서울지역 식육판매업소의 미생물 오염도 조사 (A survey of the microbial contamination level in butcher's shops in Seoul, Korea)

  • This survey was conducted to evaluate the microbial contamination level of butcher's shops in Seoul, Korea. For microbial inspections, a total of 584 samples (146 cotton work gloves, 146 utensils and equipments, 154 beef samples, 138 pork samples) were collected from butcher's shops. E. coli and pathogenic bacteria such as Staphylococcus aureus, Yersinia enterocolitica, Listeria monocytogenes, Salmonella spp., and E. coli O157:H7 were tested in the samples. As a result, the level of aerobic plate count (APC) ranged ${\leq}10^4\;CFU/cm^2$ from utensils and equipments, $10^1{\sim}10^9$ CFU/glove from cotton work gloves and ${\leq}10^6$ CFU/g from meat. The APC level of E. coli ranged ${\leq}10^1\;CFU/cm^2$ from utensils and equipments, ${\leq}10^5$ CFU/glove from cotton work gloves, and ${\leq}10^3$ CFU/g from meat, respectively. Staphylococcus aureus was detected in 2 beef samples, 1 pork sample, and 10 used cotton work gloves. Yersinia enterocolitica was detected in 3 beef samples, 1 pork sample, and 3 used cotton work gloves. Listeria monocytogenes was detected in 2 used cotton work gloves. In order to improve the sanitation status of butcher's shops, application of HACCP (Hazard Analysis Critical Control Point) or SSOP (Sanitation Standard Operating Procedure), regular hygiene education, and continuous monitoring for microorganisms will be required.

Identification of Enterococcus faecalis antigens specifically expressed in vivo

  • Objectives: Molecular mechanism of the pathogenicity of Enterococcus faecalis (E. faecalis), a suspected endodontic pathogen, has not yet been adequately elucidated due to limited information on its virulence factors. Here we report the identification of in vivo expressed antigens of E. faecalis by using a novel immunoscreening technique called change-mediated antigen technology (CMAT) and an experimental animal model of endodontic infection. Materials and Methods: Among 4,500 E. coli recombinant clones screened, 19 positive clones reacted reproducibly with hyperimmune sera obtained from rabbits immunized with E. faecalis cells isolated from an experimental endodontic infection. DNA sequences from 16 of these in vivo-induced (IVI) genes were determined. Results: Identified protein antigens of E. faecalis included enzymes involved in housekeeping functions, copper resistance protein, putative outer membrane proteins, and proteins of unknown function. Conclusions: In vivo expressed antigens of E. faecalis could be identified by using a novel immune-screening technique CMAT and an experimental animal model of endodontic infection. Detailed analysis of these IVI genes will lead to a better understanding of the molecular mechanisms involved in the endodontic infection of E. faecalis.

Does Visitation Dictate Animal Welfare in Captivity? : A Case Study of Tigers and Leopards from National Zoological Park, New Delhi

  • Zoological Parks house exclusive animal species, thus creating a source of education and awareness for visitors. Big cats like tigers and leopards are among the most visited species in zoos globally. However, they often display stressful or stereotypic behaviours. Such behaviours are influenced by multiple factors including visitors, animal history, and captive environment. To understand this impact, we investigated the behavioural response of tigers and leopards to visitation, captive, and biological factors. The behaviour of eight big cats housed in the National Zoological Park, New Delhi, was monitored using focal sampling technique during May and June 2019. We recorded the captive and biological factors and visitor density for the subjects. The study revealed high proportions of inactive and stereotypic behaviours amongst the species. Tigers and leopards were found to perform stereotypic behaviours for 22% and 28% of their time, respectively. Generalised Linear Models revealed a significant variation of stereotypy in association with the factors. Stereotypy was influenced by visitor density, age, sex, breeding history, coat colour, and enclosure design. Adults, males, white-coated, previously bred, and those housed in smaller and simple enclosures display more stereotypy than young, females, normal-coated, unbred, and those housed in larger and complex enclosures, respectively. A high density of visitors induced more stereotypic behaviours amongst the big cats. As providing entertainment and awareness amongst the public is one of the fundamental objectives of the zoo, visitors can not be avoided. Thus, we suggest providing appropriate enrichments that would reduce stereotypies and promote naturalistic behaviours.

