The Effects of Vitamin C on Lipid Contents and Fatty Acid Compositions of Serum and Liver in Rats Treated with Radiation or Aflatoxin B1 (Vitamin C가 방사선과 Aflatoxin B1을 투여한 흰쥐의 혈청과 간장의 지질성분 및 지방산 조성에 미치는 영향)

  • Lipid peroxidation is one of the main manifestations of oxidative damage and has been found to play an important role in the toxicity and carcinogenesis of many carcinogens. This study was carried out to determine the effects of vitamin C on lipid contents and fatty acid compositions of serum and liver in male rats treated with radiation or aflatoxin $B_1\;(AFB_1)$. Six week-old male Sprague-Dawley rats were randomly divided into 7 groups; control group, radiation exposed group, $AFB_1$ treated group, X-ray and $AFB_1$ co-treated group. Three groups, except control group, were each further divided into vitamin C administered group and not administered groups. For this study, vitamin C was injected with 10 mg/kg of body weight by intraperitoneal injection and 1 hr later, 0.4 mg/kg of $AFB_1$ was injected by the same method. These administrations were repeated every 3 days over a period of 15 days. Only one time, X-ray was irradiated on whole liver with 1,500 cGy. Then vitamin C and AFB1 were administered by the same level and same method described above. On the 16th day of treatments, the animals were sacrificed. From the analysis of the serum lipid patterns, significant decrease (p<0.01) in triglyceride (TG) and total cholesterol levels were observed in X-ray and $AFB_1$ co treated group administered with vitamin C (group 7). In liver lipids, the levels of free cholesterol and total cholesterol were also decreased in X-ray and $AFB_1$ co treated group administered with vitamin C (group 7). The levels of serum free cholesterol and hepatic TG were not significantly different among all groups according to vitamin C administrations. The high density lipoprotein (HDL)-cholesterol level of serum was significantly (p<0.01) increased while the low density lipoprotein (LDL)-cholesterol level was decreased in X-ray and $AFB_1$ co treated group administered with vitamin C (group 7). In the phospholipid fatty-acid compositions of serum and liver tissue, group 3, 5 and 7 showed an increase in polyunsaturated fatty-acid (PUFA) but a decrease in saturated fatty acid (SFA) when compared to the control group. The composition ratio of fatty acid varied according to vitamin C administration. These results suggested that vitamin C has partly suppressive effects on lipid contents and fatty acid composition of serum and liver in rats treated by radiation and $AFB_1$.

Biochemical Study on the Environmental Preservation of Lake -Distribution of Chemical Compounds and $Vitamin\;B_{12}$ of Okjeong Lake- (호수의 환경보전에 있어서의 생물화학적 연구 -옥정호의 영양염과 $Vitamin\;B_{12}$의 함량-)

  • Concentration of dissolved $vitamin\;B_{12}$ in the water of Okjeong lake have been determined by microbiological assay methods on the 20th of August, 1983. Also the relations between the $vitamin\;B_{12}$ and other physical, chemical and biological environmental factors were investigated. $Vitamin\;B_{12}$ was assayed with Euglena gracilis Z strain. The results obtained in this investigation were as follows : 1. Generally, the values of COD and $Cl^-$ were higher than values reported in 1981. 2. The concentration of nutrient salts in the lake water was found to be at a higher level: above the general criterion in a eutrophic lake viz., phosphorous 0.175-0.225 mg/l. 3. Chlorophyll a contents in this lake were $1.1-8.7{\mu}g/l$, 4. The concentration of $vitamin\;B_{12}$ in the water fairly high: 1.25-6.0 ng. It is apparent that concentrations of the $vitamin\;B_{12}$ as well as nutrient salts were high in the water of Okjeong lake. It was found that Okjeong lake was in eutrophic.

  • Yeolgujatang is traditional casserole with meat, fish and vegetables in special pot. Name of Yeolgujatang was in 1800s, as yeolgujatang, yeolgujatangbang, yeolguja, in 1900s, Yeolgujatang, royal feast food in Yi Dynasty was yeolgujatang, Meon-sinseolro, tangsinseolro. Yeolgujatang frame has legs and a lid, and a cylinder that can contain charcoal at the center of its rounded pot. with this charcoal, food can be cooked. Its material has changed from brazier to brassware, stainless steel, and silver. Nowdays electric sinseolro was also launched, which uses electric power instead of charcoal. Materials in yeolgujatang are beef, intestines, pork, chicken, pheasant, fish, sea bream, abalone, shrimps, vegetables, mushroom, ddock, guksu, cooked rice, seasening and garnish. Nutrition of Yeolgujatang per capita contains 221.5kal of calory, 17.3g of protein, 16.5g of fat, 6.1g of carbobydrates, 2g of fiber, 57.6mg of calcium, 208mg of phosphorus, 4.3mg of ferrum, $2177{\mu}gRE$ of vitaminA, 1.58mg of vitamin $B_1$, 0.3mg vitamin $B_2$, 6.6mg of vitaminC and 5.26mgNE of niacin. Yeolgujatang is excellent in nutrition, except for calcium and vitaminC.

