Quality Characteristics and Changes in GABA Content and Antioxidant Activity of Noodle Prepared with Germinated Brown Rice (발아현미 첨가에 따른 국수의 제조 특성과 GABA 함량 및 항산화 활성 변화)

  • This study was to investigate the quality characteristics of noodles prepared with germinated brown rice and rice bran, and to determine the changes in GABA content and antioxidant activity of the noodles. These noodles were evaluated for cooking properties (cooking loss, swelling index, water holding capacity), total polyphenols, antioxidant activities (radical scavenging of ABTS and DPPH), and GABA contents. The addition of germinated rice bran in the range of 5, 10, and 15% showed a significant influence on cooking loss, although no effect on swelling index was observed. The raw and cooked noodles containing germinated brown rice and rice bran contained more GABA contents (2751.6~4176.7 and 5522.0~9617.8 nmol/20 g of fresh noodle, respectively) than those of non-germinated brown rice and rice bran. Noodles made with germinated brown rice and rice bran had similar total polyphenolics contents and antioxidant activities compared to noodles made with non-germinated brown rice and rice bran. Overall, the germinated brown rice could be used as a good ingredient to increase the nutritional value and antioxidant properties of wheat flour noodle without affecting the cooking properties.

The Fruiting Body Formation of Oudemansiella radicata in the Sawdust of Oak (Quercus variabilis) Mixed with Rice Bran

  • To screen additives and their mixed ratio suitable for the mycelial growth and fruiting body formation of Oudemansiella radicata in the oak sawdust, additives such as rice bran, fermented soybean powder and wheat bran were used. Generally, the mycelial growth of O. radicata has been stable on oak sawdust mixed with rice bran of $5{\sim}20%$. In case that O. radicata was cultured for about 30 days at $22{\pm}1^{\circ}C$ under the illumination (350 lux) of 12 hours and moisture condition of $90{\pm}5%$, the primordia have been formed gradually from red-brown crusts covering the surface of oak sawdust media. Based on the experimental results from 9 strains of O. radicata, fruiting bodies were produced widely on oak sawdust medium mixed with rice bran of 5 to 30%. Even though fruiting bodies of O. radicata have been produced well on oak sawdust media mixed with rice bran, fruiting bodies of O. radicata were produced intensively on oak sawdust media mixed with rice bran of 10%. Therefore, this result will provide a basic information for commercial production of fruiting body of wild O. radicata. This result is the first report associated with an artificial fruiting body formation of O. radicata in Korea.

Performance and carcass characteristics of Australian purebred and crossbred lambs supplemented with Rice Bran

  • Background: This study examined the effects of dietary supplementation with rice bran, sire breed and gender on live animal performance and carcass characteristics in Australian crossbred and purebred Merino lambs. Methods: Forty-eight lambs balanced by sire breed (Dorset, White Suffolk, Merino) and gender (ewe, wether) were randomly allocated into three dietary supplementation groups (Control- 24 lambs fed wheat/barley-based pellets, Low- 12 animals fed a 50/50 ratio of wheat-based/rice bran pellets, and High- 12 lambs fed rice bran pellets). The Rice bran pellets replaced 19 % of the barley component of the feed. Animals were group-fed at the rate of 1000 g of the supplement per head per day with ad libitum access to lucerne hay as the basal diet and water. The duration of the feeding trial was 49 days with an initial 21-day adjustment period. Results: Sire breed differences were evident for initial (p < 0.0002) and final (p < 0.0016) liveweights, hot carcass (p < 0.0030) and cold carcass (p < 0.0031) weights, as well as dressing percentage (p < 0.0078), fat thickness (p < 0.0467), yield grade (p < 0.0470) and rib eye area (p < 0.0022) with purebred Merino under-performing compared to the crossbreds. Concentrate feed conversion efficiency, costs per unit of liveweight gain and over the hooks income were comparable between treatments regardless of the observed trend where the high supplementation group tended to show lower feed intake (745.8 g/day) compared to both the control (939.9 g/day) and low supplementation groups (909.6 g/day). No significant differences (p > 0.05) were observed between treatments for live animal performance, carcass characteristics, gender and their second-order interactions. Conclusions: Results indicate that Rice bran can be utilised as a cost-effective supplementary feed source in genetically divergent sheep over a 49-day feeding period without detrimental effects on overall live animal performance or carcass characteristics.

