Viscum Album Var Hot Water Extract Mediates Anti-cancer Effects through G1 Phase Cell Cycle Arrest in SK-Hep1 Human Hepatocarcinoma cells

  • Viscum album var (VAV) also known as mistletoe, has long been categorized as a traditional herbal medicine in Asia. In addition to its immunomodulating activities, mistletoe has also been used in the treatment of chronic hepatic disorders in China and Korea. There are numerous reports showing that VAV possesses anti-cancer effects, however influence on human hepatocarcinoma has never been elucidated. In the present study, hot water extracts of VAV was evaluated for its potential anti-cancer effect in vitro. SK-Hep1 cells were treated with VAV (50-400ug/ml) for both 24 and 48 hours then cell viability was measured by cell counting kit-8 (CCK-8). Flow cytometry analysis was used to measure the proportion of SK-Hep1 in the different stages of cell cycle. RT-PCR and Western blot analysis were conducted to measure expression of cell cycle arrest related genes and proteins respectively. VAV dose dependently inhibited the proliferation of SK-Hep1 cells without any cytotoxicity with normal Chang liver cell (CCL-13). Flow cytometry analysis showed that VAV extract inhibited the cell cycle of SK-Hep1 cells via G1 phase arrest. RT-PCR and Western blot analysis both revealed that cyclin dependent kinase 2 (Cdk2) and cyclin D1 gene expression were significantly down regulated while p21 was upregulated dose dependently by VAV treatment. Combined down regulation of Cdk2, Cyclin D1 and up regulation of p21 can result in cell death. These results indicate that VAV showed evidence of anti-cancer activity through G1 phase cell cycle arrest in SK-Hep1 cells.

약용 식물을 첨가한 머루주의 이화학적 특성과 발효 품질 평가 (Evaluation of Physicochemical and Fermentation Qualities of Moru Wines Supplemented with Pine Needles or Medicinal Herbs)

  • For the development of Moru wine with enhanced sensory qualities, Moru (Vitis amurensis) wines were fermented with pine needles from Pinus densiflora Siebold et Zuccarini to make Moru-pine wine or medicinal herbs from Astragali Radix and Viscum album to make Moru-herb wine. Moru without pine needles or medicinal herbs was included as a control. Pine needles and Astragali Radix/Viscum album delayed the fermentation of Moru wine, but after 40 days of fermentation and aging, final ethanol contents, pH, acidity, and the sugar/organic acid content in these kinds of Moru wines had no differences. The final ethanol level and acidity of Moru wines were 11.5~12.9% and 1.1~1.2%, respectively, but any sugars (glucose, fructose and sucrose) were not detected in all Moru wines. These results are consistent with the general characteristics of Moru wines, which have a high acidity and low sugar contents. Relatively low browness and antocyanins were detected in Moru-herb wine decreasing its chromaticity in a sensory test. Overall, the supplements of Astragali Radix/Viscum album reduced the color of normal Moru wine, which might be applied toward the development of conventional Moru wines.

겨우살이 종자 발아 및 화아 배양에 의한 캘러스 형성 (In vitro seed germination and callus formation on flower bud of Korean mistletoe ( Viscum album L. var. cololatum [Kom.] Ohwi))

  • 겨우살이의 캘러스 형성 및 종자 발아 와 haustorium 발달에 요구되는 환경요인 및 생장조절제의 영향을 조사하였다. 아울러 종자의 인위부착을 통한 식물체 재생 체계를 확립하였다. 겨우살이 조직관찰 결과 종자에는 1개 또는 2개의 막대모양의 접합자배가 존재하며 종자의 외부에 위치한 유근 부위에 주로 세포분열이 왕성한 세포층이 존재하며 반대로 자엽은 종자 내부에 위치하며 상대적으로 분열능을 가진 세포수가 감소하였다. 겨우살이 종자 발아 와 haustorium 발달에 요구되는 여러 요인 중에서 광의 요구도가 절대적이었다. 본 실험에 조사된 다른 배양 환경요인 및 생장조절제들은 광의 효과를 대체하지 못하였으며 ethylene의 경우 발아촉진 효과가 3배 이상 증가함을 알 수 있었다. 여러 조직 중에서 오로지 겨우살이 화아로부터 캘러스 형성이 가능하였으며 캘러스 형성빈도는 $0.1\;mgl^{-1}$ IAA가 첨가된 B5 배지에 배양 시 27.3%로 가장 높았다. 본 연구에서 확립된 겨우살이 캘러스 배양 및 종자 인위 부착을 통한 식물체 재생체계는 겨우살이의 유용물질 생산 연구 및 대량증식 연구분야에 활용이 가능할 것으로 기대된다.

