오미자 추출물의 추출특성 및 항산화 효과 (Extraction Characteristics and Antioxidative Activity of Schiznadra chinensis Extracts)

  • 오미자 추출액의 추출 특성과 추출액의 항산화성을 검토하여 오미자의 추출 조건을 결정함으로써 오미자 유효성분의 식품 소재화를 위한 기초자료를 얻고자 하였다. 용매와의 비율을 결정하기 위해 8배, 10배, 12배, 14배의 water를 가하여 추출 수율을 비교한 결과 water와의 비율이 10배일 때 추출 수율이 가장 좋은 것으로 나타났다. 탁도는 Water 추출액에서 가장 높았으며, 추출용매의 ethanol 농도가 증가할수록 낮아졌다. 적정산도는 추출용매의 50% ethanol에서 가장 높았으나, 전체적으로 산도가 매우 낮았다. 유리당 함량은 fructose $1.24{\sim}4.42mg%$, glucose $3.65{\sim}4.71mg%$, sucrose $0.89{\sim}1.69mg%$로 glucose의 함량이 가장 높았으며, 당질의 대부분을 glucose와 fructose인 것으로 나타났다. 총 폴리페놀 함량을 나타낸 것으로 50% ethanol 추출액은 0.98%, 75% ethanol 추출액은 0.89%, water 추출액은 0.58%로 나타나 50% ethanol 추출액에서 가장 높은 함량을 나타내었으며. water에 비해 유기용매 추출시 높은 함량을 보였다. 전자공여능은 water 추출액 60.87%, 50% ethanol 추출액 57.24%, 75% ethanol 추출액 55.61%로 나타나 용매별 전자공여능 효과가 큰 차이는 나타나지 않았으며. water 추출액이 다른 용매에 비해 높은 전자공여 효과를 보여 주었다. SOD 유사환성 측정 결과는 50% ethanol 추출액에서 33.65%로 가장 높은 활성을 나타냈었으며, 50% ethanol 추출액이 다른 두 용매 보다 우수한 것으로 나타나 총 폴리페놀 함량과 전자공여능 측정 결과 와 비슷한 경향을 보여주었다.

심재, 변재, 형성층 및 사부를 포함한 뽕나무 심부 고온추출물의 항염증 활성 (Anti-Inflammatory Activity of High Temperature Extract from the Central Part Including Heartwood, Sapwood, Cambiun, and Phloem in Morus alba L.)

  • Background: Inflammation plays an important role in various diseases, including ulcerative colitis, Behcet's disease, and rheumatoid arthritis. In this study, we investigated the anti-inflammatory effects of Morus alba L. extracts obtained using different extraction methods (water extraction or high temperature extraction) on RAW264.7 cells. Methods and Results: Extracts from the central part (including the heartwood, sapwood, cambiun, and phloem) and bark (including the periderm and cortex) of Morus alba L. were obtained using either water or high temperature extraction. The following extract were obtained: MA1, water extract from the central part of Morus alba L., MA2, high temperature extract from the central part of Morus alba L., MA3, water extract from the bark of Morus alba L., and MA4, high temperature extract from the bark of Morus alba L. None of these extracts was observed to be cytotoxic to RAW264.7 cells. The MA2 extract reduced the production of LPS-induced NO (nitric oxide), $PGE_2$ (prostaglandin $E_2$), $TNF-{\alpha}$, IL-6, and $IL-1{\beta}$ production in LPS-stimulated RAW264.7 cells. Conclusions: These results indicated that the inflammatory response was moderated by MA2. Treatment with MA2 could be used as a natural medicine for treating diseases involving inflammation. However, further experiments are required to determine how the high temperature extraction method alters the active ingredients in the extract and influences the anti-inflammatory effects of Morus alba L..

환경시료 중 폴리브롬화비페닐(Polybrominated Biphenyls) 분석법 (Method for the Analysis of Polybrominated Biphenyls(PBBs) in Environmental Samples)

  • 수질 및 토양 시료 중의 PBBs의 분석방법 확립을 위해 표준물질이 준비된 PBBs 22종을 대상으로 추출 및 정제 등의 전처리 회수율 및 검출한계를 환경시료로부터 얻었다. PBBs의 시료 추출법으로 액/액, 속실렛, 초음파 추출법을 적용하였고, 실리카겔, 플로리실 정제컬럼의 적용성 검토 후 GC/MS로 분석하였다. 수질 시료의 경우 검출한계는 브롬화된 정도에 따라 약간의 차이를 보였고 5~10 ng/L이었으며, n-hexane, dichloromethane 및 toluene 등의 단일 용매의 평균회수율은 96~107%, n-hexane:acetone (1:1) 및 (9:1)의 혼합용매의 평균 회수율은 60~80%로 나타났다. 또한, 토양 시료의 경우 검출한계는 0.5~3.5 ng/g이었으며, 선정 용매의 평균 회수율은 속실렛 추출이 67~97% 및 초음파 추출이 64~76%로 나타났다.

