In vitro Glutathione Production using Mixed Cells in an Aerated Slurry Bioreactor (혼합세포를 이용한 Aerated Slurry Bioreactor에서의 in vitro Glutathione 생산)

  • Glutathione production was carried out using mixed cells of E. coli TG1/pDG7 $\alpha$ and bakers yeast in an Aerated Slurry Bioreactor. Glutathione-producing enzymes were stable for 34 hours, yielding 4.6 mM glutathione in suspension reaction. Glutahione production with high density mixed cells was studied as a function of flow rate in an Aereated Slurry Bioreactor. Glutathione concentration was higher than that in suspension reaction for 32 hours at the substrate feeding rate of 5.2 mL/hr with cell recycle in continuous Aerated Slurry Bioreactor. It was for 42 hours at 2.6 mL/hr and 22 hours at 5.2 mL/hr without cell recycle. Glutahione productivity was 25.7 mg/g wet $cell{\cdot}hr$ at the substrate feeding rate of 10.4 mL/hr with cell recycle, but 5.28 mg/g wet $cell{\cdot}hr$ at 5.2 mL/hr and 1.65 mg/g wet $cell{\cdot}hr$ at 2.6 mL/hr without cell recycle. Effective production time increased from 25 to 45 hours, by using a surfactant, tween 80. As a purfing gas, nitrogen was tested instead of air to avoid a possible oxidizing effect on glutathione-producing enzymes, resulting in the increase of effective production time to 40 hours.

Probiotic Properties of Enterococcus durans LP44 Isolated from Pigs Feces (돼지의 분변에서 분리한 Enterococcus durans LP44의 생균제로서의 특성)

  • We have screened the microorganisms from pigs feces for the development of probiotics which have acid-and bile-tolerance. Among them, a strain which was identified as Enterococcu durans LP44 was selected. Sixty Duroc$\times$Yorkshire$\times$Landrace pigs (6.30$\pm$0.04 kg average initial body weight) were used in a 14 days growth assay to determine the effects of Enterococco duran Lf44 culture on growth performance of weaned pigs. Dietary treatments included 1) DF (dry feeding),2) W (wet feeding),3) WF+5ED (wet feeding+5% Enterocucu duran LP44 culture),4) WF+ 10ED (wet feeding +10% Enterococcus durans LP44 culture). For overall periods, average daily gain in pigs fed % diet tended to increase compared to pigs fed DF diet without significant difference. Also, there was no significant difference in gain/feed. At 5 days after the onset of the experiment, pigs fed WF+5ED and WF+10ED diets were significantly increased in dry matter and nitrogen digestibilities compared to pigs fed DF and W diets (p<0.05). At 14 days after the onset of the experiment, pigs fed WF+ 10ED diet were significantly increased in dry matter digestibility compared to pigs fed DF diet (p<0.05). Also, pigs fed WF + 10ED diet were significantly increased in nitrogen digestibility compared to other treatments (p<0.05). In conclusion, supplemental Enterococcus durans LP44 was an effective means of improving ADG and nutrient digestibility.

Predation, Feeding Preference of Threadsail Filefish Stephanolepis cirrhifer on Different Life Stages of Moon Jellyfish Aurelia aurita s.l. in the Laboratory (전 생활사 단계의 보름달물해파리(Aurelia aurita s.l.)에 대한 쥐치(Stephanolepis cirrhifer)의 포식과 먹이 선호도 실험)

