생선초밥용 냉동횟감의 미생물학적 조사 (Microbiological Investigation of the Frozen-Raw Sliced Fishes for Sushi Manufacturing)

  • This study assessed the contamination levels of total aerobic bacteria, fungi, coliforms, Escherichia coli, and Staphylococcus aureus, and qualitative analysis of Bacillus cereus, Salmonella spp., Listeria spp., and Vibrio spp. in four frozen-raw sliced fishes (cuttlefish, flatfish, salmon, and shrimp) for sushi production. The total aerobic bacteria, fungi, and coliforms were 2.95-3.38, 1.96-2.88, and 0.92-1.29 log CFU/g, respectively. In particular, shrimp was highly contaminated with total aerobic bacteria (3.38 log CFU/g) and fungi (2.88 log CFU/g). Over 3 log CFU/g of total aerobic bacteria was also detected in cuttlefish, flatfish, and salmon. Less than 1-2 log CFU/g of E. coli was detected in all frozen samples. S. aureus was detected at 2.25-3.13 log CFU/g in most samples. B. cereus was qualitatively detected at 25% in most samples, except for salmon (0%). Salmonella spp., L. monocytogenes, and Vibrio spp. were qualitatively detected at 25-50% of all four samples. The microbial contamination levels determined in the current study may be potentially used as basic data to perform microbial risk assessments of frozen-raw sliced fishes.

심해 해양미생물의 분리 및 분리균의 생태적 특징에 관한 연구 (Studies on the Ecological Characteristics of Marine Bacteria Isolated from Deep Sea)

  • 일본의 Sagami bay와 Suruga bay의 810-4,000m되는 심해에서 채취한 시료에서 Flavvobacterium sp., Pseudomonas sp., Vibrio sp. 균종을 분리하였다. 이들 중 barophilic과 psychrophilic bacteria인 sp.-86과 sp.-87를 분리동정하고 압력단계별 성장속도를 조사한 결과 400기압에서도 잘 성장하였고 특히 100기압에서 가장 성장속도가 좋았다. 균의 형태는 1기압의 균 형태에 비하여 400기압에서 성장한 간균이 30-50배 이상 긴 간균 형태였으나(Fig. 3), 다시 1기압에 방치하면 단간균으로 균괴(flock)를 형성한다(Fig. 4) 이들 균의 발육온도 범위는 $5-37^{\circ}C$까지 발육하며, NaCl 농도의 범위는 sp.-86은 $0.5-9\%$이고 최적성장 농도가 $3\%$, sp.-87 균주의 지방산 조성은 $C_{20-22}\;C_{16:1},\;C_{16:0}$ 순으로 높은 함량이었고 400기압에서는 $C_{18:1}\;C_{18:0},\;C_{20-22:0}$ 의 함량순을 보였다. sp.-87균주는 $C_{16:1}\;C_{14:0},\;C_{20-22:0}$의 순이었고 400기압에서는 $C_{14:1}\;C_{12:0},\;C_{16:1}$의의 순으로 함량조성이 변하였다. 아미노산 조성은 sp.-86의 깅우 aspartic acid, methionine, glutamic acid 함량순이었고 400기압에서는methionine, glutamic acid, aspartic acid 함량순이었다. sp.-87의 균주는 methionine, glutamic acid, aspratic acid 순이었고 400기압에서는 methionine이 가장 높은 함량이고 glutamic acid, isoleucine 순으로 높은 함량이었다.

A Retrospective Study on the Comparison of Outbreaks of Food Poisoning for Food Hygiene in Korea and Japan

  • Retrospective study on the comparison of outbreaks of food poisoning for food hygiene in Korea and Japan. The average value of morbidity rate by year in Korea during the period of 1971 to 1992 was 2.9 per 100,000 population, and that of Japan was 29.1. The mean value of mortality rates in case of food poisoning by year in Korea was 2.33%, and that of Japan was 0.07%. When compared the rates of morbidity and mortality between Korea and Japan during the same period, the morbidity rates of Japan were much higher than those of Korea (p<0.01). However, mortality rate of patients in Korea were much higher then those of Japan(p<0.01). Resulting from comparative observation of food poisoning by preparing facilities between Korea and Japan. The highest list the places where the outbreaks occurred was home-made foods accounted for 48.8% of the total cases in Korea and that of Japan was restaurants accounted for 33.0%. Causative foods in Korea, the most common incrimination vehicles were seafood, meat and animal products and grain and vegetables, including mushroom. However, in the case of the common incrimination vehicles Japan were unknown and other foods, seafood, vegetables and meat and animal products etc.. Food poisoning of pathogenic substance in Korea were 60.9% of bacterial food poisoning of the total cases showing that Vibrio species, Salmonella spp., Staphylococcus spp., pathogenic E. coli, Clostridium spp. and other spp. were 33.3%, 26.2%, 16.3%, 5.3%, 0.4% and 18.5%, respectively. On the other hand, in Japan, major causes were Vibrio spp. (45.7%), Staphylococcus spp. (23.7%), Salmonella spp. (16.8%), pathogenic E. coli (3.8%), Clostridium spp. (0.2%) and other spp. (9.6%).