Studies on the optimal conditions for the high yield in the production of Ganoderma lucidum (영지버섯 다수확 생산을 위한 최적조건에 관한 연구)

  • The results were as follows; 3 additives(rice bran, wheat bran and soybean bran) to the Quercus dentata sawdust were applied at 10, 20 and 30%. High yield was recorded at 20% of rice bran and wheat bran and 30% of soybean bran. Among various sawdust media, the cultivation with the Quercus dentata sawdust alone showed the most favored growth index, such as 22 days of full mycelial growth, 11 days of pinheading from spawning and 23.5g per $1,200m{\ell}$ bottle. In the size of bottles, the lowest infection ratio was 5 % at $1,200m{\ell}$ bottle. In the growth characteristics with the bottle size, Reishi at $1,600m{\ell}$ bottle showed best result when it is observed for 13~15 days in pinheading from spawing, 36~37 days in fruit bodies growing and 26.7~29.3g yield per bottle. The highest yield per area was 6.4kg at horizontal bed with 218 bottles per $3.3m^2$. The Reishi produced in $1,600m{\ell}$ bottle with horizontal cultivation had less number of fruit bodies and best yield, thus it is determined to be the most economic method.

The Effect of Bifidobacteria and Various Oligosaccharides Consumption on the Risk of Colon Cancer in Rats

  • This study examined the effect of viable bifidobacteria and non-digestible carbohydrates on the cecal pH, colonic neoplastic lesion (aberrant crypt) and proliferating cell nuclear antigen (PCNA) labeling index in carcinogen-treated mts. Animals received s.c. injection of dimethylhydrazine (DMH) (15 mg/kg body weight) twice 3 days apart. Three days after the second carcinogen administration, the treatments were begun. 1he treatments were basal diet (AIN-76) with skim milk (Basal/skim), or the following diets with daily gavage of $10^8$ bifidobacteria: basal (Basal/bifido), $2\%$ fructo-oligosaccharide (FOS/bifido), $2\%$ soybean oligosaccharide (SBO/bifido), $2\%$ wheat bran oligosaccharide (WBO/bifido) and $8.4\%$ wheat bran (WB/bifido). After 4 weeks of treatment, cecal pH was measured using a pH probe. The number of aberrant crypt (AC), aberrant crypt foci (ACF) and crypt multiplicity were enumerated and colonic PCNA labeling index was determined using immunohistochemistry. Cecal pH was significantly reduced in SBO/bifido and FOS/bifido groups compared to control group. However, there were no significant differences in either number of AC or rates of cell proliferation as shown by PCNA labeling index among the groups, although mts fed FOS/bifido reduced the numbers of ACF compared to Basal/skim group. The SBO/bifido group did not reduce the number of ACF or PCNA labeling index. Also, other oligosaccharides did not reduce the risk of colon cancer compared to control group. The concomitant reduction of cecal pH and number of ACF suggest that the combination of bifidobacteria and FOS may reduce the risk of colon cancer.

Studies on the Protopectinase Produced by Verticillium sp. (Part 1) Optimum Conditions for the Protopectinase Production and Utilization (Verticillium sp.가 생산하는 Protopectin 용해효소에 관한 연구 (제 1 보) Protopectin 용해효소의 생산조건 및 이용)

  • A fungus with the highest protopectinase productivity was selected among 205 strains isolated from the soil and identified as a Verticillium sp. The Verticillium sp. was cultivated on wheat bran and the crude extruct of its culture medium showed the highest protopectinase activity on the following conditions: 3 days of cultivation time, 27$^{\circ}C$ of cultivation temperature, 1.2 $m\ell$/g wheat bran of water content, and reinforcement of ammonium nitrate and calcium chloride at the concentration of 0.5 and 0.02%, respectively. The optimum conditions for pectin production from Citrus peel pulp by the protopectinase were consequently obtained as follows: 20$m\ell$/g of liquid volume-to-pulp weight ratio, 4$0^{\circ}C$ of reaction temperature, and 4 of reaction pH. The higher the enzyme concentration, the better the yield of pectin and the shorter the reaction time. Total 45.6mg of pectin/g peel was produced by 1 hour reaction at the enzyme concentration of 10.5 units/$m\ell$. Molecular weight of the pectin produced by the enzyme was estimated to be about 62,000 by Smit and Bryant's method.

