Various Biological Activities of Ramie (Boehmeria nivea)

  • The purpose of this study was to evaluate the biological activities of extracts of ramie (Boehmeria nivea (L.) Gaud.), hereafter referred to as Bn. Bn extracts from various collecting area were extracted with methanol. Two extracts from our study, Bn-13 and -82, showed significant antioxidant properties, likely due to their ability to scavenge free radicals. In addition, Bn extracts showed stronger anti-bacterial activity against Escherichia coli (Bn-40), Stapylococcus aureus (Bn-33), and Helicobacter pylori (Bn-05). In addition, this study was conducted to evaluate the anti-inflammatory effects of Bn extracts in lipopolyssacharide (LPS)- and interferon-${\gamma}$ (IFN-${\gamma}$)-stimulated RAW 264.7 macrophages cells. Bn-37 significantly inhibited the production LPS/IFN-${\gamma}$-induced nitric oxide. The most noteworthy anti-cancer effect was found in Bn-23. Bn-08 showed inhibition of aldose reductase. This study provides basic information for the development of functional foods.

Screening of Biological Activities of Grass Clippings from Turfgrass (잔디 예지물의 생리활성 탐색)

  • This study was carried out to investigate the biological activities of grass clippings from turfgrass including Zoysia japonica (TG-1), Zoysia matella (TG-2), Agrostis palustris (TG-3), and Poa pratensis (TG-4). The 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity was relatively higher in TG-1 and -2. Especially, TG-1 exerted the strongest hydroxyl radical scavenging effect, showing 90.40% at the concentration of 100 μg/ml. In addition, TG-1 inhibited the growth of Staphylococcus aureus, Escherichia coli and Helicobactor pylori. TG-1 also showed the highest inhibitory effect of AGS human gastric adenocarcinoma cell growth and nitric oxide production against lipopolysaccharide in RAW 264.7 macrophage cells. In conclusion, among the TG extracts, TG-1 has anti-oxidative, anti-microbial, anti-cancer and anti-inflammatory effect, indicating that TG-1 may be the potential source of functional food.

  • CFT(Concrete-Filled Tube) structures have problems at processing cause closed section and concrete filling problems. In this study, a CFT structure that uses different types of diaphragms in its upper and lower connections to improve the concrete filling was tested and analyzed via the FEM program. Implementation of variable analysis of EP-T type to find out the reason that effect on the resistance force of the connection. As a result, through experiments and analysis investigated the structural characteristics of circular CFT beam-to-column connection.

  • The objective of this study is to investigate the influence that how does contact surface between concrete and steel influence the steel encased composite column by push-out test. Also nominal bond stress indicated by design standard such as Eurocode 4 is underestimated in small scale steel encased composite column. The other objective of this study is to investigate how does the number and space of shear connector influence the H-shaped steel encased composite column. The shear behavior of shear connectors is investigated by push-out test.

  • β amyloid protein (Aβ) plays a critical role in the pathogenesis of Alzheimer's disease (AD) and possibly in Aβ-induced mitochondrial dysfunction and oxidative stress. Aβ can directly cause reactive oxygen species (ROS) production. Overproduction of ROS is considered to be involved in the pathogenesis of neurodegeneration of AD. Here, we investigated 9 kinds of ramie (Boehmeria nivea, (L.) Gaud., BN; hereafter denoted as BN) for their protective action against oxidative stress in a cellular system using C6 glial cells. We observed loss of cell viability and high levels of ROS generation after treatment with hydrogen peroxide (H2O2) and Aβ25-35. However, treatments with BN extracts led to an increase in cell viability and decrease in ROS production induced by H2O2 and Aβ25-35. In particular, the extracts of BN-01 (seobang variety from Seocheon) and BN-09 (local variety from Yeonggwang) showed excellent anti-oxidative properties. This indicates that BN extracts could prevent neurodegeneration by reducing oxidative stress in cells.

Application of Artificial Neural Network Model for Environmental Load Estimation of Pre-Stressed Concrete Beam Bridge (PSC Beam교 환경부하량 추정을 위한 인공신경망 모델 적용 연구)

  • Considering that earlier stage of construction project has a great influence on the possibility of lowering of environmental load, it is important to build and utilize system that can support effective decision making at the initial stage of the project. In this study, we constructed an environmental load estimation model that can be used at the early stage of the project using basic design factors. The model was constructed by using the artificial neural network to estimate environmental load by applying to planning stage (ANN-1), basic design stage (ANN-2). The result of test, shows that average of absolute measuring efficiency and standard deviation of ANN-1 and ANN-2 were 11.19% / 5.30% and 9.59% / 3.09% each. This result indicates that the model using the input variables extended with the project progress has high reliability and it is considered to be effective in decision support at the initial design stage of the project.