비브리오의 병원성 인자에 관한 연구 (The Virulence Factors of Vibrio spp.)

  • A total of 113 Vibrio sp. strains were examined for plasmid content which were subjected to digestion with restriction enzymes. About the 55% Vibrio spp. have the plasmid more than one. Most of these plasmid various derivatives ranged from $2.4\;kb{\sim}23\;kb$, especially two strains of V. mimicus and one strain of V. furnissii carried one high-molecular weight plasmid (molecular weight ranging between $70\;kb{\sim}100\;kb$). Results of restriction analysis for plasmid of this three strains were by no means the rule. For detection of tdh and ctx gene, the virulence factor involved in the pathogenesis, we carried out the TDH and CT assay, PCR amplification, and hybridization. A total 11 strains were produced TDH, involved in 9 strains of V. parahaemolyticus and 1 strain of V. alginolyticus from clinical isolates and 1 strains of V. mimicus from environmental isolates. In the experiments of tdh gene detection, in all, 3 strains of V. parahaemolyticus from clinical isolates and 2 strains from environmental isolates could be successfully amplified in 400 bp by PCR. The PCR results were consistent with DNA hybridization tests. In the experiments of CT assay, in all, 3 strains of V. cholerae from clinical isolate and 1 strain of V. cholerae from environmental isolates were observed CT-producing. These CT-producing strains amplified in 302 bp by PCR for the detection of ctx gene. All CT-producing strains hybridized with digoxigenin-labeled DNA probe, while CT-negative strains did not hybridize. Also hybridization tests results for detection of ctx gene consistent with PCR.

경북 동해안 환경에서 분리된 V. parahaemolyticus 및 V. vulnificus의 생태학적 및 항생제 감수성 특성 (Environmental and Antimicrobial Characteristics of Vibrio spp. Isolated from Fish, Shellfish, Seawater and Brackish water samples in Gyeongbuk Eastern Coast)

  • This study was carried out to investigated the distribution and characteristics of Vibrio spp. isolated from fish and shellfish, seawater and brackish water samples collected from Pohang, Uljin, Yeongduk and Gyeongju in Gyeongbuk Province from April 2000 to October 2000. Total 155 strains of genus Vibrio were isolated from 439 collected samples, and numbers of isolated strains of V. parahaemolyticus and V. vulnificus were 140 and 15, respectively. The isolation rate from the samples collected in Pohang was the highest as 41.5% (76 strains), and wat the highest as 71.4% (30 strains) in brackish water, and was the highest as 55.7% (34 strains) in August. And the optimal pH, temperature, and NaCl concentration for growth of V. parahaemolyticus and V. cholerae were 8.0, 3$0^{\circ}C$ and 2.0%, respectively. In a resistance test for environmental factors, heat and cold resistants of V. parahaemolyticus were higher than those of V. vulnificus, withstanding for 15 minutes at 6$0^{\circ}C$ and 6 days at -18$^{\circ}C$. The pH range for existence of V. parahaemolyticus and V. vulnificus were 4.5~l1.0 and 4.5~10.0, showing the similar resistance to pH. V. parahaemolyticus and V. vulnificus could grow in media containing up to 10.0% and 7.0% NaCl, respectively, Salt-tolerance of V. parahaemolyticus was higher than that of V. vulnificus. In an antibiotics sensitivity test against 16 strains of V. parahaemolyticus, twelve strains were resistant to ampicillin, eight strains were resistant to cephalothin. one strain was resistant to streptomycin, and one strain was resistant to ticarcillin.

  • No established method can be used to select effective antibiotics in antibiotic susceptibility tests for fish bacterial pathogens quickly and accurately. Here, we established the optimal conditions for determining the minimal inhibitory concentration (MIC) of major fish bacterial pathogens (Streptococcus spp., Edwardsiella tarda, Vibrio spp., Aeromonas spp., and Pseudomonas spp.) using the KRAQ1 and CAMPY2 panels. The MIC panel used 18 antibiotics of two types and we conducted experiments to establish the optimal culture medium and temperature for each species. The optimal conditions for incubating Streptococcus spp. were in cation-adjusted Mueller-Hinton broth with TES buffer (CAMHBT) at 28℃, using 5% lysed horse blood (LHB) as recommended by the Clinical Laboratory Standards Institute. For Vibrio spp., the optimal culture conditions were 28℃ in CAMHBT supplemented with 1% NaCl. The optimal conditions for culturing E. tarda, Aeromonas spp., and Pseudomonas spp. were in CAMHBT at 28℃.

