  • 다양한 잡초로부터 유용 생리활성물질을 탐색하고자 46종 잡초의 methanol 추출액을 대상으로 $50{\sim}100\;{\mu}g/ml$ 농도로 실험한 결과, 항균 활성에 있어서는 파리풀, 까실쑥부쟁이, 중머리대가리, 큰엉겅퀴, 부처꽃 등이 antobleb 활성에 있어서는 낙지다리, 밭뚝외풀, 까실쑥부쟁이, 술패랭이꽃 등이 항암활성에 있어서는 파리풀, 골풀, 밭뚝외풀, 까실쑥부쟁이, 술패랭이꽃, 겨우살이 등이 항산화활성에 있어서는 골풀, 물레나물, 청비녀골풀, 금불초, 방울고랭이, 좀고추나물 등이 비교적 강한 활성을 나타내었다.

Effect of Korean Mistletoe Lectin on Gene Expression Profile in Human T Lymphocytes: A Microarray Study

  • Korean mistletoe has a variety of biological effects, such as immunoadjuvant activities. This study investigates the effects of Korean mistletoe lectin (Viscum album L. var. coloratum agglutinin, VCA) on human T lymphocytes to determine whether VCA acts as an immunomodulator. Purified human T-lymphocytes were cultured with VCA and RNA from each point was analyzed using Affymetrix human genome chips containing 22,500 probe sets which represents more than 18,000 transcripts derived from 14,500 human genes. As a result, there was a striking upregulation of genes coding for chemokines. Seventeen genes out of 50 coding for proteins with chemokine activity were upregulated including CXCL9 and IL-8 which are related to the treatment of cancer. In addition, 28 cytokine genes were upregulated including IL-1, IL-6, IL-8, IFN-$\gamma$, and TNF-$\alpha$. Taken together, the data suggest that Korean mistletoe lectin, in parallel with European mistletoe, has an ability to modulate human T cell function.

멸종위기식물 종 꼬리겨우살이의 기주선호성과 서식환경 특성 (Host Preference and Habitat Characteristics of the Endangered Mistletoe Species, Loranthus tanakae in Korea)

  • 본 연구는 멸종위기종 꼬리겨우살이의 기주선호성과 서식환경 특성을 살펴보기 위해 실시되었다. 꼬리겨우살이가 자생하는 강원도 7개 지역을 선정하여 기주목의 DBH, 기주 높이, 가지고사 및 기주 당 꼬리겨우살이와 겨우살이가 자생하는 수 등을 조사하였다. 조사 결과 1,270개체의 꼬리겨우살이를 420개 기주목에서 발견하였으며, 동일한 227개 기주목에서 665개의 겨우살이를 발견하였다. 기주목은 5과 6속 8종 1변종으로 참나무과에서 86.9% 빈도로 가장 높게 나타났다. 꼬리겨우살이가 자생하는 해발고는 420~1,250 m로 나타났으며 주요 해발고는 600~900 m인것으로 나타났다. 기주목의 평균 DBH와 기주 높이는 각각 38.6 cm와 13.0 m였으며, 가지고사는 300개의 기주목에서 기주목당 1.5개의 빈도로 나타났다. 본 연구결과를 통해 기주목의 특성 중 DBH가 꼬리겨우살이의 군집과 지역적 분포에 가장 큰 영향을 미치는 것으로 나타났다.

  • 이 연구는 한국산 겨우살이 엽의 생태학적 특성을 이해하고자 P-V 곡선법에 의한 계절별 수분특성 연구를 수행하였다. 생장시기에 최대포수시의 삼투포텐셜(${\Psi}_o^{sat}$)과 초기원형질분리시의 삼투포텐셜(${\Psi}_o^{tlp}$)은 감소한 반면 최대 탄성계수($E_{max}$)와 상대함수율($RWC^{tlp}$)은 증가하였다. 11월의 신갈나무와 굴참나무의 겨우살이는 가장 높은 최대팽압 (${\Psi}_{P,\;max}$)을 보였고, 신갈나무의 압포텐셜(${\Psi}_P$)과 수분포텐셜(${\Psi}_L$)은 상대함수율의 감소에 따라 급격하게 감소하였다. 11월의 상대함수율($RWC^{tlp}$)은 모두 84% 이상을 보여 삼투조절기능이 다소 좋았으며, 건중당 삼투수(Vo/DW)와 건중당 총수분함량(Vt/DW)은 계절에 따라 변화하였다. 결과적으로 신갈나무의 굴참나무 겨우살이의 ${\Psi}_o^{sat},\;{\Psi}_o^{tlp},\;E_{max},\;{\Psi}_{P,\;max},\;RWC^{tlp}$와 같은 수분특성인자는 엽령의 증가에 따라 내건성이 증가하는 경향을 보였다.