누에 나방에 있어서 한국산 겨우살이(Viscum album coloratum) 추출물의 생명연장 효과 (Effect of Korean Mistletoe (Viscum album coloratum) Extract (KME) on the Extensions of Life span in Silkworm Moth, Bombyx Mori)

  • Viscum album coloratum (Korean mistletoe; KM) is a semi-parasitic plant growing on various trees. This plant hasbeen shown to possess a variety of biological functions such as immunomodulation, apoptosis-induction and antitumor activity. However, there is no information about how the extract of KM affects life span of the host. In this study, in order to examine the effect of KM on the longevity of the host, we investigated whether a Korean mistletoe extract (KME) was able to extend the life length in an experimental model using the spring silkworm moth of Bombyx mori (Lepidoptera bombycidae). We established a model test for survival in which all of the male and female moths died within 18 days after the beginning of the experiment. Mean survival time of female moths was longer than that of male moths. Both male and female moths fed with KME containing diet (200 or 400 ${\mu}g$/head/day) showed significantly higher mean survival times than those of the control moths. Studying the effect of KME on prolongation of mean survival time showed that male moths were more susceptible than female moths. The moths fed with KME-containing diet showed a slight decrease in body weight. Interestingly, however, no difference in food intake was observed between moths fed with KME-containing diet and those with control diet. In analysis for mRNA increase in the SIRT2 gene, a member of the Sir2 gene family playing important roles in regulation of cell death and prolongation of life span, moths fed with KME-containing diet showed a significant increase in SIRT2 gene expression. These results suggest that KME has a potential to extend the life span in Bombyx mori, and its effect is partly associated with increase in SIRT2 gene expression.

겨우살이 추출물의 미백 효과 (Depigmenting Effects of Mistletoe (Viscum album var. coloratum) Extracts)

  • 멜라닌 색소는 피부색의 주요 원인이다. 멜라닌 색소는 멜라닌 세포에서 생성된 다음 각질 세포로 전달되어 결국 피부 표면에 다양한 색상을 부여한다. 많은 탈색제 및 피부 미백제가 개발되었지만, 색소 침착을 감소시키기 위한 재료에 대한 수요는 여전히 증가하고 있다. 본 연구에서 천연 화합물을 사용하여 탈색 및 피부 미백에 대한 재료를 찾으려고 시도한 결과 겨우살이(Viscum album var. coloratum) 추출물이 색소 침착을 억제할 수 있음을 발견하였다. 인간 멜라닌 세포에 겨우살이 추출물(mistletoe extracts, ME)을 처리했을 때 색소 침착이 극적으로 감소하였다. 프로모터 리포터 분석은 ME 처리가 HM3KO 흑색종 세포에서 microphthalmia-associated transcription factor (MITF), melanophilin (MLPH), tyrosinase related protein 2 (TRP-2), and tyrosinase (TYR) 유전자의 전사를 억제한다는 것을 보여주었다. 일관되게 ME는 MITF, TRP-1 및 TYR과 같은 색소 침착 관련 분자의 단백질 수준을 감소시켰다. 또한 ME는 cAMP Responsive Element Binding Protein (CREB), AKT 및 ERK의 인산화를 감소시켰다. 이러한 결과는 ME가 색소 침착과 관련된 세포 내 신호 전달의 조절을 통해 멜라닌 생성을 억제한다는 것을 시사한다. 끝으로 ME는 색소 침착에 대한 생체 내 평가 모델인 제브라피쉬 배아의 멜라닌 생성을 현저하게 억제하였다.

계절에 따른 국내 자생 꼬리겨우살이, 겨우살이 및 기주목의 광합성 특성과 엽록소 함량 (Seasonal Variation in Photosynthetic Characteristics and Chlorophyll Content of the Loranthus tanakae, Viscum album var. coloratum and its Hosts in Korea)

