Effect of DIF and Cytokinins on In Vitro Growth of 'Campbell Early' Crapes (Vitis spp.) (주야 온도차[DIF]와 시토키닌이 '캠벨얼리' 포도의 기내생장에 미치는 영향)

  • Effects of difference between day and night temperatures (DIF) and cytokinins on in vitro growth of 'Campbell Early' grape plantlets were investigated, Shoot growth was suppressed by -7DIF (18/$25^{\circ}C$). Effects of BA and TDZ were not affected by DIF. 0DIF (25/$25^{\circ}C$) enhanced the growth with increasing cytokinin level. Kinetin at 10 $\mu$M was the most effective for shoot growth with 87 mm among cytokinins used except for control. Zeatin and kinetin each at 5 $\mu$M significantly increased shoot growth to respective 162 and 110 mm. In 0DIF, two-fold increase of 162 mm was obtained by 5 $\mu$M zeatin. In control, no further shoot proliferation was observed regardless of DIF. In 0 and +7DIF (25/18$^{\circ}C$), 4 shoots were observed with 10 $\mu$M BA. Zeatin, 2iP and kinetin resulted in poor shoot proliferation while BA at 10 $\mu$M resulted in profuse branching in all DIFs.

  • Effect of cytokinins (BA, TDZ, zeatin, 2iP, and kinetin) applied either singly or in combination on in vitro growth of two grape cultivars ('Cabernet Sauvignon' and 'Campbell Early') was investigated as a serial work for mass production of grapevine nursery stocks. In single treatment, shoot growth of two cultivars was most favorable in control. Shoot proliferation was satisfactory with 10 $\mu$M BA regardless of cultivars and cytokinin combinations, followed by TDZ. Other treatments resulted in very poor or no branching. Total explants ready for subculture produced by 10 $\mu$M BA outnumbered those by other treatments. TDZ was also effective. TDZ significantly increased the fresh weight and callus formation while shoot growth was unsatisfactory. Shoot growth response of two cultivars in combined treatments was also most favorable in control as was in single treatments. When TDZ was combined with zeatin, 2iP, and kinetin which failed to induce branching, proliferous branching was induced though the shoot number was behind that of single treatments of BA and TDZ. TDZ was very effective for total number of explants and fresh weight, showing 10-fold increase.

Effects on Quality Characteristics of Pork Loin Fed with Wild Grape(Vitis amurensis Ruprecht) Wine By-product (머루주 박의 급여가 돈육 등심의 품질에 미치는 영향)

  • This study was carried out to investigate the feeding effect of wild grape wine by-products on pork qualities. The chemical composition, volatile basic nitrogen, thiobarbituric acid reactive substances and pH were not significantly different between control and wild grape pork, but the cholesterol and roast loss of wild grape polk. were lower than those of the control, and 1l1e salt soluble protein extractability of the control was lower than that of the wild grape pork. The Hunter's $a^{\ast}$ and $b^{\ast}$ value of wild grape polk. meat were higher than the control, the $a^{\ast}$ value of wild grape polk. meat was higher than the control, but the $L^{\ast}$ value of meat and fat were not significantly different between control and wild grape pork. The cohesiveness, gumminess and chewiness of control(respectively $66.2\%$ 428kg and 189g) were higher than wild grape polk. (respectively $61.4\%$ 357 kg and 154 g). The total amino acid composition of polk. were not significantly different between control and wild grape wine, The glutamic acid, leucine, arginine and aspartic acid were major amino acids in control and wild grape pork. The arachidonic acid $(C_{20:4})$, EPA $(C_{20:5})$ and DHA $(C_{22:6})$ of wild grape polk. were higher than those of the control. The taste (p<0.001), aroma (p<0.05), flavor (p<0.001), juiciness (p<0.01) and palatability (p<0.01) of wild grape polk. were higher than those of the control, but the texture between control and wild grape polk. were not significantly different.

