The Properties of Yellow Layer Cakes Made by Different Substituting Levels of Waxy Maize Starch for Shortening (Waxy Maize Starch의 대체율을 달리하여 제조한 옐로우 레이어 케이크의 특성)

  • Waxy maize starches are inherently stable in soluble status and can be chemically modified to improve stability along with heat, acid and shear resistance. This study was carried out to investigate the effect of theological and sensory characteristics of the yellow layer cake made by adding different levels of waxy maize starch as a fat substitute for shortening. By increasing the substitution level of waxy maize starch for shortening, the specific gravity of cake batter increased and the viscosity decreased. The microstructures of cake crumb observed by the scanning electron microscope were not different significantly, and the size of air cells and fat particles also were not substantially decreased by increasing fat substitution level. The texture profile analysis using texture analyzer decreased by increasing the different substituting levels of waxy maize starch. Among various sensory properties, the color value of layer cake increased by increasing the level of waxy maize starch. However, the appearance, flavor, taste, texture and overall preference decreased.

Analysis of Genetic Diversity and Population Structure for Core Set of Waxy and Normal Maize Inbred Lines using SSR Markers (SSR 분자마커를 이용한 찰옥수수 및 종실용 옥수수 자식계통들의 핵심집단에 대한 유전적 다양성 및 집단구조 분석)

  • Maize is divided into two types based on the starch composition of the endosperm in the seed, normal maize(or non-waxy maize) and waxy maize. In this study, genetic diversity and population structure were investigated among 80 waxy maize and normal inbred lines(40 waxy maize inbred lines and 40 normal maize inbred lines) using 50 SSR markers. A total of 242 alleles were identified at all the loci with an average of 4.84 and a range between 2 and 9 alleles per locus. The gene diversity values varied from 0.420 to 0.854 with an average of 0.654. The PIC values varied from 0.332 to 0.838 with an average of 0.602. To evaluate the population structure, STRUCTURE 2.2 program was employed to confirm genetic structure. The 80 waxy and normal maize inbred lines were separated with based on the membership probability threshold 0.8, and divided into groups I, II and admixed group. The 13 waxy maize inbred lines were assigned to group I. The 45 maize inbred lines including 7 waxy maize inbred lines and 38 normal maize inbred lines were assigned to group II. The 22 maize inbred lines with 20 waxy maize inbred lines and 2 normal maize inbred lines were contained in the admixed group. The cluster tree generated using the described SSR markers recognized three major groups at 31.7% genetic similarity. Group I included 40 waxy maize inbred lines and 11 normal maize inbred lines, and Group II included 27 normal maize inbred lines. Group III consist of only 2 normal maize inbred lines. The present study has demonstrated the utility of SSR analysis for the study of genetic diversity and the population structure among waxy and normal maize inbred lines. The information obtained from the present studies would be very useful for designing efficient maize breeding programs in Maize Experiment Station, Kangwon Agricultural Research and Extension Services.

The Stable Production of Organic Seed to Distribute Certified Seed of Waxy Corn Hybrid

  • In order to produce the organic foods in accord with international standard, organic seeds should be used in organic farming. This study was conducted to establish the stable production of organic seed of waxy com by examining the growing characteristics, seed yield, and the economy for seed production by organic farming. The optimal sowing timing for organic seed production of waxy com hybrid was within 10 days of the $1^{st}$ of May with yield of 88~90% of conventional seed production. The optimal planting density was 41,600 plants/ha ($80{\times}30$ cm) for organic seed production of waxy com. The weight of 100 seeds and seed productivity increased at the planting ratio of 2:1 mother plant:male plant. Growth and seed production were improved by removing male plant at 7~10 days after silking. Organic fertilizer (mixed oil cake) was applied at a rate of 4~6 Mg/ha before sowing. Black plastic mulching was used for weed control. In addition, sex pheromone trap and bio-control agents were used for safe pest control and low labour cost.