  • Small and medium-sized enterprises (SMEs) account for a large proportion of the total number of enterprises in many countries. The development of SMEs has contributed to job creation and economic benefits. Every government has formulated active diversification strategies to promote the export market of SMEs, but the performance of export capabilities remains insufficient. The primary purpose of this study is to examine the effectiveness of export credit insurance in promoting SME export performance in Canada. Using data from 2008-2017, the augmented Dickey-Fuller (ADF) model to test the stationarity of the concerned variables and the error correction model (ECM) and autoregressive distributed lag (ARDL) cointegration test to empirically investigate the cointegration relationship between the research targets. The results represent the positive and critical impact of export relative price and domestic demand pressure on Canada's export performance, and the negative impact of the export volume index at a significant level. Regrettably, the impact of export credit insurance on the export performance of Canadian SMEs is considered exaggerated overall. In view of this result, it is necessary for the Canadian government to enact policies based on the current market status. And enhance confidence among SMEs to begin exports and diversify their markets rather than focusing only on the domestic or US market, especially given the impact of COVID-19. From the case of Canada, Korean government can attempt to learn from them to conduct more efficient strategies for SMEs.

Tissue Fluid Enzyme-Linked Immunosorbant Assay for Piglets Experimentally Infected with Toxoplasma gondii and Survey on Local and Imported Pork in Korean Retail Meat Markets

  • To investigate the prevalence of Toxoplasma gondii in pork on the market in Korea, an in-house enzyme-linked immunosorbent assay for tissue fluid (CAU-tf-ELISA) was developed using a soluble extract of T. gondii RH strain tachyzoites. As the standard positive controls, the piglets were experimentally infected with T. gondii: Group A (1,000 cysts-containing bradyzoites), Group B (500 cysts-containing bradyzoites) and Group C ($1.0{\times}10^3$ or $1.0{\times}10^4$ tachyzoites). The CAU-tf-ELISA demonstrated infection intensity-dependent positivity toward tissue fluids with average cut-off value 0.15: 100% for Group A, 93.8% for Group B and 40.6% for Group C. When tissue-specific cut-off values 0.066-0.199 were applied, CAU-tf-ELISA showed 96.7% sensitivity, 100% specificity, 100% positive and 90.0% negative predictive values. When compared with the same tissue fluids, performance of CAU-tf-ELISA was better than that of a commercial ELISA kit. Of the 583 Korea domestic pork samples tested, anti-T. gondii antibodies were detected from 9.1% of whole samples and 37.9% from skirt meat highest among pork parts. In the 386 imported frozen pork samples, 1.8% (skirt meat and shoulder blade) were positive for anti-T. gondii antibodies. In Korea, prevalence of anti-T. gondii antibodies in the pork on retail markets appeared high, suggesting that regulations on pig farming and facilities are necessary to supply safe pork on the tables.

Apportionment of Liquidated Damages and Compensation for Delay Damages in Domestic Construction Project : Analysis and Improvement (국내 현행 공기지연 책임에 따른 지체상금 및 손실보상의 문제점 및 개선방안)

  • To calculate the amount of owner-caused and contractor-caused delays based on a simplified delay analysis, which has been customarily used in Korea, has a limitation in reflecting the impact of the concurrent delay and the acceleration work. It also resulted in the apportionment of liquidated damages by applying the ratio of the number of delays between the owner and the contractor. This study analyzes that the conventional method does not meet the international standards. In order to improve the problem of construction delay analysis and the apportionment of liquidated damages based on it, owner delays, contractor delays, concurrent delays, and the impact of acceleration should be analyzed together. This study suggests that in the apportionment of liquidated damages, the extension of time should be extended by the sum of concurrent delays and the owner-caused delays, and liquidated damages should be imposed on delays incurred after the extension of time. It can be seen that it conforms to the international standards. The results of this study are expected to contribute to improving the problems of delay analysis and liquidated damages calculation, which have been conventionally accepted.

A systematic study of nuclear interactome of C-terminal domain small phosphatase-like 2 using inducible expression system and shotgun proteomics

  • RNA polymerase II C-terminal domain phosphatases are newly emerging family of phosphatases that contain FCPH domain with Mg+2-binding DXDX(T/V) signature motif. Its subfamily includes small CTD phosphatases (SCPs). Recently, we identified several interacting partners of human SCP1 with appearance of dephosphorylation and O-GlcNAcylation. In this study, using an established cell line with inducible CTDSPL2 protein (a member of the new phosphatase family), proteomic screening was conducted to identify binding partners of CTDSPL2 in nuclear extract through immunoprecipitation of CTDSPL2 with its associated. This approach led to the identification of several interacting partners of CTDSPL2. This will provide a better understanding on CTDSPL2.
