Analysis and Improvement Practise of Drainage Problem on Soil Profile at the Golf Course Fairway (골프코스 페어웨이 지반 토양의 배수불량 원인과 개선방안)

  • Research was focused on the improvement of poor drainage problems on golf course fairway which had not been performed soil test or properly amended during the course construction. The analysis of the drainage problem basically was caused by a deterioration of soil physical properties by the top layer compaction. The soil hardness reached about 3,000 Kpa around 5~6 cm of soil profile. The slow infiltration speed to subsoil by the compaction was caused directly a poor drainage capacity. However, the properly amended sand soil showed an apparent value of 1,500 Kpa through the subsoil. The water content test showed a similar result that higher rate of 20~30% and ideal rate of 8~12% at poor drainage area and successfully amended area, respectively. However, an imported topsoil media which had higher content of silt and clay from a trans-planted sod had made a heterogeneous soil profile and that caused a poor drain capacity by a low infiltration rate. Those drainage problems triggered to buildup a reduced soil layer by poor soil gas exchange. The soil environment of deoxidation enhanced anaerobic microbial population and induced methane gas build-up to 55 ppm, and that resulted an adverse effect on turf growth by root growth retardation, consequently.

Purification, and Biochemical and Biophysical Characterization of Cellobiohydrolase I from Trichoderma harzianum IOC 3844

  • Because of its elevated cellulolytic activity, the filamentous fungus Trichoderma harzianum has a considerable potential in biomass hydrolysis applications. Trichoderma harzianum cellobiohydrolase I (ThCBHI), an exoglucanase, is an important enzyme in the process of cellulose degradation. Here, we report an easy single-step ion-exchange chromatographic method for purification of ThCBHI and its initial biophysical and biochemical characterization. The ThCBHI produced by induction with microcrystalline cellulose under submerged fermentation was purified on DEAE-Sephadex A-50 media and its identity was confirmed by mass spectrometry. The ThCBHI biochemical characterization showed that the protein has a molecular mass of 66 kDa and pI of 5.23. As confirmed by smallangle X-ray scattering (SAXS), both full-length ThCBHI and its catalytic core domain (CCD) obtained by digestion with papain are monomeric in solution. Secondary structure analysis of ThCBHI by circular dichroism revealed ${\alpha}$- helices and ${\beta}$-strands contents in the 28% and 38% range, respectively. The intrinsic fluorescence emission maximum of 337 nm was accounted for as different degrees of exposure of ThCBHI tryptophan residues to water. Moreover, ThCBHI displayed maximum activity at pH 5.0 and temperature of $50^{\circ}C$ with specific activities against Avicel and p-nitrophenyl-${\beta}$-D-cellobioside of 1.25 U/mg and 1.53 U/mg, respectively.

Effects of Irrigation Interval, Medium Composition and Drainage on the Growth Response of Korea Native Fern Rumohra aristata at Indoor (실내에서 관수주기, 상토종류, 배수층의 유$\cdot$무에 따른 자생 가는쇠고사리의 생육반응)

  • It was aimed to promote as a material for interior landscape by validating Rumohra aristata, in an indoor environment, especially irrigation interval, medium composition and drainage at indoor. 1. The result of physico-chemical analysis of medium composition showed that porosity, pH and Ex-Ca, Ex-Mg and Ex-K were high with peatmoss: vermiculite: perlite(1:1:1) and water contents, organic matter content, total nitrogen and cation exchange capacity were high with sand: leaf mold(1:1). 2. Growth and indoor adaptability of Rumohra aristata were better with irrigation at 2 interval per week than irrigation at 7 interval per week regardless of drainage. 3. In the case of medium composition, of growth and indoor adaptability were higher with sud: leaf mold (1:1) than peatmoss: vermiculite: perlite(1:1:1). 4. Fronds fresh weight and dry weight decreased when irrigation interval increased and were higher with sand: leaf mold(1:1) than peatmoss: vermiculite: perlite(1:1:1) treatment.

Sugar composition and glycemic indices of frequently consumed fruits in Korea (우리나라 다빈도 섭취 과일의 당 함량 및 혈당지수에 관한 연구)