Effect of Hydrothermal and Enzymatic Treatments on the Physicochemical Properties of Waxy Maize Flour (열수 및 효소 처리에 의한 찰옥수수가루의 물리화학적 특성)

  • Physicochemical properties of waxy maize flours prepared by hydrothermal and enzymatic treatments were evaluated. Waxy maize flours were hydrothermally treated using heat-moisture treatment (HMT) and annealing (ANN) and enzymatically treated using commercial enzymes (cellulase, proteinase, and pectinase). The HMT-modified waxy maize flours had low water absorption index (WAI), melting enthalpy, viscosity, and crystallinity. However, ANN-modified and enzymatically modified waxy maize flours had high WAI, melting enthalpy, and viscosity. X-ray diffraction analysis of ANN-modified and enzymatically modified waxy maize flours revealed a typical A-type pattern and displayed sharper crystalline peaks than those observed for the control groups (native waxy maize flours). In contrast, the crystallinity of HMT-modified waxy maize flours were decreased by hydrothermal treatment.

Physicochemical Properties of Oxidized Waxy Maize Starches with Sodium Hypochlorite (찰옥수수 산화전분의 이화학적 특성)

  • Physicochemical properties of waxy maize starch and oxidized waxy maize starch with sodium hypochlorite $(0{\sim}60\;mg\;CL_2/g\;starch,40^{\circ}C,\;pH\;10,\;3.0\;hr)$ were studied. As sodium hypochlorite concentration was increased, the content of crude lipid and crude protein of the oxidized starch were decreased. And crude protein content and whiteness was considered to show negative regression. However, the crude ash content of the oxidized starch increased significantly with oxidation and bore a positive regression to the chlorine content. There was a progressive increase in the carboxyl content with increasing oxidant level. After pasting in hot water and cooling, viscosity of the oxidized starches were drastically lower than that of native starch . As carboxyl contents of the oxidized starch increased, the solubility and swelling power was increased. When waxy maize starch treated with 0, 1.5, 3.0 and 6.0% sodium hypochlorite, temperature of initial gelatinization of oxidized starch was shown to 65, 65, 60 and $50^{\circ}C$, respectively. The oxidized waxy maize starches also form clearer pastes. Water binding capacity of the oxidized starch decreased as the degree of carboxyl group substitution increased. Waxy maize starch has polygonal and some round granules which range from about 3.7 to $20\;{\mu}m$ in diameter. Surface appearance of the waxy maize starch became rough when oxidized with sodium hypochlorite. When homogenate of the oxidized waxy maize starch solution and corn germ oil was stored under room temperature for 24 hours, the emulsion stability was considered to depend on starch concentration and degree of substitution.

The Effect of Mild-Acid Treated Waxy Starches on the Yield of Resistant Starch (약산 처리가 찰전분의 저항전분수율에 미치는 영향)

  • The characteristics and yield of resistnat starch (RS) in waxy starches which were treated with 0.1N HCl were investigated. Hydrolysis rate of waxy starches were 0.32-0.88%. The yields of RS in waxy maize and waxy barley starches increased with acid treatment time up to 12hr and then decreased. The yield of RS in waxy rice starches increased with acid treatment up to 8hr and then decreased. The hydrolysis and yield of RS were different depending upon the varieties of starch source. Autoclaved normal maize starch showed a gel-like structure, but autoclaved normal maize starch showed crystal particle on the surface by SEM. The shape of autoclaved resistant starch was not different from acid treated ones. X-ray diffraction patterns of native and acid treated starches were all A-types. X-ray diffraction patterns of autoclaved waxy maize starches showed amorphous structures, however, those of RS showed all completely crystalline structure.

Genetic Diversity Among Waxy Corn Accessions in Korea Revealed by Microsatellite Markers

