미치는 영향 (Effect of Vitamin A on the Immune Response in Mice)

  • The effect of vitamin A on the immune response in ICR mice was studied. The effects were evaluated by immuno organ weight, peripheral circulating white blood cells, HA and HY titer, peritoneal exudate cells, RFC, Arthus reaction and DTH in mice. The spleen of mice was significantly hypertrophied by deficiency or over doses of vitamin A as compared with control group (50IU/kg). Arthus reaction, RFC and peritoneal exudate cells were sharply decreased according to the increase of vitamin A doses. The number of white blood cell was decreased according to the increase of vitamin A doses, but in the case of vitamin A 50,000 IU/kg treated group, it was significantly increased. These results suggest that deficiency or over dose of vitamin A decrease humoral and cellular immune response.

과량의 미나리 생즙 투여가 HK phenotype 진도견의 혈액상의 미치는 영향 (The Change of Blood Properties of HK Phenotype Jindo dogs Administrated with Excessive Amount of Water Celery Extracts)

  • This study was conducted to investigate changes in blood properties of high potassium (HK) phenotype Jindo dogs (15kg$\pm$2kg) after daily oral administration with water celery extracts (10 ml/kg) for 7 days. Blood samples were collected for three days in a row before administration of water celery extracts. After water celery extracts administration, blood samples were collected at 3h, 6h, 9h and then on daily basis until day 10 post administration (PA). At day 15, final sample was collected. Blood samples were analyzed on the basis of red blood cell (RBC), white blood cell (WBC), packed cell volume (PCV), hemoglobin (Hb) concentration, mean corpuscular volume(MCV), mean corpuscular hemoglobin concentration(MCHC), gluthathione concentration(GSH) and met-hemoglobin(Met-Hb) concentration. The significant changes (p<0.01, p<0.05) of RBCs were shown at 3 h to day 5, and days 7 and 9 after administration. PCV values were decreased form 3 h to day 10 after administration. Mean Hb concentration showed significant increase as 3 h to day 3, and day 6 to day 9 after administration. The significant changes (p<0.05) of WBCs were shown at 9 h and day 1 after administration. The increased numbers of MCV were detected at days 6 to 9 after administration (p<0.05, p<0.01). The significant changes of MCHC were shown at 9h and day 1 after administration. The significant increases (p<0.01, p<0.05) of GSH concentration were detected at days 1, 6 and 7 after administration. In Met-Hb concentration, the significant increases (p<0.05) occurred at only 9h and day 7 after administration, The significant increases (p<0.01, p<0.05) of reticulocyte were detected at days 2, 4, 5, 6 and 7. Data from blood samples collected at day 15 after administration showed that all of blood analysis results returned to normal level, compared to controls.

The Effects of Heat Application on the Immune Activities of the Human Body

  • The purpose of this study was to determine the effects of heat application on the immune activities of the human body. To exam, furthermore, the immune effect from the healthy volunteer(male:15, female:15) by monitoring changes of immune substances such as various leukocytes[total white blood cell(WBC), eosinophil, neutrophil, basophil, monocyte, and lymphocyte], a comparative study with warm water immersion($40.8{\pm}0.3^{\circ}C$) and infrared(250W) was carried out. The plasma analysis showed that the count of white blood cell, eosinophil, and neutrophil were elevated in warm water immersion- or infrared. stimulated group compared with control group. However, the count of basophil was decreased in both warm water immersion- and infrared-stimulated group than control group. Therefore, these results suggest that the thermostimulation improved immune activity.

가미자운고(加味紫雲膏)가 mouse의 피부화상 치료에 대한 분자생물학적 효과 및 기전연구 (Effects of Gamijaungo on the burn mice model and the study of hematologic, pathologic and molecular mechanism)

  • Objective: The aim of this study was to investigate the wound healing effect of herbal ointment, Gamijaungo, on the burn-induced model. Reports about Gamijaungo on the wound healing effect by local application in mice model or human study have published in the several domestic or internationally, but most are anecdotal and lack solid scientific evidence. Method: We observed the morphologic and histologic changes in the burn-induced mice model. we counted white blood cell and platelet changes. we confirmed VEGF, PI3K and pAkt protein expression by Western blot analysis. Result: In this study, we observed that Gamijaungo showed strong wound healing effects in the morphologic and histologic changes in the burn-induced mice model. Also we found that the significant changes of white blood cell and platelet changes by the treatment of Gamijaungo. In molecular mechanism, we got the strong positive effect by Gamijaungo treatment on angiogenesis, a key process in the formation of the granulation tissue during wound healing. Conclusion: These findings suggest the potential use of Gamijaungo as a therapeutic in thermal burn-induced skin injuries.

