Resveratrol과 Methoxylated Resveratrol이 닭 다리육의 저장 품질에 미치는 영향 (Dietary Supplementation of Resveratrol and Methoxylated Resveratrol Affects on Chicken Thigh Meat Quality)

  • 본 연구는 뽕나무 가지에서 추출한 resveratrol과 methoxylated resveratrol의 급여가 닭고기 다리육의 품질에 미치는 영향을 알아보기 위해 실시하였다. 1일령 Ross 320마리 육계 병아리를 8처리구로 나누어 대조구(BD), 항생제 처리구(AB), 비타민 E 20 ppm 처리구(VE 20), 비타민 E 200 ppm 처리구(VE 200), resveratrol 20 ppm(RV 20), resveratrol 200 ppm (RV 200), methoxylated resveratrol 20ppm(MR 20), methoxylated resveratrol 200 ppm(MR 200) 처리구로 나누어 35일간 급여하였다. 급여 후 36일째 도계하여 다리육만 분리하였고, 1, 3 및 5일 냉장 저장 기간 동안의 pH, 보수력, 육색 및 관능적 특성을 조사하였다. pH는 저장 1일에 AB, VE 및 MR 처리구에서 유의적으로 높은 수준을 나타내었다(p<0.05). 저장3일과 5일에 각 RV와 MR의 20과 200 처리는 닭다리 육의 pH 수준을 대조군보다 높게 유지하는 것으로 판단된다(p<0.05). 명도($L^*$)는 저장 1일과 5일에 모든 처리구에서 BD 처리구와의 유의적인 차이를 보이지 않았다. VE 200 처리구와 MR 20 처리구는 저장 기간 동안 적색도의 수준의 유의적인 차이를 보이지 않았으며(p<0.05), 육색의 안정화에 영향을 미치는 것으로 나타내었다. 보수력은 저장 3일의 경우는 VE 20, RV 200, MR 20, MR 200 처리구에서 BD 처리구에 비해 유의적으로 높은 값을 나타내었다(p<0.05). 관능적 특성 중 연도는 저장 1일과 5일에는 VE 20을 제외하고는 유의적인 차이를 보이지 않았으나 저장 3일에 항생제 처리구, VE 20, RV 20과 MR 200 처리구에서 유의적으로 높은 점수를 보여 관능요원들의 선호도 증진에 영향함을 나타내었다. 본 연구 결과 resveratrol과 methoxylated resveratrol은 육계 다리육의 품질을 증진시키기 위한 육계 사료 첨가제로서 이용 가능성이 있을 것으로 기대된다.

Effects of functional nutrients on chicken intestinal epithelial cells induced with oxidative stress

  • The objective of this study was to investigate the protective effects of functional nutrients including various functional amino acids, vitamins, and minerals on chicken intestinal epithelial cells (cIECs) treated with oxidative stress. The cIECs were isolated from specific pathogen free eggs. Cells were exposed to 0 mM supplement (control), 20 mM threonine (Thr), 0.4 mM tryptophan (Trp), 1 mM glycine (Gly), 10 μM vitamin C (VC), 40 μM vitamin E (VE), 5 μM vitamin A (VA), 34 μM chromium (Cr), 0.42 μM selenium (Se), and 50 μM zinc (Zn) for 24 h with 6 replicates for each treatment. After 24 h, cells were further incubated with fresh culture medium (positive control, PC) or 1 mM H2O2 with different supplements (negative control, NC and each treatment). Oxidative stress was measured by cell proliferation, whereas tight junction barrier function was analyzed by fluorescein isothiocyanate (FITC)-dextran permeability and transepithelial electrical resistance (TEER). Results indicated that cell viability and TEER values were less (p < 0.05) in NC treatments with oxidative stress than in PC treatments. In addition, FITC-dextran values were greater (p < 0.05) in NC treatments with oxidative stress than in PC treatments. The supplementations of Thr, Trp, Gly, VC, and VE in cells treated with H2O2 showed greater (p < 0.05) cell viability than the supplementation of VA, Cr, Se, and Zn. The supplementations of Trp, Gly, VC, and Se in cells treated with H2O2 showed the least (p < 0.05) cellular permeability. In addition, the supplementation of Thr, VE, VA, Cr, and Zn in cells treated with H2O2 decreased (p < 0.05) cellular permeability. At 48 h, the supplementations of Thr, Trp, and Gly in cells treated with H2O2 showed the greatest (p < 0.05) TEER values among all treatments, and the supplementations of VC and VE in cells treated with H2O2 showed greater (p < 0.05) TEER values than the supplementations of VA, Cr, Se, and Zn in cells treated with H2O2. In conclusion, Thr, Trp, Gly, and VC supplements were effective in improving cell viability and intestinal barrier function of cIECs exposed to oxidative stress.

