녹차와 로즈마리 추출물을 첨가한 두유의 항산화 활성 (Antioxidant activities of soymilk added with green tea and rosemary extract)

  • 제주산 녹차, 로즈마리, 레몬 잎, 조릿대 잎을 물과 에탄올을 이용해 다양한 조건에서 추출물을 제조하여 각 추출물에 대한 항산화 활성을 측정하였다. 그 결과, 녹차와 로즈마리 추출물이 레몬과 조릿대 잎 추출물과 비교하여 상대적으로 높은 항산화 활성을 보였다. 항산화 활성이 상대적으로 우수한 녹차와 로즈마리 추출물들을 두유에 첨가한 뒤, 혼합물의 항산화 활성을 측정하였다. 녹차 증류수 가압 가열 추출물과 상온 정치 추출물 10% 첨가한 두유에서 상대적으로 가장 높은 활성을 보였으며, 녹차 증류수 가압 가열 추출물과 상온 정치 추출물의 DPPH 라디칼 소거능은 각각 $85.1{\pm}2.3%$, $84.4{\pm}2.0%$을, ABTS 라디칼 소거능은 각각 $73.4{\pm}2.8%$, $81.3{\pm}4.8%$로 확인되었다. 한편, 로즈마리 증류수 가압 가열 추출물 10% 첨가한 두유에서 무첨가 두유에 비해 더 높은 DPPH 라디칼 소거능이 확인되었으며, ABTS 라디칼 소거능은 무첨가 두유와 유사했다. 따라서 녹차 증류수 가압 가열추출물과 상온 정치 추출물, 그리고 로즈마리 증류수 가압 가열 추출물을 두유에 첨가 시 항산화 활성이 향상된 고부가가치 두유제품 개발 제발이 가능할 것으로 기대된다.

추출 조건을 달리한 감잎과 녹차의 물 및 에탄올 추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향 (Effect of Water and Ethanol Extracts of Persimmon Leaf and Green Tea Different Conditions on Lipid Metabolism and Antioxidative Capacity in 12-month-old Rats)

  • This study was performed to investigate effects of dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in 12-month-old rats. Fifty-four male Sprague-Dawley rats weighing 542$\pm$4.5g were blocked into groups according to their body weight and were raised for four weeks with the diets containing 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) and green tea(Camellia Sinensis O. Ktze), water or 75% and 95% ethanol extracts from same amount of each dried tea powder. Food intake was not significantly different among all groups, but weight gain of green tea powder group was significantly lower than that of control group. Plasma and liver lipid levels of all the tea diet groups were lower than those of control group. Especially, 75% ethanol extract of persimmon leaf decreased total lipid and triglyceride concentrations in plasma and 95% ethanol extract of persimmon leaf decreased liver total lipid level. However, there was no difference between 75% ethanol extracts groups and 95% ethanol extracts groups in lipid metabolism. Superoxide dismutase(SOD) and catalase activities in erythrocyte were remarkably increased by all the green tea diets. SOD, catalase and glutathione peroxidase activities in liver were increased by the feeding of ethanol extracts from green tea and persimmon leaf powder. Liver xanthine oxidase activity was not different among all groups. Plasma Thiobarbirutic acid reactive substance(TBARS) concentrations of all the green tea diet groups were significantly low. It was thought that high flavonoids in green tea inhibited plasma lipid peroxidation by promoting SOD, catalase activities in erythrocyte. 95% ethanol extract of persimmon leaf also inhibited plasma lipid peroxidation by high vitamin E and beta-carotene. Persimmon leaf powder decreased liver TBARS concentration by vitamin E, betacarotene and vitamin C and by increasing activities of antioxidative enzymes with flavonoids. In conclusion, dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea were effective in lowering lipid levels and inhibiting lipid peroxidation in 12-month-old rats. Above all, ethanol extracts of persimmon leaf decreased plasma and liver lipid levels and persimmon leaf powder effectively inhibited liver lipid peroxidation. Extracts of green tea leaf inhibited plasma lipid peroxidation. In lowering lipid levels and inhibiting lipid peroxidation, ethanol extracts were more effective than water extracts, but there was no difference between 75% ethanol extracts and 95% ethanol extracts in lipid metabolism. 항생제(抗生劑) 내성(耐性) 화농균(化膿菌)의 생육(生育)을 억제(抑制)하는 한약재(韓藥材) 탐색(探索)에 관(關)한 硏究(연구) (A Study on Screening of Oriental Medicines Against Antibiotics-resistant Staphylococcus aureus)

