Antimicrobial Activity of Korean Traditional Prescription (Kumkue-Yoryak) and Herb Simplexes Extracts to Vibrio Parahemolyticus (Vibrio parahemolyticus에 대한 한방처방(금궤요략) 및 그 구성약재의 항균활성에 관한 연구)

  • Vibrio become prevailing if superficial temperature of ocean is raised and their activities of area are expanded and most of ocean creatures(fishes, oysters etc) are polluted with vibrio. The one who has taken these polluted fishes and oysters uncooked caused foodpoisoning and diarrhea from Vibrio. Frequently these diseases breakout is disposed in westsea shore of Korea. According to ancient and traditional Korean medical book - Kumkue Yorak - and their single, we carried out experiment check the activities of natural medicinal effects on Vibrio parahemolyticus. The prescriptions of trial materials are processed from extraction boiling water and 80% methanol and followed by freeze dried and adsorbed to every discs in dosage of 10mg. Gentamycin of 10mg were used for control. The result of compound prescription displayed special diseases in antimicrobial activities of boiling water and MeOH extraction compared with control. In single prescription, extraction of boiling water and MeOH of Schisandra sphenanthera(clear zone : 15mm) presented extraordinal antimicrobial activity. The MBC of Schisandra sphenanthera extracts was expressed 2.56mg/ml in boiling water and MeOH.

A Profile of Naturally Occurring Plasmids from Selected Strains of Vibrios

  • The naturally occurring plasmids of Vibrio species have been isolated in part to investigate their genetic traits. Among six different Vibrio species tested, Vibrio anguillarum, Vibrio fluvialis, Vibrio vulnificus, Vibrio mimicus and Vibrio furnissi did not show any presence of plasmid. One environmental isolate of Vibrio pnrahemolyticus harboring plasmid was observed. The isolated plasmid was 8.7 kb by analysis with restriction endonuclease digestion. No common feature was shown relationships between the presence of plasmid and resistance against commonly used antibiotic compounds from the tested Vibrios. In compound prescription, extraction MeOH of Sashinhwan(clear zone : 17mm) presented extraordinaire antimicrobial activity. In single prescription, extraction of boiling water(clear zone : 16mm) and MeOH(clear zone : 18mm) of Fructus Chebulae presented extraordinaire antimicrobial activity. The MBC of Fructus Chebulae extracts was expressed in boiling water(1.28mg/ml) and MeOH(0.64mg/ml).

Detection of Food Poisoning Toxin Genes Produced by Vibrio sp. (비브리오 균속이 생산하는 식중독 유발 Toxin 유전자의 검출)

  • Recently Vibrio sp. are the most frequently isolated microorganism, which causing food poisoning. We investigated the detection of toxin genes and effect of chitosan to toxin genes with PCR. Thirty strains of Vibrio sp. were isolated from sea water and sea products through biological and biochemical tests. Out of 30 strains, 8 were identified as V. parahemolyticus, 7 as V. mimcus, 6 as V. damsela, 5 as V. vulnificus, 4 as V. alginoyticus. In detection of ctx, tdh, and t고 as food poisoning-causing toxin genes, ctx from 7 strains, trh from 4 strains and tdh from 6 strains were detected. Among toxin genes detected strains, we selected V. vulnificus-2 V. damsela-1 and V. parahemolyticus-7. As adding chitosan solution to PCR product of 3 strains, the amplified DNA bands were not detected over 450$\mu\textrm{g}$/$m\ell$ concentrantion of chitosan. Over the result, chitosan is thought to influence the detection of toxin gene.