항산화비타민이 Aflatoxin $B_1$ 투여 마우스의 간세포에서 Aflatoxin $B_{1}-DNA$ Adduct 형성에 미치는 영향 (The Effect of Antioxidant Vitamins on Aflatoxin $B_{1}-DNA$ Adduct the Formation in Aflatoxin $B_1$ Administered Mice Liver)

  • 본 연구는 aflatoxin $B_1(AFB_1)$에 의하여 유발된 마우스 간세포에서의 $AFB_1-DNA$ 부가체 형성과 세포의 산화적 손상에 대한 항산화 비타민의 효과를 조사하기 위하여 수행되었다. 먼저, 비타민 C와 비타민 E를 6 주령 수컷 ICR 마우스에 10mg/kg, 63.8mg/kg의 농도로 복강내에 각각 주사(i.p.; intraperitoneal injection) 하였고, $AFB_1$과 비타민 혼합 투여군은 비타민을 주사한 후 1시간이 경과한 다음 0.4mg/kg의 $AFB_1$을 투여하였다. 투여 횟수는 2일 간격으로 동일한 방법에 의해 4회 반복 투여하였다. 반면, $AFB_1$ 단독 투여군은 위와 같은 방법으로 비타민 없이 $AFB_1$만 투여하였다. ELISA에 의한 마우스 혈청내 $AFB_1$의 잔여 농도는 $AFB_1$ 단독 투여군에서 12.28, 18.78 ng/mL이 검출되었다. 그러나 항산화비타민 C 혼합 투여군에서는 7.60 ng/mL, 항산화 비타민 E 혼합 투여군에서는 4.85 ng/mL이 각각 검출되었다. 마우스의간에서 분석된 $AFB_1-DNA$ 부가체 농도에 의하면, $AFB_1$ 단독 투여군에서는 23.78, 25.48 ng/mL이었으며, 항산화 비타민 C 혼합 투여된 군에서는 5.26 ng/mL, 항산화 비타민 E 혼합투여군에서 7.81 ng/mL로 각각 조사되었다. 이들 결과에 대한 통계적 유의성은 p<0.01이었다. 또한 면역조직화학적 관찰에서 $AFB_1$ 단독 투여군에서는 중심정맥과 동양혈관 주변에 갈색의 침전이 두드러지게 나타났으며 이러한 현상은 항산화비타민 혼합 투여군에서 유의하게 감소한 것으로 나타났다.

Degradation kinetics of vitamins in premixes for pig: effects of choline, high concentrations of copper and zinc, and storage time