Reanalysis of 2007 Korean National Health and Nutrition Examination Survey (2007 KNHANES) Results by CAN-Pro 3.0 Nutrient Database (2007년도 국민건강영양조사 결과 재분석 : CAN-Pro 3.0 식품영양가표의 활용)

  • This study aimed to reanalyze energy and nutrient intakes of 2007 Korean Nutrition and Health Examination Survey (KNHANES) using CAN-Pro 3.0, a commonly used nutrient analysis software in Korea. Food items and their codes were selected from 2007 KNHANES dietary intake file and converted to food codes of CAN-Pro 3.0 nutrient database (NDB). Of the 1,324 total food items, 1,155 items were converted by direct matching, 123 items were matched using other items in CAN-Pro 3.0 NDB and 42 items were matched using external sources. Consumption frequencies of items converted by direct matching contributed 94.5% of total consumption. Nutrient intakes of 4,091 participants of 2007 KNHANES, over 1 year old, were recalculated using CAN-Pro 3.0 NDB and compared with intakes in 2007 KNHANES dietary intake file. Intakes for energy and all nutrients except protein and Vitamin C calculated by two NDBs were significantly different by paired t-test (p < 0.001), but significantly correlated by Pearson' correlation coefficients (p < 0.001). Percent differences between the NDBs ranged from 0.3% to 15.1%, low for protein, energy, vitamin C, iron, vitamin B$_2$ (below 5%) but high for phosphorus, retinol, vitamin A, and $\beta$-carotene (over 10%). Age group, sex, and their interactions significantly influenced six nutrients (p < 0.05). Intake levels of zinc, vitamin B6, vitamin E, folate and cholesterol were not available in 2007 KNHANES but were calculated by CAN-Pro 3.0. Mean intake levels of zinc, vitamin B$_6$, vitamin E, and folate by age and sex groups revealed that some groups had mean levels below RI (Recommended Intake) or AI (Adequate Intake) levels. Intake level of cholesterol was higher than the recommended level (below 300 mg/day) in some groups, especially males. Results of the present study indicate the need for comparable and more comprehensive NDB to be used for dietary assessment of KNHANES and other researches. More rigorous evaluation of nutrients which have not been reported in KNHANES is needed.

Recipe Standardization and Nutrient Analysis of Local Foods of Cheollabuk-do Province(The Second Report) (전라북도 향토음식의 조리법 표준화 및 영양성분 분석(제 2 보) - 추어탕, 다슬기탕, 민물고기매운탕, 어죽, 백합죽-)

  • Five representative local foods of Cheollabuk-do Province such as chuotang, ochuk, baekhapchuk, dasulgitang and minmulgokimaeuntang were subjected to recipe standardization and nutrient analysis. To derive a standard recipe, first a test recipe was prepared with the consideration of information obtained from literature survey, personal interview and survey of restaurant recipe. This test recipe was modified three times after sensory evaluation. Then the modified test recipe was accepted as the standard recipe when all characteristics of food was assessed over 5 points and the percentage of judges who gave a score over five exceeded 70% in seven-point hedonic scale. Actually, all characteristics of each food were judged as "satisfactory". Nutrient analysis was performed in food cooked according to the decided standard recipe. In general, it appeared that energy content was rather low. However protein contents in chuotang, ochuk and minmulgokimaeuntang were higher than the recommended value per meal. Vitamin A, vitamin C, vitamin B$_1$, vitamin B, niacin, calcium, phosphorus and iron were rich in chwotang and minmulgokimaeuntag. Onhuk contains plenty of vitamin C, vitamin B$_1$, vitamin B$_2$and the contents of vitamin A, vitamin B$_1$, and niacin in baekhapapchuk were over the recommended values per meal. The foods contained large percentage of aspartic acid and glutamic acid, and major essential amino acids appeared to be leucine and lysine. On the other hand, major fatty acids were oleic acid, linoleic acid and plamitic acid. Among them the content of oleic acid was the highest in chuotang, ochuk and baekhapchuk, whereas linoleic acid and palmitic acid were the most rich fatty acids in baekhapchuk and dasulgitang respectively.