Microbilolgical Studies on the Takju (Makguly) Brewing : The Korean Local Wine (한국 고유주의 일종인 막걸리에 대한 미생물학적 연구)

  • This study included two parts of investigation, the microfloral changes during the brewing process with the changes of pH, total acidity, temperature and alcoholic contents, as well as determination of survival times of major enteric pathogens in Takju. 1. Maximum number of Saccharomyces cerevisiae was $4.3{\times}10^7$ per milliliter on the 5th day of fermentation and gradually decreased. Saccharomyces cerevisiae was one of the predominant strains of the fermentation process. The number of Saccharomyces cerevisiae was $4.3{\times}10^6$ per milliliter at the completion of the brewing and human consumption. In a few days after the completion of the brewing. Bacillus subtilis and some species of Staphylococcus spp. began to grow and those organisms were responsible for the spoilage. 2. Maximum pH, during the brewing, was 5.8 on the first day of fermentation and rapidly decreased until 6th day of fermentation at pH 4.3. 3. Maximum alcholic content was 14.5 degree on the 4th day of fermentation, 10.3 degree on the 5th day and this degree was continued during the experimentation. 4. Maximum temperature, during Takju brewing was 34.deg.C on the 3rd day of fermentation and rapidly decreased up to 23.deg.C on the 6th day and this temperature was continued until the brewing process was finished. 5. Maximum total acidity was 0.57 percent on the 4th day of fermentation and gradually decreased by brewing process was completed. 6. Survival time of major enteric pathogenic bacteria in Takju was as follows : Shigella dysenteriae and Escherichia coli were isolated in two hours and 14 hours respectively, but Salmonella typhi, Vibrio parahemolyticus were not isolated even in an hour after the inoculation of those organisms in undiluted Takju. In diluted Takju, Salmonella typhi, Vibrio parahemolyticus were not isolated even in an hour after the inoculation of those organisms in undiluted Takju. In diluted Takju, Salmonella typhi, Shigella dysenteriae, and Escherichia coli were survived for 50-60 hours, but Vibrio cholerae and Vibrio parahemolyticus were not isolated even if treated within one hour.

Susceptibility of Vibrio vulnificus to Human Serum Bactericidal Activity and Effect of Vibrio Infections on Hematocrit Value in Mice (Vibrio vulnificus의 인혈청살균력에 대한 감수성과 Vibrio 감염이 마우스의 Hematocrit치에 미치는 영향)

  • Vibrio vulnificus, a halophilic Vibrio has gained worldwide attention as a cause of severe and frequently fatal wound infections and life-threatening septicemia. For this reason V. vulnificus is thought to produce extreme hemoconcentration and rapid death after infection, and because V. vulnificus is thought to be less susceptible to bactericidal activity of normal human serum, the present study was undertaken to assess the susceptibility of V. vulnificus to human serum bactericidal activity with that of V. parahemolyticus and V. cholerae and to assess the effect of Vibrio species, Salmonella typhimurium and E. coli on hematocrit values in experimentally infected mice. Serum bactericidal activity against both V. vulnificus and V. cholerae was higher than against V. parahemoltyicus. Survival of the test strains in heat-inactivated human serum was greater than that in heat-uninactiveted serum. Both V. parahemolyticus and V. cholerae produced slight hemoconcentration within 2 to 6 hr after intraperitoneal injection of $10^7$ viable bacteria into mice. In contrast, V. vulnificus, S. typhimurium and E. coli produced hemodilution rather than hemoconcentration after 4 or 6 hr after infection. With these results the author can conclude that V. vulnificus is more susceptible to serum bactericidal activity than other Vibrio species, and V. vulnificus did not produce hemoconcentration.