  • Objectives : This study was carried out to analyze the trends of clinical studies of allergic contact dermatitis published in the Journal of Korean Medical Ophthalmology & Otolaryngology & Dermatology. Methods : We studied 10 research papers on allergic contact dermatitis published in JKOOD. We analyzed about experiment methods, pharmacodynamic effects, therapeutic effects in published papers. Results : 1. The number of searched journal is 10 papers. 2. All experiments was performed using DNCB primed mouse. To demonstrate therapeutic effect of drugs, previous researches observed dermatological and histopathogical conditions, erythema, melasma, white blood cell count, Lymphocyte ratio to white blood cell. 3. Most of drugs have heat-clearing and detoxifying effects. 4. Every drugs have positive effect in treating allergic contact dermatitis. Conclusions : Figuring out how experiment proceeded and tendency of drugs in researching allergic contact dermatitis is significance of this paper. Further research about drugs and their treating methods should be carried out to improve curative effectiveness for allergic contact dermatitis.

한국산 잣기름이 정상토끼 및 동맥경화증 유발 토기의 적혈구막 지방산조성에 미치는 영향 (Effect on Fatty Acid Composition of red Blood Cell Membranes of supplement of Korean Pinenut Oil, rich in 5-Olefinic acids, in Normocholesterolemic and Atherosclerotic New Zealand White Rabbits)

  • The present study was carried out in normal and atherosclerotic New Zealand white(NZW) rabbits, to evaluate the effect of dietary supplementation with Korean pinenut oil, on the total fatty acid composition of red blood cell membranes. Erythrocyte fatty acids of chain lengths ranging fro 12 : 0 to 22 : 6$\omega$3 were identified. In study I, palmitic acid(16 : 0) was found to be the main erythrocyte membranes fatty acid, followed in turn by stearic(18 : 0), oleic(18 : 1), and linoleic acids(18 : 2$\omega$6). There was a large increase in the proportion of 20 : 4$\omega$6 in 10% pinenut oil group on days 40 and 80(compared with control), although the increase was not statistically significant. Most erythrocyte fatty acids were less affected by the types or levels of oils supplemented for periods fo 40 and 80days. In study II,at the end of the dietary treatment, the levels of 12 : 0, 14 : 0, 15 : 0 and 18 : 0 were significantly decreased in the 5% soybean oil/cholesterol group, whereas those of 12 : 0, 14 : 0 and 18 : 0 were significantly decreased, but that of 18 : 2$\omega$6 was significantly increased in the 10% soybean oil/cholesterol group, compared to the cholesterol group. No significant differences in fatty acid composition were seen between the pinenut oil/cholesterol group, whereas the 10% soybean oil/cholesterol group had significantly elevated 18 : 2$\omega$6, compared to the cholesterol group. The proportion of 18 : 4$\omega$6 was significantly raised in the 5% soybean oil/cholesterol and 5% pinenut oil/cholesterol group, and the content of 16 : 1$\omega$7 was significantly lowered in the 5% and 10% soybean/cholesterol groups, compared to the cholesterol group at 10 weeks supplementation. In studies I,II and III, the total proportions of saturated fatty acids were not significantly affected by the types of levels of oils supplemented throughout the study period.

Immuno-enhancement effects of Korean Red Ginseng in healthy adults: a randomized, double-blind, placebo-controlled trial

  • Background: Most clinical studies of immune responses activated by Korean Red Ginseng (KRG) have been conducted exclusively in patients. However, there is still a lack of clinical research on immune-boosting benefits of KRG for healthy persons. This study aims to confirm how KRG boosts the immune system of healthy subjects. Methods: A total of 100 healthy adult subjects were randomly divided into two groups that took either a 2 g KRG tablet or a placebo per day for 8 weeks. The primary efficacy evaluation variables included changes in T cells, B cells, and white blood cells (WBCs) before and after eight weeks of KRG ingestion. Cytokines (TNF-α, INF-γ, IL-2 and IL-4), WBC differential count, and incidence of colds were measured in the secondary efficacy evaluation variables. Safety evaluation variables were used to identify changes in laboratory test results that incorporated adverse reactions, vital signs, hematological tests, blood chemistry tests, and urinalysis. Results: Compared to the placebo group, the KRG intake group showed a significant increase in the number of T cells (CD3) and its subtypes (CD4 and CD8), B cells, and the WBC count before and after eight weeks of the intake. There were no clinically significant adverse reactions or other notable results in the safety evaluation factors observed. Conclusion: This study has proven through its eight-week intake test and subsequent analysis that KRG boosts the immune system through an increase in T cells, B cells, and WBCs, and that it is safe according to the study's safety evaluation.