  • When pyrazinamide is used in the treatment of tuberculosis, the measurement of pyrazinoic acid which is an intermediate metabolite of pyrazinamide in body is required in order to prevent its associated side-effects, especially that of hyperuricemia. Effects of vitamin B$_6$ on pyrazinoic acid metabolism were studied in this experiment. The activity of hepatic pyrazinoic acid oxidizing enzyme in the presence of pyridoxal was powerfully inhibited, and the pattern was competitive inhibition type. Whereas, its enzyme activity was significantly increased by the treatment of pyridoxal, and the characteristics of the increase may include induction of enzyme proteins. As mice received pyrazinoic acid(300mg/kg) after pyridoxal-pretreatment(40mg/kg) once daily for 4 days, the blood level of pyrazinoic acid and uric acid was decreased significantly.

Evaluation of Nutrient Intakes and Diet Quality of Rural Middle School Students (농촌지역 급식교 및 비급식교 중학생의 영양소 섭취 적절성에 관한 연구)

  • This study was performed to assess the nutrient intakes and diet quality of rural middle school students provided with and without the national school lunch program(NSLP). The data on dietary intakes were obtained by using food record method questionnaires which were collected from 340 rural middle school students provided with(n=177) and without (n=163) NSLP. Diet quality was accessed by NAR(nutrient adequacy ration), MAR(mean adequacy ration), and INQ( index of nutritional quality). Total energy intakes of the boys were 2123 kcal with NSLP and 1857 kcal without NSLP. Total energy intakes of the girls were 1913 kcal with NSLP and 1814 kcal without NSLP. Phosphorus, vitamin A, vitamin E, ascorbic acid, thiamin, riboflavin, and niacin intakes were above the RDA in the NSLP group. For those without NLSP, phosphorus, vitamin E, thiamin intakes were above RDA, however, calcium, iron vitamin A, vitamin $B_6$ were less than the RDA. The mean adequacy ration(MAR), an index of dietary quality was 0.69(with) 0.62(without) for males and 0.62(without) for females The indexes of nutritional quality(INQ) were over 1 for most nutrients except calcium, vitamin A, vitamin B$_2$ and zinc. The study showed that total daily energy and nutrient intakes were significantly higher in students provided with than those without school-lunches. Thus, the school-lunch program is recommenced and necessary to improve the nutritional status of middle school students.

Evaluation of nutrient intake and diet quality of gastric cancer patients in Korea

  • This study was conducted to identify dietary factors that may affect the occurrence of gastric cancer in Koreans. Preoperative daily nutrition intake and diet quality of patients diagnosed with gastric cancer were evaluated. Collected data were comparatively analyzed by gender. The results were then used to prepare basic materials to aid in the creation of a desirable postoperative nutrition management program. The subjects of this study were 812 patients (562 men and 250 women) who were diagnosed with gastric cancer and admitted for surgery at Soonchunhyang University Hospital between January 2003 and December 2010. Nutrition intake and diet quality were evaluated by the 24-hr recall method, the nutrient adequacy ratio, mean adequacy ratio (MAR), nutrient density (ND), index of nutritional quality (INQ), dietary variety score (DVS), and dietary diversity score (DDS). The rate of skipping meals and eating fast, alcohol consumption, and smoking were significantly higher in males than those in females. The levels of energy, protein, fat, carbohydrate, phosphorous, sodium, potassium, vitamin $B_1$, vitamin $B_2$, niacin, and cholesterol consumption were significantly higher in males than those in females. Intake of fiber, zinc, vitamin A, retinol, carotene, folic acid were significantly higher in females than those in males. MAR in males was significantly higher (0.83) than that in females (0.79). INQ values were higher in females for zinc, vitamin A, vitamin $B_2$, vitamin $B_6$, and folic acid than those in males. The average DVS was 17.63 for females and 13.19 for males. The average DDS was 3.68 and the male's average score was 3.44, whereas the female's average score was 3.92. In conclusion, males had more dietary habit problems and poor nutritional balance than those of females. Our findings suggest that proper nutritional management and adequate dietary education for the primary prevention of gastric cancer should be emphasized in men.

