This study was conducted to investigate the physiological activity of acai berry (Euterpe oleracea Mart.) extracts from three different solvents (water, methanol, and ethanol). We measured total polyphenol and total flavonoid content, DPPH radical scavenging activity, nitrite scavenging activity, metal chelating effect, and reducing power. The extraction yield from water, methanol, and ethanol was 17.10, 9.50, and 37.51%, respectively. The highest total polyphenol content (10.54 mg/100 g) and total flavonoid contents (1.88 mg/100 g) was observed in water extract. DPPH radical scavenging activity was the highest in both water extract (72.03%) and methanol extract (74.79%) at levels of 5 mg/mL, which was similar to that of BHT (78.90%). Water extract yielded the highest metal chelating effect (92.54%) and reducing power (1.09) at levels of 5 mg/mL. Taken together, these findings suggest that extracts of acai berry can be used as functional food materials with antioxidative and nitrite scavenging activities.
In this work, caffeine and some catechin compounds + C, EC, EGC, and EGCG were extracted from green tea by using molecular imprinted polymers (MIP) as sorbent materials in a solid-phase extraction (SPE) process known as MISPE (molecular imprinted solid-phase extraction). For synthesis of MIP, caffeine was employed as the template, MAA as the monomer, EGDMA as the crosslinker, and AIBN as the initiator. A solution of caffeine (0.2 mg/mL in methanol) was utilized in the solid extraction cartridges following loading, washing, and elution procedures with acetonitrile, methanol, and methanol-acetic acid (90/10, %v/v) as the solvents, respectively. This solid-phase extraction protocol was applied for the extraction of caffeine and some catechin compounds from green tea. A comparison was made between the results obtained with the MIP cartridges and a traditional C18 reversed-phase cartridge. It was thereupon found that the recovery of caffeine by the MIPbased sorbent used in this work was almost two and four times greater than that by a commercially available C18 material. A quantitative analysis was conducted by high performance liquid chromatography (HPLC) using a C18 column (5 μm, 250 × 4.6 mm) with methanol/water (40/60, %v/v) as the mobile phase at a flow rate of 0.5 mL/min.
오죽(Phyllostachys nigra Munro)을 이용하여 추출용매에 따른 총 페놀함량, 항산화 활성, superoxide dismutase (SOD) 유사활성, 아질산 소거능, tyrosinase활성 저해능, angiotensin-converting enzyme (ACE) 저해활성을 in vitro에서 검토 했다. Ethyl acetate와 hexane을 추출용매로 이용할 때 총 페놀화합물 함량은 각각 $44.1{\pm}2.3mg/kg$와 $47.3{\pm}2.4mg/kg$였다. 항산화 효과는 추출용매 종류에 따라 각각 다르게 나타났고 hexane > ethyl acetate > n-butanol > methanol > water순이었다. 그러나 SOD 활성은 물 > methanol > n-butanol> hexane> ethyl acetate 추출 순이었다. 아질산 소거능은 pH 1.2에서 추출용매에 따라 약간의 차이는 있었으나 54~69.2% 범위였다. 그러나 pH가 증가함에 따라 추출용매에 관계없이 모두 감소하였다. 특히 pH가 1.2에서 6.0으로 증가 할 경우 ethyl acetate 추출물의 아질산 소가능은 $69.2{\pm}3.1%$에서 $7.8{\pm}2.4%$로 감소하였다. 오죽 추출물의 tyrosinase의 저해 활성은 추출용매에 따라 약간 차이가 있으나 전반적으로 15.2~21.3% 범위였다. 추출용매에 따른 tyrosinase의 저해 활성은 water > methanol > n-butanol > hexane > ethyl acetate 추출 순이었다.