Forest Vegetation Classification and Species Composition of Mt. Ilwol, Yeongyang-Gun, Korea (일월산 산림식생의 종구성적 특성)

  • Forest vegetation classification and species composition of Mt. Ilwol, Yeongyang-Gun, Korea, were studied combining the Braun-Blanquet approach with numerical syntaxonomical analyses (TWINSPAN). Vegetation types and various ecological characteristics such as flora, constancy classes, species ratio of life-form, species diversity and importance value were analyzed. Sixty-eight samples were taken from a $100m^2$ square plot each. Forest communities were identified as two great types: arid landform of mountainside (AM) and humid fertility of piedmont and valley (HP). The former was divided into 3 communities (Rhododendron mucronulatum, Quercus variabilis, Hosta capitat community) and 2groups, and the latter into 3 communities (Tilia amurensis, Vitis coignetiae, Philadelphus schrenckii community) and 2 groups. Vegetation was classified into 8 units. Floristically, the most represented family was Compositae with 26 species. Species with percentage constance degree of more than 61% was Quercus mongolica (72.1%, IV); Carex siderosticat (III) and Fraxinus rhynchophylla (III) were 50.0 and 41.1%, respectively. Life-forms species ratios for trees, subtrees, shrub, vines, grominoids, forbs and ferns were 18.5, 5.7, 14.9, 6.6, 8.8, 42.4 and 3.1%, respectively, PH type showed from $1.70{\pm}0.50\;to\;1.97{\pm}0.57$ and AM type was from $1.40{\pm}0.18\;to\;1.62{\pm}0.20$ in species diversity; therefore, the former type showed higher species diversity than the latter, According to importance value analysis, Pinus densiflora, Quercus mongolica and Q. variabilis were higher in the tree layer, Q. mongolica in the subtree layer, Fraxinus sieboldiana, R. schlippenbachii, etc. in the shrub layer and Carex siderosticta, Carex humilis, etc. in the herb layer.

Physicochemical Analysis and Antioxidative Effects of Wild Grape (Vitis coignetiea) Juice and Its Wine (머루즙과 머루주의 이화학적 분석 및 항산화 효과)

  • Physicochemical analysis of the wild grape juice and its wine extracted with ethanol were assessed. The contents of moisture in wild grape juice and its wine were 78.7${\pm}$0.2% and 74.4${\pm}$O.4%, respectively. Total anthocyanin and flavonoid contents of wild grape juice were higher 4 times and 10 times than those of wild grape wine, respectively. Of the 17 amino acids, the glutamic acid in wild gape juice and threonine in wild grape wine were the highest, which were 43.7${\pm}$1.4mg/100 g and 14.1${\pm}$0.7 mg/100 g, respectively. Electron donating abilities of wild grape juice and its wine at concentration of 1,000 ${\mu}$g/ml were 93.1${\pm}$1.2% and 88.9${\pm}$O.8%, respectively. The nitrite scavenging abilities of wild grape juice and its wine were gradually increased by increasing levels of the extracts to 79.6${\pm}$1.27% and 72.8${\pm}$1.01% at concentration of 1,000 ${\mu}$g/ml under pH 2.5.

Fermentation Characteristics of Moru Wine Fermented with Rose rugoga Thun (해당화와 머루를 함께 발효한 해당화 머루주의 발효특성)

  • Four different mixing ratios of Rose rugosa Thun and Vitis amurensis (Moru) were prepared. These included Rose rugosa Thun two vs. Moru one (Moru 33), Rose rugosa Thun one vs. Moru two (Moru 67), Rose rugosa Thun one vs. Moru five (Moru 83), and Moru (Moru 100). Their physiochemical changes were investigated during 28 days of fermentation followed by aging. The final brix, pH, and total titratable acidity values of the four experiments were in the following ranges: 6.0-8.2$^{\circ}$Bx, 3.87-4.03, and 0.94-1.18%, respectively. Final ethanol contents were 6.5% in Moru 33, 11.8% in Moru 67, 11.1% in Moru 83, and 11.4% in Moru 100. As the amount of Rose rugosa Thun increased, anthocyanin content, color intensity, organic acid, and free amino acid concentrations were reduced. These findings demonstrate that the supplementation of Rose rugosa Thun to Moru prior to alcohol fermentation may help change the acidity, colour, and taste in the final product.

