Effects of Different Levels of CP Intake on Protein Utilization and N Excretion in Varying Growth Stages of Hanwoo Steers (한우 거세우의 성장단계별 CP 급여수준이 단백질 체내 이용성 및 N 배설량에 미치는 효과)

  • Three experiments with different stage of Hanwoo steers (average BW of 167kg, 355kg and 449kg, respectively) were conducted to examine the effect of different quantities of CP intake with similar DM intake on protein balance and N excretion. Twelve Hanwoo steers in each experiment were offered three diets containing three levels of protein (9, 14 and 18% on DM basis) in the concentrate formulated with using different amount of ground corn grain or corn gluten meal. There were significant effects of increasing CP intake on CP digestibility but not on DM digestibility. Despite different CP intake, apparent fecal N outputs were not significantly affected. However, amounts of N excreted into urine increased (p<0.05) with increasing CP intake. There was an obvious effect of live body weight on efficiency of body protein conversion of CP consumed in excess of animal requirement for maintenance, with slopes of 70% and 46~39% for growing and fattening stage, respectively. Nitrogen excretion into feces and urine in this experiment was 70% of the daily N consumption for fattening stage, compared with 60% for growing stage.

Diurnal Patterns in the Flow of Escapable Soluble Non-Ammonia Nitrogen Fractions in Omasal Digesta as Influenced by Barley and Rapeseed Meal Supplementation in Cows Fed Grass Silage Based Diet (목초 사일리지 급여 시 보리와 채종박 보충급여에 의한 제 3위 소화액내 Soluble Non-ammonia Nitrogen Fraction의 Flow 패턴 변화)

  • The present study was conducted to measure diurnal patterns in the flow of soluble non-ammonia nitrogen (SNAN) fractions in the liquid phase of digesta entering the omasum of cows fed grass-red clover silage supplemented with barley and rapeseed meal. Four ruminally cannulated cows were fed, in a 4×4 Latin square design, grass-red clover silage alone (GS) or supplemented with (on a DM basis) 6.0kg/d of barley grain, 2.1kg/d of rapeseed meal or 6.0kg/d of barley and 2.1kg/d rapeseed meal. Omasal digesta was taken using an omasal sampling system at 1.5h intervals during a 12h feeding cycle, and SNAN fractions (free AA, peptide and soluble protein) in the omasal digesta were assessed using ninhydrin assay. Dietary supplementation numerically increased the mean flow of SNAN fractions relative to GS diet despite the lack of statistical significance. Diurnal patterns in the flow of peptide entering the omasum during a 12h feeding cycle appeared to be highest immediately after feeding, declined by 10.0h post-feeding and slightly increased thereafter. In SNAN fractions, the flow of peptide was higher for supplemented diets than for GS diet throughout the feeding cycle. Based on the microbial contribution to total SNAN using 15N, diurnal patterns in the flow of dietary SNAN for dietary supplemented diets appeared to be higher compared with GS diets. Present results may conclude that peptide flow is quantitatively the most important N in SNAN fractions and that dietary supplementation can increase peptide flow entering the omasal canal.

QTL analysis of agronomic traits in recombinant inbred lines of sunflower under partial irrigation

  • The objective of the present research was to map QTLs associated with agronomic traits such as days from sowing to flowering, plant height, yield and leaf-related traits in a population of recombinant inbred lines (RILs) of sunflower (Helianthus annuus). Two field experiments were conducted with well-irrigated and partially irrigated conditions in randomized complete block design with three replications. A map with 304 AFLP and 191 SSR markers with a mean density of 1 marker per 3.7 cM was used to identify QTLs related to the studied traits. The difference among RILs was significant for all studied traits in both conditions. Three to seven QTLs were found for each studied trait in both conditions. The percentage of phenotypic variance ($R^2$) explained by QTLs ranged from 4 to 49%. Three to six QTLs were found for each yield-related trait in both conditions. The most important QTL for grain yield per plant on linkage group 13 (GYP-P-13-1) under partial-irrigated condition controls 49% of phenotypic variance ($R^2$). The most important QTL for 1,000-grain weight (TGW-P-11-1) was identified on linkage group 11. Favorable alleles for this QTL come from RHA266. The major QTL for days from sowing to flowering (DSF-P-14-1) were observed on linkage group 14 and explained 38% of the phenotypic variance. The positive alleles for this QTL come from RHA266. The major QTL for HD (HD-P-13-1) was also identified on linkage group 13 and explained 37% of the phenotypic variance. Both parents (PAC2 and RHA266) contributed to QTLs controlling leaf-related traits in both conditions. Common QTL for leaf area at flowering (LAF-P-12-1, LAF-W-12-1) was detected in linkage group 12. The results emphasise the importance of the role of linkage groups 2, 10 and 13 for studied traits. Genomic regions on the linkage groups 9 and 12 are specific for QTLs of leaf-related traits in sunflower.