  • Knowledge of genetic diversity and of the genetic relationships among elite breeding materials has had a significant impact on the improvement of crops. In maize, this information is particularly useful in i) planning crosses for hybrid and line development, ii) in assigning lines to heterotic groups and iii) in plant variety protection. We have used the SSR technique to study the genetic diversity and genetic relationships among 76 Korean waxy corn accessions, representing a diverse collection from throughout Korea. Assessment of genetic diversity among members of this group was conducted using 30 microsatellite markers. Among these 30 microsatellite markers, we identified a total of 127 alleles (with an average of 4.2 and a range of between 2 and 9 alleles per locus). Gene diversity at these 30 microsatellite loci varied from 0.125 to 0.795 with an average of 0.507. The cluster tree generated with the described microsatellite markers recognized two major groups with 36.5% genetic similarity. Group I includes 63 inbred lines, with similarity coefficients of between 0.365 and 0.99. Group II includes 13 inbred lines, with similarity coefficients of between 0.45 and 0.85. The present study indicates that the 30 microsatellite loci chosen for this analysis are effective molecular markers for the assessment of genetic diversity and genetic relationships between Korean waxy corn accessions. Specifically, this study's assessment of genetic diversity and relationships between a set of 76 Korean waxy corn inbred lines will be helpful for such activities as planning crosses for hybrid and line development and association mapping analyses of maize breeding programs in Korea.

Morphological Studies on Ear Characteristics of Korean Indigenous Corn Lines Collected in Pusan and Kyungnam

  • In order to get abundant germplasm for develop- ing new corn varieties, major morphological characteristics of ear were evaluated with a total of 210 Korean indigenous maize lines collected from various parts of Pusan City and Kyungnam Province, Korea. The ear shape of Korean indigenous maize was mainly cone type or similar to cone type. Cylinder type or similar to cylinder type of corn was collected from the north-west mountainous. region of Kyungnam Province, whereas cone type or similar to cone type was collected from the plain region. In the kernel colors of each ear, ears with mono colored kernels were 55.7% of the total, and ears with two to four mixed colors were 44.3%. Among the mono colored ears, brown was the most abundant at 16.8%. There was an average of 12∼14 rows per ear. Kernal rows per ear varied from 8∼20. Waxy corn was predominant at 67.2%, mixed corn with waxy kernel and common kernel at 11.0%, and common corn at 21.8%.

Electron Donating Ability and Contents of Phenolic Compounds, Tocopherols and Carotenoids in Waxy Corn (Zea mays L.) (찰옥수수의 전자공여작용과 페놀성화합물, Tocopherols 및 Carotenoids의 함량)

  • The antioxidative activity measured by electron donating ability was investigated for the breeding of the highest antioxidative waxy corn (Zea mays L.) and the research for the most effective antioxidant in waxy corn. The electron donating ability was $15.5{\sim}65.0%$. The contents of phenolic compounds and tocopherols and the absorbance at 450 nm were $102.3{\sim}323.5\;{\mu}g/mL$, $15.6{\sim}144.2\;{\mu}g/mL$ and $0.047{\sim}0.206$, respectively. The mean values of electron donating ability and contents of phenolic compounds and tocopherols of four black waxy corn were comparatively high, that is, 48.7%, $267.0\;{\mu}g/mL$ and $87.0\;{\mu}g/mL$, respectively. The electron donating ability was significantly correlated with the level of phenolic compounds and tocopherols but not with the content of carotenoids.

Natural variation of functional components between Korean maize types (국내 옥수수 품종에 따른 기능성 성분의 자연 변이 분석)

  • Maize is one of the major crops consumed in worldwide, which nutrients accounts for a large amount of starch, but also functional components, and phenolic acid is known to have a high content. Maize is divided into waxy maize, sweet maize, and normal maize with its shape and use, therefore there is also a difference in nutritional composition. This study was conducted to analyze the content of functional components according to the type of maize and to produce natural variation data in consideration of environmental factors. 3 shapes of maize (waxy maize, sweet maize, and normal maize) samples cultivated in 3 regions (Suwon, Daegu, and Hongcheon) were analyzed using HPLC and GC-TOF-MS. Comparing with type through ANOVA, multivariate statistical analysis, Pearson correlation analysis, 28 components, including carotenoids and tocopherols, showed significant differences among a total of 32 components (p <0.05), 15 of them showed very significant differences (p <0.001). When comparing with regions, 15 components showed significant differences and only vanillate, syringate, C23-ol of them showed most significant differences (p <0.001). As a result of principal component analysis, cluster classification was distinguished by shape than by region, with α-carotene, cholesterol for waxy maize, vanillate and stigmasterol for sweet maize, lutein and β-carotene for normal maize had a great effect on cluster formation. It suggests that the content of functional components is more affected by genetic factors than environmental factors.