  • Fruits are generally recommended for a balanced meal, as they are good sources of vitamins, minerals, and fiber, which may improve blood glucose control. However, fruits have simple sugars with a wide glycemic index (GI) range. The purpose of this study was to analyze the sugar content and composition and to determine the glycemic indices of the most frequently consumed fruits in Korea, including apple, tangerine, pear, water melon, persimmon, grape, oriental melon, and peach. The sugar content and composition of the fruits were analyzed by high performance anion-exchange chromatography (Dinonex model DX-600). The GI of the fruits was measured in 13 healthy subjects (seven females and six males) after permission was received from the University Hospital institutional review board (KHU-IRB 1114-06). The subjects consumed 50 g of glucose as a reference and carbohydrate portions of eight fruits. Blood samples were collected at 0, 30, 60, 90, and 120 min after consuming the fruits. The GI values for the fruits were calculated by expressing the increase in the area under the blood glucose response curve for each subject. As a result, the total sugar contents of 100 g fruits were: grape (13.9 g), apple (12.3 g), persimmon (11.9 g), oriental melon (11.2 g), watermelon (9.3 g), tangerine (8.9 g), peach (8.6 g), and pear (8.3 g). The GI values of the fruits were as follows: GI value of peach ($56.5{\pm}14.17$), watermelon ($53.5{\pm}18.07$), oriental melon ($51.2{\pm}18.14$), tangerine ($50.4{\pm}15.16$), grape ($48.1{\pm}14.05$), persimmon ($42.9{\pm}18.92$), pear ($35.7{\pm}14.38$), and apple ($33.5{\pm}11.92$). These findings will help individuals choose fruit for controlling blood sugar.

A Study on Improvement Measures for the Design Documents Delivery System in Public Road Construction Projects (공공 도로건설사업에서의 설계도서 납품체계 개선방안에 관한 연구)

  • PURPOSES : This study aims to survey and analyze the status of the design and completion documents output delivery system in public construction projects in order to assess the problems of the Ministry of Land, Infrastructure and Transport (MOLIT)'s electronic delivery system, with a focus on roads and rivers, and to offer improvement measures. METHODS : This study first surveys and analyzes laws and existing study trends with regard to the design and completion documents output delivery system in public construction projects. It further analyzes the status of the output delivery system in public construction projects, including roads, rivers, expressways, water resources, and railroads. In addition, a comparative analysis of the current electronic delivery system of MOLIT and Korea Expressway Corporation in the road field is conducted, and the problems and improvement measures for MOLIT's electronic output delivery system are presented. This study seeks to analyze problems and prepare improvement measures with regard to road and river public construction projects, as ordered by MOLIT's five regional offices. RESULTS : This study sought to prepare the electronic output delivery system with regard to public construction projects and present the analysis of its major problems and improvement measures in four categories: "the preparation of electronic design documents output delivery system improvement measures including inspection and delivery," "preparation of guidelines, systems, etc., concerning electronic design documents," "preparation of improvement measures for the construction project management system and electronic design document support tools", and "linking of MOLIT's electronic output delivery system with relevant agencies and the expansion thereof to local governments." CONCLUSIONS : This study analyzed the current status and problems of the MOLIT's electronic output delivery system in the field of road and river construction projects, and presented corresponding improvement measures. This study is expected to address the problems of the MOLIT's electronic output delivery system, and to provide the foundation for the preparation of an efficient electronic output delivery system for design and completion documents. In addition, this study is expected to boost the quality and utilization of the electronic design documents output, and to exchange, share, and link construction information among relevant agencies so as to prepare the foundation for sharing construction information.

Preparation and Characterization of Proton Conducting Crosslinked Membranes Based On Poly(vinyl chloride) Graft Copolymer (Poly(vinyl chloride) 가지형 공중합체를 이용한 수소이온 전도성 가교형 전해질막의 제조와 분석)

  • A graft copolymer consisting of poly(vinyl chloride) (PVC) backbone and poly(hydroxyethyl acrylate) (PHEA) side chains was synthesized via atom transfer radical polymerization (ATRP). Direct initiation of the secondary chlorines of PVC facilitates grafting of hydrophilic PHEA monomer. This graft copolymer, i.e. PVC-g-PHEA was cross-linked with sulfosuccinic acid (SA) via the esterification reaction between -OH of the graft copolymer and -COOH of SA, as confirmed by FT-IR spectroscopy. Ion exchange capacity (IEC) continuously increased to 0.87meq/g with increasing concentrations of SA, due to the increasing portion of charged groups in the membrane. However, the water uptake increased up to 20.0wt% of SA concentration above which it decreased monotonically. The membrane also exhibited a maximum proton conductivity of 0.025 S/cm at 20.0 wt% of SA concentration, which is presumably due to competitive effect between the increase of ionic sites and the crosslinking reaction.

Perfluorinated Sulfonic Acid based Composite Membranes for Vanadium Redox Flow Battery (바나듐 레독스 흐름 전지를 위한 과불소화 술폰산 복합막)

