Prokaryotic Selectivity, Anti-endotoxic Activity and Protease Stability of Diastereomeric and Enantiomeric Analogs of Human Antimicrobial Peptide LL-37

  • LL-37 is the only antimicrobial peptide (AMP) of the human cathelicidin family. In addition to potent antimicrobial activity, LL-37 is known to have the potential to inhibit lipolysaccharide (LPS)-induced endotoxic effects. To provide the stability to proteolytic digestion and increase prokaryotic selectivity and/or anti-endotoxic activity of two Lys/Trp-substituted 19-meric antimicrobial peptides (a4-W1 and a4-W2) designed from IG-19 (residues 13-31 of LL-37), we synthesized the diastereomeric peptides (a4-W1-D and a4-W2-D) with D-amino acid substitution at positions 3, 7, 10, 13 and 17 of a4-W1 and a4-W2, respectively and the enantiomeric peptides (a4-W1-E and a4-W2-E) composed D-amino acids. The diastereomeric peptides exhibited the best prokaryotic selectivity and effective protease stability, but no or less anti-endotoxic activity. In contrast, the enantiomeric peptides had not only prokaryotic selectivity and anti-endotoxic activity but also protease stability. Our results suggest that the hydrophobicity and ${\alpha}$-helicity of the peptide is important for anti-endotoxic activity. In particular, the enantiomeric peptides showed potent anti-endotoxic and LPS-neutralizing activities comparable to that of LL-37. Taken together, both a4-W1-E and a4-W2-E holds promise as a template for the development of peptide antibiotics for the treatment of endotoxic shock and sepsis.

Essential role of tryptophan residues in toxicity of binary toxin from Bacillus sphaericus

  • Bacillus sphaericus produces mosquito-larvicidal binary toxin composed of BinA and BinB. While BinB is expected to bind to a specific receptor on the cell membrane, BinA interacts to BinB or BinB receptor complex and translocates into the cytosol to exert its activity via unknown mechanism. To investigate functional roles of aromatic cluster in BinA, amino acids at positions Y213, Y214, Y215, W222 and W226 were substituted by leucine. All mutant proteins were highly produced and their secondary structures were not affected by these substitutions. All mutants are able to insert into lipid monolayers as observed by Langmuir-Blodgett trough and could permeabilize the liposomes in a similar manner as the wild type. However, mosquito-larvicidal activity was abolished for W222L and W226L mutants suggesting that tryptophan residues at both positions play an important role in the toxicity of BinA, possibly involved in the cytopathological process after toxin entry into the cells.

Dielectric Properties of Strontium-substituted Lead Magnesium Tungstate up to Microwave Frequencies

  • $Pb_{1-x}Sr_x(Mg-{1}2}W_{1}2})O_3$$ ceramics for application in the microwave frequency range were investigated by dielectric constant and X-ray diffraction measurements. The dielectric constant curves showed two concentration dependent characteristics in the $$Pb(Mg-{1}2}W_{1}2})O_3$-rich$ region. As the Sr constant further increases to x=0.3 the dielectric curve levels off. In the concentration range between x=0.4 and x=1 in which dielectric constant dependence on temperature is negligible, it decreases and Qf value increases in the microwave frequency with increasing Sr. The temperature coefficient (${\tau}_{\varepsilon} $) of the dielectric constant changes from the negative to positive value between x=0.9 and x=1. The dielectric constant, Qf and $\tau\varepsilon$ are correlated with tolerance factor(t). From the X-ray diffraction results for $0.1{\le}x{\le}1$ the cell parameter is found to decrease as x increases and B-site ordering is observed in all the composition ranges.

