Mexican tetra (Astyanax fasciatus)의 녹색 시간 색소포 유전자의 염기서열 다형화 현상 (Nucleotide Polymorphism of Green-like Visual Pigment Gene from Eyed and Blind Forms of the Mexican Tetra, Astyanax fasciatus)

  • Since the end of the Pliocene, ancestral strains of Astyanax fasciatus have been accidently washed into different caves at the time of flooding and have lost their eyes and body pigments. Availability of this independently derived cave fish and their ancestral form within a single species provided a unique opportunity for studying the process of molecular evolution of the visual pigment gene. The nucleotide sequence comparisons of an ancestral river fish and two cave fish showed that nucleotide polymorphism of a green-like visual pigment gene between the eyed and blind form of A. fasciatus was much higher than that between the same blind form. Considering the number of nucleotide substitutions per nucleotide site and the direction of the nucleotide substitutions, more nucleotide substituions between the different forms of fish rater than the same one were probably due to more frequent mutations in the eyed river form. Nucleotide substitutions per site at the intron have been ocurred more than three times faster than those at the exon. This result indicates that the functional constraint has affected the green-like visual pigment gene of the blind cave fish although its eye sight is no longer required.

THE EFFECTS OF ZINC DURING VISUAL ADAPTATION OF VERTEBRATE EYE

  • Zinc plays a key role in genetic expression, cell division, and growth and is essential for the function of more than 200 enzymes; effects of zinc deficiency induce many syndromes, including abnormal visual adaptation. The pigment epithelium (EP) contains high concentrations of zinc in humans and in animals and it participates in threshold elevation, visual sensitivity increment, and acceleration of rhodopsin regeration during visual adaptation. The origin of c-wave of electroretinogram(ERG) is not only pigment epithelium as shown in present research, but also other cell layers, perhaps the photoreceptors. We propose zinc as a candidate for an internal messenger which participates in signal amplification.

An Analysis Study of Wall Painting Pigment Excavated at Iksan Jeseoksaji Dumpsite

  • The fragments of a wall painting excavated from among the historical remains of Jeseoksa Temple in Iksan. The extent of the damage to the fragments was examined and an analysis of the components of the pigment was conducted. The results of the component analysis of the pigment confirmed that the white pigment consisted of alkali feldspar and mullite. Although the results of the visual inspection revealed differences in color in the red and black pigments, the main component of the two colors was confirmed to be iron oxide. Red and black pigments are found at the same position. Although differences of color is obvious, those are identified as hematite and magnetite of oxidized steel's affiliation. It is judged that Differences of ingredients happened by external environment's factors.

진도홍주색소의 사용기준에 관한 연구 (Studies on the standard method of Jindo Hongju pigments)

  • Jindo Hongju is a traditional liquor in Jindo island of Korea. The characteristics of Hongju are its unique flavour by fermetation and red color of gromwell(Lithospermum erythrorhizon) root. However, the evaluation of red pigment is different from one manufactures to other manufactures and from place to place, also the standard method is not established. An attempt has made to compare the quality of gromwell root from different places and to standardize the extracting method. The results obtained from this study are summerized as follow, The chemical properties and composition of gromwell root from Jindo and other areas were compared. There were no difference among the samples in moisture content, content of naphtoquinone derivatives and absoption spectra. These results indicate that the pigments from Jindo and other region products seems to be the same quality. For efficient extraction of gromwell pigment, more than 40% ethanol as solvent and at least 10 hours extraction time was required. According to the visual test for Hongju pigment, the most preferable color was that it shows absorbance of 1.0 (contents of shikonin was 3.90 mg/45% EtOH 20 ml). From this visual test it can be proposed that the may be applied absorbance at 1.0 for the quality control of pigment.

Whole Structure of the Photoreceptors in the Ascidian Larva Visualized by an Antibody Against Arrestin (Ci-Arr)

  • The anterior brain vesicle of ascidian larvae contains two distinct pigment cells. Ultrastructure of these pigment cells has been shown that the anterior pigment cell is an otolith for perception of gravity and the posterior pigment cell is an ocellus for light reception. The larva has remarkably simple central nervous system (CNS) composed of about 330 cells. We focused to study neural networks of visual systems. In the present paper, we report the whole structure of the photoreceptors of the ascidian larva visualized by an antibody against arrestin. Visual arrestin is the key protein for the termination of phototransduction and one of the abundant proteins in photoreceptors. Recently, we cloned an arrestin homologue gene, Ci-arr and the expression of Ci-arr was found to be restricted to the photoreceptors in the ocellus. To study the whole structure of the photoreceptors in the larva, we prepared an antibody against Ci-Arr. It is found that anti Ci-Arr antibody specifically stains the photoreceptors, including the cell bodies, the axons, and the nerve terminals. The photoreceptor cell bodies lies in row outside the pigment cup which penetrate the pigment cell and is continuous with the outer segments of the photoreceptor cell, inside the concavity of the pigments. The axons form bundle into a single tract. The tract extends toward the midline, where the nerve terminals diverge and seem to form synapses