Application of Synergistic Solvent Extraction by Formation of Ternary Complex for Determination of Trace Zn(II) in Water Samples

  • The application of a synergistic solvent extraction by the formation of ternary complex with pyrocatechol violet (PV) and benzalkonium chloride (BC) was studied for determination of trace Zn(II) in water samples. The pH of sample solution and the amount of PV and BC added were optimized for the formation of the stable complex, a proper solvent was selected for the effective extraction, and the concentration of nitric acid was fixed for the back extraction of the complex from the solvent. After the ionic strength of 100 mL sample solution was adjusted to 0.1 M by adding NaCl and the pH was fixed at 9 with a carbonate buffer, 1.0 mL of 2% PV solution was added to form Zn(II)-PV complex then the Zn(II)-PV/BC ternary complex was made by adding 1.0 mL of 10% BC solution. The ternary complex was extracted into 10 mL of MIBK. And the ternary complex was back-extracted with 10 mL of 1.0 mol/L nitric acid to determine Zn(II) by a flame atomic absorption spectrophotometer (flame-AAS). The interference of concomitant ions on the extraction of Zn(II) was investigated. This procedure was applied to the analysis of three real samples such as Dalbang-dam water, laboratory tap water and Jungnajin seawater. The recoveries of Zn(II) in spiked samples were 86.58-104.1%.

Speciation of Chromium in Water Samples with Homogeneous Liquid-Liquid Extraction and Determination by Flame Atomic Absorption Spectrometry

  • A novel method was developed for the speciation of chromium in natural water samples based on homogeneous liquid-liquid extraction and determination by flame atomic absorption spectrometry (FAAS). In this method, Cr(III) reacts with a new Schiff's base ligand to form the hydrophobic complex, which is subsequently entrapped in the sediment phase, whereas Cr(VI) remained in aqueous phase. The Cr(VI) assay is based on its reduction to Cr(III) by the addition of sodium sulfite to the sample solution. Thus, separation of Cr(III) and Cr(VI) could be realized. Homogeneous liquid-liquid extraction based on the pH-independent phase-separation process was investigated using a ternary solvent system (water-tetrabutylammonium ion ($TBA^+$)-chloroform) for the preconcentration of chromium. The phase separation phenomenon occurred by an ion-pair formation of TBA and perchlorate ion. Then sedimented phase was separated using a $100\;{\mu}L$ micro-syringe and diluted to 1.0 mL with ethanol. The sample was introduced into the flame by conventional aspiration. After the optimization of complexation and extraction conditions such as pH = 9.5, [ligand] = $1.0{\times}10^{-4}\;M$, [$TBA^+$] = $2.0{\times}10^{-2}\;M$, [$CHCl_3$] = $100.0\;{\mu}L$ and [$ClO_4$] = $2.0{\times}10{-2}\;M$, a preconcentration factor (Va/Vs) of 100 was obtained for only 10 mL of the sample. The relative standard deviation was 2.8% (n = 10). The limit of detection was sufficiently low and lie at ppb level. The proposed method was applied for the extraction and determination of chromium in natural water samples with satisfactory results.

식품첨가제용 뽕잎 농축액의 제조 및 특성평가 (Manufacturing and Characterization evaluation of mulberry concentrate for food additive)

  • 뽕잎을 다양한 방법으로 추출하여 수율을 측정한 결과 저온 초음파 추출법과 알콜 추출법이 효율적이었다. 식품첨가제로 활용하기 위한 뽕잎 농축액의 특성을 조사한 결과, 알콜 추출 농축액이 농축액별 곰팡이 증식에 대해 가장 우수한 억제력을 보였다. 색상의 변화는 비교적 안정적이었으나 뽕잎 열수 추출액의 경우는 전처리 과정이 없을 경우 갈변현상이 발생하였으며, 탁도의 변화조사와 비슷한 결과를 통해 열수 추출의 경우는 식품첨가제로서 그대로 이용하기엔 부족함을 인지할 수 있었다. 시간에 따른 농축액별 침전물 변화를 조사한 결과를 본 결과 필터를 통한 여과과정만 거친다면 알콜 추출과 당 추출의 경우는 거의 유사한 결과를 얻을 수 있으므로 식품첨가제 활용하기 위해 두 추출법은 모두 용이하다 할 수 있다. 그러나 농축액의 수율과 특성을 동시에 고려하여 농가에서 활용할 수 있는 가장 손쉬운 방법은 알콜 추출법으로 판단되었다.

