Effects of Cryoprotectants on In Vitro Development of Vitrified Immature Porcine Oocytes Following ICSI

  • In the present study, effects of concentration of cryoprotectant solutions on the nuclear maturation of vitrified-thawed porcine oocytes were examined. Also, the developmental capacity of vitrified-thawed immature porcine oocytes following ICSI was investigated. Oocytes were cultured in NCSU-23 medium supplemented with 5% FBS at $38^{\circ}C$ in 5% $CO_2$ and air. The in vitro maturation rate of vitrified-thawed oocytes ($24.1{\pm}2.5%$) was lower than that of the control ($46.0{\pm}3.2%$, p<0.05). The in vitro maturation rate of vitrified-thawed oocytes treated with $1.0{\sim}5.0\;ug$ CB + NCSU- 23 medium were $22.2{\pm}3.0%$, $30.7{\pm}3.2$, $46.3{\pm}3.1%$, $38.5{\pm}3.2%$, respectively. The in vitro maturation rate ($46.3{\pm}3.4%$) of the vitrified-thawed oocytes treated with $3.0\;{\mu}g$ CB for 30 min was the highest of all vitrification groups. When the in vitro developmental rates of the vitrified-thawed (with EDS and EDT) oocytes following ICSI were $18.5{\pm}2.5%$, $16.4{\pm}2.1%$, respectively. This results were lower than the control group ($24.0{\pm}2.5%$).

  • In this study, experiments were carried out to investigate the characteristics of grinding and wear process of diamond wheel during grinding ceramic materials. Normal component of grinding resistance was decreasing while increase of spindle speed. The resistance of vitrified bond wheel was less then that of resinoid bond wheel because of imbedded large holes on the surface of cutting edge. Surface roughness was decreasing while increase of spindle speed. The surface roughness using vitrified bond wheel was less than that of resinoid bond wheel because of small elastic deformation. After continuous finding of ceramics, cutting edge ratio of resinoid bond wheel decreased. For the case of vitrified bond wheel, cutting edge ratio did not change.

  • In order to examine and compare the characteristics of two vitrified forms (AG8W1 and DG2) simulated for the operation of a commercial vitrification facility being constructed in Ulchin nuclear power plant, the vitrified forms were cooled by the natural cooling and annealing methods respectively. And the Product Consistency Test (PCT), compressive strength, thermal conductivity, specific heat, phase stability, softening point and Coefficient of Thermal Expansion (CTE) of the vitrified farms were experimented. Consequently, it was shown that there were no significant differences on the physiochemical properties of the vitrified forms performed the natural cooling and annealing.

The Cytoskeletal and Chromosomal Constitution of Vitrified Immature Mouse Oocytes (초자화동결된 생쥐 미성숙란의 세포골격과 염색체성상)

  • This study was to confirm whether the vitrification method using EFS40 freezing solution has detrimental effect on the cytoskeleton and chromosome constitution of the immature mouse oocytes by indirect immunocytochemistry and chromosome analysis. Immature mouse oocytes were vitrified using EFS40 (40% EG, 18% ficoll, 0.5 M sucrose diluted in M2 medium), thawed and then survived oocytes were in vitro matured for 16 hr. When the microtubule morphology and micro filament distribution in vitrified-thawed immature mouse oocytes were examined, normal percentage of two cytoskeleton in vitrified group (93.9 and 100.0%) was not significantly different from that in control (100.0 and 100.0%) and exposed group (94.4 and 100.0%). The rate of oocytes containing a normal chromosome number in vitrified group was 65.8%, this result was not significantly different from that in control (79.6%) and exposed group (69.0%). These results indicated that exposure to cryoprotectant or freezing has not effect on the alteration of cytoskeleton morphology and the chromosome constitution of mouse oocytes and that our vitrification methods using EFS40 freezing solution was suitable for the cryopreservation of immature mouse oocytes.

A Study for Recoverability of Iron Resource in Red Mud (레드머드 내 철 자원 회수 가능성 고찰)

  • The red mud generated from bauxite during the Bayer alumina production process has been regarded as an industrial waste due to the high alkaline property and high content of Na. Despite of its environmental problem, various studies for recovery of the valuable resources from red mud has been also carried out because of high content (25.7 wt.% as Fe2O3 in this study) of iron in red mud. In order to recover the iron resource in the red mud, microwave heating experiments were performed with adding of activated carbon and elemental sulfur to the red mud. Through the microwave heating the powdered red mud mixtures converted to porous and vitrified solid aggregates. The vitrified aggregates produced by microwave heating are composed of goethite, zero valent iron (Fe0), pyrrhotite and pyrite. And then, the microwave heating samples were dissolved in the aqua regia solution, and Fe precipitates were obtained as a Fe-chlorides by adding of NaCl salt in the aqua regia solution. The Fe recovery rates in the Fe-chloride precipitates showed differences depending on the experimental mixture conditions, and Fe grades of the end products are 49.0 wt.%, 58.0 wt.% and 59.5 wt.% under mixture conditions of red mud, red mud + activated carbon, and red mud + activated carbon + elemental S, respectively. The Fe content of 56.0 wt.% is generally known as the grade value of Fe in a iron ore for iron production, and the Fe grades of microwave heating samples with adding activated carbon and elemental S in this study are higher than the grade value of 56.0 wt.%.

Sow Transfer of Cultured Embryos : Embryo Recovery, Oocyte Collection, IVM-IVF-IVC Treatment, Vitrification/Thaw, and Surgical and Nonsurgical Transfer

  • The aims of this study are 1) to test oocytes and embryos collected from in-vivo and in-vitro to achieving the valuable protocol by culturing, vitrifying and thawing of oocytes/embryos, and 2) to transfer them to recipient, and finally have resulted in pregnancies from recipient females after surgical or nonsurgical transfer. In vitro maturation and fertilization were performed according to the procedures of Funahashi et al. Fertilized oocytes were cultured in glucose-free NCSU 23 supplemented with 5 mM sodium pyruvate, 0.5 mM sodium lactate and 4 mg/ml bovine serum albumin for 2 days at 39$^{\circ}C$, and 10% fetal bovine serum was added to the culture medium thereafter. Embryos were treated with 7.5$\square$g/ml cytochalasin-B for 30 min, centrifuged at 13,000 ${\times}$ g for 13 min and then exposed sequentially to an ethylene glycol (EG) vitrification solution, aspirated into OPSs, and plunged/thawed into/from liquid nitrogen. In vivo embryos were surgically collected from three donors after Al. Forty-six embryos (18, 9 and 19 embryos, respectively) were washed 3 times in mPBS+10%FBS, followed treatments : cultured, centrifuged, vitrified, recovered and transferred to recipients as in vitro prepared embryos. Three recipients received surgically 34(control), 188 and 184 embryos (derived from abattoir), respectively. Another three recipients were received nonsurgically 150, 100 and 150 embryos, respectively. All recipient sows exhibited delayed returns to estrus. To our knowledge, these results suggest that required an improved techniques, more vigorous embryos preparation and cleaner uterous condition(use gilt).