  • Lactobacillus helveticus was inoculated to the fermentation liquid containing skin milk powder(SMT) plus soymilk, SMP plus isolated soybean protein(ISP), SMP plus ISP plus whey powder(WP) to increase the nutritional and economic value of commercial soy yogurt. The yogurt fermented with soymilk and SMP showed the lower acid production than of SMP and had significant beany flavor in the product. The yogurt prepared with ISP and SMP showed the higher cell number and lower acid production than that of SMP. Also, the partial substitution of SMP with ISP over 6%(w/w) produced less acceptable product due to gel production. The yogurt prepared by the partial substitution of SMP with ISP, WP and SMP showed the higher cell number and lower acid production than that of SMP and not bring about gel formation unlike the case of ISP. Sensory properties of yogurt substituted SMP with ISP and WP(38:62 mixture) below 4% were not significantly different from that of SMP and the sample containing the mixture over 6% and 0.067% artificial flavor showed lower sensory score due to beany taste than that of SMP. But increase of yogurt flavor up to 0.1% resulted in significantly high score in organoleptic acceptability. The separation of water occured in yogurt prepared by the combined mixture of ISP, WP and SMP, and this problem could be resolved by addition of Na-alginate and PGA at the concentration of 0.1%(w/w).

  • The dynamics and Durability of Permanent Form Mortar were observed. The results were as the followings. To construct New permanent form, this study constructed permanent form with fiber mortar. In this course glass fiber and polymer, fine aggregate were substituted with river sand and crush sand. To understand material quality effect, flexible mortar and inflexible mortar were tested. According to the microstructure of polymermatrix test by SEM, higher density by Co-matrix effect from polymer addiction was observed. This is considered to increase acid-resistanc, impact proof strenght. In the test of durability of freezing and thawing, polymer mortar was proved to be water proof. It seems to be improved in its characteristics in proportion to increasing W/C ratio. When the W/C ratio is 30%, it is most effective.

  • In batch culture of A. pullulans HP-2001, maximal production of pullulan was 57.90 g/l at 96 hr. The maximal production of pullulan with 76.55 g/l occurred when substituted solution was 10% (w/v) sucrose, 0.25% (w/v) yeast extract and mineral salts. The maximal production of pullulan was 75.00 g/l with a dilution rate of $0.015\;h^{-1}$. Productivity of pullulan in the continuous culture fed with the fresh medium was higher than that fed with the fresh medium without nitrogen source.

  • A 4-bit parallel-series A/D converter has been designed using a new matrix circuit and breadboarded with transister array(TPQ2483). The simple matrix circuit is substituted for D/A converter and sebtracter-multiplier. The system has been simulated with SPICE. This converter is capable of operating at clock rate of 20MHz.

  • The doubly altered mutant tryptophan synthase $\alpha$-subunits, in which Thr 24 was replaced by Ser, Leu or Lys in addition to F139W substitution, were purified. Urea-induced unfolding equilibrium curves of these proteins, monitored by fluorescence intensity of tryptophan, show that the alterations of residue 24 resulted in marked changes in folding properites, suggesting the importance of this residue in folding of this protein.

  • Recently in the construction site is required various performance of mortar to deal with reliability, economy. Especially of environment-friendly, saving resources and high performance of mortar it began to require improvement of performance. Therefore, in this study, a series of tests was designed to develop a using mud flat mortar in view of the high absorption rate of tidal flats, to fix the water cement ratio to 70%. The mixing ratio were 10:0, 9:1, 8:2, 7:3, 6:4, 5:5, 4:6 of fine aggregate and mud flats. Compared the flow value, chloride content, compressive strength, tensile strength, and workability of fine aggregate the substituted mud flat mortar and basic cement mortar, present the basic data on the characteristics and effects of mud flat mortar.

  • We have prepared 20 kinds of Mn-Zn ferrites as content of CaO(0.1 mol%), NiO(0.0~0.60 mol%) and CoO(0.0~0.8 mol%) adding by the coprecipitation method and studied the magnetic properties as content of CaO, NiO and CoO adding. Initial permeability decrease as the content of NiO and CoO adding increases, while Curie tem~ perature increase as the content of NiO and CoO adding increases. $(H_{c})$, $(B_{s})$ and $(W_{h})$ increase as content of NiO adding increases.