Activities of Essential Oils from Perilla frutescens var. acuta against Antibiotic-Susceptible and -Resistant Vibrio and Salmonella Species

  • We determined the inhibitory activity of the essential oil fraction obtained by steam distillation from the fresh and dried leaves of Perilla frutescens var. acuta against some pathogenic Salmonella and Vibrio spp. The activities of compounds isolated from the essential oils, apiol and myristicin, were also tested and the results were compared with those of the essential oil fraction. The Perilla essential oil fraction and its main components showed significant inhibition against antibiotic-susceptive and antibiotic-resistant strains of the tested Salmonella and Vibrio strains. Synergistic or additive effects were identified by combing the oils with ampicillin by checkerboard-titer tests. We conclude that essential oils from P. frutescens can be useful in the treatment of Salmonella and Vibrio infections and as safe additives to food materials for the prevention of contamination of food by these bacteria. This is especially important because of the rapid increase in antibiotic-resistant strains, which could cause severe symptoms in humans.

곰소만 및 변산 해역 해수에 병원성 비브리오균(Vibrios spp.)의 분포 (Distribution of Pathogenic Vibrio Species in Seawater in Gomso Bay and Byeonsan, West Coast of Korea)

  • The pathogenic Vibrio genus contains halophilic bacteria that are distributed in marine and freshwater environments. Vibrio cholerae, Vibrio vulnificus, and Vibrio parahaemolyticus are potent human pathogens and leading causes of septicemia, wound infection, and seafood-borne gastroenteritis. The aim of this study was to investigate the presence of pathogenic Vibrio species in seawater off the west coast of Korea. Sixty-four seawater samples were obtained from different sites in Gomso Bay and Byeonsan from April 2018 to November 2018. Pathogenic Vibrio species were detected using a combination of most probable number (MPN)-polymerase chain reaction methods. V. cholerae, V. vulnificus, and V. parahaemolyticus were found in 0.0%, 20.3%, and 65.6% of seawater samples, respectively. Quantitative results revealed 3.6-23 MPN/100 mL of V. vulnificus, and 3.6-930 MPN/100 mL of V. parahaemolyticus in the samples. Overall, these results provide new insight into the necessity for seawater sanitation in Gomso Bay and Byunsan; they also provide evidence that will help reduce outbreaks of seafood-borne illness caused by pathogenic Vibrio species.

일본산 참돔, Pagrus major 종묘생산과정중 사육수 변화로 인한 대량폐사 (Environmental Factors Related to Mass Moralities of Young Red Seabream (Pagrus major) in the Artificial Seed Production)

  • 남해안 일원에서 다양한 어류의 종묘생산 및 양식이 시도되고 있는데, 자어기에 종묘의대량폐사가 발생하여 문제의 심각성이 야기되고 있다. 본 연구에서는 일본 킨키대학에소 높은 경과를 올리고 있는 참돔의 서발육종 및 잡종강세에 의한 품종개량된 일명 킨키돔의 수정란을 국내에 이식하여 종묘생산시 대량폐사와 관련되는 수질변화 등을 조사하였다. 부화직후 자어의 평균전장은 2.93 mm이었으며, 일령 40일째에는 19.83~20.12 mm로 성장하였다. 종묘생산과정중 자어의 1차 감모현상은 일령 25~30일째(7.98~9.37 mm)에 발생하였으며, 생존율은 59.8~60.3%에서 32.1~32.5%로 감소하였다. 그 이후에는 거의 폐사 없었으며, 40일째의 생존율은 20.5~25.4%였다. 일령 2-~3-일째 수질은 II등급 이하였으며, 조사기간중 COD, $PO_4$-P, $NO_2$-N, $NO_3$-N 및 $NH_$-N은 각각 3, 7, 34, 6 및 8배씨 증가하였다. 사육수 및 자어의 총세균수는 시간이 경과함에 따라 증가하였으며, 일령 25일째의 총 세균수는 각각 $6.3{\times}10^6$~$2.3{\times}10^7$ cfu/ml, 4.3~$7.4{\times}10^6$ cfu/g였다. 한편, 일령 25일째 참돔 자어는 수면위를 선회하고 복북가 팽만되는 특징을 보이는 질병에 의하여 다수가 폐사되었으며, 병어에서 Vibrio spp.를 분리.동정하였다. 분리된 Vibrio spp. PSV-1균주는 참돔자어에 병원성이 있으며, 참돔자어의 $LD_50$dms $10^6$ cfu/ml 이상이었다. 이상이 결과로부터 참돔자어의 1차 대량폐사는 수질의 급격한 변화, 총세균수 증가 그리고 병원 생물의 복합적인 상호작용에 의한 것으로 추정된다.
