Effects of Cryoprotectant, Warming Solution and Removal of Lipid on Viability of Porcine Nuclear Transfer Embryos Vitrified by Open Pulled Straw Method

  • This study was carried out to investigate the effects of cryoprotectants, warming solution and removal of lipid on open pulled straw vitrification (OPS) method of porcine embryos produced by nuclear transfer (NT) of fetal fibroblasts. All solutions used during vitrification were prepared with holding medium consisting of 25 mM Hepes buffered TCM199 medium containing 20% fetal bovine serum (FBS) at $38.5^{\circ}C$. The blastocysts derived from NT with or without lipid were vitrified in each medium of different concentrations of dimethyl sulfoxide (DMSO) and ethylene glycol (EG). Also, blastocysts after cryopreservation were warmed into different concentrations of sucrose in warming solution. The optimal concentrations of cryoprotectants in vitrification solution were 10% DMSO + 10% EG in vitrification solution 1 (VS1) and 20% DMSO + 20% EG in vitrification solution 2 (VS2). The optimal concentrations of sucrose were 0.3 M sucrose in warming solution 1 (WS1) and 0.15 M sucrose in warming solution 2 (WS2). lipid removal from oocytes before NT enhanced the viability of NT embryos after vitrification. Our results show that use of the OPS method in conjunction with lipid removal provides effective cryopreservation of porcine nuclear transfer embryos.

Effect of Warming Temperature on the Viability of Bovine IVP Blastocyst Cryopreserved by Glass Micropipette (GMP) Vitrification

  • The purpose of this study was to investigate the warming temperature and exposed time on the post-thaw survival rate and viability of bovine blastocyst cryopreserved by GMP vitrification. Groups of three bovine IVP blastocysts were sequentially placed into vitrification solution before being loaded into the GMP straws and immersed into LN$_2$within 20 to 25 sec. The warming rate was increased 2 times of warming temperature for improvement of post-thaw survival rates. The frozen embryos were warmed either at 35 or 70$^{\circ}C$ for 1 or 2 sec and then diluted in sucrose solution. Post-thaw blastocysts were serially washed in 0.25 and 0.15 M sucrose in holding medium (HM: TCM199 supplemented with 10% FCS) and TCM-199 for each 5 min, respectively, and then cultured in TCM199 for 24 h. The rate of re-expanded blastocyst was significantly different fer 35 and 70$^{\circ}C$ warming temporature (76.4 vs. 89.3%; P<0.05). The rate of re-expanded blastocyst at 70$^{\circ}C$ for 1 sec was significantly higher than that for 2 sec (91.1 vs. 70.9%; P<0.05). The number of nuclei counted were significantly different among control, 35 and 70$^{\circ}C$ (121${\pm}$8.5 vs. 104${\pm}$11.7 vs. 114${\pm}$10.3; P<0.05). These results indicated that the increasing of warming rate can provide high survival rates of bovine IVP blastocysts. Especially, the best viability of post-thaw blastocyst could be thaw at 70$^{\circ}C$ for 1 sec. Effect of warming local anesthesia solutions before intraoral administration in dentistry: a systematic review

  • Background: The aim of the present systematic review was to evaluate and compare the efficacy of warmed and unwarmed local anesthesia solutions in reduction of pain during intraoral injection administration. Methods: PubMed, Ovid SP, and Cochrane Central Register of Controlled Trials were searched from publication years 1990 to 2020 with relevant MeSH terms. Studies were screened by titles and abstracts, followed by full-texts evaluation of the included studies. Results: A total of four studies were included in the systematic review. Outcomes evaluated were subjective and objective pain during administration of the warmed local anesthesia solution in comparison with the unwarmed local anesthesia solution. Among the four studies that evaluated the self-reported pain score, three studies showed significantly lower pain scores associated with warmed local anesthesia. Only two studies evaluated the observed pain score, and both of them reported a significantly lower pain reaction with the warmed local anesthesia solution. Conclusion: Within the limits of this systematic review, warming the local anesthesia solution to body temperature (37℃) before administration seemed to reduce the discomfort during intraoral local anaesthesia administration, and more high-quality studies should be carried out to validate the same.

  • A Thin-layer Wavier-Stokes equations are applied for the hypersonic flow over blunt bodies with applications to laminar as well as turbulent flows. The equations are expressed in the forms of flux-vector splitting and explicit algorithm. The upwind schemes of Steger-Warming and Van Leer are investigated to predict accurately the heating loads along the surface of the body. A mixed scheme has been presented for the differencing the convective terms and the mixed scheme is found to be less dissipative producing accurate solutions.