  • Vanadium redox flow batteries (VRFBs) using the electrolytes containing various vanadium ions in sulfuric acid as supporting solution are one of the energy storage devices in alternatively charging and discharging operation modes. The positive electrolyte contains $V^{5+}/V^{4+}$ and the negative electrolyte $V^{2+}/V^{3+}$ depending on the operation mode. To prevent the mixing of two solutions, proton exchange membranes are mainly used in VRFBs. Nafion 117 could be the most promising candidate due to the strong oxidative property of $V^{5+}$ ion, but causes high crossover of electroactive species to result in a decrease in coulombic efficiency. In this study, the composite membranes using Nafion ionomer and porous polyethylene substrate were prepared to keep good chemical stability and to decrease the cost of membranes, and were compared to the properties and performance of the commercially available electrolyte membrane, Nafion 117. As a result, the water uptake and ionic conductivity of the composite membranes increased as the thickness of the composite membranes increased, but those of Nafion 117 slightly decreased. The permeability of vanadium ions for the composite membranes significantly decreased compared to that for Nafion 117. In a single cell test for the composite membranes, the voltage efficiency decreased and the coulombic efficiency increased, finally resulting in the similar energy efficiency. In conclusion, the less cost of the composite membranes by decreasing 6.4 wt.% of the amount of perfluorinated sulfonic acid polymer due to the introduction of porous substrate and lower vanadium ion permeability to decrease self-discharge were achieved than Nafion 117.

Selection of Proper Medium and Amount of Applied Fertilizer for Exportable Cymbidium Young Plants Grown in Korea (수출용 심비디움 묘 생산에 적합한 배지 선발 및 시비량 구명)

  • Bark is a general medium for potted Cymbidium in Korea, but it is difficult to shake off or eliminate the medium from the roots before exporting the potted plants. This working process can injure the roots and then deteriorate plant marketability. This study was carried out to select the optimum medium easier to eliminate from the roots instead of bark and the optimum amount of applied fertilizer to improve the plant growth. Cymbidium young plants 'Honey Hot' and 'Desert Look' were planted in pots with bark, cocochip, and peatmoss. The plants were treated with 2, 4, and 6 g of slow release fertilizer. The plant growth characteristics were investigated in the first and second years during production period of three years. The medium characteristics and mineral nutrient content of the leaves were also examined in the second year. In the first year, the plant growth of 'Desert Look' was improved in all peatmoss treatments more than bark. 'Honey Hot' showed the highest plant growth values in the bark treatment. In the second year, the plant growth of the two cultivars was improved in peatmoss. Cocochip treatments showed the lower plant growth values than bark and peatmoss in the first and second year. There was no significant difference among fertilizer amounts in all the media. The higher CEC values of peatmoss medium resulted to higher capacity to hold more nutrients than bark, and the nutrient retention of the peatmoss improved the plant growth. The higher K and Ca contents in the leaves would contribute to improve the plant growth. Consequently, it would be possible to use peatmoss instead of bark for Cymbidium young plants, but there must be always attention to appropriately water the medium and manage the moisture.

Screening of Antithrombotic Peptides from Soybean Paste by the Microplate Method (Microplate법에 의한 된장유래의 항혈전 펩타이드 탐색)

  • In order to search for antithrombotic peptides from soybean paste, the inhibitory activity of water extract of soybean paste and its peptide fractions on ADP-induced aggregation of washed platelets was assayed. Soybean paste extract treated with ultrafiltration (M.W. cut off, 3,000 daltons) was found to have inhibitory activity of 90% at the dose of $96\;{\mu}g/ml$ by the method of turbidometric aggregometer. Soybean paste extract was fractionated to 19 fractions (No. B-18) by Dowex 50W X-2 ion exchange column chromatography and activity test was performed by the microplate method. All of the fractions had antithrombotic activity $(IC_{50},\;101,000\;{\mu}g/ml)$, and most fractions had higher activity than positive control, RGDS $(IC_{50}.\;205\;{\mu}g/ml)$. Especially, basic fractions No.16-18 showed higher activity than soybean paste extract $(IC_{50},\;30\;{\mu}g/ml)$. The fraction No.16 with the highest activity $(IC_{50},\;10\;{\mu}g/ml)$ was purified and analysed for amino acid composition. The results showed that histidine, arginine, and alanine were major residues in the peptide part of the fraction.

A Study of the Actual Condition for Inducing Container Cargos to Gunsan Port (군산항 컨테이너화물 유치를 위한 실태조사연구)

  • To induce container cargos to Gunsan port, we should struggle on securing the quantity of goods transported from the competitive Jeonbuk and Chungcheong areas by reducing transportation expenses by land, and make an activating plan for the international exchange between Gunsan port and nearby ports in China. As a result, if the plan to induce container cargos to Gunsan port is activated, the regular liners will stop at Gusan port. So the new sea route will be opened so that consignors can ship cargos in the proper time. The consignors will use Gunsan port frequently because of the reduction of distribution expenses. To do so, for the first, many incentives should be implemented to attract container cargos. In addition, various facilities should be fully equipped and improved. This paper is to improve the problems for which consignors, the shipping lines and forwarders may feel uncomfortable when they use Gunsan port. According to the research on the actual condition, the biggest problem in Gunsan port is the depth of water. The management, relation system, infrastructure and information system of Gunsan port are also insufficient. Therefore, this paper will make an alternative plan to improve these problems and help the container cargos induce to Gunsan port.