Resistance Patterns of Frequently Applied Antimicrobials and Occurrence of Antibiotic Resistance Genes in Edwardsiella tarda Detected in Edwardsiellosis-Infected Tilapia Species of Fish Farms of Punjab in Pakistan

  • Edwardsiella tarda is one of the most significant fish pathogens, causes edwardsiellosis in a variety of freshwater fish species, and its antibiotic resistance against multiple drugs has made it a health risk worldwide. In this study, we aimed to investigate the antibiotic resistance (ABR) genes of E. tarda and establish its antibiotic susceptibility. Thus, 540 fish (299 Oreochromis niloticus, 138 O. mossambicus, and 103 O. aureus) were collected randomly from twelve fish farms in three districts of Punjab in Pakistan. E. tarda was recovered from 147 fish showing symptoms of exophthalmia, hemorrhages, skin depigmentation, ascites, and bacteria-filled nodules in enlarged liver and kidney. Antimicrobial susceptibility testing proved chloramphenicol, ciprofloxacin, and streptomycin effective, but amoxicillin, erythromycin, and flumequine ineffective in controlling edwardsiellosis. Maximum occurrence of qnrA, blaTEM, and sul3 genes of E. tarda was detected in 45% in the liver, 58%, and 42% respectively in the intestine; 46.5%, 67.2%, and 55.9% respectively in O. niloticus; 24%, 36%, and 23% respectively in summer with respect to fish organs, species, and season, respectively. Motility, H2S, indole, methyl red, and glucose tests gave positive results. Overall, E. tarda infected 27.2% of fish, which ultimately caused 7.69% mortality. The Chi-squared test of independence showed a significant difference in the occurrence of ABR genes of E. tarda with respect to sampling sites. In conclusion, the misuse of antibacterial agents has led to the emergence of ABR genes in E. tarda, which in association with high temperatures cause multiple abnormalities in infected fish and ultimately resulting in massive mortality.

A Newly Identified Glutaminase-Free L-Asparaginase (L-ASPG86) from the Marine Bacterium Mesoflavibacter zeaxanthinifaciens

  • L-Asparaginase (E.C. 3.5.1.1) is an enzyme involved in asparagine hydrolysis and has the potential to effect leukemic cells and various other cancer cells. We identified the L-asparaginase gene (L-ASPG86) in the genus Mesoflavibacter, which consists of a 1,035 bp open reading frame encoding 344 amino acids. Following phylogenetic analysis, the deduced amino acid sequence of L-ASPG86 (L-ASPG86) was grouped as a type I asparaginase with respective homologs in Escherichia coli and Yersinia pseudotuberculosis. The L-ASPG86 gene was cloned into the pET-16b vector to express the respective protein in E. coli BL21 (DE3) cells. Recombinant L-asparaginase (r-L-ASPG86) showed optimum conditions at 37-40℃, pH 9. Moreover, r-L-ASPG86 did not exhibit glutaminase activity. In the metal ions test, its enzymatic activity was highly improved upon addition of 5 mM manganese (3.97-fold) and magnesium (3.35-fold) compared with the untreated control. The specific activity of r-L-ASPG86 was 687.1 units/mg under optimum conditions (37℃, pH 9, and 5 mM MnSO4).

기능성 유제품의 생산 현황 및 관리제도 - 미국.유럽을 중심으로 - (Current Status and Regulation Issues of the Functional Dairy Products in European Countries and the United States of America)

  • This work has been conducted as a part to set up the regulations and the scientific evaluation systems for the functional dairy products with health claims in Korea. Toward this end, current regulations, requirements, and all kinds of provisions related to the functional dairy products overseas were taken into accounts and feasible recommendations of new initiatives on the current regulations as well. By doing so, not only protecting hopefully the innocent consumers from the false labeling statements and fake advertisements, but also eventually to revitalize the stagnated domestic dairy industries. Currently, functional daily products are simply regarded as "Dietary Supplements" in the United States, and subjected to the regulations under the Dietary Supplement Health and Education Act, established m 1994. Manufacturers and sales distributors should notify their products to the FDA, which is in charge of the dietary supplements on nutrition and labeling, in advance when they start marketing a new dietary ingredient or any foods containing it in the States. For EU countries, there exists keen interests about the functional food products between the member countries of the European Union even though the products are categorized into "Food Supplements" similar to those in the United States. Therefore, they maintain a cautious attitude in applying the health claims to the functional food products. On the other hand, under the Japanese health food system, functional foods are qualified to the functional health foods in terms of legal status as long as any traditional foods can meet the legal standards in its effectiveness, safety as well as quality, along with significant scientific evidences related to the products, thus categorized into "Foods for Specified Health Uses and "Foods with Nutrient Function Claims". Through this study, we may have some expectations and potential utilizations as follows: Legal regulations of dietary supplements especially for the dairy products will be implemented by the outcomes of this research and proposed a tentative amendment of functional ingredients for the sake of consumer protection from the false advertisements and overstatement labeling. Current regulations on the animal foods processing and advertisements will be amended and supplemented in order to revitalize the current downturn dairy sales and to harmonize the international Codex recommendations. The results obtained from this study will make the consumers a wise selection of the dairy products with health claims and be utilized for consumer education and advertisement of the functional products as well.

