Pysicochemical and Bacteriological Properties of Yogurt made by Single or Mixed Cultures of L. bulgaricus and S. thermophilus (L. bulgaricus 와 S. thermophilus 의 단독 및 혼합배양에 의한 요구르트의 이화학적 미생물학적 특성)

  • The pysicochemical and bacteriological properties of yogurts made by single or mixed cultures of L. bulgaricus FRI025 and S. thermophilus CHI were investigated. L. bulgaricus FRI025 which was isolated from raw milk was selected as starter culture among 22 strains of lactic culture by measuring viscosity, flavor, growth and acid production ability. The acid production and number of viable cell were increased by using L. bulgaricus FRI025 and S. thermophilus CHI together in ratio of apporximately 1:1. The pH, titratable acidity, viable cell number and viscosity of yogurt were 4.08, 1.14%, $2.5{\times}10^{10}/ml$ and 2100 cp after 9 hours incubation at $40^{\circ}C$, respectively. The pH and viable cell number were decreased on the other hand titratable acidity and viscosity were increased after 7 days of storage at $4^{\circ}C$. The changes of quality did not show significantly after storage. The selected starter was much higher than commercial yogurt starter in the acid production and growth of starter. The yogurt nanufactured with selected starter was better than with commercial yogurt in sensory evalution such as taste, texture, flavor and overall acceptability.

  • Disinfection and molecular weight changes by electron beam irradiation has been examined. Before radiation, the viscosity of oven-dried HA decreased about 30% but the viscosity of vacuum-dried HA did not decrease. The number of viable cell depends on the relative humidity(RH). The cell number decreased about 30% in RH 0% and about 10% in RH 100%. After radiation, the decrease in the molecular weight of the hyaluronic acid was larger in the RH=0% than in the RH=100% and was larger in the vacuum-dried HA than in the oven-dried HA. The extent of disinfection were similar for all form.

  • The viability of Bifidobacterium longum KCTC 3128, entrapped in calcium alginate beds in simulated gastroenteric juices (gastric and bile salt solution), was tested to evaluate the influences of several parameters (gel concentration, bead size, and initial cell number). The death rate of B. longum in beads after being sequentially exposed to simulated gastric juices and bile salt solution decreased propertionally with increasing both the alginate gel concentration and bead size. The number of initial cell loading in beads affected the numbers of survivors after being exposed to these solutions, while the death rate of the viable cells were not affected. From the results obtained, the influence of entrapment parameters on the survival of bifidobacteria was quantitatively and systematically evaluated by using a mathematical method.

Neuroprotective Effect of Citri Pericarpium On Transient Global Ischemia in Gerbils

  • The current study was carried out to evaluate neuroprotective effects of Citri Pericarpium after transient global ischemia in gerbils. Male Mongolian gerbils weighing 60-80g were anesthetized with 2% isoflurane mixed with 30% oxygen and 70 % nitrogen. Bilateral common carotid arteries were occluded for 5 minute with microaneurysm dips. On 3 or 7 days after ischemic surgery, the gerbils were sacrificed. The brain were removed, embedded in paraffin and sectioned at 8㎛-thickness. Gerbils that received ischemic insult for 5 min showed extensive neuronal damage in the hippocampal CA1 region, and the number of viable neuronal cell was 51.0±2.5/mm, 32.2% of normal group at 7 days after ischemic surgery. In animals that underwent the extract of Citri Pericarpium treatment, the number of viable neuronal cell were significantly better preserved at 110.58±3.58/mm, 72.0% of normal group than those of ischemic group (P<0.01). In the immunohistochemistry of Bax and Bcl-2, the Citri Pericarpium treated group down-regulated the expression of Bax protein at 72hr after transient global ischemia. In contrast, Bcl-2 protein level was not changed. The appearance in TUNEL assay is similar to the pattern of Bax protein. The water extract of Citri Pericarpium significantly reduced the number of TUNEL-positive CA1 pyramidal neurons at 72hr. The results suggest that Citri Pericarpium has potential neuroprotective effects in the transient global ischemia and the increase in the ratio of Bcl-2 to Bax may contribute to the anti-apoptotic effect of Citri Pericarpium.

  • In cultivating human neuroblastoma cells maximum number of neurites per cell and length of the neurite were estimated as 5.5 and 2.2 (nm), respectively It was found that there was correlation between growth and differentiation of nerve cells. Maximum specific BDNF production rate was also calculated as 2.5$\times $10$^{-5}$(ng/cell/day) at 7$\times $ 10$^{5}$ (viable cells/ml) of maximum cell density, corresponding to 100 (ng/mL) of BDNF. The secretion of BDNF was occurred most in the later peroids of the cultivation, yielding 75 (ng/mL) of BDNF. The production of rate of BDNF was elongated in adding 1 ($\mu $g/mL) of BDNF as well as 40% increase of the length of the BDNF. It proves that BDNF can be used as one of biopharmaceuticals to treat age-related diseases such as Alzheimer's disease and Prakinson's disease. It can also provide the information of scaling-up mammalian cell cuture system to economically produce BDNF.