Keyhole Imaging Combined Phase Contrast MR Angiography Technique (Keyhole Imaging기법을 적용한 위상대조도 자기공명 혈관조영기법)

  • Phase Contrast MR Angiography(PC MRA) is excellent MRA technique for measuring the velocity of vessels in the human body. PC MRA need to at least four images for angiogram reconstruction and it caused longer scan time. Therefore, we used keyhole imaging combined PC MRA to reduce the scan time. However, keyhole imaging can lead the erroneous effects as loss of phase information or frequency discontinuous. In this study, we applied the keyhole imaging combined 2D PC MRA for improving the temporal resolution and also measured the velocity to evaluate the accuracy of phase information. We used 0.32T MRI scanner(Magfinder II, Scimedix, Korea). Using the 2D PC MRA pulse sequence, the vascular images for a human brain targeted on the Superior Sagittal Sinus(SSS) were obtained. We applied tukey window function for keyhole images to minimize the ringing artifact and erroneous factors that are induced frequency discontinuous and phase information loss. We also applied zero-padded algorithm to peripheral missing k-space lines to compare keyhole imaging results and the artifact power(AP) value was measured on the complex difference images to validate the image quality. Consider as based on our results, heavy image distortions and artifacts were shown until using at least 50% keyhole factor. Using above the 50% keyhole factors are shown well reconstructed and matched for magnitude images and velocity information measurements. In conclusion, we confirmed the image quality and velocity information of keyhole technique combined 2D PC MRA. Especially, measured velocity information through the keyhole imaging combination was similar to the velocity information of full sampled k-space image despite of frequency discontinuous and phase information loss in the keyhole imaging reconstruction process. Consequently, the keyhole imaging combined 2D PC MRA will give some clinical usefulness and advantages as improving the temporal resolution and measuring the velocity information via selecting the appropriate keyhole factor at low tesla MRI system.

  • The purpose of this study is to collect the raw data for conservation of plant ecosystem by surveying and analysing the flora of Mt. Geumgok located in Gyeongju-si, Gyeongsangbuk-do, Korea. The flora were summarized as 453 taxa including 91 families, 298 genera, 397 species, 4 subspecies, 46 varieties and 6 forms. The rare plants designated by Korea Forest Service were 3 taxa such as Eranthis byunsanensis B.Y.Sun, Potentilla discolor Bunge and Iris odaesanensis Y.N.Lee. The Korean endemic plants were 6 taxa such as Carpinus laxiflora (Siebold & Zucc.) Blume, Eranthis byunsanensis B.Y.Sun, Philadelphus schrenkii Rupr., Lespedeza maritima Nakai, Vicia chosenensis Ohwi and Weigela subsessilis (Nakai) L.H.Bailey. The specific plants by floristic region were 36 taxa such as Pinus koraiensis Siebold & Zucc., Salix chaenomeloides Kimura, Anemone raddeana Regel, Chloranthus japonicus Siebold, Euphorbia pekinensis Rupr., Ilex macropoda Miq., Ajuga multiflora Bunge, Saussurea odontolepis Sch.Bip. ex Herd, Viola orientalis (Maxim.) W.Becker, Betula davurica Pall., Vitex negundo var. incisa (Lam.) C.B.Clarke and Cimicifuga heracleifolia Kom.. The naturalized plants were 36 taxa such as Fallopia dumetorum (L.) Holub, Lepidium apetalum Willd., Robinia pseudoacacia L., Trifolium repens L., Euphorbia supina Raf., Ipomoea purpurea Roth, Veronica persica Poir., Bidens pilosa L., Carduus crispus L., Xanthium canadense Mill., Bromus unioloides H.B.K. and Festuca arundinacea Schreb.. The invasive alien plants were 2 taxa such as Ambrosia artemisiifolia L. and Aster pilosus Willd.. The ratios of the urbanized index(UI), the naturalized index(NI) and the disturbed index(DI) were 11.2%, 7.9% and 18.2% each.