본 연구는 정향추출물(물, 메탄올 및 에테르)의 항산화 효과를 리놀레인산 기질 및 리놀레인산 에멀젼기질에서 비교, 검토하였다. 물, 메탄올 및 에테르용매에 따른 정향의 총페놀 함량은 23.1, 42.8, 34.9%로 메탄올>에테르>물 추출물 순이었다. Eugenol 함량은 메탄올 추출물의 경우 6.1%이었고 에테르 추출물 8.1%이었으며 물 추출물에서는 검출되지 않았다. 정향 추출물의 DPPH 소거효과는 물 추출물 경우 36.1%, 메탄을 추출물의 경우 30.9%, 에테르 추출물의 경우 29.7%로 나타났다. 리놀레인산 기질에서의 항산화 효과는 BHT>에테르 추출물> $\alpha$-tocopherol>메탄올 추출물>물 추출물의 순이었다. 리놀레인산 에멀젼 시스템에서의 항산화 효과는 BHT>물 추출물>메탄올 추출물>에테르 추출물> $\alpha$-tocopherol의 순이었다.
This study was carried out to investigate the biological activities of star anise extracts obtained from four different solvents (water, methanol, ethanol and chloroform) by measuring total polyphenol and flavonoid contents, DPPH radical scavenging, hydroxyl radical scavenging, nitrite scavenging activity and antimicrobial activity. The methanol extract showed the highest extraction yield, followed by ethanol, water and chloroform. The properties of the extracting solvents significantly affected the total polyphenol content. Methanol extracts contained more total polyphenols than any other extracts. The highest DPPH radical scavenging activity was found in methanol extract. The hydroxyl radical and nitrite scavenging activity were highest in methanol and ethanol extracts. In antimicrobial activity, water extract showed stronger activities than methanol and ethanol extract against Micrococcus luteus and Bacillus subtilis, and no inhibitory effects on Gram negative bacteria were found in all extracts at the concentration used.
In different approaches ginsenosides were extracted from Korean ginseng roots by ammonia and for comparison with methanol-water and water. The extracts have been analyzed qualitatively and quantitatively to evaluate yield and selectivity of extractions of ginsenosides. Water supplied the lowest yield. The yields of extracts with liquid ammonia were higher than those with methanol-water. However, this is partly due to the conversion of malonyl ginsenoside to normal ginsenosides by ammonia. It was proved by HPLC that malonyl-ginsenosides $m-Rb_1,\;m-Rb_2,$ m-Rc and m-Rd were converted to the corresponding neutral ginsenosides. Furthermore, ginsenosides from ginseng roots were extracted by alkaline methanol-water $(60\%)$ solutions. Alternatively, the extracts of the methanol-water $(60\%)$ extraction were treated with sodium hydroxide solution. Both methods also convert the malonyl-ginsenosides to neutral ginsenosides.
In order to investigate the optimal conditions which affects to extraction of ginseng extract and saponin in ginseng extract, experiment was carried out varing with ethanol percentage, extraction time, temperature, sol$.$vent and Plant Parts. The results art as follows: 1. The amounts of ginseng saponin was increased according to increanation of ethanol Percentage while the amounts of ginseng extract was decreased. 2. The amounts of ginseng extract was increased as the prolongation of extraction time, on the ether hand, ginseng saponin contents increased lentil 40hr. and decreased after that. 3. By the raise of extract temperature, both of the amounts of ginseng saponin and ginseng extract was increased two times and four times. respectively. 4. The total amounts ginseng extract was obtained 22.86u when the water used as the extraction solvent, 11.28% on ethanol and 11.04U on methanol, in the order. and the saponin contents gained when the extraction solvents of water, methanol and ethanol 7.47%, 12.36% and 12.77%, respectively. 5. It showed 9.23% of ginseng extract in epidermis and 8.4% of ginseng saponin in tail Part of raw ginseng and in the case of dried ginseng, ginseng extract and saponin showed the most amounts in epidermis of 18.28% and 19.35%, respectively. 6. The ratio of panaxadiol and panaxatriol contents of ginseng saponin was almost same when it was extracted varing with ethanol percentage and extraction time (duration), and the more alcohol percentage and the longer extraction time increased, the more fractional content of ginseng saponin was extracted.