Comparison in Restriction Profile Analysis of Vibrio furnissi, Vibrio fluvialis, and Vibrio parahaemolyicus Bacteriophage from Sea Product

  • The bacteriophages lytic for Vibrio furnissi, Vibrio furniulis and Vibrio parahemolyticus were isolated from fish gills and shellfish. Nucleic acid of bacteriophage was prepared and restriction endonuclease profile was compared. All isolates contained deoxyribonucleic acid. V. fumissi bacteriophage from fish gills showed 2 bands with Bgl II, 1 with Pst, 3 with Hind III, 1 with Bm HI and 2 with EcoR I. V Puuialis phage represented 7 fragments with Bgl II, 1 with Pst, 4 with Hind III, and 2 with EcoR I. V parhemolyticn produced 13 sites with Hind III and 4 sites with EcoR I. The fragment types were varied depending on the phage isolation. All three phages were digested with Hind III and EcoR I with different sizes. V furnissi phage were digested with 5 different restriction enzymes.   • Toll-like receptors (TLRs) are a major pattern recognition receptor that recognize the structure of invading pathogen and play key roles by triggering immune response. In this study, we identified a sequence of TLR homolog and characterized at molecular level from the abalone (Haliotis discus hannai). Multiple alignments and phylogenetic analysis of abalone TLR protein belongs to the TLR 2/6. Expression level of abalone TLR 2/6 in the tissue was comparatively high in the mantle, gill, digestive duct, and hemocytes, but lowest in the muscle. Expression level of abalone TLR 2/6 mRNA in the mantle, gill, digestive duct, and hemocytes was 20-fold, 60-fold, 115-fold, 112-fold higher than in the muscle, respectively. Expression level of abalone TLR 2/6 mRNA in the mantle was steadily increased until 12 h and decreased post-infection with Vibrio parahemolyticus. While the expression level of abalone TLR 2/6 mRNA in the gill and hemocytes was drastically increased at 6 and 9 h post-infection with Vibrio parahemolyticus, respectively. These results suggest that abalone TLR 2/6 is conserved through evolution and may play roles similar to its mammalian counterparts.

Antibacterial Activity of Poncirus trifoliata Juice against Pathogenic Bacteria (병원성 세균에 대한 탱자즙의 항균효과)

  • For development of functional food, antibacterial effect of Poncirus trifoliata juice was examined. Strong antibacterial activities of Poncirus trifoliata juice were observed aginst Gram positive and negative pathogenic bacteria such as Baillus cereus, Bacillus subtilis, Corynebacterium zerosis, Listeria monocytogenes, Micrococcus luteus, Rhodococcus equi, Klebsiella pneumoniae, Pseudomonas aeruginosa, Vibrio alginolyticus, Vibrio cholerae, Vibrio parahaemolyticus, Vibrio vnlnificus and Yersinia enterocolitica. The minimum inhibitory concentration(MIC) of Poncirus trifoliata juice against Bacillus cereus. Listeria monocytogenes, Micrococcus luteus, Rhodococcus equi, Staphylococcus epidermidis, Citrobacter freundil and Pseudomonas aeruginosa was 2.5% and the MIC against Vibrio cholerae, Vibrio parahemolyticus, Vibrio vulnificus and Yersinia enterocolitica was 1.25%. Also, antibacterial activities of Poncirus trifoliata juice treated for 15 min at 121$^{\circ}C$ were confirmed to be stable.

Sensitivity Test on the Food Poisoning Bacteria of the Garlic Extract (마늘추출물의 식중독균에 대한 항균검사)

  • Garlic (Allium sativum, L.) has a long history of reputed value and actual use for its medicinal, antimicrobial, and pesticidal properties. This study was conducted to find possible developments to natural food preservatives from garlic extracts. Garlic extracts were prepared from fresh crushed garlic and diluted with sterilize distilled water. The sensitivity test was conducted to investigate the antimicrobial effect of garlic extract against six strains of food poisoning bacteria such as Staphylococcus aureus, Escherichea coli, Salmonella typhimurium, Vibrio parahemolyticus, Bacillus subtilis, and Listeria monocytogenes. The results of sensitivity test were then compared with those of Vancomycin and Imipenem which were used as treatment for G. (+) bacteria and G. (-) bacteria, respectively. All of the 6 tested strains exhibited strong or moderate activity. V. parahemolyticus exhibited susceptible only in undiluted solution, but not in diluted garlic extracts.