Effects of L-Carnitine with Different Lysine Levels on Growth and Nutrient Digestibility in Pigs Weaned at 21 Days of Age

  • This study was conducted to investigate the effects of L-carnitine with different levels of lysine on performance of pigs weaned at 21 days of age. A total of 120 pigs were allotted into a $3{\times}2$ factorial design with three different levels of lysine (1.40%, 1,60% and 1.80%) and two levels of L-carnitine (0 and 1,000 ppm). Each treatment had 4 replications with 5 pigs per replicate. Pigs of $22{\pm}1$ days (5.9 kg of body weight) were grouped into a completely randomized block design. Treatments were 1) 1.4-Crt; 1.40% of lysine with 1,000 ppm of L-carnitine, 2) 1.4-N; 1.40% of lysine without L-carnitine, 3) 1.6-Crt; 1.60% of lysine with 1,000 ppm of L-carnitine, 4) 1.6-N; 1.60% of lysine without L-carnitine, 5) 1.8-Crt; 1.80% of lysine with 1,000 ppm of L-carnitine and 6) 1.8-N; 1.80% of lysine without L-carnitine. Growth performance was optimized in pigs fed 1.6% lysine regardless of carnitine addition. For the first 7 days of the experimental period, the best ADG and F/G were found in pigs within the 1.6-Crt group. Carnitine significantly improved (p<0.05) ADG of pigs when the lysine level in the diet was 1.6%. Only in the third week carnitine had a significant influence on growth performance of pigs. A lysine-sparing effect of L-carnitine was not detected in this study. The 1.6-Crt group showed the best proximate nutrient digestibility, and the crude fat and gross energy digestibility were higher when the L-carnitine was added in the diet. Lysine level significantly affected the digestibilities of DM (p<0.001), GE (p<0.001), CP (p<0.01) and C.fat (p<0.05). Carnitine also significantly improved digestibility of nutrients. Lysine level as well as carnitine level affected the amino acids digestibility, however, in 1.8% lysine diet carnitine did not influence on amino acids digestibility. Plasma carnitine content was significant higher (p<0.05) in pigs fed L-carnitine. This indicates the increased biological availability of carnitine within the body. L-carnitine supplementation tended to improve feed utilization during the third week (p<0.10) and during the entire period (p=0.10). Lysine level significantly affected feed utilization of pigs during the third week and entire period (p<0.05). As pigs grew, the lysine requirement was reduced.

Comparative Calorimetric Evaluation of Ammoniated Straw-Based Rations Supplemented with Low Levels of Untreated and Formaldehyde Treated Groundnut Cake and Fish Meal with Respect to Growing Buffalo Calves

  • Eighteen growing male Murrah buffalo (Bubalus bubalis) calves were divided into three groups consisting of six animals each and fed three urea ammoniated wheat straw (UAS) -based rations supplemented with concentrate mixtures (roughage: concentrate ratio 58:42) containing deoiled ground nut cake, GNC (8%), formaldehyde treated GNC (8%) or fish meal (8%) to undertake comparative evaluation of these rations in terms of their $CH_4$ production and growth (285 d duration) potential. A digestibility trial (10 d duration) was followed by a comparative calorimetric study in respiration chamber. Dry matter (DM) intake (84.3 to $89.3g/kg\;W^{0.75}d^{-1}$) did not differ between treatments. The digestibility coefficient of DM, organic matter (OM), crude protein (CP), neutral and acid detergent fiber did not differ significantly in different diets. Urinary energy loss as a percent of gross energy (GE) was not affected by diets. Average values of $CH_4$ production were 84.3, 77.6 and 99.1 g/d and $CH_4$ energy losses as percent of gross energy were 5.7, 5.2 and 6.1 percent on .GNC, formaldehyde treated GNC and fishmeal, respectively, and did not differ significantly. When expressed per unit of digestible OM intake, $CH_4$ production (g) was lower (p<0.05) on formaldehyde treated GNC (30.6) than on untreated GNC (30.6) and fish meal (31.9). Total ME intake and heat production were similar and hence the energy balances on different diets were similar. Nutritive value of rations in terms of digestible CP and ME were similar. Average daily gain calculated on the basis of regression of fortnights on cumulative liveweight gain in calves fed on concentrate containing unprotected GNC, protected GNC and fish meal were 437.1, 483.9 and 481.6 g, respectively. This indicated that the intake of energy was sufficient to meet the requirement of calves growing at 400 g per d. However, CP intake was around 150% of the stipulated standard (Kearl, 1982). Feed conversion ratios on unprotected GNC, protected GNC and fish meal were 11.60, 11.10 and 10.4 respectively. It was concluded that because significantly (p<0.05) low $CH_4$ is produced on protected GNC (8%), it is very good and sustainable protein source in comparison to poor quality fish meal and untreated GNC to be used in concentrate mixture for supplementing UAS-based diets.