  • Objective: The present work was undertaken to evaluate the effects of storage time, choline chloride, and high concentrations of Cu and Zn on the kinetic behavior of vitamin degradation during storage in two vitamin premixes and four vitamin-trace mineral (VTM) premixes. Methods: Two vitamin premixes (with or without 160,000 mg/kg of choline) were stored at 25℃ and 60% humidity. Besides, four VTM premixes were used to evaluate the effects of choline (0 vs 40,000 mg/kg) and trace minerals (low CuSO4+ZnO vs high CuSO4+ZnO) on vitamin stability in VTM premixes stored in room, and the VTM premixes were stored in room temperature at 22℃. Subsamples from each vitamin and VTM premix were collected at 0, 1, 2, 3, 6, and 12 months. The retention of vitamin A (VA), vitamin D3 (VD3), vitamin E (VE), vitamin K3 (VK3), vitamin B1 (VB1), vitamin B2 (VB2), vitamin B3 (VB3), vitamin B5 (VB5), and vitamin B6 (VB6) in vitamin premixes and VTM premixes during storage was determined. The stability of vitamins in vitamin premixes and VTM premixes was determined and reported as the residual vitamin activity (% of initial) at each sampling point. Results: The effect of choline on VK3 retention was significant in vitamin premixes (p<0.05). The negative effect of storage time was significant for the retentions of VD3, VK3, VB1, VB2, VB5, and VB6 in vitamin premix (p<0.05). For VTM premixes, negative effect of storage time was significant (p<0.05) for the losses of vitamin in VTM premixes. Choline and high concentrations of Cu and Zn significantly increased VA, VK3, VB1, and VB2 loss during storage (p<0.05). The supplementation of high concentrations of Cu and Zn significantly decreased the concentrations of VD3 and VB6 (p<0.05) in VTM premixes at extended storage time. Conclusion: The maximum vitamin stability was detected in vitamin and VTM premixes containing no choline or excess Cu and Zn. The results indicated that extended storage time increased degradation of vitamin in vitamin or VTM premixes. These results may provide useful information for vitamin and VTM premixes to improve the knowledge of vitamin in terms of its stability.

당뇨 쥐의 신장 염증 단계에서 단기간의 혼합 항산화 영양소 보충 식이가 산화적 스트레스와 염증반응의 조절에 미치는 영향 (Effects of Short Term Antioxidant Cocktail Supplementation on the Oxidative Stress and Inflammatory Response of Renal Inflammation in Diabetic Mice)

  • Diabetes mellitus is a multifactorial disease. Particularly, diabetic nephropathy is a serious complication for diabetic patients, yet the precise mechanisms that underline the initial stage of diabetic renal inflammation remain unknown. However, oxidative stress induced by hyperglycemia in diabetes is implicated in diabetic renal disease. We hypothesized that dietary supplementation of antioxidants either VCE (0.5% VC + 0.5% VE) or Comb (0.5% VC + 0.5% VE + 2.5% N-acetylcysteine) improves acute diabetic renal inflammation through modulation of blood glucose levels and antioxidant and anti-inflammatory responses. Experimental animals (5.5 weeks old female ICR) used were treated with alloxan (180 mg/kg) once. When fasting blood glucose levels were higher than 250 mg/dL, mice were divided into 3 groups fed different levels of antioxidant supplementation, DM (diabetic mice fed AIN 93G purified rodent diet); VCE (diabetic mice fed 0.5% vitamin C and 0.5% vitamin E supplemented diet); Comb (diabetic mice fed 0.5% vitamin C, 0.5% vitamin E and 2.5% N-acetylcysteine supplemented diet), for 10 days and then sacrificed. Body weights were measured once a week and blood glucose levels were monitored twice a week. Lipid peroxidation products, thiobarbituric acid reacting substances were measured in kidney. NF-${\kappa}B$ activation was indirectly demonstrated by pI${\kappa}B$-${\alpna}$ and expressions of selective inflammatory and oxidative stress markers including antioxidant enzymes were also determined. Dietary antioxidant supplementation improved levels of blood glucose as well as kidney lipid peroxi-dation. Dietary antioxidant supplementation improved NF-${\kappa}B$ activation and protein expression of HO-1, but not mRNA expression levels in diabetic mice fed Comb diet. In contrast, the mRNA and protein expression of CuZnSOD was decreased in diabetic mice fed Comb diet. However, antioxidant supplementation did not improve mRNA and protein expressions of IL-$1{\beta}$ and MnSOD in diabetic mice. These findings demonstrate that acute diabetic renal inflammation was associated with altered inflammatory and antioxidant responses and suggest that antioxidant cocktail supplementation may have beneficial effects on early stage of diabetic nephropathy through modulation of blood glucose levels and antioxidant enzyme expressions.