수삼으로 제조된 백삼 제품들의 가공방법의 차이에 따른 성분 변화에 대한 조사로서 원료삼인 수삼과 전통적인 방법으로 제조한 백삼, 태극삼 A, 새로운 방법으로 제조된 태극삼 B의 일반성분, 추출수율, 사포닌 함량 및 조성, 유리당 조성을 비교 분석 하였다. 4가지 시료의 조단백질은 13.34-15.33%, 조지방은 1.04-2.05%, 조회분은 3.13-5.25%, 총 식이섬유는 9.72-15.46%, 당질은 64.75-73.48% 범위의 함량이었다. 추출수율은 열수 추출이 80% 메탄올 추출보다 전체적으로 약 2배 이상 높게 나타났으며, 가공하지 않은 수삼의 열수 추출 수율이 56.4%로 가장 높았다. 사포닌 함량은 열수 추출시 수삼의 총 사포닌이 2.40%로 가장 많이 추출되었으며, 태극삼 A는 태극삼 B보다 비율로 볼 때 약 20% 이상 적은 1.45%의 총 사포닌 함량을 나타내었다. Ginsenoside 조성은 열수 추출시 $ginsenoside-Rg_1$이 수삼은 36.52%이지만 태극삼 A와 B는 68.66, 67.89%고 약 2배의 함량을 나타내었다. $Ginsenoside-Rb_1$은 태극삼 A차 B에 18.41, 17.43%만이 함유되어 있었다. 한편 태극삼 A와 태극삼 B의 총 사포닌 양에는 큰 차이가 있었지만 ginsenosides의 조성비에 있어서는 서로 큰 차이가 없는 것으로 나타났다. 80% 메탄올 추출의 경우에도 가공방법이 다르게 제조된 백삼 제품별로 ginsenoside조성에 큰 차이를 나타내었다. 수삼과 백삼에서는 열수 추출과 80% 메탄올 추출 모두에서 함량비는 다를지라도 fructose, glucose, sucrose, maltose만이 함유되어 있었지만 열 처리된 태극삼 A와 B에는 rhamnose도 각각 2.56, 4.01%씩 함유되어 있었다.
Korea is one of the countries with a large veterinary antibiotics market, although antimicrobial resistance in bacteria is becoming a serious issue in many countries. The Korean government started to take interest in estimating the effects of livestock manure on rivers and agricultural soils and in monitoring of heavy metals, organic pollutants and antibiotics in the ambient water and soil. In this paper, pre-treatment methods to separate the selected antibiotics from solid samples were reviewed. It is essential to select an efficient and appropriate procedure for pre-treatment due to the high proportion of proteins and organics in biosolid samples. Pre-treatment consists of extraction followed by clean-up. Initially, homogenized samples were extracted by sonication, mechanical agitation or pressurized liquid extraction with methanol/acetonitrile/water mixture under acidic/basic conditions depending on the compound. However, aminoglycosides and colistin were extracted with 5% trichloroacetic acid and HCl, respectively. Since the ${\beta}-lactams$ are easily decomposed in acidic and basic conditions, they were extracted in neutral pH. Filtration with a membrane (pore size, $0.2{\mu}m$) or solid phase extraction with HLB and methanol, as eluents, was normally applied for the clean-up. At least, three different pre-treatment procedures should be adopted to screen all the selected antibiotics in solid samples.
The optimum extraction conditions of green tea used for a bath were investigated for later application to textile. The extraction was more effective in water and methanol than in ethanol. The optimum extraction temperature was determined as 80$^{\circ}C$ in water and 60$^{\circ}C$ in methanol. The extracts were more stable in acid and neutral conditions than in alkaline region. The UV satbility was better than generally expected. From the analysis of contents of active ingredients, the about 20% of effective catechin was appeared to be contained in the extract solid. The contents of polyphenol is 0.016g in the 0.1g extracts.