Preparation of Fermented Citrus Peels Extracts for Their Antimicrobial Activity against Campylobacter jejuni (감귤 진피 추출물을 이용한 발효액 제조 및 Campylobacter jejuni 에 대한 항균 활성)

  • Jeju citrus, which contains an abundance of calcium and vitamin, was used to develop fermented citrus peel extract. A total of seven probiotic strains were applied to tangerine dermis to select the best growing bacteria in citrus peel extracts. B. longum, B. bifidum, and L. mesenteroides were found to grow best in citrus peel extract culture containing glucose, yeast extracts, peptone, and potassium phosphate. Citrus peel extract culture consisting of 1% yeast extract, 5% peptone, and 0.1% phosphate was the best environment for growth of probiotics. The pH, acidity, and viable cell numbers of these fermented extracts were measured. The initial pH level of fermented extracts with nutrients was 5.25 and dropped rapidly to 3.39 after 72 hours of fermentation. The acidity of fermented extracts increased to 4.08 % after 72 hours of fermentation, and the viable cell number in fermented extracts after refrigeration for 2 weeks was $1.3{\times}10^{10}CFU/mL$. The antimicrobial activity of citrus peel fermented extracts against Campylobacter jejuni was determined, and concentrations more than 25,000 ppm showed antimicrobial activity.

High Concentrated Spore Production of Bacillus thuringliensis by Fed-Batch Processes (유가식 배양공정에 의한 Bacillus thurngiensis의 고농도 포자생산)

  • Both the production of high spore concentration and high insecticidal activity are required in the production of Bacillus thuringiensis to be used for the bacterial insecticide. In the production of high cell and spore concentrations of B. thuringiensis the continuous fed-batch culture(CFBC) and intermittent fed-batch culture(IFBC) were investigated at $28^{\circ}C$ by maintaining 40% dissolved oxygen concentration. When the final glucose concentration was 50 g/L the maximum viable cell number obtained using the CFBC with linear gradient feeding was $9.37{\times}109$ cells/mL and maximum spore concentration was $8.33{\times}109$ spores/mL which was approximately 84.4% yield of spore formation. When the final glucose concentration was 100 g/L the aximum viable cell and spore concentrations obtained using the IFBC with pH-statb were $1.38{\times}$1010 cells/mL and $1.35{\times}1010$ spores/mL respectively and the yield of spore formation was approximately 97.8%.

Effects of Daturae Flos and Daturae Semen Extract on the Immunocyte Response in Mice (만타라화(曼陀羅花) 및 만타라자(曼陀羅子) 수추출물(水抽出物)이 마우스의 면역세포기능(免疫細胞機能)에 미치는 영향(影響))

  • This study was undertaken to test the effects of Daturae Flos(DF) and Daturae Semen(DS) on the cellular and humoral immune responses, and the functions of the cells involved in immunoinflammation. Both extracts decreased the activity of superoxide dismutase, and the decrease was greater in the mouse group which was treated with DS. Both extracts decreased the phagocytic activity as measured by assessing the number of the latex particle within the phagocyte after incubation of peritoneal macrophages with fluorochrome-labelled latex particle and decreased natural killer cell activity as measured by enumerating the viable YAC-1 cells after treatment of target cells with splenic natural killer cells. Both extracts also decreased the cell-mediated immunity in vivo as assessed by measuring the ear thickness after sensitization and challenge with dinitrofluorobenzene, however, had no effects on the humoral immune responses as measured by checking hemolysin and hemagglutinin titers after immunization with sheep red blood cells(SRBC). Extracts of Semen caused decrease in the number of rosette forming cells between the splenic cells and SRBC. The results of this study suggested that both Daturae extracts could depress the immunoinflammation by affecting the various cell types involved in inflammation.

Utilization of Makgeolli sludge for growth of probiotic bacteria (Probiotic bacteria의 생장에 대한 막걸리슬러지의 이용)

  • A number of health benefits have been claimed for probiotic bacteria such as Bifidobacterium (B) spp. Lactobacillus(L) acidophilus, and Lactococcus(Lc) cremoris. Viability of probiotic bacteria is important in order to provide health benefits. Only a limited culture media for the test purpose of probiotic bacteria are commercially available (MRS broth), but the media for large-scale propagation of viable cells which are able to be used as food additive are not available. The manufacture of a low priced and preferred novel medium for probiotic bacteria was therefore, attempted using whey protein concentrate(WPC) and Makgeolli sludge as a starting material. The effect of WPC and Makgeolli sludge on the growth of four strains (B. bifidum 15696, B. longum 15707, L. acidophilus CH-2, and Lc. cremoris 20076) was investigated. Medium prepared such as WPC, Makgeolli sludge, and WPC+Makgeolli sludge(WPCMs). It was observed that the growth of 4 strains (B. bifidum 15696, B. longum 15707, L. acidophilus CH-2, and Lc. cremoris 20076) was stimulated by Makgeolli sludge, WPC, WPCMs. Especially, Viable cell number of 4 strains in the WPCMs were higher than that of the single media. These result suggest the possibility that Makgeolli and WPC, acts as a growth factor for the growth of probiotic bacteria.

Effect of Dried Powders of Pine Needle, Pine Pollen, Green Tea and Horseradish on Preservation of Kimchi-yangnyum (솔잎, 송화, 녹차 및 고추냉이 분말이 김치양념물 저장성에 미치는 영향)

  • This study was performed to investigate the effects of dried powders of pine needle, pine pollen, green tea and horseradish on preservation of Kimchi-yangnyum. The physicochemical and microbial changes of Kimchi-yangnyum were investigated during 30-days preservation. The changes of pH, total acid, and number of total viable cell, lactic acid bacteria, E. coli of the Kimchi-yangnyum were insignificant. And the addition of pine needle powder were more effective to suppress fermentation than that of pine pollen or green tea or horseradish.