Isolation of Aspergillus niger K-25 Prroducing Acid-stable ${\alpha}-amylase$ (내산성(耐酸性) 아밀라제를 생산하는 Aspergillus niger 균주의 분리)

  • One strain of Aspergillus niger K-25 producing an acid-stable ${\alpha}-amylase$ was isolated from the soil. The optimum culture conditions were investigated. The production of the acid-stable ${\alpha}-amylase$ was enhanced when the strain was incubated in a medium containing soluble starch 3.5%, peptone 2%, $KH_2PO_4$ 0.5%, $MaSO_4{\cdot}7H_2O$ 0.25% and $FeCI_3$ 1.0% at pH 3 for 7 days. However, higher activity of acid-stable ${\alpha}-amylase$ was demonstrated on wheat bran culture. Amylase production was doubled when A. niger K-25 was incubated on the wheat bran supplemented with fumaric acid buffer (pH 3).

Production of the Convertible Enzyme of Ginsenoside $Rb_1$ by Rhizopus japonicus (Rhizopus japonicus 에 의한 Ginsenoside $Rb_1$ 전환효소의 생산조건)

  • In ginseng saponin, $ginsenoside-Rb_1$ was contained the most abundantly. But ginsenoside-Rd which is similar to ginsenoside $Rb_1$ in structure was known to be superior to $ginsenoside-Rb_1$ pharmaceutically. A strain of Rhizopus japonicus is able to produce the convertible enzyme which can convert selectively $ginsenoside-Rb_1$ to ginsenoside-Rd without the change of any other ginsenoside. The strain can produce the most enzyme after 5 day-culture on wheat bran medium. The enzyme production was promoted best efficiently by addition of red ginseng powder in ginseng products, xylose in sugars, laminarin in polysaccharides, naringin in flavonoids, and potassium nitrate in nitrogen substrates.

Effect of Dietary Fiber Enrichment and Different Cooking Methods on Quality of Chicken Nuggets

  • The effect of dietary fiber enrichment (wheat bran) and cooking methods (oven, steam and microwave) on functional and physico-chemical properties of raw nuggets formulation as well as nutritional, color and textural properties of chicken nuggets were analyzed in this study. Among different cooking methods used for nuggets preparation, steam cooked nuggets had significantly (p<0.05) higher water holding capacity (56.65%), cooking yield (97.16%) and total dietary fiber content (4.32%) in comparison to oven and microwave cooked nuggets. The effect of cooking methods and wheat bran incorporation was also noticed on textural properties of the nuggets. Hardness, firmness and toughness values of oven and steam cooked nuggets were significantly (p<0.05) higher than microwave cooked nuggets. Among nuggets prepared by different cooking methods, cohesiveness of microwave cooked nuggets was found to be significantly (p<0.05) highest, whereas, oven cooked nuggets had significantly (p<0.05) highest gumminess and chewiness values. Steam cooked nuggets were found to be better among all nuggets due to their higher cooking yield and dietary fiber content.

In Situ Ruminal Digestion Kinetics of Forages and Feed Byproducts in Cattle and Buffalo

  • The relative disappearance and rate of degradation of dry matter (DM) and neutral detergent fiber (NDF) of nine different feedstuffs were determined by simultaneously suspending groups of substrates, using the nylon bags, in the rumen of males of Sahiwal cattle and Nili-Ravi buffalo. The digestion kinetics of leguminous forages (Lucerne, berseem and cowpeas) and feed byproducts (cotton seed cake, wheat bran and wheat straw) did not differ between the two species. However, the DM and NDF digestibilities and rates of digestion of grasses and wheat straw were greater in buffalo than in cow bulls, indicating that buffaloes are better converters of poor quality roughages than are Sahiwal. The lag time for DM of grasses did not differ between these two species but the NDF lag time was lower in buffalo than in cows, indicating that both the rate and lag time of digestion may be reliable indicators for assessing the NDF quality.