Ortho-phenylphenol을 주성분을 하는 훈증소독제의 Trichophyton mentagrophytes, Candida albicans 그리고 Aspergillus niger에 대한 살진균 효과 (Fungicidal Efficacy of a Fumigation Disinfectant with Ortho-phenylphenol as an Active Ingredient against Trichophyton mentagrophytes, Candida albicans and Aspergillus niger)

  • Objectives: This study evaluated the fungicidal efficacy of a fumigant containing 20% ortho-phenylphenol against Trichophytone mentagrophytes (T. mentagrophytes), Candida albicans (C. albicans) and Aspergillus niger (A. niger). Methods: Five replicates of each carrier were contaminated by depositing 0.05 mL of each fungal suspension. After drying, two carriers without exposure to the fumigant and three carriers with exposure to the fumigant were left in a sealed room ($25m^3$) at $21{\pm}0.5^{\circ}C$ and $60{\pm}10%$ relative humidity for 15 hours. Immediately after removal from the test room, each carrier was transferred into recovery diluent and suspended, diluted and inoculated. After incubation, the numbers of each colony were counted, and the parameter values (N, T, d) were calculated. Results: The working culture suspension number (N value) of T. mentagrophytes, C. albicans and A. niger were $1.0{\times}10^8$, $1.2{\times}10^8$ and $5.7{\times}10^7CFU/mL$, respectively. All the colony numbers on the carriers exposed to the fumigant (n1, n2, n3) were higher than 0.5N1 (the number of fungal test suspensions by pour plate method), 0.5N2 (the number of fungal test suspensions by filter membrane method) and 0.5N1, respectively. In addition, all mean numbers of test strains recovered on the control-carriers (T value) were over $10^6CFU/mL$. For the fungicidal effect of the fumigant, all numbers of fungal reductions after exposure of the fumigant (d value) were 4 logCFU/mL. Conclusions: The present study showed that fumigant containing 20% ortho-phenylphenol has effective fungicidal activity against T. mentagrophytes, C. albicans and A. niger.

Comparative Calorimetric Evaluation of Ammoniated Straw-Based Rations Supplemented with Low Levels of Untreated and Formaldehyde Treated Groundnut Cake and Fish Meal with Respect to Growing Buffalo Calves

  • Eighteen growing male Murrah buffalo (Bubalus bubalis) calves were divided into three groups consisting of six animals each and fed three urea ammoniated wheat straw (UAS) -based rations supplemented with concentrate mixtures (roughage: concentrate ratio 58:42) containing deoiled ground nut cake, GNC (8%), formaldehyde treated GNC (8%) or fish meal (8%) to undertake comparative evaluation of these rations in terms of their $CH_4$ production and growth (285 d duration) potential. A digestibility trial (10 d duration) was followed by a comparative calorimetric study in respiration chamber. Dry matter (DM) intake (84.3 to $89.3g/kg\;W^{0.75}d^{-1}$) did not differ between treatments. The digestibility coefficient of DM, organic matter (OM), crude protein (CP), neutral and acid detergent fiber did not differ significantly in different diets. Urinary energy loss as a percent of gross energy (GE) was not affected by diets. Average values of $CH_4$ production were 84.3, 77.6 and 99.1 g/d and $CH_4$ energy losses as percent of gross energy were 5.7, 5.2 and 6.1 percent on .GNC, formaldehyde treated GNC and fishmeal, respectively, and did not differ significantly. When expressed per unit of digestible OM intake, $CH_4$ production (g) was lower (p<0.05) on formaldehyde treated GNC (30.6) than on untreated GNC (30.6) and fish meal (31.9). Total ME intake and heat production were similar and hence the energy balances on different diets were similar. Nutritive value of rations in terms of digestible CP and ME were similar. Average daily gain calculated on the basis of regression of fortnights on cumulative liveweight gain in calves fed on concentrate containing unprotected GNC, protected GNC and fish meal were 437.1, 483.9 and 481.6 g, respectively. This indicated that the intake of energy was sufficient to meet the requirement of calves growing at 400 g per d. However, CP intake was around 150% of the stipulated standard (Kearl, 1982). Feed conversion ratios on unprotected GNC, protected GNC and fish meal were 11.60, 11.10 and 10.4 respectively. It was concluded that because significantly (p<0.05) low $CH_4$ is produced on protected GNC (8%), it is very good and sustainable protein source in comparison to poor quality fish meal and untreated GNC to be used in concentrate mixture for supplementing UAS-based diets.