Synergism Between Zinc and Taurine in the Visual Sensitivity of the Bullfrog's Eye

  • Although there are high concentrations of zinc and taurine in ocular tissue, their exact role and correlation in the visual process are not clear. The purpose of present study was to clarity this point using electroretinogram (ERG) recording and spectrophotometer measurements before and after zinc and taurine treatment in bullfrog's eye. The optimal zinc concentration used in this study was 10$^{-2}$ M ZnCl$_2$120 ${mu}ell$/12$m\ell$ ringer solution while the optimal turine concentration was 10$^{-2}$ M taurine 12${mu}ell$/12$m\ell$ ringer solution. For the effects of zinc and taurine on the retinal function, the changes of ERG parameters (especially threshold and b-wave) and absorption spectra were observed before and after treatment. It is noteworthy that high concentrations of zinc and taurine present in the retinal pigment epithelium and the retina. Our results indicate that dark-adapted ERG threshold became elevated and the peak amplitude of b-wave was increased with zinc and taurine treatment. Furthermore there are some synergism effects between zinc and taurine as a result of co-treatment. In spectral scan, absorbance increment due to zinc and taurine treatment was shown over the whole range of spectral range (300-750 nm) with some differences in absorbance increment depending on the case of treatment. As the results of above we believe that zinc and taurine, which are abundant in the retinal pigment epithelium and the retina particularly, may be essential factors for visual process, have some synergism with each other and be required to improve the visual sensitivity during visual adaptation.

Efficiency of Phototransduction Cascade in Carp Cones

  • In the vertebrate retina, rods mediate twilight vision and cones daylight vision. Rods have been purified easily from the retina, and thus the phototransduction mechanism in rods is now well documented. However, it has not been possible to purify cones in large quantities, and therefore, the knowledge on the mechanism in cones is limited. Here we report purification of carp (Cyprinus carpio) cones with a stepwise Percoll gradient. Using purified cells, we compared the phototransduction mechanism between rods and cones. The results showed that both transducin activation and phosphodiesterase activation are less effective, and visual pigment phosphorylation is faster in cones. These differences explain lower light-sensitivity and briefer photoresponse time course in cones.

진도홍주색소의 저장안정성에 관한 연구 (Studies on the Storage Stability of Jindo Hongju Pigment)

  • 진도 홍주는 전남 진도의 전통민속명주로서 자초에서 용출된 색이 아름다운 홍색을 띠어 시각적인 매력을 갖고있는데, 홍주의 특색인 홍주의 색소가 보존중 색소의 변색으로 인한 상품가치가 떨어진다. 이러한 문제점을 해결하고자 홍주저장중 변색요인의 검토와 아울러 변색 방지에 관하여 검토하였다. 홍주색소의 변색에 영향을 미치는 인자에 대해 조사한 결과, 광(가시영역), $40^{\circ}C$ 이상의 온도, pH 10 이상, 무기이온($C^{2-}$)의 조건에서 가장 변색이 현저하였다. 또한 변색방지제를 첨가하여 변색방지효과를 검토한 결과, 가장 효과적인 것은 cysten 100 ppm 첨가구였다.

  • 별늑대거미의 전측안은 전중안과 함께 전방을 향하여 앞이마 제 1 열에 위치하고 있었으며, 조직학적 조성은 각막, 렌즈, 초자체, 그리고 망막으로 이루어져 있었다. 큐티클성 각막층은 표피의 외큐티클(exocuticle)과 연결되어 있었고, 렌즈는 두개의 볼록렌즈가 내외 양방향으로 연결된 biconvex type이었고, 초자체는 단층의 원주세포로 오목렌즈 모양으로 관찰되었다. 망막을 구성하는 시세포는 단극성 신경세포 (unipolar neuron)로 확인되었으며, 크기가 시세포의 길이에 비하여 큰 세포체가 매우 불규칙 한 크기로 분포하고 있었다. 미세융모로 이루어진 감간체는 세포체 아래에 분포하고 있었으며, 이 부위의 시세포 사이에서는 전자밀도가 높은 색소과립이 관찰되었다. 교질세포는 시세포의 세포체 사이에 분포하였고, 반사층(tapetum)은 감간과 시세포 미부(intermediate segment) 부위 사이에 4-5 정도의 층을 이루어 존재하였다.