Follicular Stimulation and Laparoscopic Ovum Pick-up (LOPU) in Repeatedly Superovulated Korean Black Goats (반복적인 과배란 처치 경험이 있는 한국흑염소에서 난포 자극 및 복강경을 이용한 난포란 채취(LOPU))

  • Laparoscopic ovum pick-up (LOPU) is a convenient method for collecting oocytes in small ruminants. LOPU has the advantage of being a less invasive means of oocyte collection, thereby allowing for a repeated usage of the oocyte donor animals. A total of 25 Korean black goats were used in the winter season (December to February) and LOPU was applied to the goats which had been treated for superovulation more than two times during the last twelve months. Estrus was synchronized with an intravaginal insert containing 0.3 g progesterone for 10 to 12 days. Ovaries were hyperstimulated with eCG 1,000 IU oneshot, FSH with eCG (50 mg / 1,000 IU; 70 mg / 500 IU; 70 mg / 1,000 IU) oneshot or FSH multiple-shot with eCG oneshot ($20mg{\times}6/300IU$) given intramuscularly 72 h prior to LOPU. For these groups, the number of follicles (mean ${\pm}$ SEM) observed which developed to larger than 2 mm in diameter were $1.6{\pm}2.5$, $4.3{\pm}3.1$, $5.5{\pm}4.2$, $6.6{\pm}2.1$ and $8.8{\pm}7.8$, respectively. Oocytes were aspirated by using OPU needles and a vacuum pump. The overall oocyte retrieval rates were 41.4%. Oocytes were matured in TCM-199 supplemented with 10% (w/v) bovine serum albumin + $10{\mu}g/ml$ FSH + $1{\mu}g/ml$ $17{\beta}$-estradiol for 27 h at $39^{\circ}C$ in 5% $CO_2$ in air. Oocytes were parthenogenetically activated by ionomycin combined with 6-diethylaminopurine (6-DMAP). Total oocyte maturation and cleavage rate were 67.3% and 78.8%, respectively. In summary, LOPU is a useful oocyte collection method in Korean black goats that can provide immature oocytes for transgenesis or nuclear transfer.

Study on the Improvement of Ballast Water Risk Assessment of GloBallast (GloBallast 선박평형수 위해도평가 방법의 개선방안에 관한 연구)

  • Recently, the management of the marine invasive alien species introduced by ballast water has come to the core as an urgent task. Because, hazardous organisms cause harmful effects on the ecosystem. Hazardous organisms also negatively affect fisheries, commercial shipping activities, and marine resources. In this regard, IMO used to operate GloBallast, Ballast Water Risk Assessment System, in order to manage the introduction of invasive species by ballast water. GloBallast is constructed to assess the relative risk due to the movement of ballast water between nations, using the volume and the number of times of ballast water discharge, the environmental similarity between ports, and the abundance of hazardous species as the main factors. According to the results of the demo-operation of GloBallast, it was found out that the environmental similarity factor had affected most highly the results of risk assessment. Therefore, in this study, the correlation between the main factors used in GloBallast risk assessment were re-examined, and some methods were suggested in order to reflect these main factors in the risk assessment more reasonable.

Novel 4,7-Dithien-2-yl-2,1,3-benzothiadiazole-based Conjugated Copolymers with Cyano Group in Vinylene Unit for Photovoltaic Applications

  • Two novel conjugated copolymers utilizing 4,7-dithien-2-yl-2,1,3-benzothiadiazole (DTBT) coupled with cyano (-CN) substituted vinylene, as the electron deficient moeity, have been synthesized and evaluated in bulk heterojunction solar cell. The electron deficient moeity was coupled with carbazole and fluorene unit by Knoevenagel condition to provide poly(bis-2,7-((Z)-1-cyano-2-(5-(7-(2-thienyl)-2,1,3-benzothiadiazol-4-yl)-2-thienyl)ethenyl)-alt-9-(1-octylnonyl)-9H-carbazol-2-yl-2-butenenitrile) (PCVCNDTBT) and poly(bis-2,7-((Z)-1-cyano-2-(5-(7-(2-thienyl)-2,1,3-benzothiadiazol-4-yl)-2-thienyl)ethenyl)-alt-9,9-dihexyl-9H-fluoren-2-yl) (PFVCNDTBT). The optical band gaps of PCVCNDTBT (1.74 eV) and PFVCNDTBT (1.80 eV) are lower than those of PCDTBT (1.88 eV) and PFVDTBT (2.13 eV), which is advantageous to provide better coverage of the solar spectrum in the longer wavelength region. The high $V_{oc}$ value of the PSC of PCVCNDTBT (~0.91 V) is attributed to its lower HOMO energy level ( 5.6 eV) as compared to PCDTBT ( 5.5 eV). Bulk heterojunction solar cells based on the blends of the polymers with [6,6]phenyl-$C_{61}$-butyric acid methyl ester ($PC_{61}BM$) gave power conversion efficiencies of 0.76% for PCVCNDTBT under AM 1.5, 100 mW/$cm^2$.