Study of the Impact of Light Through the Vitamin $B_{12}$/Folate Inspection (Vitamin $B_{12}$/Folate 검사 시 빛의 영향에 대한 고찰)

  • Purpose : Vitamin $B_{12}$ and Folate are for anemia work-up which is well known for its sensitivity of light; the screening manual also specifies to be careful with light conditions. According to this, our laboratory minimized the exposure of light when inspecting the Vitamin $B_{12}$ and Folate, but the exposure cannot be wholly blocked due to other various factors such as when conducting specimen segregation. Thus, this inspection is to identify to what extent light can influence and whether the exclusion of light is mandatory during the Vitamin $B_{12}$/Folate test. Materials and Methods : We have conducted two experiments of identifying the extent of light's influence when conducting the Vitamin $B_{12}$/Folate test and also when specimens are under preservation. These experiments were progressed with various concentrations of patients' specimens which were requested to our hospital in March 2012. The first experiment is to verify the results on Vitamin $B_{12}$/Folate dependent on light exposure during the experiment. In the process, we have compared the results of light exposure/exclusion during the incubation process after the reagent division. The second experiment is about the impact of light exposure on the results on Vitamin $B_{12}$/Folate during the preservation. For 1, 2, 7 days the light on the specimen were wholly blocked and were preserved under $-15^{\circ}C$ temperature refrigeration. Then, we compared the results of light-excluded specimen and the exposed one. Results : When conducting first experiment, there were no noticeable changes in the Standard and specimen's cpm, but for Vitamin $B_{12}$, the average result of specimen exposed to light increased 7.8% compare to that of excluded one's. Furthermore, in the significant level 0.05, the significance probability or the p-value was 0.251 which means it has no impact. For Folate, the result being exposed to light decreased 5.4%, the significance probability was 0.033 which means it has little impact. For the second preservation, the result was dependent on the light exposure. The first day of preservation of Vitamin $B_{12}$, the clinical material exposed to light was 11.6%, second day clinical material exposed to light was 10.8%, seventh day clinical material exposed to light increased 3.8%, the significance probability of the $1^{st}$, $2^{nd}$, $7^{th}$ day is 0.372, 0.033, 0.144 respectively, and which indicates that the $1^{st}$ and $7^{th}$ day seems to have no impact. For Folate's case, the clinical material exposed to light has increased 1.4% but hardly had impact, $2^{nd}$ day clinical material being exposed to light was 6.1%, $7^{Th}$ day clinical material being exposed to light decreased 5.2%. The significance probability of Folate on the $1^{st}$, $2^{nd}$, $7^{th}$ day is 0.378, 0.037, 0.217 respectively, and the $1^{st}$ day and the $7^{th}$ day seems to have no impact. Conclusion : After scrutinizing the impact of light exposure/exclusion, Vitamin $B_{12}$ has no impact, while Folate seems to have no noticeable influence but light exclusion is recommended due to its significance probability of 0.033 when conducting experiment. During the preservation, the $2^{nd}$ day result depend on the light exclusion seems to have impact or influence. However, to consider the complication of the experimental process, the experiment including technical errors is predictable. Hence, it is likely to have no impact of light. Nevertheless, it is recommendable to exclude the light during the long preservation as per the significance probability (p-value) of $1^{st}$ and $7^{th}$ day has been diminished.

Quantitative analysis of water-soluble vitamins and polyphenolic compounds in tomato varieties (Solanum lycopersicum L.) (토마토(Solanum lycopersicum L.) 품종 간 수용성 비타민과 폴리페놀계 성분 함량 변이 분석)

  • Tomato fruit quality is determined by the contents of various functional metabolites in addition to fruit appearance. To develop tomato cultivars with higher amounts of functional compounds, an efficient quantification method is required to identify the natural variations in the compounds in the tomato germplasm. In this study, we investigated tomato varieties, which included 23 inbred lines and 12 commercial F1 cultivars, for their contents of seven watersoluble vitamins (vitamin C, vitamins B1, B2, B3, B5, B6, and B9) and five polyphenolic compounds (quercetin, rutin, kaempferol, myricetin, and naringenin chalcone). The results of high performance liquid chromatography and liquid chromatography-mass spectrometry showed that vitamin C and naringenin chalcone were the major water-soluble vitamins and polyphenolic compounds, respectively, and their abundance was highly variable depending on the cultivar. By contrast, the contents of vitamin B1, quercetin, and kaempferol were lowest among the cultivars. With regard to the relationship between metabolic compounds and fruit characteristics, a significant association was found in fruit size, indicating that cherry tomato varieties contain higher amounts of the compounds compared to large fresh-type varieties. However, no direct association was detected in fruit color, except for naringenin chalcone. The results of this study provide new insights on the quantification of metabolic compounds and the selection of breeding materials, which are prerequisites for the development of functional tomato varieties.