  • The various oriental herbal medicines, which have usually been used for treatment of reducing fever, purging intense heat and detoxication, were screened to determine the antibacterial activity and the minimal inhibitory concentration against antibiotics-resistant Staphylococcus aureus. The results obtained were as follows: 1. Among the 23 oriental medicines tested, the water-soluble extract of Coptis japonica showed very excellent antibacterial activity against antibiotics-sensitive and -resistant S. aureus. A little activity was found in the extracts of Scutellaria baicalensis, Eriobotrya japonica and Prunus mume against antibiotics-sensitive S. aureus and in that extracts of P. mume, Schizandra chinensis and S. baicalensis against antibiotics-resistant S. aureus. When C japonica was used, the high antibacterial activity was shown against Bacillus subtilis and other extracts showed a little or no antibacterial activity against B. subtilis and E. coli as a control. 2. The highest antibacterial activities against antibiotics-resistant S. aureus were found in the ethanol-soluble extract of C japonica. Other ethanol-soluble extracts of S, baicalensis, S. chinensis and P. mume showed a little activity against both antibiotics-sensitive and antibiotics-resistant S. aureus. 3. Among the medicinal herbs tested, the water- and ethanol extracts of C. japonica showed very excellent antibacterial activity against the pathogenic S. aureus and both Escherichia coli and Bacillus subtilis used as a control. 4. When the water-soluble extracts of C. japonica and S. baicalensis, minimal inhibitory concentrations (MICs) against antibiotics-sensitive S. aureus were $20mg/m{\ell}\;and\;30mg/m{\ell}$, respectively. The MICs of the ethanol-soluble extracts of C. japonica and P. scabriosaefolia were $7.5mg/m{\ell}\;and\;12mg/m{\ell}$, respectively. 5. For the MICs against antibiotics-resistant S. aureus, the water-soluble extracts of C. japonica and P. mume showed $15mg/m{\ell}\;and\;10mg/m{\ell}$, respectively. The MICs of the ethanol-soluble extracts of C. japonica and P. mume were $13mg/m{\ell}\;and\;20mg/m{\ell}$, respectively. As a result, the highest antibacterial activity was found in both water- and ethanol-soluble extracts of C. japonica against antibiotics-resistant S. aureus. And the water- and ethanol-soluble extracts of S. baicalensis, S. chinensis and P. mume showed comparatively high antibacterial activities against both antibiotics-sensitive and -resistant S. aureus.

오미자의 과육과 종자의 물추출이 약물대사에 미치는 효과 (Effects of Water Extracts of Endocarps and Seeds of Omija(Schizandra chinensis Baillon)on Drugs metabolism)

  • Effects of water extracts of endocarps and seeds of omija(Schizandra chinensis Baillon)on alloxan and CCl4 induced liver injuries were determined by measuring the contents of metabolites and enzyme activities both in liver and serum in male Sprague-Dawley rats. The treatment with water extracts in parts of omija showed low levels of glucose, GOT, LDH and FFA of serum but high level of hepatic G-6-P DH in alloxan treated rats. CCl4 treatment with water extracts of parts of omija resulted in low levels of GOT, LDH, UN of serum and hepatic pyruvate but hepatic cytosolic protein has been increased.

산채류가 장내세균의 In Vitro 생육에 미치는 영향 (Effects of Edible Herbs on the Growth of In Vitro Intestinal Microorganisms)

  • This study was aimed to screen edible herbs which control the composition of intestinal microflora. With in vitro experiments, we screened the water or ethanol extracts of about 60 edible herbs and wild plants in terms of the inhibition activity on the growth of the harmful Clostridium perfringens and growth promoting activity for the beneficial Bifidobacteria. The water extracts of mugwort and small water dropwort inhibited the growth of Cl.perfringens both in agar diffusion method and broth culture. On the other hand, the water extracts of petasites, mugwort, yellow day-lily and bitter cress have shown the promotion effect on the growth of Bifidobacterium longum. In the culture test using human feces as starter, the extracts of the above selected herbs increased the population of Bifidobacteria and Lactobacillus while they reduced the numbers of Cl.perfringens and E.coli.

Negative Effects of Water Extracts from Pinus densiflora Sawdust on Mycelium Growth of the Shiitake Mushroom Lentinula edodes

  • Extracts from sawdust of Pinus densiflora were showed antifungal activity against Lentinula edodes. It was extracted by hot water and then successively extracted by n-hexane, ethyl acetate, and methanol. The yields of the n-hexane-soluble, ethyl acetatesoluble, methanol-soluble and methanol-insoluble fractions of water extracts were 8.2%, 10.6%, 32.0%, and 49.2%, respectively. The ethyl acetate-soluble fraction showed the greatest antifungal activity against L. edodes: 41.5% inhibition at 1,000 ppm. However, there were not significant differences of antifungal activities between n-hexane-soluble fraction and methanol-soluble fraction at a concentration of 1,000 ppm. The hot water extracts showed 23.5% of antifungal activity against L. edodes at a concentration of 1000 ppm. The four antifungal compounds were separated from ethyl acetate fraction by thin layer chromatography.