  • To determine the predation rate of threadsail filefish, we conducted feeding experiment of threadsail filefish, Stephanolepis cirrhifer on different life stages (medusae, ephyrae, and polyps) of Aurelia aurita s.l. in the laboratory. All size groups of threadsail filefish actively preyed on moon jellyfish Aurelia aurita s.l. and predation rate was proportional to the wet weight of threadsail filefish. Average wet weight 0.5, 1, 2.2 g of threadsail filefish preyed on average $0.77{\pm}0.51g\;fish^{-1}h^{-1}$ of medusae. Average wet weight 150 and 344 g of threadsail filefish preyed on $9.26{\pm}1.61$ and $15.89{\pm}3.67g\;fish^{-1}h^{-1}$ of medusae respectively. In ephyrae and polyps, average wet weight 3.7 g of juvenile of threadsail filefish preyed on 100 ephyrae within 10 minutes and average wet weight 2.2 g of juvenile of threadsail filefish preyed on $5.73{\pm}0.59$ polyps $g^{-1}$ of fish $h^{-1}$. Whereas adults of threadsail filefish did not prey on polyps. In feeding preference experiments to elucidate whether threadsail filefish prey on moon jellyfish in the sea, adults of threadsail filefish preyed on mostly oysters (ca. 60%) and polychaetes (ca. 80%). Medusae were consumed only 2% of total. However, juvenile of threadsail filefish preyed on ephyrae and polyps as well as mysid shrimps. Besides, polyps were consistently consumed during the experiment. Although juvenile of threadsail filefish consumed all kinds of prey, they ceased feeding at temperatures below $11^{\circ}C$, which suggests that possibility of predation on ephyrae may be very low in the sea because ephyrae are released at low temperatures below $11^{\circ}C$ from March to April. Whereas polyps are inhabitable in all seasons in Korean coastal waters, juvenile of threadsail filefish may prey on polyps in the sea. These results suggest that removing moon jellyfish at polyp stage by using juvenile of threadsail filefish is the most effective way for extermination of moon jellyfish.

Improvement of adhesion of Polyester Fabric and Knit by Corona Treatment (코로나 처리를 이용한 폴리에스테르 편·직물의 접착력 향상)

  • Polyester fabrics and knits were corona treated with different current intensity (5, 10, 15, 20 A) and feeding speed (5, 10, 15 m/min). We confirmed the surface change of polyester fabrics and knits using scanning electron microscopy (SEM) and X-ray photoelectron spectrometer (XPS). And the change of physical properties through measuring the tensile strength, dry and wet bonding strength. Thermosetting reactive polyurethane hot melt adhesive was used in the adhesion of polyester fabrics and knits. Functional groups were introduced on PET fabrics and knits surface by treating the PET fabrics and knits in air atmosphere with corona discharge, and take placed unevenness, the result adhesion was improved. When consider both dry and wet bonding strength, we think that appropriate conditions in the PET fabrics corona treatment are feeding speed 10 m/min, current intensity 15 A and appropriate conditions in the mesh type PET knits corona treatment are feeding speed 10 m/min, current intensity above 10 A.

The Comparison of Adhesion Properties on Polyester Fabric by AC and DC Corona Treatment (AC와 DC 코로나 처리에 따른 폴리에스테르 직물의 접착성질 비교)

  • Compare AC corona and DC corona treatment effects, polyester fabrics were AC corona treated with different current intensity (5, 10, 15 A) and feeding speed (5, 10, 15 m/min) as the same DC corona treatment conditions of the preceding research. We confirmed the surface change of polyester fabrics using scanning electron microscopy (SEM) and X-ray photoelectron spectrometer (XPS), and the change of physical properties through measuring the dry and wet bonding strength. The surface changes of polyester fabrics by treating in air atmosphere with AC corona discharge are shown similar tendency with DC corona discharge. Generally dry bonding strength were increased with increasing current intensity and feeding speed, but wet bonding strength were increased with increasing current intensity and decreasing feeding speed in both AC and DC corona treatment. When the current is 20 A, carbonization occurs in DC corona discharge but carbonization does not occur in DC corona discharge.

  • In this paper, we described the characterization of High-Tc Superconducting(HTS) microstrip antenna using non-radiating edge feeding technique and reported the microwave properties of HTS antennas with temperature. To do this, we prepared the $YBa_2Cu_3O_{7-x}$ superconducting thin film on MgO substrate using pulse-laser deposition techniques. The HTS microstrip antenna using non-radiating feeding technique was fabricated using chemical wet-etching. Then it was compared with identical antenna patterned with evaporated gold. The diverse measured results have been reported in terms of the input impedance, resonant frequency and return loss. In additional, at around the critical temperature, the effect of kinetic inductance which affect the resonant characteristic of the HTS microstrip antenna was reported.