  • Studies were carried out to find the freezing media which gives no ice crystals in single(glycerol, ethylene glycol, dimethyl sulfoxide(DMSO)) and mixture solutions(glycerol+propylene glycol, glycerol+ethylene glycol) of permeable cryoprotectants in vitrification solution and to study effects of VS on the survival of vitrified mouse morulae. The results are summarized as follows: 1. In toxicity test of permeable cryoprotectants, 30% glycerol of single solution showed the highest FDA-score(4.1) in mouse morulae frozen compared among other single solutions. The FDA-score(4.1) of 30% glycerol was higher than 30% ethylene glycol(3.6) and DMSO(1.4( (P<0.05). 2. 20, 30 or 40% single solution of permeable cryoprotectants containing m-PBS with 10% sucrose and 20% BSA was not crystallized during cooling, but crystallized during warming. However, the 30% mixture solution of the two permeable cryoprotectants was not crystallized both during cooling and warming.3. When mouse morulae were frozen in 30% mixture solutions of two permeable cryoprotectants(glycerol and propylene glycol, glycerol and ethylene glycol), highest FDA-score(4.5) was obtained in a mixture solution of 20% glycerol and 10% ethylene glycol(20G10E) than other 30% mixture solutions(10G20E, 15G15E, 20G10P, 15G15P, 10G20P) and there was significant difference between 20G10E and 10G20E(P<0.05).

  • Green ocean is one of the main issues internationally. Most governments are leading the role in boosting the economy through creating new green market and establishing system of coping with increasing international environmental regulation. Green ocean, which is a solution for the environmental issue of global warming, is applied throughout the industry. Domestic transportation industry including railway is also developing technology for creating green ocean. Transportation must decrease energy consumption at running stages because it shows high environmental loads expecially on using stages during its life cycle. Therefore, There are some tries for developing technologies; new engine through alternative energies, hybrid and lightweight. Railway transportation can not be exception. it is intended for this paper to suggest the direction of manufacturing environmentally friendly railway vehicle through the global warming effect evaluation of several materials being applied to vehicle and comparison of the results.

The Global Warming Hiatus Simulated in HadGEM2-AO Based on RCP8.5 (HadGEM2-AO RCP8.5 모의에서 나타난 지구온난화 멈춤)

  • Despite the greenhouse gases like carbon dioxide have steadily increased in atmosphere, the overall trend of the global average surface air temperature has stalled during the last decade (2002-present). This phenomenon is often called hiatus or warming pause, which is challenging the prevailing view that anthropogenic forcing causes warming environment. Our study characterized the hiatus by analyzing the HadGEM2-AO (95 yrs) simulation data based on RCP8.5 scenario. The PC2 time series from the EOF of the zonal mean vertical ocean temperature has been defined as the index that represents the warming pause. The relationship between the hiatus, ENSO and the changes in climate system are identified by utilizing the newly defined PC2. Since the La Nina index (defined as the negative of NINO3 index) leads PC2 by about 11 months, it may be possible that the La Nina causes the warming to be interrupted. We also show that the cooling of the climate system closed tied to the heat penetration into the deep ocean, indicating the weakening the warming rate is due to the oceanic heat uptake. Finally, the global warming hiatus is characterized by the anomalous warming in Arctic region as well as the intensification of the trade wind in the equatorial Pacific.

  • Many people all over the world have been conducting a great deal of research to solve the problem of global warming since the great majority consider reduction of CO2 as the only solution for that. That is why the production and conservation of energy is thought to be highly crucial. while it is important to produce energy with the high efficiency, the efficient use of the energy is also important. This paper focused on the development of devices for the reducing electricity which is a primary energy source used in homes, shops, buildings, factories and so on. Also the objective of this paper is to develop the inference mechanism as the core component of the devices. Therefore, in this paper, we propose the inference algorithm for reducing the electricity consumption using SARIMA mode and present the feasibility of the procedure.

  • Recently large capacity of electric equipment and increasing in atomic power generation are shown. One of the reason is shortage of the electric power supply for air conditioning load during summer. And every consumer is concerning about economical refrigeration and air conditioning system to decreases electric power consumption and decrease in global warming. For these necessities, ice making thermal storage system is required. Therefore, in this paper, the possibility of continuous slurry ice making using flowing organic water solution in cooled circular tube has been investigated. The experiments was carried out under some parameters of concentration and velocity of water solution, temperature of cooled tube wall, and control pressure in tube, As a result, four types of operating conditions in the pipe, that was supercooling, continuous ice making, intermittent ice making and ice blockage, were classified . And it was found that the critical condition for continuous ice making was acquired as a function of these experimental parameters.

Effect of S-Adenosylmethionine on Hepatic Injury from Sequential Cold and Warm Ischemia

  • We investigated whether S-adenosylmethionine (SAM) treatment improved ischemic injury using perfused rat liver after sequential periods of 24 h cold and 20 min re-warming ischemia. SAM (100 $\mu\textrm{mol/L}$) was added to University of Wisconsin (UW) solution and Ringers lactate solution. After cold and sequential warm ischemia, releases of lactate dehydrogenase (LDH) and purine nucleoside phosphorylase (PNP) markedly increased during repefusion. The increase in PNP was significantly reduced by SAM treatment. While the concentration of reduced glutathione (GSH) in ischemic livers significantly decreased, the concentration of glutathione disulfide (GSSG) increased. This decrease in GSH and increase in GSSG were suppressed by SAM treatment. Lipid peroxidation was elevated in cold and warm ischemic and reperfused livers, but this elevation was also prevented by SAM treatment. Hepatic ATP levels were decreased in the ischemic and reperfused livers to 42% of the control levels. However, treatment with SAM resulted in significantly higher ATP levels and preserved the concentration of AMP in ischemic livers. Our findings suggest that SAM prevents oxidative stress and lipid peroxidation and helps preserve hepatic energy metabolism.