Tyrosinase Inhibitory and Antioxidant Activities of Korean Mistletoe (Viscum album var. coloratum) Extract and Its Fractions

  • The copper-containing enzyme, tyrosinase, catalyzes the oxidation of tyrosine into dihydroxy phenylalanine (DOPA) and subsequently DOPAquinone. It is responsible, not only for the pigment melanin biosynthesis in human skin, but also for browning in foods. In the present study, tyrosinase inhibitory and antioxidant activities of Korean mistletoe extract and its fractions were investigated. As a result, both water and methanol (MeOn) extracts inhibited the tyrosinase activity. Among the fractions, the fraction eluted with $95\%$ MeOn significantly inhibited the tyrosinase activity. The fraction was further purified, and the purified fraction C strongly inhibited the enzyme activity up to $92\%$. In addition, water and methanol extracts exerted radical scavenging effects. The fractions eluted with $70\%\;MeOn\;and\;95\%$ Me on showed high radical scavenging activities. In conclusion, these results suggest that Korean mistletoe extract and its fractions might be useful for the treatment of various dermatological disorders such as epidermal hyperpigmentation and for improving food quality.

한국산 겨우살이 분획물의 면역세포의 생육증진 및 세포독성 (Growth enhancement and cytotoxicity of Korean mistletoe fractions on human cell lines)

  • 기주목(상수리나무, 밤나무, 자작나무, 버드나무)별 겨우살이 ethanol 추출물의 5가지 분획물(hexane fr., chloroform fr., ethyl acetate fr., butanol fr., aqueous fr.)을 이용하여 in vitro에서 사람 면역세포주에 대한 생육활성, 사람 정상 세포주에 대한 세포독성, 사람의 각종 암 세포주에 대한 생육 저해능을 조사하였다. 4종의 한국산 겨우살이 분획물들의 정상세포에 대한 세포독성은 밤나무 겨우살이 chloroform fr.(28%)을 제외한 모든 겨우살이 분획물 0.5mg/ml이하의 농도로 투여시 정상세포에 대한 세포독성이 20%이하로 나타났다. 또한 암세포 생육 저해능의 경우, 폐암 세포주 A549와 위암세포주 AGS에 대하여 상수리나무, 밤나무, 버드나무 겨우살이 분획물에서 80%이상의 생육억제 효과를 보였다. Human B 세포주와 T 세포주에 대한 생육촉진 실험에서, B 세포주에 대하여 상수리나무 겨우살이 hexane fr. buthanol fr., 밤나무 겨우살이 hexane fr., 자작나무 겨우살이 ethyl acetate fr., 버드나무 겨우살이 hexane fr. chloroform fr. ethyl acetate fr.은 배양 3일에서 5일 사이 control에 비하여 약 $1.5{\sim}3$배의 세포 생육촉진을 보였다. T 세포주에 대하여 상수리나무 겨우살이 chloroform fr. ethyl acetate fr., 밤나무 겨우살이 chloroform fr. ethyl acetate fr., 자작나무 겨우살이 hexane fr. chloroform fr. ethyl acetate fr., 버드나무 겨우살이 hexane fr. chloroform fr. ethyl acetate fr.은 배양 3일에서 5일 사이 control에 비하여 약 $1.5{\sim}2.3$배의 세포 생육촉진을 보였다. 한국산 겨우살이 분획물들은 정상세포 대한 독성이 약하고, 면역세포의 생육을 촉진하며, 암세포 생육을 저해하는 효과가 있는 결과로부터 겨우살이는 신체질병에 대한 효과가 기대되며, 앞으로 보다 상세한 연구가 필요할 것으로 사료된다.

Microencapsulation of Korean Mistletoe (Viscum album var. coloratum) Extract and Its Application into Milk

  • This study was designed to develop microencapsulated Korean mistletoe extract, to determine the stability in vitro and to examine its application in milk. Coating materials used were polyglycerol monostearate (PGMS) and medium-chain triacylglyderol (MCT). The highest efficiency of microencapsulation was 78.3% with 15:1:40 (w/w/v) as PGMS : mistletoe extract : distilled water and 66.1% with 15:1 (w/w) as MCT : mistletoe extract. The size of microcapsule was about 30.0 and $19.5{\mu}m$ with PGMS and MCT, respectively. When microcapsules of mistletoe extract were incubated in simulated gastric fluid at pH 2 for 60 min, 14.8 and 17.2% of lectin was released from capsules which were coated with PGMS and MCT, respectively. Comparatively, 83.2 and 87.3% of lectin was released in simulated intestinal fluid (pH 8) after 60 min incubation of capsules coated with PGMS and MCT, respectively. The subsequent study determined the changes of physicochemical and sensory characteristics of milk with fortification of the mistletoe extract microcapsules during 12 day storage. TBA value was significantly lower in microcapsule-added groups than in the uncapsulated mistletoe extract-added group during the storage. When 100 ppm microencapsulated mistletoe extract was added, the L-, a- and b- values and viscosity were not significantly different from those of the control. In addition, the release of lectin from mistletoe extract over 12 days was 8.3 and 9.5 mg/100 ml in milk containing microcapsules made by PGMS and MCT, respectively. All sensory attributes showed a significant difference in uncapsulated mistletoe extract-added milk compared with other groups. The present study indicated that microcapsules of Korean mistletoe extract could be applied to milk and microcapsules coated with PGMS were effectively released in a simulated intestinal environment.