  • 본 연구는 희귀기생식물인 꼬리겨우살이(Loranthus tanakae)의 보전을 위한 기초자료 제공을 목적으로 꼬리겨우살이와 겨우살이(Viscum album var. coloratum)를 대상으로 계절에 따라 자생지에서 어떠한 생리특성을 나타내고, 기주목과는 어떠한 차이를 보이는지, 광합성 속도, 증산 속도, 수분이용효율, 엽록소 형광반응 및 엽록소 함량을 조사함으로써 살펴보았다. 꼬리겨우살이의 최대 광합성 속도는 6월 중순 광량 $941{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$일 때 $9.36{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, 최대 증산 속도는 7월 하순 광량 $1,596{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$일 때 $5.06{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$로 나타났다. 겨우살이의 최대 광합성 속도는 9월 중순 광량 $418{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$일 때 $6.51{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, 최대 증산 속도는 9월 초 광량 $1,735{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 일 때 $3.91{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$로 나타났다. 겨우살이는 기주목과 꼬리겨우살이 모두 낙엽이 떨어진 11월에도 광합성이 가능하였다. 특이하게 꼬리겨우살이의 증산 속도가 기주목보다 항상 높은 값을 유지하였다. 최대 엽록소 함량(엽록소 a+b)은 꼬리겨우살이가 7월 하순 $8.23mg{\cdot}g^{-1}$, 겨우살이는 6월 중순 $10.27mg{\cdot}g^{-1}$로 나타났으며, 엽록소 a/b 비율은 계절에 따라 꼬리겨우살이 1.7~3.7, 겨우살이 1.1~4.5로 큰 차이를 보였다.

한국산(韓國産) 겨울살이류(類)의 당류(糖類)와 triterpenoids의 화학적(化學的) 조성(組成) 분석(分析) (Analysis of Chemical Constituents of Saccharides and Triterpenoids in the Korean Native Mistletoes - I. Triterpenoids -)

  • The aim of this research was to investigate the chemical components of C30 compounds, especially triterpenoids in Korean native mistletoes of Korthalsella japonicus Engler parasiting to Camellia japonica L., Viscum album var. coloratum (Kom.) Ohmi, to Quercus acutissima Carruth. and Loran-thus yadoriko Sieb. to Neolitsea sericea (BI.) Koidz. For the identification of triterpenoidal components, alkaline hydrolyzates of mistletoes meals were analyzed by TLC, GC, and GC/MS. The content of oleanolic acid and ursolic acid derivatives were highest in K. japonica. In V. album, there was no big difference between leaves and twigs in content. but oleanolic acid in leaves. and olean-12-en-$3{\beta}$-ol and lup-20(29)-en-3-one in twigs were prominent. Similiar to V. album in L. yadoriki there was no difference between leaves and twigs in content, and both olean-12-en-$3{\beta}$-ol, lup-20(29)-en-3-one and urs-12-en-$3{\beta}$-ol in leaves, lup-20(29)-en-3-one in twigs were abundant. Triterpenoids as olea-12-en-$3{\beta}$-ol, lupe-20(29)-en-3-one, 3-oxo-urs-12-en-24-oic acid, and $21{\beta}$-A'-neogam-macer-22(29)-en-3-ol acetate were common in all samples tested. whereas ursolic acid only in P. japonicus and ursenol in L. yadoriki were detected. And P. japonicus had the largest number of triterpenoids and showed the highest in biological activity. So it is noted that Korean mistletoes tested in the study had three types of triterpenoid, oleanane, lupane, and ursane, irrespective of hosts, sampling positions and species.

겨우살이 분말 첨가 식빵의 품질특성 및 항산화 활성 (Quality Characteristics and Antioxidant Activities of White Bread Added with Mistletoe (Viscum album var.) Powder)

  • This study was carried out to investigate the quality characteristics and antioxidant activities of bread containing mistletoe (Viscum album var.) powder (at ratios of 0, 3, 5, 7, and 10% of total flour quantity). As mistletoe powder (MP) content increased, the pH levels of dough and bread and lightness decreased, whereas total titratable acidity increased. The fermentation power of the dough expansion by fermentation decreased with an increase in MP content added. Although bread weight decreased, there were no significant differences in baking loss as MP content increased. Volume and specific volume significantly decreased as MP content increased. Moisture content was highest at a substitution level of 10% and increased with increasing MP content. Lightness of crumbs significantly decreased with increasing MP content, whereas redness and yellowness showed the reverse effect. As MP content increased, changes in hardness, gumminess, and brittleness were all significant. In the sensory evaluation, color, flavor, softness, and overall acceptability were highest at MP 3% and were lowest at MP 10%. Total polyphenol contents and DPPH radical scavenging activity of bread significantly increased with increasing MP content. In conclusion, addition of MP 3% showed the best performance in terms of the nutritional and functional aspects of the bread.