The Evaluation of Nutritive Value of Three Tropical Browse Species for Sheep Using in Vitro and in Vivo Digestibility

  • In vitro and in vivo experiments were carried out to evaluate the nutritive value of three tropical browse species as assessed by DM and CP digestibility, and NDF and ADF degradability with incubation times (T) of 6, 12, 24 and 48h. During the in vivo digestibility experiment three male castrated sheep (age 16 - 25 months) with a mean liveweight of $11.5{\pm}0.9kg$ were placed in individual metabolism stalls and were allocated to one of the three browse species in a $3{\times}3$ Latin square design. The browse species were all leguminous and consisted of: Acacia sieberina (A. sieberina), Ficus polita (F. polita), and Ficus sycomorus (F. sycomorus). The mean DM and CP contents of F. polita were higher than for A. sieberina and F. sycomorus (p<0.05). In contrast the NDF and ADF contents of F. sycomorus were higher compared to the other species examined (p<0.05). The in vitro DM and CP digestibility, and NDF and ADF degradability observed at different stages of incubation were higher in F. polita followed by A. sieberina and F. sycomorus. The DM and CP digestibility at 48 h incubation were 72.92, 74.84 and 53.52% and 77.38, 77.68 and 63.64% for A. seiberina, F. polita and F. sycomorus, respectively. This shows that F. polita contains more soluble materials which ruminant can benefit from and hence has more feeding value. The fermentation of F. sycomorus was slower for all the nutrients evaluated due to the presence of more fibre. Similarly, higher in vivo digestibility coefficient of DM, CP, NDF, ADF and hemicellulose were observed for F. polita reflecting its higher values of CP, ether extract (EE) and hemicellulose associated with lower values of NDF and ADF. Higher DMI and daily gain were recorded in sheep during feeding of F. polita compared to the other species evaluated. The digestibility of all the nutrients examined were higher in the in vivo than in the in vitro trial except for CP and DM. Sheep showed no visual signs of toxicity throughout the study periods. These results showed A. seiberina, F. polita and F. sycomorus can sustain sheep on a maintenance diet and could as well be used as a supplementary feed to low producing animals during the tropical dry season. Further research is needed to ascertain the viability of using these browse species on a long-term basis.

우리나라 중소기업의 스마트 팩토리 수용 및 활성화 제고를 위한 정책 방향에 대한 연구 (A Study on the Policy Direction for the Introduction and Activation of Smart Factories by Korean SMEs)

  • 본 연구는 우리나라의 중소기업들에 대하여 스마트 팩토리 관련 기술의 수용 및 사용의 수준을 제고하기 위한 관련 정책의 수립에 도움을 제공하기 위한 것이다. 이를 위하여 통합기술수용모델(UTAUT)을 확장하여 기술 수용의도에 영향을 미칠 수 있는 추가적인 영향요인들을 선정하였으며, 이를 실증하는 것이다. 연구목적의 달성을 위하여 리커트 7점 척도로 구성된 설문지를 작성하였으며, 제조 관련 기업을 대상으로 설문조사를 시행하였다. 통계처리에 활용된 설문지는 총 136부이다. 가설검정 결과 성과기대와 사회적 영향은 수(사)용의도에 정(+)의 유의한 영향을 미치지만, 노력기대와 촉진조건은 유의한 영향을 미치지 않는 것으로 검정되었다. 확장요인으로서 네트워크 효과, 조직특성은 정(+)의 유의한 영향을 미치며, 혁신저항은 부(-)의 유의한 영향을 미치지만 인지된 위험은 유의한 영향을 미치지 않는 것으로 검정되었다. 기업의 규모가 클수록 인지된 위험과 혁신저항의 수준은 낮으며, 이를 제외한 수(사)용의도 영향요인과, 수(사)용의도, 수(사)용행동의 수준은 높은 것으로 검정되었다. 본 연구를 통하여 스마트 팩토리 관련 기술의 수(사)용에 긍정적, 부정적 영향을 미칠 수 있는 요인들을 규명함으로서 제고되어야 하는 요인과 감소시켜야 하는 요인을 제시하였으며, 비교적 기업의 규모가 큰 기업들부터 우선적으로 스마트 팩토리 관련 기술이 수용될 수 있도록 하여야 한다는 연구결과를 제시한다.