흡착제를 이용한 수중의 미량 이취물질 분석법 개발에 관한 연구 (A Study on the Development of Analytical Method for Micro-Odorous Compounds in Water)

  • Even if odorous compounds remained very low concentration in water, it caused strong odor. Because Geosmin and most of odorous compound had very vaporization, those were difficult to analyze with GC/MSD and Purge & Trap. So, we needed pre-treatment method for decreasing amounts of extracting solvents, improving recovery efficiencies and increasing analytical efficiencies. This study developed efficient technology for analyzing odorous compounds, using various adsorbents and extracting solvents. The optimum adsorbent was XAD resins. Especially, XAD-2, XAD-7 and XAD-2010 were superior, but XAD-2 of these and MTBE was the optimum extraction solvent. Other extraction solvent's efficiency was in order of MTBE>Dichloromethane>n-Hexane>Diethylether. The optimum NaCl dosage for increasing efficiency was 5g in liquid-liquid extraction method. The shaking time(0∼24hr) had no concern with adsorption efficiency. The optimum adsorbent was XAD-2 resin and extraction solvent was MTBE. Dosing NaCl, adsorption efficiency was increased in liquid-liquid extraction method, but NaCl has no effect on liquid-solid extraction method. In this experimental results, this method will apply to not only Geosmin but other well-known odorous compounds (2-MIB, IBMP, IPMP, TCA) and algae toxins (Mycrocystin, Anatoxin etc)

프로폴리스 농도가 항균활성에 미치는 영향 (The effect of propolis concentration on the antibacterial activity)

  • The objetives of this study are to set up optimum extraction temperature, time and organic solvent for propolis extraction, to investigate chemical properties, and to develop health foods from propolis preparation. In this study, ethanol and ultrasonic extracts method performed to optimum extraction temperature was at 60, $20^{\circ}C$, optimum extraction time was at 12, 4 hours and optimum extraction amount of solvent was at 20, 15 times of propolis weight. When various ethanol solutions were used, whereas flavonoid content was highest in 70, 80% aqueous ethanol, respectively. So the ultrasonic extracts method used gave better results than the ethanol extracts method in this work. Extraction of propolis with etanol and ultrasonic extracts method was performed by using the water and various concentrations of aqueous ethanol as solvent. Sensitivity of propolis samples to Staphylococcus aureus was investigated and the results were shown. Samples of water extract did not inhibit microbial growth, where as 50% aqueous ethanol extract the largest inhibitory zone for Staphylococcus aureus, then decreased inhibition with increasing ethanol concentrations.

Effects of Physiological Active Substance Extracted from Silkworm Fece

  • Silkworm (Bombyx mori ) feces have long been used in the pharmaceutical and food industries as a natural colorant. However, there is limited data on the bioactive compounds that constitute silkworm feces. This research emphasizes the antioxidant activity of different solvent and flavonoid extracts of silkworm feces. The solvents were ethanol, butanol, and water, while the methods utilized included ultrasonification, stirrer, reflux, and reflux after ultrasonification extraction. Results showed that butanol ultrasonification extraction (BUE) yield the lowest extraction (1.75%), while the other methods yielded 7 to 14%. The total polyphenol content utilizing BUE was 3.3 mg TAE/g, while water ultrasonification extraction (WUE) yielded the highest extraction rate with 51.6 mg TAE/g. The total flavonoid content was significantly higher using ethanol reflux extraction (EUE) at 266.8 mg QRE/g BUE, which was 158.3 and 151.3 mg QRE/g. Both DPPH radical scavenging activity and SOD-like (superoxide dismutase) activity, showed significant antioxidant effects. Finally, all other extracts except for BUE had a-glucosidase inhibition at 60%. Therefore, an effective extraction method for physiologically active substances must be selected.

유출조건에 따른 인삼중의 Ginseng Extract와 Saponin 합량변화에 관한 연구 (Studies on the Extracting Methods of Ginseng Extract and Saponins in Panax Ginseng)

  • In order to investigate the optimal conditions which affects to extraction of ginseng extract and saponin in ginseng extract, experiment was carried out varing with ethanol percentage, extraction time, temperature, sol$.$vent and Plant Parts. The results art as follows: 1. The amounts of ginseng saponin was increased according to increanation of ethanol Percentage while the amounts of ginseng extract was decreased. 2. The amounts of ginseng extract was increased as the prolongation of extraction time, on the ether hand, ginseng saponin contents increased lentil 40hr. and decreased after that. 3. By the raise of extract temperature, both of the amounts of ginseng saponin and ginseng extract was increased two times and four times. respectively. 4. The total amounts ginseng extract was obtained 22.86u when the water used as the extraction solvent, 11.28% on ethanol and 11.04U on methanol, in the order. and the saponin contents gained when the extraction solvents of water, methanol and ethanol 7.47%, 12.36% and 12.77%, respectively. 5. It showed 9.23% of ginseng extract in epidermis and 8.4% of ginseng saponin in tail Part of raw ginseng and in the case of dried ginseng, ginseng extract and saponin showed the most amounts in epidermis of 18.28% and 19.35%, respectively. 6. The ratio of panaxadiol and panaxatriol contents of ginseng saponin was almost same when it was extracted varing with ethanol percentage and extraction time (duration), and the more alcohol percentage and the longer extraction time increased, the more fractional content of ginseng saponin was extracted.