Developmental Rate Equations for Predicting Bud Bursting Date of 'Campbell Early' (Vitis labrusca) Grapevines (발육 속도 모델을 이용한 포도 '캠벨얼리'의 발아기 예측)

  • To predict the bud bursting date of 'Campbell Early' grapevines, the bud developmental rate (DVR) models were constructed. The DVRs for bud bursting were calculated from the demanded times at controlled air temperatures. The DVRs were examined on the 'Campbell Early' grapevines incubated in three different temperatures at 4.6, 11.8, and $16.6^{\circ}C$. The DVR increased exponentially or linearly on the air temperature with a slope of about 0.0019. The DVR equations were computed as $DVR=0.0249+0.0020e^{0.1654x}$ or DVR = 0.0019x + 0.0187. These DVR equations offered developmental indices and predicted dates for bud bursting with air temperature data. The DVR equations were validated to the bud bursting data observed in the field. When bud bursting dates were calculated with daily temperature data, the root mean squared error (RMSE) between the observed and the predicted dates was less than 4 days. When those were calculated with hourly temperature data, on the other hand, the RMSE was less than 3 days. These results suggest that the DVR models are useful to predict bud bursting date of 'Campbell Early' grapevines.

Selection of Hypo- and Hyper-tetraploid Seedlings from Abnormal Cotyledons Seedlings Obtained during Crossing of Tetraploid Grapes (Vitis Complexes) (4배체 포도간 교배된 이상자엽실생으로부터 저·고4배체 식물의 선발)

  • We observed abnormal morphology of cotyledons occurring in seedlings derived from open-pollinated and cross-pollinated tetraploid grapes and selected aneuploids, especially hypo- and hyper-tetraploid in seedlings with abnormal morphology of cotyledons. Five types of morphologically abnormal cotyledons were observed. In open-pollination of four tetraploid grapes, the frequency of abnormal cotyledons was 1.6% (49 of 3029 seeds). Percentage of aneuploids in the seedlings of abnormal cotyledons was 20.4% (10 of 49 seedlings). Aneuploids in open-pollination consisted of three (4n = 4x-2), four (4n = 4x-1), and three (4n = 4x+1) seedlings. In cross-pollination of tetraploids, the frequency of abnormal cotyledons was 3.4% (59 of 1729 seeds). Percentage of aneuploids in the seedlings with abnormal cotyledons was 22.0% (13 of 59 seedlings). Aneuploids from cross-pollination of tetraploids consisted of two (4n = 4x-2), nine (4n = 4x-1), one (4n = 4x+1), and one (4n = 4x+3) seedlings. According to the results, although the abnormal cotyledon morphology of seedlings obtained from crossing between tetraploid grapes appeared at low rate (2.3%), aneuploid seedlings occurred at high rate (22.0%); therefore, it indicated that this selection strategy might be very efficient in the initial seedling stage.

Characteristics of Ice Wine Fermentation of Freeze-Concentrated Campbell Early Grape Juice by S. cerevisiae S13 and D8 Isolated from Korean Grapes (포도로부터 분리한 S. cerevisiae S13 및 D8에 의한 캠벨 얼리 동결농축 과즙의 아이스와인 발효 특성)

  • Cryoextraction (a freeze concentration using an instrument) can increase the sugar concentration in grape juice by reducing its water content, similar to the natural freezing of grapes for natural ice wine. In this study, fermentation of freeze-concentrated Campbell Early grape (Vitis labruscana) juice to 36 $^{\circ}Bx$ was carried out using Saccharomyces cerevisiae strains D8 and S13 isolated from Korean grapes. During the fermentation, strains S13 and D8 showed rapid sugar reduction and alcohol production compared with S. cerevisiae Fermivin$^{(R)}$ used as a control. After nine-day fermentation, the residual sugar contents were lower in W13(9.77%) and D8 wine(9.07) than that in Fermivin$^{(R)}$ wine(14.0%). Total acid content was high in the D8>S13>Fermivin$^{(R)}$ wine, in that order. The acetaldehyde content was highest in the D8 wine and lowest in the Fermivin$^{(R)}$ wine, among the three. The methanol content was slightly higher in the S13 and D8 wines than in the Fermivin$^{(R)}$ wine. In the sensory evaluation, the S13 wine exhibited the highest score in flavor and taste among the three wines. Both the two S13 and D8 wines exhibited higher scores than Fermivin$^{(R)}$ wine in overall preference.