Development of Vaccinium uliginosum L. extracts for whitening & anti-wrinkle functional food

  • This study is peformed to investigate the effect of water extract from Vaccinium uliginosum L., on melanin production in B 16 melanoma cells, procollagen production and matrix metalloproteinase-1(MMP-1) inhibition in human fibroblast cells. One hundred grams of the Vaccinium uliginosum L. was extracted with 2000 mL of water($90^{\circ}C$, 16h, 2times). The water extracts were lyophilized and stored at $4^{\circ}C$ until used. Dry weight yields of extracts of Vaccinium uliginosum L. were $3\%$(w/w). Extracts from Vaccinium uliginosum L. showed scavenger activities on DPPH radical, superoxide anion radical, hydroxyl radical, hydrogen peroxide and singlet oxygen radical. And these substances inhibited release of cyiokines from human keratinocyte after UV B exposure. Therefore we confirmed that extracts from Vaccinium uliginosum L. had antioxidative effect. These substances inhibited purified tyrosinase activity and melanogenesis in B 16 melanoma cells treated/untreated IL-$1{\alpha}$. Moreover this extract stimulated procollagen production and inhibited MMP-1 production in human fibroblast cells treated/untreated IL-$1{\beta}$. Therefore we confirmed that extracts from Vaccinium uliginosum L. had whitening effect. And these substances decreased degree of wrinkle in hairless mouse skin that induced by UV B irradiation. Therefore we confirmed that extracts from Vaccinium uliginosum L. had anti-wrinkle effect. From the above results, it is possible that Vaccinium uliginosum L. may be developed to be an anti-melanogenesis agent and anti-wrinkle agent.

In Vitro Antiviral Activity of Aqueous Extracts from Korean Medicinal Plants Against Influenza Virus Type A

  • Boiled-water extracts from 101 Korean medicinal plants were tested in vitro for their inhibitory activity against influenza virus type A by means of a modified hemagglutination inhibition test. Thirteen of the 101 extracts exhibited strong anti-influenza virus type A activity at concentrations of less than $780\;{\mu}g/ml$. Out of the above 13 extracts, MW-40 (Chaenomeles speciosa), MW-88 (Citrus junos), and MW-100 (Zingiber officinale) exhibited marked antiviral activity in the concentration range of $0.195\;{\mu}g/ml$ to 100 mg/ml, $0.0487\;{\mu}g/ml$ to 100 mg/ml, and $0.0487\;{\mu}g/ml$ to 100 mg/ml, respectively. The extracts MW-88 and MW-100 were not cytotoxic to red blood cells, whereas MW-40 showed very weak cytotoxicity in the concentration range of 50 mg/ml to 100 mg/ml. Therefore, the present results demonstrate that boiled water extracts of 2 Korean medicinal plants, MW-88 and MW-100, have strong anti-influenza virus type A activity and no cytotoxic effects, and they may inhibit attachment of the virus to the cell and may be used for prophylaxis.

Extracts of Codonopsis lanceolata Decrease $CCl_4$-Induced Hepatotoxicity on High Fat Diet in Rats

  • This study was designed to investigate the effect of Codonopsis lanceolata extracts on carbon tetrachloride (CCl₄)­induced hepatotoxicity in rats fed high, medium or low fat diet. The liver weight was increased not by the level of fat in diet but by administration of CCl₄. Activities of serum aspartate aminotransferase (AST) and alkaline phosphatase (ALP) were significantly increased by CCl₄, the activities of AST and alanine aminotransferase (ALT) were significantly decreased in case of CCl₄ treatment with C. lanceolata extracts. In high fat diet with CCl₄ administration, the content of total lipid in liver was significantly increased, which was significantly decreased by the administration of C. lanceolata extracts. Administration of C. lanceolata water extracts remarkably prevented the tissue injury such as lipid degeneration, infiltration of inflanunatory cells and necrosis caused by CCl₄, but those methanol extracts did not affect histopathological changes of the liver as water extract. The data suggest that intake of C. lanceolata extracts would prevent lipid accumulation and hepatotoxicity induced by high fat diet and CCl₄ abministratim, and the water extract of C. lanceolata is supposed to be more effective than those methanol extract.

대나무 추출액의 항산화 작용과 미백작용 (Anti-oxidant Activity and Whitening Activity of Bamboo Extracts)

  • To investigate the possibility of development as a whitening agent using bamboo extracts (Phyllostachys nigra var. henonis), we measured DPPH assay, NBT/XO assay, intracellular ROS scavenging assay, tyrosinase assay and MSH-induced melanin production in B-16 cells. Bamboo extracts had dose-dependently anti-oxidant activity in DPPH, NBT/XO and intracellular ROS assay. Bamboo extracts appear to inhibit xanthine oxidase directly. Bamboo extracts inhibited not only purified tyrosinase activity but also inhibited tyrosinase activity and melanin production in B16 melanoma cells stimulated by $1{\mu}M\;{\alpha}$-MSH. Anti-oxidant activity and cytotoxicity of ethyl acetate fraction was more potent than those of water fraction, whereas whitening effect of water fraction was stronger than ethyl acetate fraction. Therefore, these results suggest that water fraction of bamboo extracts may be useful for the development as whitening agents reducing cytotoxicity.