한국산 겨우살이 열수추출액이 본태성 고혈압쥐의 지질성분 및 혈압 저하에 미치는 영향 (Effects of the Korean Mistletoe Hot-Water Extract on the Lipid Components and Blood Pressure Level in Spontaneously Hypertensive Rats)

  • This study was designed to observe the effects of Korean mistletoe(Viscum album L. var. coloratum Ohwi) hot-water extract administration on the improvement on the lipid components, apolipoprotein and blood pressure level in spontaneously hypertensive rats (SHR, Wistar strain, male) fed the experimental diets for 33 days. Concentrations of total cholesterol, triglyceride (TG), LDL-cholesterol, free cholesterol and atheroscieotic index in serum were significantly lower in the Korean mistletoe extract administration group [group KM(Korean mistletoe 10.0 g% hot-water extract)] than those in the control group. In the ratio of HDL-cholesterol concentration Korean mistletoe 10.0% hot-water extract administration group were higher percentage than in the control group. However, concentrations of total cholesterol and TG in liver and brain were significantly lower in the group KM than those in the control group. But the components of apolipoprotein (Apo) A-I and Apo A-II in serum were significantly higher in the KM group than in the control group. However, components of Apo C-II, Apo C-III, Apo E and ratio of Apo B to Apo A-I in serum were fairly reduced in the group KM than in the control group. The levels of systolic and diastolic blood pressure were significantly lower in KM cup than control group. From these results, Korean mistletoe hot-water extract administration were effective on the improvement of the lipid components, coronary heart disease and hypertension in spontaneously hypertensive rats. And particularly, Korean mistletoe extract administration were more effective as a therapeutic regimen for the control of blood pressure in hypertension.

Mistletoe Lectin Induces Apoptosis and Telomerase Inhibition in Human A253 Cancer Cells through Dephosphorylation of Akt

  • Mistletoe lectin has been reported to induce apoptosis in different cancer cell lines in vitro and to show antitumor activity against a variety of tumors in animal models. We previously demonstrated the Korean mistletoe lectin (Viscum album var. coloratum, VCA)-induced apoptosis by down-regulation of Bcl-2 and telomerase activity and by up-regulation of Bax through p53- and p21-independent pathway in hepatoma cells. In the present study, we observed the induction of apoptotic cell death through activation of caspase-3 and the inhibition of telomerase activity through transcriptional down-regulation of hTERT in the VCA-treated A253 cells. We also observed the inhibition of telomerase activity and induction of apoptosis resulted from dephosphorylation of Akt in the survival signaling pathways. In addition, combining VCA with the inhibitors of phosphatidylinositol 3-kinase (PI3-kinase) upstream of Akt, wortmannin and LY294002 showed an additive inhibitory effect of telomerase activity. In contrast, the inhibitor of protein phosphatase 2A (PP2A), okadaic acid inhibited VCA-induced dephosphorylation of Akt and inhibition of telomerase activity. Taken together, VCA induces apoptotic cell death through Akt signaling pathway in correlated with the inhibition of telomerase activity and the activation of caspase-3. From these results, together with our previous studies, we suggest that VCA triggers molecular changes that resulting in the inhibition of cell growth and the induction of apoptotic cell death of cancer cells, which suggest that VCA may be useful as chemotherapeutic agent for cancer cells.

기주목, 온도 및 습도가 멸종위기 종 꼬리겨우살이 종자의 기외발아에 미치는 영향 (Effects of Host Tree Species, Temperature and Humidity on ex vitro Seed Germination in Endangered Species of Loranthus tanakae)

  • 본 연구는 멸종위기에 처한 꼬리겨우살이 종자의 형태적 특징과 발아과정 및 종자발아에 미치는 기주목, 온도, 습도의 영향 및 꼬리겨우살이 종자의 형태적 특징을 관찰하고 일반 겨우살이 종자와의 비교를 위하여 연구를 수행하였다. 꼬리겨우살이 종자발아는 기주목에 접종 1주일 후 원기근을 형성하기 시작하였으며 대부분의 발아조건에서 80~95%의 발아율을 보였다. 종자발아 후 흡기근 침투율은 $20^{\circ}C$ 조건에서 다양한 기주식물 중 신강포플러에서 가장 높은 흡기근 침투율(72%)을 보였으며, 이 외에도 $20^{\circ}C$ 조건에서 산뽕나무, 단풍나무 및 밤나무에서 흡기근 침투율이 각각 57%, 55% 및 42%를 보여 수종과 온도에 따라 흡기근 침투율의 차이가 나타났다. 관수가 없는 조건에서도 종자발아가 진행되었으나 접종 12주 후 꼬리겨우살이 종자와 기주목이 모두 고사하였다. 꼬리겨우살이 종자발아 과정은 원기근 유도${\rightarrow}$흡착기관 발달${\rightarrow}$흡기근 형성 및 침투단계로 총 14주의 기간이 소요되며 꼬리겨우살이가 종자발아를 위해서는 단계별 과정과 긴 기간이 필요한 것으로 보였다.