Effect of Cutting off Processing SCM415 on Surface Roughness and Thickness of Materials (SCM415의 절단가공이 표면조도와 재료 두께에 미치는 영향)

  • This study focused on how it affects the surface roughness of work piece in cutting SCM415 steel, widely used steel in industry, by TiCN and TiN tools. Following conclusion was drawn from several experiments. The surface roughness of heat treated workpiece was better than that of non heat-treated materials. Moreover, the roughness of surface roughness(Ra 0.25) on feeding rate of 0.05 was better when it was in wet process, rather than dry process. As the feeding rate increases, TiCN coating tool shows better roughness of surface than TiN tool. Also, in heated treatment, TiCN coating tool shows the least straightness dimension deviation at feeding rate of 0.05, 0.15mm/rev, and concave-like R shape appears by the feeding rate orders of 0.05, 0.15, 0.1, 0.125 and 0.075mm/rev.

Effects of Feeding Methods of Total Mixed Ration on Behavior Patterns of Growing Hanwoo Steers

  • A study was conducted to investigate the effects of methods of feeding a total mixed ration (TMR) on behavior patterns of growing Hanwoo steers. A total of 15 growing steers (13 months old) were assigned to the control (fed roughage and concentrate mix separately), TMR1 (fed restricted TMR), and TMR2 (fed TMR ad libitum) groups. Individual behaviors of steers were observed for 48 hours. Compared with the control, feeding restricted TMR (TMR1) resulted in short eating time, long ruminating time, short chewing time, high frequencies of defecation, urination, and drinking of water, great numbers of boluses and chews, long ruminating time per bolus, low feed value index, high eating and chewing efficiencies (p<0.05). Compared with feeding restricted TMR (TMR1), feeding TMR ad libitum (TMR2) resulted in 1.2 kg more daily feed DM intake, long eating and chewing times, short resting time, great frequencies of defecation, urination and drinking of water, more numbers of boluses and chews, long ruminating time per bolus, low feed value index, low eating and high ruminating efficiencies (p<0.05) and similar chewing efficiency (p>0.05). Considering all these results, the wet TMR feeding system induced generally more desirable eating and ruminating behaviors of growing Hanwoo steers, but made the barn floor wetter due to more defecation and urination.

  • Ultra-fine copper powders with particle size about 150 nm were synthesized from copper hydroxide slurry by wet method using hydrazine as reduction agent and several sur factants at below $80^{\circ}C$. The particle size distribution and dispersion of synthesized powders as function of temperature, feeding rate of reduction and especially, sur factants were character ized by XRD, BET, PSA and SEM by this process.

Study on Cone Type Thresher (I) (원추형(圓錐型) 탈곡기(脱糓機)에 관(關)한 연구(硏究))

  • The major limiting factor on the determination of combine capacity is the frequent occurence of clogging over the some parts of machine when the crop is wet in the case of Japanese self-feeding type combine. And in the case of American conventional combine having big separating parts, the great grain loss and damage occur when the machine is used for rice harvesting. This experiment was carried out to develop the new type threshing and separating equipment. Proto-type thresher which consist of a conical threshing drum and a conical separating sieve rotating around the threshing cone was constructed and tested. In the case of 800 rpm of threshing cone speed, average threshing loss was below 1 percent, separating loss was about 1 percent, grain damage was about 0.4 percent, and average total power required was about 2.6 PS. This design has some problems such as higher power required or wrapping problems under the conditions of feeding long damp straw. But, compared with the conventional combine or thresher, this machine certainly has some potentials for this approach to combine development. The crop feed rate must be increased through improvement of the feeding portion of the threshing cone. And it is required to investigate further about some parameters causing wrapping phenomena.
